The experiments were conducted to determine the factors such as light and darkness, phytohormone and seed coat, influencing on seed germination of Bidens tripartita L. and B. frondosa. The seeds of both species were germinated when seed coat was damaged and weakened. $GA_3$ and BA stimulated germination of both species but ABA and IAA had no effect on germination of them, which ranged 50.0% to 80.0%. In B. forndosa, when inner layer of seed coat was removed, germination was highly promoted up to 96.7% compared with 10.0% germination rate in another treatments.
Crotalaria sessiflora is one of a few medicinal herbs among the legumes used as antitumor herb medicine but has lower seed germination and afterward seedling emergence. This study was carried out to elucidate the effects of cold stratification as presown treatment as well as $GA_3$, $KNO_3$ and acetone on its seed germination and seedling emergence. Cold stratification, $GA_3$, $KNO_3$ and acetone treatments were performed with different concentrations and period levels. Cold treatment was done under 14 hour red light illumination a day or darkness and the two others under darkness. On the basis of the best germination rate of each treatment, $GA_3$ and cold stratification treatments showed the highest germination rates at 0.1 mM but 12 hours and 2 week periods under darkness, respectively. $KNO_3$ and acetone treatments came out at 400 mM and 200 mM lasted for, respectively. The best germination rates from $KNO_3$ and acetone were comparatively higher than those from the former $GA_3$ and cold treatments. In evaluation of seedling emergence using the last two treatments, $KNO_3$ treated seeds were better performed than acetone treated seeds showed somewhat higher germination rate, implying that presown seed treatments must be evaluated by the combination with indoor and field tests.

Gibberellin(GA) 3-$\beta$ hydroxylation is very important for the shoot elogation in the higher plants, since only 3$\beta$-hydryoxylated GAs promote shoot elogation in several plants. Fluctuation of 3$\beta$-hydryoxylase activity was examined during seed maturation using two cultivars of , P. vulgaris, Kentucky Wonder (normal) and Masterpiece (dwarf). Very immature seeds of both cultivars contain high level of 3$\beta$-hydroxylase activity (per mg protein). Both cultivars showed maximum of enzyme activity (per seed) in the middle of their maturation process. Gibberellin 3$\beta$-hydroxylase catalyzing the hydroxylation of GA20 to GA1 was purified 313-fold from very early immature seeds of P. vulgaris. Crude soluble enzyme extracts were purified by 15% methanol precipitation, hydrophobic interaction chromatogrphy, DEAE ion exchange column chromatography and gel filtration HPLC. The 3$\beta$-hydroxylase activity was unstable and lost much of its activity duting the purification. The molecular weight of purified enzyme was extimated to be 42, 000 by gel filtration HPLC and SDS-PAGE. The enzyme exhibited maximum activity at pH 7.7. The Km values for [2.3-3H] GA20 and [2.3-3H]GA9 were 0.29 $\mu$M and 0.33 $\mu$M, respectively. The enzyme requires 2-oxoglutarate as a cosubstrate; the Km value for 2-oxoglutarate was 250 $\mu$M using 3H GA20 as a substrate. Fe2+ and ascorbate significantly activated the enzyme at all purification steps, while catalase and BSA activated the purified enzyme only. The enzyme was inhibited by divalent cations Mn2+, Co2+, Ni2+, Cu2+, Zn2+, Cd2+ and Hg2+. Effects of several GAs and GA anaogues on the putrified 3$\beta$-hydroxylase were examined using [3H]GA9 and GA20 as a substrates. Among them, GA5, GA9, GA15, GA20 and GA44 inhibited the enzyme activity. [13C, 3H] GA20 was converted by the partially purified enzyme preparation to [13C, 3H]GA1, GA5 and GA6, which were identified by GC-MS, GA9 was converted only GA4, GA15 and GA44 were converted to GA37 and GA38, respectively. GA5 was epoxidized to GA6 by the preparation. This suggests that 3$\beta$-hydroxylation of GA20 and epoxidation of GA5 are catalyzed by the same enzyme in P, vulgaris.

