Amphibian Biotope Planning to Research on Reproduction of Bombina orientalis (무당개구리(Bombina orientalis) 번식 연구를 위한 양서류 바이오톱(amphibian biotope) 조성)

  • Bombina orientalis, or Korean fire-bellied toad is belonging to Class Amphinia, Order Anura, Family Bombinatoridae, Genus Bombina. Ecological characteristics, such as small size, slow moving and swimming, gentle attitude to other species, under laboratory and/or wild condition lead the toad as suitable experimental animals with easy handling. Seasonal reproductive biology except breeding season (usually from Apr. to late Jun.) are unknown still caused by sampling methods for hibernating specimens have not been established yet. Most species of amphibian, include B. orientalis, are declined around the world related with global warming, therefore, wild caught specimen for any purposes must be limited controled soon. Even though, some pre-studies of biotope for increasing biodiversity have been reported, construction of an artificial biotope to get natural breeding cycling specimen for study of amphibian reproduction is unique modelling and only trial. In an effort to set year-round sampling for study of reproduction of B. orientalis, we suggest protocols for planning, construction manual and SOP(Standard Operation Process) to manage amphibian biotope. During pilot operation of a designed and constructed biotope, the seasonal acclimation during winter hibernation of B. orientalis were successful. It will be helpful for studying the reproductive biology of wild amphibians in temperate region and allow to get reasonable research conditions in amphibian laboratory.

Characteristics of Salted Radish Cubes at Different Season (계절별 절임 무의 특성)

  • Characteristics (salt concentration, weight loss, soluble solid content, moisture content, and textural properties) of radish cube $(2{\times}2{\times}2\;cm)$ were evaluated during salting. Three different summer radish cultivars harvested in high land were immersed into 15% brine solution (radish : brine solution = 1 : 2) at $15^{\circ}C$ for 4 h. The time required to reach the optimum salt concentration (3%) was different among cultivars; 3 h for 'Bakbong', 4 h for 'Kwandong' and '92343'. Similar weight loss (about 24-26%) of salted radish cubes was found among cultivars. While the highest value of weight loss of salted radish cubes was observed with '92343', the lowest, 'Bakbong'. Hardness decreased during salting; the highest value of hardness was observer with 'Kwandong', the lowest, 'Bankbong'. The sample of '92343' has the lowest moisture content but has the highest soluble solid content and hardness, while 'Bakbong' has the highest moisture content, but has the lowest soluble solid content and '92343'. In separate experiments, seasonal variations in characteristics of salted radish cubes at optimum salt concentration (3%) were observed: for 'Bakbong', salt concentration, weight loss, soluble solid content, and hardness, and for 'Bakwang', salt concentration, weight loss, and hardness were higher in autumn cultivars than in spring or summer one, while moisture contents of both cultivars were higher in spring or summer cultivars than in autumn one.

  • CONSTANS (CO) induces the expression of FLOWERING LOCUS T (FT) in the photoperiodic pathway, and thereby regulates the seasonal timing of flowering. CO expression is induced and CO protein is stabilized by FLAVIN-BINDING KELCH REPEAT F-BOX PROTEIN 1 (FKF1) in the late afternoon, while CO is degraded by CONSTITUTIVE PHOTOMORPHOGENIC 1 (COP1) during the night. These regulatory cascades were thought to act independently. In our study, we investigated the relationship between FKF1 and COP1 in the regulation of CO stability in response to ambient light conditions. A genetic analysis revealed that FKF1 acts as a direct upstream negative regulator of COP1, in which cop1 mutation is epistatic to fkf1 mutation in the photoperiodic regulation of flowering. COP1 activity requires the formation of a hetero-tetramer with SUPPRESSOR OF PHYA-105 (SPA1), [$(COP1)_2(SPA1)_2$]. Light-activated FKF1 has an increased binding capacity for COP1, forming a FKF1-COP1 hetero-dimer, and inhibiting COP1 homo-dimerization at its coiled-coil (CC) domain. Mutations in the CC domain result in poor COP1 dimerization and misregulation of photoperiodic floral induction. We propose that FKF1 represses COP1 activity by inhibiting COP1 dimerization in the late afternoon under long-day conditions, resulting in early flowering.

Effect of seasonal changes on fertility parameters of Holstein dairy cows in subtropical climate of Taiwan

  • Objective: The purpose of this retrospective study was to investigate the relationship between temperature-humidity index (THI), season, and conception rate (CR) of Holstein cows in central Taiwan. Methods: The mean performance and number of observations were statistically evaluated for various parameters, including age at first service, number of days open, gestation length, CR, and calving interval for different parities. Results: The results indicate that the mean age at first service was 493.2 days; the gestation length was similar across all cows of different parities, ranging from 275.1 to 280.7 days. The overall CR of all inseminations was significantly lower in multiparous cows ($47.26%{\pm}0.22%$) than in heifers ($57.14%{\pm}0.11%$) (p<0.05). At THI>72 and during the hot season (from June to November), CRs for multiparous cows were significantly reduced compared to that for heifers, while the ratio remained unchanged among heifers for all seasons. Conclusion: To achieve a high CR, lactating cows should be bred in winter and spring (from December to May) from the start of the seasonal breeding program, whereas the heifer should be allowed to breed in summer and fall under the subtropical climate in Taiwan.

