비알코올성 지방간질환의 이수(利水) 약재 포함 한약 치료에 대한 최신 임상 연구 동향 - CAJ 검색을 중심으로 (Review of Clinical Research about Herbal Medicine Treatment Containing Diuretic Herbs on Non-Alcoholic Fatty Liver Disease - Research on China Academic Journal)

  • Objective: This study reviewed recent clinical research trends regarding the effects of herbal medicine treatment containing diuretic herbs on nonalcoholic fatty liver disease (NAFLD) in China. Methods: We reviewed clinical studies on NAFLD from the China Academic Journal of the China National Knowledge Infrastructure from January 1, 2016, to September 10, 2018. Results: Thirty-nine papers from 86 studies were reviewed. The most commonly used herbal prescriptions included Alismatis Rhizoma(澤瀉), Salviae miltiorrhizae Radix (丹蔘), Crataegii Fructus (山査), Polia Sclerotium (茯?), Bupleuri Radix (柴胡), and Cassiae Semen (決明子). The treatment effective rate, liver function test, serum lipid test, radiological examination, and symptom comparisons were used to analyze the treatments. In 32 of the papers, the effective treatment rate was 76.9%-100%, and the effectiveness in the experimental group was significantly higher than that in the control group (p<0.05). The risk ratio for the treatment effective rate was 1.26. The mean difference of ALT was -13.44 U/L (95% confidential interval, -24.45 U/L, -2.44 U/L, p=0.0166). Conclusion: Herbal medicine containing diuretic herbs has been demonstrated to be an effective and useful treatment for NAFLD. Clinical studies that are more scientific and systematic should be actively conducted in the future, and the results of the current study could be used as basic data in future clinical studies on herbal medicine treatment for NAFLD.

LC-ESI-MS에 의한 사군자탕의 지표성분 분석 (Analysis of the Marker Compounds in Sagunja-tang by LC-ESI-MS)

  • One of the oriental medicine prescriptions, Sagunja-tang consists of four herbal medicines (Ginseng Radix, Poria Sclerotium, Atractylodis Rhizoma Alba, and Glycyrrhiziae Radix et Rhizoma) and has been used as a medicine to enhance tonify the function of spleen and stomach in Korea. In this study, we conducted simultaneous analysis of the 9 marker components, liquiritin apioside, liquiritin, ginsenoside Rg1, liquiritigenin, ginsenoside Rb1, glycyrrhizin, atractylenolide III, atractylenolide II, and atractylenolide I in Sagunja-tang using a liquid chromatography-electrospray ionization-mass spectrometry (LC-ESI-MS/MS). Marker compounds were separated on a Waters Acquity UPLC BEH $C_{18}$ analytical column ($2.1{\times}100mm$, 1.7 mm) and the column was maintained at $45^{\circ}C$. The mobile phase consists of 0.1% (v/v) aqueous formic acid and acetonitrile with gradient condition. The LC-MS analysis was performed using a Waters ACQUITY TQD LC-MS/MS system with multiple reaction monitoring (MRM) method in the positive and negative modes. The calibration curves of the nine marker components showed good linearity with coefficient of determination ${\geq}0.9984$ within tested range. The limits of detection and limits of quantification values were 0.27-2.42 ng/mL and 0.81-7.27 ng/mL, respectively. The concentrations of tested 9 analytes in the lyophilized Sagunja-tang sample using the established LC-ESI-MS/MS MRM method were detected up to 16.593 mg/g. These results can be useful as a basic data for the quality control of an oriental medicine prescriptions.

CNKI 검색을 통한 중국에서의 성장통의 한의학적 치료에 대한 임상연구 동향 (Review of Clinical Studies of Traditional Chinese Medicine Treatment for Growing Pains in China using CNKI Database)

  • Objectives The purpose of this study is to investigate clinical studies on the effectiveness of Traditional Chinese Medicine treatment on growing pains in China, and to seek better methods to treat and study for growing pains in South Korea. Methods The clinical studies from the China Academic Journal (CAJ) in China National Knowledge Infrastructure (CNKI) that were published up to January 2019 were reviewed. The key word used were '生長痛', '發育期痛', '治療', '中?治療', '中藥'. Then, the literature were analyzed in regards to the treatment methods and results. Results Among the 35 searched studies, 1 randomized controlled trials, 16 case series, and 3 case reports were selected and analyzed. In most of the studies, the effectiveness of Traditional Chinese Medicine treatment on growing pains was significantly positive. The most commonly used herbs for oral administration were Glycyrrhizae Radix et Rhizoma (甘草), Achyranthis Bidentatae Radix (牛膝), Paeoniae Radix Alba (白芍藥), Dioscoreae Rhizoma (山藥), Angelicae Gigantis Radix (當歸), Astragali Radix (黃?), Atractylodis Rhizoma Alba (白朮), Poria Sclerotium (茯?) Rehmanniae Radix Preparata (熟地黃), Lycopodii Herba (伸筋草). Other treatment methods such as, external application of herb medicine, massage, acupuncture were also used to treat growing pains. Conclusion Based on the results of the clinical studies from China, the use of chinese medicine for treatment of growing pains has been shown to be effective. Additional clinical studies are needed to confirm these findings.

  • The purpose of this study was to analyze the Korean medicine treatment methods for Postherpetic Neuralgia (PHN) in Korea. There were 5 online databases searched (OASIS, NDSL, RISS, KISS and KMBASE) for studies which were related to PHN. A total of 12 studies were selected. Various treatments such as acupuncture, herbal medicine, and moxibustion were reported treatments for PHN, some of which included Western medicine and Korean medicine treatment. Korean herbal medicine was the most frequently used treatment method, followed by manual acupuncture. Sipjeondaebotanggami was the most frequently used prescription. Poria Sclerotium was the most frequently used principal herb, followed by Angelicae Gigantis Radix, Zingiberis Rhizoma, and Ginseng Radix. GB44, LI4, LR3 were the common acupoints used for the treatment of PHN. CV12 was the most frequently used moxibustion point, and gabapentin was the most frequently used concomitant Western drug. Additional research on Korean medicine treatment of PHN is expected in the future.

다낭신의 중의치료에 대한 임상연구 동향 - CNKI검색을 중심으로 (A Review of Clinical Studies of Chinese Medicine Treatment of Polycystic Kidney Disease Using the CNKI Database)

  • Objective: This research reviews and investigates the trends in recent clinical studies of polycystic kidney disease (PKD) in China. Method: We searched for clinical studies discussing Oriental medicine-based treatments for PKD in the China National Knowledge Infrastructure (CNKI) database. Thirteen clinical articles published from 2001 to 2019 were analyzed. The search focused on the authors, publication year, type of study, purposes of study, method and duration of treatment, evaluation criteria, and results of the selected articles. Results: Of the articles from the database, 9 case series and 4 randomized controlled trials (RCTs) were analyzed. Ten articles used herbal medicine; 4 used herbal medicine for external use. Gamigyejibokryeong-hwan was the most common herbal prescription. The most frequently used herb was Polia Sclerotium (茯苓), and Cnidii Rhizoma (川芎) was employed in all the external uses. All 13 studies confirmed the efficacy of Oriental medicine treatments. Conclusion: 1. Scientifically designed and more varied clinical studies are required to develop treatments for PKD. 2. The current study could be used as basic data in future clinical studies on treatment and further studies of PKD.

  • Objectives: This study investigated the latest clinical studies on Korean medicine treatment of idiopathic edema in the Chinese National Knowledge Infrastructure Database (CNKI). Methods: We searched the last 9 years of clinical studies discussing Oriental medicine-based treatments for idiopathic edema in the CNKI database. The search focused on the authors, publication year, type of study, purposes of study, method and duration of treatment, evaluation criteria, and results of the selected articles. Results: The search identified 7 randomized controlled trials (RCTs) and 9 others. All studies used oral herbal medicine for treatment. The most commonly used herbs were Poria Sclerotium (茯苓), Atractylodis Rhizoma Alba (白朮), and Alismatis Rhizoma (澤瀉), which are the constituent herbs of Oryeong-san (五苓散). All 16 studies confirmed the efficacy of Oriental medicine treatments. Conclusion: More varied and scientifically designed clinical studies are required to develop treatments for idiopathic edema. The results of this study could be used as basic data for further studies on idiopathic edema.

오령산 구성약재 성분의 Drug-likeness와 Oral bioavailability (Drug-likeness and Oral bioavailability for Chemical Compounds of Medicinal Materials Constituting Oryeong-san)

  • Objectives : Oryeong-san was composed of Alismatis Rhizoma, Atractylodis Rhizoma Alba, Poria Sclerotium, Polyporus, Cinnamomi Cortex, and known to have hundreds of chemical compounds. The aim of this study was to screen chemical compounds constituting Oryeong-san with the drug-likeness and oral bioavailability from the analysis of their physicochemical properties. Methods : A list of chemical compounds of Oryeong-san was obtained from TM-MC(database of medicinal materials and chemical compounds in Northeast Asian traditional medicine). To remove redundant compounds, the SMILES (Simplified Molecular Input Line Entry System) strings of each compound were identified. All of the physicochemical properties for the compounds were calculated using the DruLiTo(Drug Likeness Tool). Drug-likeness was estimated by QED(Quantitative Estimate of Druglikeness) and OB(Oral bioavailability) was checked based on the Veber's rules. Results : A total of 475 compounds were obtained by eliminating duplication among 544 compounds of 5 medicinal materials. Analysis of the physicochemical properties revealed that the most common values were MW(molecular weight) 200~300 g/mol, ALOGP(octanol-water partition coefficient) 1~2, HBA(number of hydrogen bond acceptors) 0~1, HBD(number of hydrogen bond donors) 0, PSA(polar surface area) 0~50 angstrom, ROTB(number of rotatable bonds) 1, AROM(number of aromatic rings) 0, and ALERT(number of structural alerts) 1. QED had 93% of the values between 0.2 and 0.7, and OB had 90% of the value of TRUE. Conclusions : We in this paper screened the candidate active compounds of Oryeong-san using the QED and Veber's rules. In the future, we will use the screening results to analyze the mechanism of Oryeong-san based on systems pharmacology.