This study was conducted to improve the postharvest storage techniques of managing and storing seeds, to test qualities and viabilities of the seeds and to examine the germination rate and the protein expression of Achyranthes japonica Nakai. The seeds collected from different areas of Je-Cheon and Gwang-Ju were stored with different temperatures and durations. Two plant growth regulators and two seed priming were treated to investigate their effect on the germination rates and the days required for germination. The weight of one hundred seed collected in Gwang-Ju was heavier than those in Je-Cheon. Seed length collected in Gwang-Ju was also longer about 5.12 mm than those in Je-Cheon about 4.90 mm and seed width was longer in Gwang-Ju than those in Je-Cheon. The rates of seed germination in two different collection areas were higher about 2.9 to 13.0% in Gwang-Ju compared to those in Je-Cheon. Comparing its rates with the storing temperatures and durations, they were not clearly different in between $4^{\circ}C$ and $25^{\circ}C$ and they also were gradually decreased with getting longer storing durations. The germination rates treated by plant growth regulators were higher with $GA_3$ than those with Kinetin. The highest seed germination rate was appeared at 50 ppm of $GA_3$. Comparing its rates with different seed priming, they were relatively higher with $KNO_3$ than those with PEG6000. In protein expression patterns between before the germinating and after the germinating of seeds, more and clear bands were appeared in the seed after the germination compared to those before the germination of seeds, especially 10 ~ 20 kDa. These results showing more and clear bands were more clearly appeared in Gwang-Ju compared to Je-Cheon. Comparing the protein expression with plant growth regulators and seed primings, $GA_3$ was better expression than those with Kinetin and $KNO_3$ was better than those with PEG6000. More and clear bands were closely related to the germination rates of seeds and more detailed studies would be required.
This study is aimed to understand the effects of temperature, light and GA soaking treatment on the seed germination of perennial ryegrass 'Dandy', creeping hentgrass 'Penlinks', tall fescue 'Arid' and Kentucky hluegrass 'Nasaw'. These experiments are carried in vitro, and their results obtained are summarized as the followings ; The optimum temperature for seed germination of perennial ryegrass 'Dandy' was found to he ranged at 20~30$^{\circ}C$, and the optimum temperature for germination of creeping bentgrass 'Penlinks', tall fescue 'Arid' and kentucky bluegrass 'Nasaw' seeds was found to be ranged at 20~25$^{\circ}C$, whereas the germination percentage under the below or above temperature of this range decreased. The germination of perennial ryegrass 'Dandy' and tall fescue 'Arid' seeds showed no effect on light, and while the germination percentage of creeping hentgrass 'Penlinks' increased more highly under the dark condition than under the light condition. The germination percentage decreased according to the higher temperature. The germination of Kentucky bluegrass 'Nasaw' seeds increased more highly under the dark condition than under the light condition at 20~25$^{\circ}C$, whereas the germination percentage under the dark condition at 15$^{\circ}C$ and 30$^{\circ}C$ decreased. In case of GA soaking treatment of seeds, all the cool season turfgrasses tested showed their own promoting effects on seed germination. The early germination of tall fescue 'Arid' was good without the soaking times. Creeping bentgrass 'Penlinks' and perennial ryegrass 'Dandy' had re-spectively the highest germination percentage at 10min and 1hr soaking treatment, and the longer the soaking time, the lesser the germination percentage. Kentucky bluegrass 'Nasaw' obtained the best results at 24has soaking treatment, and the longer the soaking time, the better the ger-mination percentage.
The infant seedling of rice(8-to 10-day-old) has often suffered from submergence just after transplanting due to the too short seedling height, 5 to 8cm, This experiment was conducted to elucidate the effect of gibberellic acid($GA_3 : 25% soluble liquid) with the application methods of foliar spray and seed-soaking on the elongation of seedling height of infant rice seedling in machine transplanting, We used a japonica rice variety, Hwaseongbyeo, and the seeding rate was 200g per seed tray(30$\times60$\times3cm). Generally,$GA_3 and Metalaxyl increased the elongation of seedling height of infant seedling while Tetracycle decreased the seedling height, To increase the seedling height of infant seedling, the foliar spary application of $GA_3 was better than seed-soaking treatment. The optimum concentration of $GA_3 for the foliar spray was about l0ppm and the optimum application time of $GA_3 was 3 to 4 days after sowing.
