Sclerotium rolfsii에 의한 설악초 흰비단병 (Stem Rot of Euphorbia marginata Caused by Sclerotium rolfsii)

  • 경상남도농업기술원 화단에서 S. rolfsii에 의한 설악초 흰비단병이 발생되었다. 병징은 줄기가 수침상으로 물러지고 썩으면서 서서히 시들어 말라 죽었다. 병든 줄기와 지제부, 토양 표면에 흰색의 곰팡이가 발생하고 갈색의 작은 둥근균핵을 많이 형성하였다. 균핵의 크기는 1~3mm이며 균사의 폭은 4~9 ${\mu}m$였다. 균사특유의 clamp connection이 관찰되었다. 균사생육과 균핵형성 적온은 $30^{\circ}C$이었다. 설악초에서 발생한 병징과 병원균의 균학적 특징을 조사한 결과, 이 병을 Sclerotium rolfsii Saccardo에 의한 설악초 흰비단병으로 명명하고자 제안한다.

  • 2002년 경상남도 진주시 도로변 화단에 재배중인 팬지에서 줄기가 수침상으로 물러지고 부패하는 증상이 관찰되었다. 이 병의 병징은 지제부가 암갈색으로 변하고 그위에 흰색의 곰팡이가 솜털처럼 밀생하며, 병반 부위뿐만 아니라 지제부의 토양 표면에도 갈색의 둥근 균핵이 많이 형성된다. 병원균의 균사생장 및 균핵형성은 PDA배지에 배양시 3$0^{\circ}C$에서 가장 양호하였다. 균사에는 특유의 clamp connection이 관찰되었고 균사 폭은 3.9~10.4 $\mu\textrm{m}$이었다. 균핵은 갈색 또는 암갈색으로 크기는 1.3~4.2 mm 이었다. 이상의 특징으로 이 병원균은 Sclerorium rolfsii로 동정하였으며 팬지 흰비단병으로 명명할 것을 제안한다.

마늘의 흑색썩음균핵병에 대한 품종저항성의 역학적 평가 (Pathogenicity of Two Species of Sclerotium to Nine Cultivars of Garlic Cultivated Widely in Korea)

  • 마늘흑색썩음균핵병에 대한 국내재배 마늘의 품종저항성을 구명하기 위하여 발병에 관여하는 Sclerotium cepivorum과 대균핵을 형성하는 Sclerotium sp.에 대한 invitro clove inoculation법에 의한 균사생장과 균핵형성소요 일수 그리고 이병포장에서의 품종별 발병정도를 조사하였다. 마늘인편 절편상에서의 병원균의 생장과 균핵형성은 품종 또는 병원균간에 차이를 보였다. 대서종에서는 두 가지 병원균 모두 가장 높은 균사생장을 보였다. S. cepivorum과 Sclerotium sp.의 균핵은 고당종에서 가장 빠르게 형성되었고, 남도종에서 가장 늦게 형성되었다. 포장에서의 품종별 병 저항성에 있어서는 S. cepivorum에 대해서는 모든 마늘에서 이병주율이 50% 이상으로 시험 품종 모두 감수성인 것으로 판명되었으며, 난지형마늘에 비해 한지형마늘에서 발병이 적었다. Sclerotium sp.에 대해서는 난지형 마늘 모두 고도의 감수성이었고, 한지형마늘은 서산종, 단양종 그리고 예천종이 중도저항성이었으며 의성종은 저항성인 것으로 나타났다. 마늘의 품종저항성을 정량적으로 평가하기 위하여 병진전곡선을 1차 회귀직선화하여 얻은 초기발병치(intercept), 병진전속도 및 병진전곡선하면적(AUDPC)을 비교한 결과 초기발병과 병진전이 함께 고려된 AUDPC가 품종별 병저항성을 평가하는데 적당한 것으로 나타났다. In vitro 조건하에서의 균사생장 및 균핵형성과 포장에서의 역학지수(초기발병치, 최종발병치, 병진전속도, AUDPC)와의 상관관계를 조사한 결과, S. cepivorum에 대하여 균사생장과 최종발병치, 균사생장과 병진전속도간에는 정의상관을, 그리고 균핵형성과 역학지수간에는 부의 상관을 보였으나 5% 수준에서 유의성은 없었다.

Phenolic Acid Changes in Mycelia of Sclerotium rolfsii After Garlic and Onion Supplementation in a Broth Medium

  • High performance liquid chromatographic (HPLC) analysis of mycelia of Sclerotium rolfsii grown in broth medium supplemented with garlic (Allium sativum) and onion (Allium cepa) was carried out to estimate qualitative and quantitative changes in phenolic acids. Several phenolic acids, such as gallic, chlorogenic; ferulic, o-coumaric and cinnamic acids were detected in varied amounts in mycelia grown on such media as compared to control. Phenolic acids represents a wide range of secondary metabolite found in the cells of plants and microbes including fungi. The growth characters of S. rolfsii in various supplements also varied from thin and transparent to thick and opaque.

Stem Rot of Tomato Caused by Sclerotium rolfsii in Korea

  • A destructive stem rot of tomato(Lycopersicon esculentum) occurred sporadically some farmers' fields in Jinju City, Gyeongnam province in Korea. The infected plants also showed stem, crown rot or whole plant blight. White mycelium spread over stems of infected plants and formed sclerotia on the old lesions nearby soil surface. The fungus showed maximum mycelial growth around $30^{\circ}C$. The fungus formed white colony on PDA, usually with many narrow mycelial strands in the aerial mycelium and the width were $4.0{\sim}9.8{\mu}m$. The typical clamp connections were formed on the mycelium. Numerous sclerotia was formed on PDA at $30^{\circ}C$. The shape of sclerotia was globoid and $1.0{\sim}3.0$ mm in size. The fungus was isolated repeatedly from the infected tissues and the pathogenicity was confirmed to tomato and identified as Sclerotium rolfsii. This is the first report on the stem rot of tomato caused by S. rolfsii in Korea.

Stem Rot of Tawny Daylily(Hemerocallis fulva) Caused by Sclerotium rolfsii in Korea

  • In July 2002, a destructive stem rot of tawny daylily(Hemerocallis fulva) was occurred sporadically in exhibition farm of Gyeongsangnam-do Agricultural Research and Extension Services located in Hamyang-gun, Korea. The fungus also caused collar and crown rot, and systemic wilt or blight of whole plant. White mycelium spread over stems and petioles of infected plants and sclerotia were formed on the old lesions and near the soil surface. The optimum temperature for mycelial growth and scierotial formations was $30^{\circ}C$ on PDA. The mycelial width ranged $4.2{\sim}10.4{\mu}m$ and the color was white, usually many narrow mycelial strand grew in the aerial mycelium and formed clamp connection. The shape of sclerotia was spherical and $1.0{\sim}3.2$ mm in diameter. The fungus was isolated repeatedly from the infected tissues and confirmed its pathogenicity to Hemerocallis fulva and identified as Sclerotium rolfsii. This is the first report on the stem rot of H. fulva caused by S. rolfsii in Korea.

Sclerotirum cepivorum에 대한 마늘 재배종의 저항성 검정 (Screening for Resistance of Garlic Cultivars to White Rot Caused by Sclerotium cepivorum)

  • The optimal quantity of inoculum was determined to screen resistance of garlic cultivars against Sclerotium cepivorum and 30 cultivars was tested. The growth of the pathogen in detached roots, Disease incidence was increased when the inoculum density was raised form 10 to 100 sclerotia. The optimal inoculum density to differentiate resistance or susceptibility of garlic cultivars was seemed to be 50 sclerotia. The cultivars collected from England, Japan, Nepal and Turkey, and cultivars such as common red, PI1356104 and PI135693 were less than the other cultivars in their disease incidence. The growth of S. cepivorum in detached roots varied from 23 to 33 mm according to garlic cultivars. There was no relationship between the disease incidence and the growth in detached roots. The sclerotial germination was increased significantly when root extract was extract was added. The addition of only distilled water resulted in 13% germination, but the addition of 0.25 g of root extract in 100 ml distilled water resulted in more than 85% germination. There was no difference in the stimulation of sclerotial germination among cultivars which showed different resistance.

Stem Rot of Strawberry Caused by Sclerotium rolfsii in Korea

  • A destructive stem rot of strawberry (Fragaria x ananassa cv. Akihime) sporadically occurred in farmers' fields in Daegok-myon, Jinju city, Gyeongnam province in Korea. The infected plants showed stem and crown rot, with occasional blighting of the whole plant. White mycelia appeared on stems of infected clones and sclerotia formed on the old lesions near soil surface. The fungus formed white colony on PDA and showed maximum mycelial growth and sclerotial formation at $30^{\circ}C$. The fungus usually have many narrow hyphal strands, 2.6-10.0 $\mu\textrm{m}$ in width, in the aerial mycelium. Typical clamp connections were formed on the mycelium. Sclerotia were spherical and 1.0-2.4 mm in size. The fungus was repeatedly isolated from infected tissues and identified as Sclerotium rolfsii. Its patho-genicity was confirmed when inoculated onto straw-berry. This is the first report on the stem rot of strawberry caused by S. rolfsii in Korea.

Stem Rot of Garlic (Allium sativum) Caused by Sclerotium rolfsii

  • Stem rot disease was found in garlic (Allium sativum L.) cultivated from 2008 to 2010 in the vegetable gardens of some farmers in Geumsan-myon, Jinju City, Gyeongnam province in Korea. The initial symptoms of the disease were typical water-soaked spots, which progressed to rotting, wilting, blighting, and eventually death. White mycelial mats had spread over the lesions near the soil line, and sclerotia had formed over the mycelial mats on the stem. The sclerotia were globoid in shape, 1~3 mm in size, and tan to brown in color. The optimum temperature for growth and sclerotia formation on potato dextrose agar (PDA) medium was $30^{\circ}C$. The diameter of the hyphae ranged from approximately 4 to $8\;{\mu}m$. Typical clamp connection structures were observed in the hyphae of the fungus, which was grown on PDA medium for 4 days. On the basis of the mycological characteristics and pathogenicity of the fungus on the host plants, the causal agent was identified as Sclerotium rolfsii Saccardo. This is the first report of stem rot disease in garlic caused by S. rolfsii in Korea.

Effect of Tricho-compost against Seedling Blight Disease of Wheat Caused by Sclerotium rolfsii

  • The efficacy of formulated Trichoderma harzianum-based Tricho-compost, seed treatment with Tricho-inocula, and chemical fungicide Provax 200 WP against foot and root rot diseases of wheat caused by Sclerotium rolfsii was tested in the pot house and in the research field of Plant Pathology Division, Bangladesh Agricultural Research Institute, Gazipur, Bangladesh. Tricho-compost was prepared with a mixed substrate of cow dung, rice bran, and poultry refuse colonized by T. harzianum. Seedling mortality of wheat was significantly reduced by the Tricho-compost, Tricho-inocula, and Provax 200 WP both in the pot house as well as in the field experiments. The yield of wheat was sharply increased over the control due to the T. harzianum formulations and Provax 200 WP. Among the treatments, soil application of Tricho-compost was more efficient in reducing seedling mortality and accelerating plant growth with an increased yield of wheat with S. rolfsii-inoculated pot cultures and field experiments.