≪상한론≫ 탕제에서 한약 하루 먹는 량 최대와 최소 비율에 근거한 복용량 1 량(兩) 교정 (Proofreading of one Ryang based on the Ratio of Maximum and Minimum Dose in the Decoction of ≪Treatise on Cold Damage Diseases≫)

  • Objectives : The purpose of this study was to proofread 'one ryang' in the Decoction of ${\ll}$Treatise on Cold Damage Diseases${\gg}$. Methods : I found out the ratio of maximum dose and minimum dose in this book. On the basis of the ratio, I corrected 'one ryang' in diverse decoctions. Results : In any decoction, maximum dose of medicinal medica in one decoction could not exceed four times minimum dose. Specifically, in the case that maximum dose in one decoction is sixteen ryang, it could not exceed eight times minimum dose in the same decoction. Any medicinal medica used in two decoctions or more, its maximum dose could not exceed four times minimum dose in other decoctions. On the basis of these results, it should be changed into three ryangs that are one ryang dose of 'Haematitum' of Seonbokdaeja Tang, 'Ginger' of Bujageongang Tang, Baektong Tang, Baektonggajeodamjep Tang and Senggangsasim Tang. Furthermore it should be changed into two ryangs that are one ryang dose of 'Coptidis Rhizoma' of Sohamhyung Tang, 'Ginger' of Dowha Tang, 'Ginseng Radix' of Whubaksenggangbanhagamchoinsam Tang, 'Polyporus, Poria Sclerotium, Alismatis Rhizoma, Talcum and Asini Corii Colla' of Jeoryeong Tang, 'Cimicifugae Rhizoma, Atractylodis Rhizoma Alba and Anemarrhenae Rhizoma' of Mahuangshengma Tang and 'Cassiae Cortex Interior' of Gyejigamchoryonggolmoryeo Tang. Conclusions : These results suggest that one ryang of thirteen medicinal medica such as Haematitum or Ginger of eleven decoctions such as Seonbokdaeja Tang or Bujageongang Tang should be changed into two or three ryangs.

  • 본 연구는 액체 정치배양한 Inonotus obliquus의 균사체 조성분을 분석하였으며, I. obliquus의 균핵과 진탕배양한 균사체의 성분조성을 비교하였다. I. obliquus 균의 정치배양 균사체가 생산하는 화학성분을 분석한 결과, 단리된 화합물은 lanosterol(1), inotodiol(2), trametenolic acid(3), 3β,22,25-trihydroxy-lanosta-8-ene(6), 3β,22-dihydroxy-lanosta-8,24-diene(A), 3β-hydroxy-lanosta-8,24-dien-21-al(B) 및 methyl trametenolate(C)이 확인되었다. 액체진탕배양 균사체가 생산하는 화합물과의 비교에 의해, 공통화합물은 lanosterol(1)이었고, ergosterol(7)과 ergosterol peroxide(8)는 액체진탕배양, lanosterol(1)의 C-21치환화합물과 C-22치환화합물은 정치배양함으로써 얻어짐을 알 수 있었다.

Evaluation and Genome Mining of Bacillus stercoris Isolate B.PNR1 as Potential Agent for Fusarium Wilt Control and Growth Promotion of Tomato

  • Recently, strategies for controlling Fusarium oxysporum f. sp. lycopersici (Fol), the causal agent of Fusarium wilt of tomato, focus on using effective biocontrol agents. In this study, an analysis of the biocontrol and plant growth promoting (PGP) attributes of 11 isolates of loamy soil Bacillus spp. has been conducted. Among them, the isolates B.PNR1 and B.PNR2 inhibited the mycelial growth of Fol by inducing abnormal fungal cell wall structures and cell wall collapse. Moreover, broad-spectrum activity against four other plant pathogenic fungi, F. oxysporum f. sp. cubense race 1 (Foc), Sclerotium rolfsii, Colletotrichum musae, and C. gloeosporioides were noted for these isolates. These two Bacillus isolates produced indole acetic acid, phosphate solubilization enzymes, and amylolytic and cellulolytic enzymes. In the pot experiment, the culture filtrate from B.PNR1 showed greater inhibition of the fungal pathogens and significantly promoted the growth of tomato plants more than those of the other treatments. Isolate B.PNR1, the best biocontrol and PGP, was identified as Bacillus stercoris by its 16S rRNA gene sequence and whole genome sequencing analysis (WGS). The WGS, through genome mining, confirmed that the B.PNR1 genome contained genes/gene cluster of a nonribosomal peptide synthetase/polyketide synthase, such as fengycin, surfactin, bacillaene, subtilosin A, bacilysin, and bacillibactin, which are involved in antagonistic and PGP activities. Therefore, our finding demonstrates the effectiveness of B. stercoris strain B.PNR1 as an antagonist and for plant growth promotion, highlighting the use of this microorganism as a biocontrol agent against the Fusarium wilt pathogen and PGP abilities in tomatoes.
