Isolation and characterization of BrMDR1 a novel MDR-type ATP-binding cassette (ABC) transporter in Brassica rapa L.

  • A cDNA clone encoding a MDR-like ABC transporter protein was isolated from Brassica rapa seedlings, through rapid amplification of cDNA ends (RACE). This gene (named as Brmdr 1; GenBank accession no.: DQ296184 ) had a total length of 4222 bp with an open reading frame of 3900 bp, and encoded a predicted polypeptide of 1300 amino acids with a molecular weight of 143.1 kDa. The BrMDR1 protein shared 71.0, 62.5, 60.0 and 58.2% identity with other MDR proteins isolated from Arabidopsis thaliana (AAN28720), Coptis japonica (CjMDR), Gossypium hirsutum (GhMDR) and Triticum aestivum (TaMDR) at amino acid level, respectively. Southern blot analysis showed that Brmdr1 was a low-copy gene. Expression pattern analysis revealed that Brmdr1 constitutively expressed in the root, stem petals and stamens, but with lower expression in leaves and open flowers. The domains analysis showed that BrMDR1 protein possessed two transmembrane domains (TMDs) and two nucleotide binding domains (NBDs) arranging in "TMD1-NBD1-TMD2-NBD2" direction, which is consistent with other MDR transporters. Within NBDs three characteristic motifs common to all ABC transporters, "Walker A", "Walker B" and C motif, were found. These results indicate that BrMDR1 is a MDR-like ABC transporter protein that may be involved in the transport and accumulation of secondary metabolites.

Tissue culture of medicinal plants: micropropagation, transformation and production of useful secondary metabolites

  • Plant tissue culture studies have been done for the preservation of medicinal plant resources and efficient production of pharmaceutically important secondary metabolites. Micropropagation methods for Cephaelis ipecacuanha have been established and these methods enabled much more efficient propagation of the plants than the conventional methods using seedling or layering. The C. ipecacuanha plants derived from tissue culture grew uniformly in the field and they showed higher alkaloid contents compared to the plants grown from seedlings. Hairy root cultures of C. ipecacuanha and Panax ginseng have been established by infection with Agrobacterium rhizogenes, and the production of important pharmaceuticals by these cultures have been successfully demonstrated. In the case of C. ipecacuanha, the highest alkaloid yields from the hairy roots cultured for 8 weeks were 2.75-fold cephaeline (5.5 mg) and one third emetine (0.7 mg) compared with those from the roots of one-year old plant propagated through shoot-tip culture and cultivated in a greenhouse (2.0 mg cephaeline and 2.0 mg emetine). In the case of P. ginseng, ginsenoside contents in the hairy roots optimally cultured for 4 weeks were much higher than those in the roots of 4-year old field-grown plant. Thus our medicinal plant tissue cultures demonstrate desirable properties. However, they are always exposed to danger of microbial contamination or unexpected trouble of culture facilities. Cryopreservation of plant tissue cultures is a reliable method for long-term preservation. Cryopreservation studies on these cultures are also presented.

Secondary System Initialization Protocol Using FFT-based Correlation Matching for Cognitive Radio Ad-hoc Networks

  • Due to the increasing demand for spectrum resources, cognitive radio networks and dynamic spectrum access draw a lot of research into efficiently utilizing limited spectrum resources. To set up cluster-based CR ad-hoc common channels, conventional methods require a relatively long time to successfully exchange the initialization messages. In this paper, we propose a fast and reliable common channel initialization protocol for CR ad-hoc networks. In the proposed method, the cluster head sequentially broadcasts a system activation signal through its available channels with a predetermined correlation pattern. To detect the cluster head's broadcasting channels and to join the cluster, each member node implements fast Fourier transform (FFT) and computes autocorrelation of an FFT bin sequence for each available channel of the member node. This is compared to the predetermined reference pattern. The join request and channel decision procedures are also presented in this paper. In a simulation study, the performance of the proposed method is evaluated.

Application of the PSTAR Method to a Thermally Massive Passive Solar House (PSTAR기법을 이용한 자연형 주택의 열 성능 연구)

  • This paper reports the application of the PSTAR(Primary and Secondary Terms Analysis and Henormalization) method to a thermally massive passive solar house located in Daejeon, Korea. The house has approximately $156m^2$ of living area with 3 bedrooms and a living room, which embodies many passive solar features for energy conservation. The primary concern of this work was to properly evaluate the thermal behavior of a thermally massive building structure using the PSTAR method. Results show close agreements between the measured and renormalized values in most cases in which the simulation results from the audit description of the house deviate somewhat considerably.

  • Numerical simulation of turbulence flow in a circulating channel was performed. The RNG $k-\varepsilon$ model and Reynolds stress model of the FLUENT was used for evaluating the flow mechanisms. The simulation results were compared with the experimental data measured by a ADV (Acoustic Doppler Velocitmeter). The distribution of chironomids was analyzed by the computational results. They distributed at the region of lower velocities and lower turbulence intensity. In the case of a hemisphere structure being located on the straight section, chironomids lived in the upstream and downstream area of the hemisphere. The secondary currents also affected the distribution of chironomids. In conclusion, the computational fluid dynamic techniques can be inexpensively applied for analysing the relationship between flow characteristics and distribution of benthic macroinvertebrates.

  • In order to estimate the pattern of medical care utilization and recognition for health factor among the inhabitants in Wan-do district which is located off the southern seashore from mainland Korea, household interview survey was performed from January 15 to 30, 1990 in Wan-do Gun area. The results observed were following: 1. Among the users of medical facilities 40.8% used public health facility as first contact facility. Lower the income level was and longer the residency duration was, the utilization rate of public health facilities was higher. By age groups and medical security status the utilization pattern was likely to disperse to local clinic and secondary contact medical resources. 2. Medical expense and access time were significantly different between primary contact and secondary contact medical resources. 3. Public health facility was recognized as the favoured institute having advantage of geographic and economic accessibility. 4. Statistically significant determinants in public health facility utilization among total medical service were the region, the medical expense, and the access time.

Secondary Literature Analysis: The Marketing Practice to Attract Potential Customers into Leisure and Sports Industry

  • Purpose: The marketing practice for the leisure and sports industry is a complex process that requires a thorough understanding of the audience, their needs and motivations. Thus, this niche market is focused on specific products, services, or experiences. The present research explores and suggests meaningful strategies based on the literature textual dataset to provide how to attract consumers in this sector. Research design, data and methodology: We have conducted the 'Secondary Literature Analysis', reviewing and summarizing numerous findings in the relevant prior studies. As a result, we could obtain a total of 15 significant textual resources which are from only peer-reviewed journal article. All resources had a high quality of the instrument to prove their results. Results: The findings of this research pointed out that marketers in leisure sports sector need to communicate via following methods: (1) Understanding the Customers' Needs and Wants, (2) Social Media, (3) Advertising, (4) Promoting Brand Affinity, (5) Offering Discounts, and (6) Providing Value-Added Services. Conclusions: The present research concludes that the marketing practice in the leisure and sports industry should be performed using various channels. In addition, marketing practitioners are supposed to check if tailored marketing messages are compatible with products, services, and events that relate to their target audience's interests.

Cloning, Expression, and Functional Characterization of the Dunaliella salina 5-enolpyruvylshikimate-3-phosphate Synthase Gene in Escherichia coli

  • 5-enolpyruvylshikimate-3-phosphate synthase (EPSP synthase, EC 2.5.1.19) is the sixth enzyme in the shikimate pathway which is essential for the synthesis of aromatic amino acids and many secondary metabolites. The enzyme is widely involved in glyphosate tolerant transgenic plants because it is the primary target of the nonselective herbicide glyphosate. In this study, the Dunaliella salina EPSP synthase gene was cloned by RT-PCR approach. It contains an open reading frame encoding a protein of 514 amino acids with a calculated molecular weight of 54.6 KDa. The derived amino acid sequence showed high homology with other EPSP synthases. The Dunaliella salina EPSP synthase gene was expressed in Escherichia coli and the recombinant EPSP synthase were identified by functional complementation assay.

A Study on the Menu Type of Instrument Cluster IVIS

  • Objective: This research was carried out to study the menu type design of instrument cluster IVIS(In Vehicle Information System) for efficient navigation under deconcentrated situations. Background: A driver controls the IVIS menu using the rest of cognitive resources while driving a car. Although a driver controls the IVIS using extra cognition resources, his or her distraction can still cause some safety problems while driving. Menu type design of instrument cluster is absolutely important for safe and efficient navigation. Method: Four menu types including paging, flow, icon, and list type were identified through reviewing the existing IVIS of vehicle and the menu structure of cellular phone. Four menu types were evaluated through experiment. The experiment consisted of primary and secondary task, which the primary task was to simulate a driving and the secondary task was to control an IVIS menu prototype. Task performances, menu type preferences, and eye-movement patterns were measured in this experiment. Results: The result shows that icon type was the best design in aspect of task performance and preference. A clue for next menu item provided a positive effect for efficient menu navigation. It was identified that most of subjects gazed the middle-top area of IVIS screen from eye-movement pattern. Conclusion: A basic design of Instrument Cluster IVIS was proposed considering the result of this study in the final. Application: The results of this study can be effectively used in the design of Instrument Cluster IVIS.

3D Flow Simulation in the Meandering Natural Channel (사행 자연수로에서의 3차원 흐름 모의)

  • In a natural river, cross sections of a channel vary according to inner or outer parts of meandering. Generally, depth of outer parts is deeper than that of inner parts. This kind of cross section change by meandering can be demonstrated by Beta distribution. The objects of this research is a 3D simulation of primary and secondary flow in the meandering natural channel. FLOW-3D program, a numerical model using CFD technique, and LES method was used for this research. 3D simulations were conducted in the channels having Beta distribution cross sections which have beds of mortar, gravel and vegetation. Two types of water stages and discharge were applied to each channel. In this research, primary flows are located in the outer parts of a top of bend and secondary flows rotate in the bottom on outer parts.