This study was carried out to investigate the effect of some plant growth regulators on the germination and seedling growth of Wasabia japonica Matsum seeds. The seeds were soaked in different concentration of indolacetic acid(LAA), indolbutyric acid(IBA), $GA_3\;GA_4$, Benzyladenine(BA) and kinetin, and the germination percent, the average germination day and the growth of seedlings were measured. The seeds were released from dormancy and the germination percent was considerably increased when the seedcoat was removed. $GA_3,\;GA_4,$ BA and kinetin were effective in breaking the dormancy, but LAA and IBA were not effective. $GA_4$, BA and kinetin were more effective in accelerating germination of dormancy seed than $GA_3$. The applications of BA combined with $GA_3$ l00ppm showed higher germination percent than that of BA alone within the range 50ppm and 100ppm. Root, hypocotyl and petiol of seedling overgrew by the application of $GA_3\;and\;GA_4$. But effects of BA and kinetin were good on seedling growth compared with those of $GA_3\;and\;GA_4$. Therefore, BA and kinetin could be use for dormancy breaking of seed instead of $GA_3$.

This study aimed to investigate the morphological characteristics and optimal germination conditions in seeds of Arabis pendula L., a traditionally edible and medicinal plant. The external seed shape was circular-obovate with narrow wings and dark brown. The seed length and width were 1.25 mm and 1.47 mm, respectively. The seeds were exalbuminous and the embryo was a bent type. Seed germination was the highest (49.7%) at $20^{\circ}C$ under dark conditions among the various temperature and light conditions applied. However, under the dark condition, the seedling was weak, overgrown, and the cotyledons were small and folded. To improve the germination and growth of seedlings, the seeds were pre-treated with $GA_3$ solutions of varying concentrations (0, 200, 500, and 1,000 mg/L). The seed germination and seedling growth were effectively improved by $GA_3$ pre-treatment. The germination rate was the highest (97.3%), mean germination time was the shortest (8.1 days), and a vigorous growth of seedlings was observed upon pre-soaking the seeds in 500 mg/L $GA_3$ solution. In conclusion, the best method for germination was pre-soaking in 500 mg/L $GA_3$ ($4^{\circ}C$, dark, 24 h) and incubating the seeds at $20^{\circ}C$ for 15 days.
This study was conducted to establish mass propagation system from seeds of Lavandula angustiolia Mill. Only 4% of dry seeds were germinated when they were incubated in $4^{\circ}C$. Germination rate of stratified seeds for 9 weeks was 15% higher in light than darkness. Soaking with 1000mg/l of $GA_3$ was helpful to overcome the effect of darkness. The optimal temperature for germination was $25^{\circ}C$ for the seeds that were treated with$GA_3$, solution. For the improvement of germination rates, pretreatment of $GA_3$ at 500-2000mg/l showed about 75% of germination, and in the combination treatment of $GA_3$, and BA, germination rate increased by about 10% in the treatment of 1000mg/l $GA_3+10mg/l$ BA compared with the 1000mg/l $GA_3$ treatment. Cold stratification treatment was very effective for seed germination, and over 70% of seeds were germinated when they were incubated in $4^{\circ}C$ for 9-12 weeks. In addition, there was synergic effect on the seed germination subject to stratification and 1000mg/l $GA_3$, treatment for 6 months. In the mixture of vermiculite: peatmoss (1 : 1, v/v), emergence rate was 6.7% in control and 65% in 1000mg/l $GA_3$ treatment, respectively.
GA4/7 spray, injection and several cultural treatments were applied to the greenhouse-grown potted hybrid larch(Larix decidua${\times}$leptolepis) grafts and field-grown European larch seedlings to induce early flowering. A treatment consisting of repeat-ed GA4/7 sprays, alone, was the most effective flower induction treatment for greenhouse-grown, potted larch grafts. Root pruning as a adjunct treatment did not show synergistic effects. Injection for potted grafts with GA4/7 was not useful approach in this study and it resulted in increased mortality. In the field experiment with 10-Year-old larch seedlings, repeated GA4/7 sprays in combination with root pruning or with plastic mulching appears to be useful and practical means for inducing larch flowers:
