Origin of Milled Wood Lignin : An Immunocytochemical Approaches in Combination with Transmission Electron Microscopy (면역전현법(免疫電顯法)에 의한 마쇄(磨碎) 리그닌의 기원(起源)에 관한 연구(硏究))

  • Present work was undertaken to investigate the origin of milled wood lignin(MWL) in the wood cell wall using immunocytochemical techniques, which can provide the information on the localization of specific antigens(MWL in the present study) to be examined. Spruce MWL dissolved in DMSO and emulsified with Freund adjuvant was injected directly into the mouse spleen. The animals were boostered at two-week intervals after the initial immunization. Blood samples were purified in standard procedures. The characteristics of antibodies against MWL were tested by indirect ELISA. Visualization of MWL was carried out using conventional indirect immunogold-labelling methods on the ultrathin sections of spruce wood. Immuno-TEM observations showed that the immunogold probes were selectively attached to secondary cell walls of spruce wood. The most intense labelling was frequently observed in the S2 layer. In contrast, gold labelling in the lignin-rich regions, such as middle lamella and cell corner was not found. The immuno-TEM provides an indication that spruce MWL originates from the S2 layer.

Ultrastructural Observation of Bacterial Attacks on the Waterlogged Archaeological Woods (세균에 의한 수침고목재 피해양태의 초미시구조적 관찰)

  • Micromorphological changes in waterlogged archaeological woods excavated from Sweden and Germany were investigated. Especially bacterial attacks on those wood samples under near anaerobic conditions were examined by transmission electron microscopy(TEM). The major feature of micromorphological alterations in those wood samples was the preferential destruction of secondary wood cell wall. In contrast, the middle lamella was not extensively degraded. Three distinct degradation patterns by bacteria were observed : erosion, cavitation and tunnelling bacteria. Erosion and cavitation bacteria attacked primarily $S_2$ layer, whereas tunnelling bacteria made the tunnel-like degradation along the $S_1$ layer. Tunnelling bacteria, in some samples, were able to degrade tunnel in the lignin-rich areas, such as middle lamella, suggesting that these bacteria had the capacity to degrade the lignin. IR spectra indicate that hemicellulose and cellulose in the waterlogged woods were preferentially decomposed. Breakdown of the lignin, on the other hand, was much slower.

Molecular Characterization of an Apple cDNA Encoding Cinnamyl Alcohol Dehydrogenase

  • The study of lignin, a major component of secondary cell wall, has been partly focused on its removal from the woody part in the kraft pulping industry. Cinnamyl alcohol dehydrogenase (CAD; EC 1.1.l95) catalyzes the synthesis of cinnamyl alcohols from corresponding cinnamaldehydes. A cDNA clone, MdCADl, encoding putative CAD from apples (Malus domestica Borkh. cv Fuji) was characterized in this study. The clone contains an open reading frame of 325 amino acid residues, which shows a greater than 80% identity with Eucalyptus CADl. MdCADl mRNA was detectable in vegetative tissues and was strongly expressed in the fruit. The expression pattern of MdCADl mRNA in the fruit peel after light exposure was also examined. The mRNA was rapidly increased until 1 day after light exposure and remained stable thereafter, suggesting that MdCADl is light inducible. The inducibility of the MdCADl gene was examined using several environmental stresses. Mechanical wounding of leaves increased the MdCADl mRNA level and the induction was further increased by salicylic acid. Southern blot hybridization showed that there is either one or a few copies of CAD genes in apples. To our knowledge, it is believed that MdCADl is the first CAD clone expressed predominantly in fruit.

  • A 61-years old man with diabetes mellitus(DM) was admitted to our hospital, complaining of progressive right periorbital swelling, headache and toothache on the right lower second molar. On the first visit, moderate swelling was noticed from the right periorbital region with exophthalmos and subconjunctival effusion. Intraorally, right lower second molar had a severe periodontal disease and fistular formation on its distal area. From 3 days after hospitalization, the visual acuity of his right eye was gradually worsen and we performed CT scan. CT scan demonstrated an inflammatory change at the right orbit with subperiosteal abscess at the inferior orbital wall, which was extended from the right infratemporal, parapharyngeal and internal pterygoid space. Patient was treated by mean of intraoral(right upper vestibular and retromolar) and extraoral(infraorbital) incision and drainage, massive anti-therapy and DM control. The patient improved gradually and finally was discharged from the hospital, but his visual loss of right side was not recovered.

Concave surface curvature effect on heat transfer from a turbulent round impinging jet (오목표면곡률이 난류원형충돌제트의 열전달에 미치는영향)

  • The effects of concave hemispherical surface curvature on the local heat transfer from a turbulent round impinging jet were experimentally investigated. The liquid crystal transient method was used for these measurements. This method, which is a variation on the transient method, suddenly exposes a preheated wall to an impinging jet while video recording the response of liquid crystals for the measurement of the surface temperature. The Reynolds number ranges from Re=11,000 to 50,000, the nozzle-to- surface distance from L/d=2 to 10, and the surface curvature from D/d=6 to 12.The present results are also compared to those for the flat plate case. In the experiment, the local Nusselt numbers tend to increase in all regions with an increasing surface curvature. The maximum Nusselt number for all Reynolds numbers occurred at L/d .ident. 6 and a second maximum in the Nusselt number occurred at R/d .ident. 2 for both Re=23,000 and Re=50,000 in the case of L/d=2 and for Re=50,000 only in the case of L/d=4. Meanwhile, as the surface curvature increases, the value of the secondary maximum Nusselt number decreases. All the other cases exhibit monotonically decreasing values of the Nusselt number along the curved surface. The stagnation point Nusselt numbers are well correlated with Re, L/d, and D/d.

Computational Analysis of the Flowfield of a Mixer-Ejector Nozzle (Mixer-Ejector 노즐 유동장에 관한 수치해석)

  • A time-iterative compressible Navier-Stokes code is developed to analyze the flowfield of a two-dimensional ejector nozzle system. A parametric study has been made for two controlling parameters, duct to nozzle area ratio and nozzle pressure ratio. Results show that there is an optimum area ratio for an efficient pumping of secondary flow. At high area ratios, a freestream flow directly passes through the mixing duct without giving adequate pumping. While at low area ratios, jet boundary is acting as a blockage to incoming flow. The nozzle pressure ratio variation shows that the pumping rate increases as the pressure ratio increases provided there is no interaction between the shroud wall and the shock cell structure.

Structure of Secondary Xylem and Cambial Initials in Pinus koraiensis S. et Z. Grown in Arid Soil (건지에서 자란 잣나무(Pinus koraiensis S. et Z.)의 이기목부 및 형성층구조의 변화)

  • This study deals with the effect of dwarf growth for the wood and the structural changes of cambium in Pinus koraiensis subjected to water stress. The length and wall thickness of tracheary elements in dwarf trees was shorter and thicker than that in normal trees. The redial width and cell number of the annual rings are narrower and smaller in dwarf trees than those in normal trees. In serial tangential sections, the frequency of anticlinal division is low and loss of cambial initials is small in dwarf trees. The length of cambial initials in water deficit, trees are slightly shorter than that in normal trees, and the instructive growth of cambial initials during differentiation of xylem mother cells is inactive in dwarf trees. Thus, it is interpreted that the shortening of tracheary elements in dwarf trees is due to the fact that the length of cambial initials are shortened and the intrusive growth of those during differentiation of xylem mother cells is inactive. The structural variations in the wood of naturally occurring dwarf trees are similar to those of trees subjected to artificial water stress. Therefore, it is suggested that the variation of xylem element in dwarf trees are related to water conditions.

Kosinostatin, a Major Secondary Metabolite Isolated from the Culture Filtrate of Streptomyces violaceusniger Strain HAL64

  • During a screening program, an actinomycete strain isolated from the Egyptian soil was investigated for its potential to show antimicrobial activity. The identification of this isolate was performed according to spore morphology and cell wall chemo-type, which suggested that this strain is a streptomycete. Further cultural, physiological characteristics and the analysis of the nucleotide sequence of the 16S rRNA gene (1480 bp) of this isolate indicated that this strain is identical to Streptomyces violaceusniger (accession number EF063682) and then designated S. violaceusniger strain HAL64. In its culture supernatant, this organism could produce one major compound strongly inhibits the growth of Gram-positive but the inhibition of Gram-negative indicator bacteria was lower. The antibiotic was separated by silica gel column chromatography and then purified on a sephadex LH-20 column and finally the purity was checked by HPLC. The chemical structure of the purified compound was determined using spectroscopic analyses (molecular formula of $C_{33}H_{32}N_{2}O_{10}$ and molecular weight of 617.21) and found to be identical to the kosinostatin, a quinocycline antibiotic which is known to be produced by Micromonspora sp. TP-A0468 (Igarashi et al., 2002) and to quinocycline B isolated from Streptomyces aureofaciens (Celmer et al., 1958). Although the antibiotic is known, the newly isolated strain was able to produce the antibiotic as a major product providing an important biotechnological downstream advantage.

Heat Transfer on a Heated Flat Plate by an Impinging Round Jet Using Liquid Crystal (Liquid Crystal을 이용한 원형충돌분류의 전열특성 연구)

  • Local heat transfer characteristics for a round air jet impinging normally on a heated flat plate were experimentally investigated. The problem parameters investigated were jet Reynolds number, Re=4000,10000, and 20000, and nozzle-to-plate spacing(L/D) of 2,6, and 10. The temperature variations on the flat uniform heat flux surface were mapped using a thermo-sensitive liquid crytal sheet. The isochromatic images corresponding to the characteristic temperature of liquid crystal were analyzed with the help of a digital image processing system. The local Nusselt number, Nu decreased rapidly in the impingement region and exhibited a similar profiles in the wall jet region independent of the nozzle-to-plate spacing L/D. In the case of large Reynolds number, heat transfer rate (Nu) was proportional to 0.5 power of the Reynolds number. For L/D=2, a secondary peak in the heat transfer rate was seen in the region of X/D=1.5~3 due to the transition from laminar to turbulent boundary layer.

Fabrication of Atmospheric Coplanar Dielectric Barrier Discharge and Analysis of its Driving Characteristics (평면형 대기압 유전장벽방전장치의 제작 및 동작특성분석)

  • The discharge characteristics of Surface Dielectric Barrier Discharge (SDBD) reactor are investigated to find optimal driving condition with adjusting various parameter. When the high voltage with sine wave form is applied to SDBD source, successive pulsed current waveforms are observed owing to multiple ignitions through the long discharge channel and wall charge accumulation on the dielectric surface. The discharge voltage, total charge between dielectrics, mean energy and power are calculated from measured current and voltage according to electrode gap and dielectric thickness. Discharge mode transition from filamentary to diffusive glow is observed for narrow gap and high applied voltage case. However, when the diffusive discharge is occurred with high applied voltage, the actual firing voltage is always lower than that with low driving voltage. The $Si_3N_4$, $MgF_2$, $Al_2O_3$ and $TiO_2$ are considered for dielectric protection and high secondary electron emission coefficient. SDBD with $MgF_2$ shows the lowest breakdown voltage. $MgF_2$ thin film is proposed as a protection layer for low voltage atmospheric dielectric barrier discharge devices.
