제주지역 갯메꽃(Calystegia soldanella L.) 유래 부정근 재분화 및 생물반응기 시스템 이용 대량증식법 (Regeneration of adventitious root from Calystegia soldanella L. in Jeju island and mass proliferation method using bioreactor system)

  • Calystegia soldanella L. is a perennial herbaceous halophyte belonging to the convolvulaceae family, which mainly grows in coastal sand dunes in Korea. Shoots and rhizomes are edible, and roots called 'Hyoseon Chogeun' are known to have medicinal effects such as antipyretic, sterilization, and diuretic. In addition, physiological activities of antioxidant, anti-inflammatory, antiviral, antifungal and PTP-1B (protein tyrosine phosphate-1B) inhibition have been reported. In this study, in vitro induction cell lines of C. soldanella L. collected from the coastal sand dunes in Jeju island was redifferentiated into adventitious roots that can be used as medicinal resources. Also the biomass of mass-proliferated adventitious roots using a bioreactor were evaluated. Plants of C. soldanella L. were collected from the crevice of the seashore in the coastal area of Taeheung 2-ri, Namwon-eup, Seogwipo-si. Then, it was separated into leaves, stems, rhizomes, and roots, and surface sterilized with 70% ethyl alcohol and 2% NaOCl (sodium hypochlorite). After washing with sterilized water, each organ section was cultured in Hormone-free MS medium (Murashige & Skoog Medium). As a result, the induction response rates were evaluated at 85% and 55%, respectively, in terms of callus formation and shoot generation in the rhizome segment. In the case of the adventitious roots morphological characteristics induced by single-use treatment of auxin-based plant growth regulators IBA and NAA from redifferentiated shoots were compared. Most efficient adventitious root culture method as a rooting rate, number, length, and biomass proliferation in the bioreactor system was confirmed when treated by culturing in MS salts, Sucrose 30 g·L-1, and IBA 1mg·L-1 for 4 weeks. In this study, the medium composition and culture period were confirmed using a bioreactor system to mass-proliferate adventitious roots derived from C. soldanella L. in Jeju island. Also this adventitious root line developed a new medicinal material could increase value of the bio-industry ingredient through quantitative and qualitative screening of phyto-bioactive compounds.

부산 내 미세먼지 고농도 지역 탐색 및 체내 휘발성유기화합물 바이오모니터링 조사 (Exploration of an Area with High Concentrations of Particulate Matter and Biomonitoring Survey of Volatile Organic Compounds among the Residents)

  • Background: With its developed port and related industries, the concentration of fine dust is high in Busan compared to other cities in South Korea. Many studies have reported the health effects of fine dust, but there has been a lack of information regarding concentrations of volatile organic compounds among those who exposed to high levels of fine dust. Objectives: This study aimed to define an area with high concentrations of particulate matter and perform biomonitoring surveys among the residents of the area. Methods: Air quality data was collected and the mean level of each district in Busan was derived. We then defined the area with the highest concentrations of PM10 as a target site. Urine samples were collected from the 400 participants and analyzed for VOCs metabolites - trans,trans-Muconic Acid (t,t-MA) and N-AcetylS-(benzyl)-L-cysteine (BMA). Interviews were conducted by trained investigators to examine demographic information. The levels of t,t-MA and BMA were compared with representative South Korean population data (Korean National Environmental Health Survey). The association of the VOC metabolites and fine dust were analyzed by general linear regression analysis. Results: The mean of PM10 in the target site was 42.50 ㎍/m3 from 2018 to 2020. Among the 400 participants in the target site, 74.8% were female and the average age of the participants was 66 years. The geometric mean of t,t-MA was 71.15 ㎍/g creatinine and the BMA was 7.00 ㎍/g creatinine among the residents. The levels were higher than the geometric mean from the 4th KoNEHS. The levels of t,t-MA showed significance in BMI, smoking status, and household income. BMA showed significance in gender and age. Conclusions: Compared to the general population of South Korea, the target site's residents had higher biomonitoring levels. Based on this study, continuous screening for high risk areas, including the target site, and biomonitoring of the residents are required.