  • The aim of the current study was to compare the effects of season and breed on the reproductive performance of male and female sheep using 12 rams and 318 ewes of Assaf and Awassi breeds under the seasonal environmental condition of United Arab Emirates for two years. The blood level of testosterone hormone was measured monthly. Semen was collected twice a month from each male using artificial vagina and evaluated for volume, motility, livability, abnormality and concentration. The scrotal circumference and thickness as well as the left testicular length, width, height and volume were measured at one-month intervals. The level of testosterone in Assaf breed was significantly higher in autumn than winter and summer. The scrotal circumference and thickness as well as the left testicular length were significantly higher in Assaf breed than Awassi breed. While, left testicular width and volume were significantly higher in Awassi breed than Assaf breed. Scrotal circumference which was higher in spring and summer than in autumn and winter season in both breeds. The SCC of semen was significantly higher in autumn than in other seasons in both breeds. The sperm abnormality was significantly higher in summer than other seasons in both breeds. The livability was significantly lower in summer in both breeds. Fecundity and prolificacy were significantly higher in Assaf than Awassi breed during autumn season. Assaf breed showed the superior reproductive performance in the autumn season when compared with Awassi breed in the same season and other seasons. The Assaf breed tolerated the climatic conditions in UAE and kept the litter size of 1.72 in comparison to Awassi breed which showed litter size of 1.09. in conclusion, the results showed the superiority of Assaf over Awassi breed and offer a good model of breeding with increased fecundity and prolificacy specially in autumn season.

Testicular Cycles in the Korean Frogs: Annual Spermatogenic Patterns, Seasonal Changes in the Steroidogenic Competence, and Responsiveness Gonadotropins in vitro

  • Using three species of Korean frogs (Rana dybowskii, R. rugosa and R. nigromaculata), the annual spermatogenic pattern, the seasonal changes in the steroidogenic competence, and responsiveness of testis to gonadotropins in terms of testosterone secretion in vitro were examined. The spermatogenic pattern of R. dybowskii was classified as a discontinuous type since spermatogenesis stops completely after spawning in late winter (February) until mid-summer (July). In contrast, the pattern of R. nigromaculata and R. rugosa was classified as a potent continuous type since sperm was always present in the seminiferous tubules all year round. In all three species, the levels of testicular testosterone and that of testosterone secreted by testis following in vitro culture were very low in late summer (August), but increased thereafter until winter (hibernation period). Interestingly, responsiveness of testis in vitro to gonadotropins in terms of testosterone secretion increased markedly in November (early hibernation period). Specifically, bullfrog LH was more effective than FSH in stimulating the secretion of testosterone by frog testis in vitro during hibernation period. This fact suggests that testosterone secretion by testis during hibernation is at least regulated by the pituitary gonadotropin rather than environmental factors. Taken together, the data presented here suggest that testicular cycles of three species of Korean frogs are closely linked to their females breeding cycles, and are eventually controlled by various environmental cues.

Seasonal Prevalence of Mosquitoes Collected from Light Traps in Gyeongsangnam Province, Republic of Korea (2013-2014)

  • Adult mosquito surveillance was conducted from 2013 through 2014 at four cattle sheds, a wild bird refuge, and two residential areas located in Gyeongnam Province in the Republic of Korea. Adult mosquitoes were collected in black light traps from April 1, through November 30. Mosquito surveillance was conducted to figure out population densities of vector mosquitoes, possibly invaded mosquitoes and identify various virus infections at the selected sites. A total of 107,466 females comprising 14 species and 7 genera were collected from 2013 to 2014. The most common species collected were Culex tritaeniorhynchus Giles (63.8%), Anopheles sinensis s.l. (18.9%), Aedes vexans nipponii (Theobald) (7.7%), and Culex pipiens Coquillett (5.1%). Trap indices (TIs) varied widely for species over their range, due to geographical distribution and degree of association with rural and urban communities. The most collected An. sinensiss.l. and Cx. tritaeniorhynchus appeared at a cow shed in Hapcheon (TI 347.5) and a pigsty in Daejeo-1-dong, Busan (TI 1,040.8), respectively, due in part to their situation near breeding sites such as rice paddies. The bi-weekly population densities for mosquito species were variable for each of the years, apparently as a result of variable annual weather conditions. None of the mosquito species collected tested for the flavivirus including Japanese Encephalitis Virus, West Nile Virus, Dengue Virus, and Zika Virus infections by polymerase chain reaction (PCR) assay were positive.

