Comparison of Fermentation Characteristics of Korean Traditional Alcoholic Beverage with Different Input Step and Treatment of Rice and Nuruk (Korean-Style Bran Koji) (원료 쌀과 누룩의 처리 및 첨가방법이 다른 전통주의 발효특성 비교)

  • Physicochemical and sensory characteristics of Korean traditional alcoholic beverages, such as Kwahaju, Sokokju, Baikhaju, Samhaeju and Hosanchun were investigated during fermentation with the difference of input step and treatment of rice and Nuruk (Korean-style bran koji). pH values in all treatments were similar to be ranged $3.1{\sim}3.2$, but acidity of the treatment brewed without seed mash was $1.5{\sim}2$ times higher than that of the treatment brewed with seed mash. The concentration of alcohol in all treatments increased as the concentration of the Nuruk increased. The alcohol concentrations of Samhaeju and Hosanchun ranging from 6.5% to 7.7% were higher than those of Kwahaju, Sokokju and Baikhaju ranging from 7.9% to 10.6%. Residual reducing sugar contents of Kwahaju, Sokokju and Baikhaju ranging from 4.6 g/L to 9.7 g/L were lower than those of Samhaeju and Hosanchun ranging from 14.2 g/L to 15.5 g/L. The consumed total sugar contents of Kwahaju, Sokokju and Baikhaju ranging from 125.4 g/L to 159.2 g/L were higher than those of Samhaeju and Hosanchun ranging from 111.2 g/L to 123.8 g/L, and the treatments brewed with seed mash showed more sugar consumption than the treatments brewed without seed mash. Sensory quality in the treatments brewed with seed mash was better than those of without seed mash. Concentration of Nuruk and preparation of seed mash from it were critical factors compared to input step and treatment of rice in Korean traditional alcoholic beverage brewing.

Efficacy of Different Seed Kernels against Root Knot Nematode Meloidogyne incognita in Mulberry

  • Five seed kernel namely, Neem (Azadirachta indica A. Juss.), Pongamia (Pongamia glabra L. Pierre), Tamarind (Tamarindus indica L.), Mahua (Madhuca indica Gmel.) and Shikakai (Acacia cancinna De.) were tested against hatching of eggs and larval mortality of Meloidogyne incognita causing root knot disease in mulberry along with Furadan (Carbofuran) and Bionema (a bioformulation developed from Verticillium chlamydosporium) for comparison. Results revealed that highest hush-up of hatching was observed in Neem (77.40%) and Pongamia (75.99%) seed kernel extracts at 100% concentration over the check. Similarly, highest larval mortality was observed in Neem and Pongamia by 76.00% and 74.50%, respectively at 100% concentration after 72 hrs of exposure period. Pot culture studies revealed that pre-application of seed kernel powders (20 days before inoculation of nematode) found to be more effective in controlling the root knot disease than post application. In pre application of seed kernel powders, maximum reduction of root knots was observed in case of Neem seed kernel powder (54.85%) followed by Pongamia (51.9%). Similar trend was also observed in reduction of egg masses/plant and nematode population /250 cc soil. Rest of the seed kernel extracts was found to be less effective in suppression of hatching, enhancing the larval mortality and controlling the root knot disease. However, application of Furadan and Bionema tested for the comparison were found to be more effective than seed kernel powders. The generated information seems to be useful in developing an ecofriendly integrated approach for the control of root knot nematode disease in mulberry.

Effects of Addition of Herbs on Pork Patties during Storage

  • This study was conducted to investigate the effects of herbs (dill weed, fennel seed and rosemary) on color stability and lipid oxidation of ground pork patties during storage at $2^{\circ}C$ and $-20^{\circ}C$. Lipid oxidation was determined by the thiobarbituric acid reactive substances (TBARS) method. Fennel seed was identified as being the most effective ingredient in terms of meat redness (Hunter a-values) during storage at $2^{\circ}C$ (p<0.05). Patties treated with rosemary had lower Hunter a-values than control, and dill weed was the least beneficial in influencing meat redness, having significantly lower a-values than control patties. During frozen storage at $-20^{\circ}C$, no significant differences in a-values were observed between the control patties and those treated with fennel seed. Patties treated with rosemary had lower a-values than control and patties with dill weed had negative a-values throughout storage. The cooking process increased Hunter L-values and decreased a-values in all patty samples. TBARS values increased during storage. Rosemary displayed excellent antioxidative activity as evidenced by the completely inhibition of TBARS formation of cooked pork patties. Dill weed and fennel seed also exerted an intermediate but significant antioxidant effect during storage (p<0.05).

  • This paper introduces to a simple culture method and growth of axenic (bacteria-free) rotifer Brachionus rotundiformis for seed stock of rotifer mass culture. This rotifer axenic culture method is based on the washing and transferring with sterilized sea water and modified antibiotic mixture AM9 solution. Population growth (final density on day 16) of axenic cultured rotifer were maintained with a high density and stable growth compared with the control of non-axenic culture (general culture style) through the 3 times-rerunning experiments (trial 1, 2 and 3). But the egg carrying rates of amictic females were not different between the axenic-and non-axenic culture condition. Although, rotifer density was higher in axenic culture, the food (Nannochloropsis oculata) was still remained unutilized than that of the non-axenic culture in third trial culture. These results suggest the possible existence of harmful bacteria for the rotifer population growth in the trial 1 and 3 of non-axenic culture compared to the trial 2. This axenic rotifer culture method is valuable for seed stock of the stable rotifer mass cultures.

  • We, Tong Yang Moolsan Co. Ltd. (TYM) set up the mass-production system for virus-free seed garlic via tissue culture technique. TYM's tissue culture technique is called as 'Multiple shoot propagation technique'. This technique can lead mass propagation of genetically homogeneous seed garlic in a short period because of its highly proliferation ,ate of in vitro shoots $(15^{10}/year)$. TYM researchers applied the technique to some selected garlic cultivars with superior characteristics and carried out Held test of productivity in the inside and outside of the country for several years. According to the yearly results of Held test with virus-free seed garlic, we ascertained that virus-free seed garlic can produce the highly yield increase (max. above 50%) and also can enhance the product quality. Consequently, we estimated that TYM's seed garlic will contribute to farmers with increase of income and can elevate the national position of garlic market in the world for its competitive power of technical and production cost.

  • A high seed yield '232' was evaluated for morphological characteristics and tissue culture re-sponse. The effect of mowing and urea application on coverage in '232' was also studied and the results were as follows ;1.'232' produced high seed yield than any other lines, but its other characteristics such as coverage, establishment rate, stolon length, stolon number and colour change were not good. 2.Callus induction of '232'(52.9%) was relatively high, and higher than that of Z. sinica(49.0%). However, embryogenic callus formation of '232' was not as good as that of Z. japonica and Z. sinica. 3.Coverage of '232' was very much enhanced by mowing on July 18, but not at all by mowing on May 29 and September 4. 4.Urea application did not increase the coverage in '232',Therefore, it can be used as a breeding material rather than direct use.

Studies on the Precipitation of Lead Ion and the Inhibition of Plant Growth (연(Pb) 이온의 침전과 식물생장의 억제에 관한 연구)

  • This study was carried out to investigate the formation of precipitates between lead ion and the essential anions of plants, the effects of lead concentration on seed germination and plant growth in water and soil culture, and the germinating and growing recovery of inhibited seed germination and plant growth by lead. Four kinds of the seeds (Glycine max M., Triticum vulgare V., Setaria viridis (L) P. De Beauvois, and Digitoria sanguinalis (L) Scopoli var) were germinated and growth in water and soil culture included the different concentrations of lead for five days. The seeds and plants inhibited germination and growth by lead were transferred to lead free Hoagland solution and the growing recovery was observed. The precipitates of lead ion were observed in the solution of both acidity and alkalinity included each anion of $H_2PO_4^-, HPO_4^{2-}, PO_4^{3-}, SO_4^{2-} and MoO_4^{2-}$ in a room temperature, whereas the precipitates between lead ion and other anions were observed largely in the solution of alkalinity, so that it seemed that lead could be remained in the state of non-soluble in plant and soil. The inhibition of germination and growth in the water culture was observed in 100ppm of lead, whereas the inhibition in the case of the soil culture was observed in 10000ppm of lead. The difference of the effected concentration between water and soil culture in germination and the growth was 100 times. When the seed and plant inhibited the growth in 5000ppm or 10000ppm of lead for five days were transferred to lead free Hoagland solution, the recovery of germination and growth was observed in three days. This growing recovery was different according to the kinds of plant and concentrations of lead. It seemed that plant growth could be inhibited by the inhibition of the metabolism concerned with the precipitates between lead iion and other anions.

A Bacterial Endophyte, Pseudomonas brassicacearum YC5480, Isolated from the Root of Artemisia sp. Producing Antifungal and Phytotoxic Compounds

  • An endophytic bacterial strain YC5480 producing antifungal and phytotoxic compounds simultaneously was isolated from the surface sterilized root of Artemisia sp. collected at Jinju area, Korea. The bacterial strain was identified as a species of Pseudomonas brassicacearum based on its 16S rRNA gene sequence analysis and physiological and biochemical characteristics. The seed germination and growth of monocot and dicot plants were inhibited by culture filtrate (1/10-strength Tryptic Soy Broth) of the strain. The germination rate of radish seeds in the culture filtrate differed in various culture media. Only 20% of radish seeds germinated in the culture media of 1/2 TSB for 5 days incubation. Mycelial growth of fungal pathogens, Colletotrichum gloeosporioides, Fusarium oxysporum and Phytophthora capsici was also inhibited by the culture filtrate of the strain YC5480. An antifungal compound, KS-1 with slight inhibitory activity of radish seed germination at 1,000 ppm and a seed germination inhibitory compound, KS-2 without suppression of fungal growth were produced simultaneously in TSB. The compounds KS-1 and KS-2 were identified to be 2,4-diacetylphloroglucinol (DAPG) and 2,4,6-trihydroxyacetophenone (THA), respectively.

Effect of Chlorella Culture Solution Using Anaerobic digestate on Seed Germination in Perennial Ryegrass (혐기소화액을 배지로 이용한 클로렐라 배양액 처리가 페레니얼라이그라스 종자 발아에 미치는 영향)

  • This experiments were conducted to evaluate the influence of Chlorella culture solution using anaerobic digestate as medium on seed germination of perennial ryegrass seeds. Four treatments were compared: control with distilled water, anaerobic digestate, Chlorella culture solution and Chlorella culture filtrate. The germination percentage of perennial ryegrass seeds was highest in the Chlorella culture solution treatment. Days required for 50, 70% seed germination were faster at 1.7 day in Chlorella culture solution compared to control. Root length of perennial ryegrass seeds was longer by 1~2cm in the Chlorella culture solution compared with control. The relative root length was by 40% longer in the Chlorella culture solution treatment compared to control. The germination index (GI) of perennial ryegrass seeds was higher by 180~202% in the Chlorella culture solution treatment compared to control. The decay rate was low as 50.0% in Chlorella culture solution, but decay rate of perennial ryegrass seeds showed 86.7~83.3% in control plot and in anaerobic digestate, respectively. Chlorella culture solution have shown stimulatory effects in germination and development of root. Overall, Chlorella culture solution could be useful to apply for promotion of germination and root elongation of seeds.