사람의 O-linked-N-acetyl-${\beta}$-D-glucosaminidase 유전자를 함유한 대장균의 배양조건과 효소학적 특성 (Culture Conditions of E. coli Harboring Human O-Linked N-Acetyl-${\beta}$-Glucosaminidase Gene and Enzymatic Properties)

  • 단백질의 serine이나 threonine의 수산기에 N-acetyl-${\beta}$-D-glucosamine (O-GlcNAc)으로 변형되는 당쇄는 대부분의 진핵세포에서 광범위하게 일어나는 번역 후 수식의 일종으로 여러 세포 내 현상에 관여하고 있다. O-GlcNAc의 변형정도는 O-GlcNAc transfearse (OGT)와 O-linked N-acetyl-${\beta}$-D-glucosaminidase (O-GlcNAcase)에 의해 조절된다. O-GlcNAcase의 효소활성 조절물질을 탐색하기 위한 시험관 내 검정계를 확립하기 위해 재조합 O-GlcNAcase의 생산을 시도하였다. O-GlcNAcase 발현을 최적화하기 위한 배양조건을 검토한 결과 유도 배양온도 $30^{\circ}C$, 유도제인 L-arabinose 0.02%, 유도 배양시간 5시간 등으로 나타났다. 위의 조건에서 대장균 형질전환체를 배양하면서 한 시간 간격으로 배양액을 채취하여 세포를 파괴하고 얻은 효소용액의 활성은 배양 후 3시간에서 5시간까지는 급격히 증가하였으나 그 이후는 거의 증가하지 않았다. Western blot으로 발현된 단백질 양을 조사한 결과는 활성 그래프와 비슷한 양상을 보였다. 이렇게 생산한 O-GlcNAcase의 최적 반응조건은 pH 6.5, 반응온도 $45^{\circ}C$, 기질의 농도 2 mM, pH 6.5로 나타났다.

관상동맥 석회화 평가에서 저선량 흉부 CT와 관상동맥 석회화검사의 일치도 (Low-dose Chest CT in Evaluation of Coronary Artery Calcification: Correlation with Coronary Artery Calcium Score CT)

  • 폐 스크리닝 검사로 이용되고 있는 저선량 흉부 CT는 Scan 범위 내에 관상동맥 석회화에 대한 정보도 함께 포함하고 있어 이를 이용한 관상동맥 석회화 판별의 유용성을 알아보고자 한다. 저선량 흉부 CT 검사와 관상동맥 석회화 점수(CACS) 검사를 같은 날 시행 받은 자들을 대상으로 하였다. 관상동맥 석회화 점수 검사 결과를 Coronary artery calcium score categories and risks 분류법을 참고하여 4개 그룹(Low: 1〈CACS〈10, Mild: 10〈CACS〈100, Moderate: 100〈CACS〈400, High: 400〈CACS)으로 각각 30명을 선정한 후 관상동맥 석회화 수치 측정 업무에 종사하고 있는 경력 15년차 이상 5명의 방사선사가 저선량 흉부 CT 영상에서 관상동맥 석회화 유무를 후향적으로 분석하였다. 저선량 흉부 CT 영상에서 5명의 관찰자가 통일되게 판독한 결과가 관상동맥 석회화점수 CT 검사 결과와 일치한 경우는 Low 그룹: 56%, Mild 그룹: 96.6%, Moderate 그룹: 100%, High 그룹: 100%로 나타났다. Low 그룹에서 5명의 관찰자 모두가 석회화를 관찰한 것은 30건 중 17건이었으며, 5명이 모두 판별 불가로 결정한 경우 7건이었다. 무증상 성인을 대상으로 저선량 흉부 CT 검사에서 석회화 점수가 15 이상인 경우에는 관상동맥 석회화를 100% 관찰할 수 있었다. 판별이 가능한 최소 석회화 수치는 1로, 피검자의 체형이 작거나 심장의 움직임이 최소가 되는 시점에서 스캔이 이루어지는 경우 매우 작은 석회화까지도 판별할 수 있다는 것을 알 수 있었다.

유방 초음파 검사에서 석회화 유발 환경에 따른 반짝 허상과 팸텀을 활용한 영상 변화에 관한 연구 (A Study on the Image Change Using Twinkle Artifact Images and Phantom according to Calcification-Inducing Environment in Breast Ultrasonography)

  • 유방 초음파 검사는 지방형 유방에서 영상화가 어렵고 미세 석회화를 찾는 데 어려움이 있지만, 미세 석회화의 발견은 유방암 선별 검사에 매우 중요하다. 초음파의 컬러 도플러 허상 중 반짝 허상은 영상에서 주로 결석이나 석회화 같은 강한 반사체에 발생되며 이를 이용한 평가 방법이 임상적으로 사용 중이다. 본 연구는 유방 석회화 중 가장 많이 차지하는 인산칼슘을 이용한 유방 모의 팬텀을 제작하여 초음파 장비의 컬러 도플러 설정 변수인 펄스 반복 주파수, 앙상블, 퍼시스트, 월 필터, 평활화, 선밀도, 임계값에 대해 실험을 진행했고 이를 통해 유방 초음파 검사에서 반짝 허상의 대조도를 개선하는 조건과 임상에서 이를 최대한 활용하기 위한 방안을 연구하고자 하였다. 그 결과 펄스 반복 주파수가 3.6 kHz ~ 7.2 kHz 범위에서 반짝 허상이 발생하며 10.5 kHz 이상에서는 발생하지 않았다. 앙상블의 경우 낮은 조건에서는 모든 크기의 석회화에 반짝 허상이 발생하였고 임계값 설정에서는 80에서 100 조건에서만 반짝 허상이 조금 증가했으며 1 mm 크기의 석회화에서는 발생하지 않았다. 퍼시스트, 월 필터, 평활화, 선밀도 설정은 반짝 허상이 발생하긴 했으나 조건별로 증가하지는 않아 설정 변수에 큰 의미가 없었으며 반짝 허상에 가장 큰 영향을 미친 것은 펄스 반복 주파수, 앙상블, 임계값으로 나타났다. 본 연구는 검사자가 컬러 도플러 설정을 조절하여 반짝 허상을 효과적으로 증가시키는 최적의 조건을 선택하는 데 도움이 될 것으로 사료된다.

Effects of Contrast Phases on Automated Measurements of Muscle Quantity and Quality Using CT

  • Objective: Muscle quantity and quality can be measured with an automated system on CT. However, the effects of contrast phases on the muscle measurements have not been established, which we aimed to investigate in this study. Materials and Methods: Muscle quantity was measured according to the skeletal muscle area (SMA) measured by a convolutional neural network-based automated system at the L3 level in 89 subjects undergoing multiphasic abdominal CT comprising unenhanced phase, arterial phase, portal venous phase (PVP), or delayed phase imaging. Muscle quality was analyzed using the mean muscle density and the muscle quality map, which comprises normal and low-attenuation muscle areas (NAMA and LAMA, respectively) based on the muscle attenuation threshold. The SMA, mean muscle density, NAMA, and LAMA were compared between PVP and other phases using paired t tests. Bland-Altman analysis was used to evaluate the inter-phase variability between PVP and other phases. Based on the cutoffs for low muscle quantity and quality, the counts of individuals who scored lower than the cutoff values were compared between PVP and other phases. Results: All indices showed significant differences between PVP and other phases (p < 0.001 for all). The SMA, mean muscle density, and NAMA increased during the later phases, whereas LAMA decreased during the later phases. Bland-Altman analysis showed that the mean differences between PVP and other phases ranged -2.1 to 0.3 cm2 for SMA, -12.0 to 2.6 cm2 for NAMA, and -2.2 to 9.9 cm2 for LAMA.The number of patients who were categorized as low muscle quantity did not significant differ between PVP and other phases (p ≥ 0.5), whereas the number of patients with low muscle quality significantly differed (p ≤ 0.002). Conclusion: SMA was less affected by the contrast phases. However, the muscle quality measurements changed with the contrast phases to greater extents and would require a standardization of the contrast phase for reliable measurement.

Reliability of Skeletal Muscle Area Measurement on CT with Different Parameters: A Phantom Study

  • Objective: To evaluate the reliability of CT measurements of muscle quantity and quality using variable CT parameters. Materials and Methods: A phantom, simulating the L2-4 vertebral levels, was used for this study. CT images were repeatedly acquired with modulation of tube voltage, tube current, slice thickness, and the image reconstruction algorithm. Reference standard muscle compartments were obtained from the reference maps of the phantom. Cross-sectional area based on the Hounsfield unit (HU) thresholds of muscle and its components, and the mean density of the reference standard muscle compartment, were used to measure the muscle quantity and quality using different CT protocols. Signal-to-noise ratios (SNRs) were calculated in the images acquired with different settings. Results: The skeletal muscle area (threshold, -29 to 150 HU) was constant, regardless of the protocol, occupying at least 91.7% of the reference standard muscle compartment. Conversely, normal attenuation muscle area (30-150 HU) was not constant in the different protocols, varying between 59.7% and 81.7% of the reference standard muscle compartment. The mean density was lower than the target density stated by the manufacturer (45 HU) in all cases (range, 39.0-44.9 HU). The SNR decreased with low tube voltage, low tube current, and in sections with thin slices, whereas it increased when the iterative reconstruction algorithm was used. Conclusion: Measurement of muscle quantity using HU threshold was reliable, regardless of the CT protocol used. Conversely, the measurement of muscle quality using the mean density and narrow HU thresholds were inconsistent and inaccurate across different CT protocols. Therefore, further studies are warranted in future to determine the optimal CT protocols for reliable measurements of muscle quality.

Validation of Deep-Learning Image Reconstruction for Low-Dose Chest Computed Tomography Scan: Emphasis on Image Quality and Noise

  • Objective: Iterative reconstruction degrades image quality. Thus, further advances in image reconstruction are necessary to overcome some limitations of this technique in low-dose computed tomography (LDCT) scan of the chest. Deep-learning image reconstruction (DLIR) is a new method used to reduce dose while maintaining image quality. The purposes of this study was to evaluate image quality and noise of LDCT scan images reconstructed with DLIR and compare with those of images reconstructed with the adaptive statistical iterative reconstruction-Veo at a level of 30% (ASiR-V 30%). Materials and Methods: This retrospective study included 58 patients who underwent LDCT scan for lung cancer screening. Datasets were reconstructed with ASiR-V 30% and DLIR at medium and high levels (DLIR-M and DLIR-H, respectively). The objective image signal and noise, which represented mean attenuation value and standard deviation in Hounsfield units for the lungs, mediastinum, liver, and background air, and subjective image contrast, image noise, and conspicuity of structures were evaluated. The differences between CT scan images subjected to ASiR-V 30%, DLIR-M, and DLIR-H were evaluated. Results: Based on the objective analysis, the image signals did not significantly differ among ASiR-V 30%, DLIR-M, and DLIR-H (p = 0.949, 0.737, 0.366, and 0.358 in the lungs, mediastinum, liver, and background air, respectively). However, the noise was significantly lower in DLIR-M and DLIR-H than in ASiR-V 30% (all p < 0.001). DLIR had higher signal-to-noise ratio (SNR) and contrast-to-noise ratio (CNR) than ASiR-V 30% (p = 0.027, < 0.001, and < 0.001 in the SNR of the lungs, mediastinum, and liver, respectively; all p < 0.001 in the CNR). According to the subjective analysis, DLIR had higher image contrast and lower image noise than ASiR-V 30% (all p < 0.001). DLIR was superior to ASiR-V 30% in identifying the pulmonary arteries and veins, trachea and bronchi, lymph nodes, and pleura and pericardium (all p < 0.001). Conclusion: DLIR significantly reduced the image noise in chest LDCT scan images compared with ASiR-V 30% while maintaining superior image quality.

  • Percutaneous transthoracic needle biopsy (PTNB) is one of the essential diagnostic procedures for pulmonary lesions. Its role is increasing in the era of CT screening for lung cancer and precision medicine. The Korean Society of Thoracic Radiology developed the first evidence-based clinical guideline for PTNB in Korea by adapting pre-existing guidelines. The guideline provides 39 recommendations for the following four main domains of 12 key questions: the indications for PTNB, pre-procedural evaluation, procedural technique of PTNB and its accuracy, and management of post-biopsy complications. We hope that these recommendations can improve the diagnostic accuracy and safety of PTNB in clinical practice and promote standardization of the procedure nationwide.

공공측량 성과심사 표본추출 자동화 가능성 분석 (Automation of Sampling for Public Survey Performance Assessment)

  • 공간정보품질관리원에서 수행되는 공공측량 성과심사는 규정에 따른 심사 비율로 수행하며, 추출된 표본을 기반으로 제출된 성과의 전체적인 경향을 심사자가 직접 판단한다. 그러나 국토지리정보원고시 '측량성과 심사수탁기관의 심사업무 및 지정절차 등에 관한 규정'에는 무작위 추출(Random Collection)이라 명시되어 있으나 공공측량 성과심사 표본추출은 심사 대상별로 심사자가 직접 판단으로 표본을 추출하고 있어 성과심사 표본추출의 공정성 확보가 필요하다. 본 연구에서는 실제 현장의 여건을 확인하고 실제 성과심사 데이터 확보를 통해 심사자들의 세부적인 기준을 분석하였다. 또한, 다양한 현장 여건에 따른 고려사항을 분석하여 공공측량 성과심사 표본추출 알고리즘 적용 방안에 대해 연구하였다. 따라서 성과심사자들의 세부적인 표본추출 기준 분석이 필요하다. 실제 성과심사가 완료된 데이터와 Python을 활용한 자동화 프로그램의 결과 비교를 통해 상대적으로 비교가능하도록 하였으며, 본 자동화 프로그램은 향후 공공측량 성과심사 표본추출의 자동화 적용을 위한 기반 프로그램으로 활용이 가능할 것으로 판단된다.