Reproductive Responses of Awassi Ewes Treated with either Naturally Occurring Progesterone or Synthetic Progestagen

  • The objective was to identify the appropriate form of progesterone, which exhibits compact reproductive responses in Awassi ewes during mid to late seasonal anestrous period. Forty-eight Awassi ewes were randomly allocated into four groups to be treated with 60 mg medroxyprogesterone acetate (MAP), 30 mg fluorogestone acetate (FGA), 40 mg FGA, or 600 mg progesterone sponges. After a 12 day period, sponges were removed and ewes were administered i.m. with 600 IU PMSG (d 0, 0 h). Five harnessed Awassi rams were turned-in with the ewes to detect heat. Ewes were checked for breeding marks at 6 h intervals for 5 days. Blood samples were collected from all ewes for analysis of progesterone concentrations. Pretreatment (d -13 and -12) progesterone concentrations were ${\leq}0.2ng/mL$ among all ewes and were indicative of seasonal anestrous period. On d 0, progesterone concentrations were elevated to $1.4{\pm}0.1ng/mL$ in ewes received progesterone sponges only and were higher (p<0.0001) than those (${\leq}0.2ng/mL$) administered MAP or FGA sponges. Progesterone concentrations returned to their basal values of <0.2 ng/mL within 24 h of sponge removal and were similar (p>0.1) among all ewes. Incidence of estrus was similar (p>0.1) among the four groups and occurred in 75% (9/12), 82% (9/11), 67% (8/12) and 58% (7/12) of the ewes receiving MAP, 30 mg FGA, 40 mg FGA and progesterone sponges, respectively. Estrous responses occurred 14.7, 20 and 13.6 h earlier in progesterone-sponge-treated ewes than those of MAP- (p<0.04), 30 mg FGA- (p<0.01) and 40 mg FGA-treated (p=0.06) ewes, respectively. Induced estrus conception rates were 50% (6/12), 55% (6/11), 50% (6/12) and 42% (5/12), out of which 4/6, 4/6, 3/6 and 3/5 lambed 151 days following d 0, and were similar (p>0.1) among ewes of the four treatment groups. Ewes that returned to estrus 16 to 20 days following d 0 were 5/12, 5/11, 6/12 and 4/12 ewes treated with MAP, 30 mg FGA, 40 mg FGA and progesterone sponges, respectively, and all lambed 169 days later. Overall lambing rates were 75% (9/12), 82% (9/11), 75% (9/12) and 58% (7/12) ewes treated with MAP, 30 mg FGA, 40 mg FGA and progesterone sponges, respectively. Results demonstrate that applications of MAP, 30 mg FGA, 40 mg FGA and progesterone sponges Awassi ewes were equally effective in induction of estrus and tended to favor both types of FGA and MAP in overall lambing rates over progesterone sponges during the seasonal anestrous period.

A Study on Seasonal Prevalence of the Populations of the Mosquito Larvae and Other Aquatic Invertebrates in Rice Fields in Korea (水畓棲息 모기 幼蟲 및 其他 無脊椎動物 個體群密度에 關한 調査)

  • The field studies on the seasonal population prevalences of the vector mosquito larvae and other aquatic invertebrates were weekly carried out in the rice fields located in front of a village of Kwangtan-samri, Byeogje-myeon, Goyang-gun, Gyeonggido throughout mosquito breeding season (June-September) in 1980, and the results are as follows. The population of C. tritaeniorhynchus larvae increased soon after heavy rainfall, and decreased to some extent during the period of the insecticide application. The seasonal prevalence of A. sinensis was rather stable, not being affected by insecticide pressure at all. The population densities of other aquatic invertebrates in rice fields were seriously suppressed by the pesticide application, and their recovery was not as same as the previous level or not at all: (1) The populations of Odonata and Ephemeroptera nymphs were drastically decreased by the first application of insecticides and never recovered through out season. (2) Coleoptera seemed very susceptible to the insecticide application, as densities were markedly decreased whenever the insecticides were applied. (3) The high density of Hemiptera shown in early June were decreased sharply after the first application of pesticides, and thereasfter, some degree of recovery was shown, but suppressed by successive each application of pesticides. (4) A native species of planaria was exceptionally not influenced by the insecticide pressure, and two peaks of the density were appeared during the heavy rainfall.

Relationship between Temperature Distributions and Outbreaks of Harmful Algal Blooms in Korean Waters

  • Harmful algal blooms (HABs) of Cochlodinium polykrikoides frequently occur around the South Sea of Korea, causing. economic losses in coastal breeding grounds. HAB outbreak scale usually changes each year depending on physical, biological and environmental conditions. Relatively large-scale HABs occurred in 1995, 1997, 1999, 2001, 2002 and 2003 with respect to spatial scale, duration and maximum density. Considering HAB scale and temperature distributions around the South Sea, we found that low coastal temperatures in August correspond to enormous HAB outbreaks. Cold waters created by coastal upwellings around the southeastern coast of Korea also corresponded to these outbreaks. Serial oceanographic investigations in August in the South Sea revealed that sea surface temperature anomalies had distinctively negative values when large-scale HAB outbreaks appeared. With regard to temperature differences between the surface and the 30-m layer, there was a tendency for large-scale outbreaks when temperature gradients around the seasonal thermocline weakened.
