Protective Effect of HP08-0106 on Ligature-induced Periodontitis in Rats

  • Periodontitis is an inflammatory disorder of the periodontium, characterized by destruction of the tooth supporting tissues including alveolar bone and mediated by various pro-inflammatory mediators. Here, we demonstrated that HP08-0106, composed of four crude drugs-Gardenia jasminoides Grandiflora, Angelica gigas Nakai, Rehmannia glutinosa, and Schizonepeta tenuifolia in a weight ratio of 2:2:1:2, perturbs inflammatory responses, osteoclast formation in LPS-induced RAW 264.7 cells and alveolar bone resorption in ligature-induced periodontitis. HP08-0106 decreased the protein level of iNOS and COX2 as well as the secreted level of IL-$1{\beta}$, indicating that HP08-0106 has antiinflammatory effects. HP08-0106 also inhibited the expression of genes associated with osteoclastogenesis including c-Fos, MMP-9 and TRAP. Moreover, HP08-0106 exhibited a protective effect from alveolar bone loss in ligature-induced periodontitis animal models. Our results strongly suggest that HP08-0106 represent an important therapeutic tool to treat inflammatory disorders associated with bone loss such as periodontitis.

  • Anticoagulant activities were tested for the fifteen kinds of medicinal plants by measuring activated partial thromboplastin time (aPTT). Of them five kinds or species (Artemisia princeps, Sanguisorba officinalis, Artemisia apiacea, Eclipa alba, Schizonepeta tenuifolia) were selected and fractionated for the preparation of acidic polysaccharides. They were extracted with water by refluxing and the extracts were precipitated with ethanol. The precipitates were separated based on charge using a DEAE-Sephadex. The low salt and high salt fractions were sulfated with anhydrous pyridine and chlorosulfonic acid complex. In vitro anticoagulant activities of sulfated polysaccharides were tested by measuring aPTT, prothrombin time (PT), and factor Xa clotting time using normal human plasma. No relationship was found between the amount of uronic acids and anticoagulant activities, but the sulfated ones show the increase of activities. In vivo anticoagulant properties of the sulfated polysaccharide from Artemisia apiacea were also tested by the intraveneous administration of three different doses (3,5 and 10 mg/kg) to rats. APTT and PT were increased significantly and the action of factor Xa and thrombin mediated through antithrombin III were inhibited slightly.

  • Sopungchungyoung-tang (SCT) has been widely used in Korea as a treatment of atopic dermatitis. SCT consists of Talcum, Rehmannia glutinosa, Angelica sinensis, Paeonia lactiflora, Cnidium officinale, Ledebouriella divaricata, Schizonepeta tenuifolia, Scutellaria baicalensis, Glycyrrhiza uralensis, Mentha arvensis, Cordyceps cicadae. We examined the immunological effect of SCT in vitro. We studied about the effect of SCT on Th cells' differentiation. In the case of CD4 T cells under neutral condition where there was only rIL-2 stimulus, SCT inhibited IFN-γ secretion by 70-80 %. Likewise, SCT also inhibited the IL-4 secretion of neutral Th cells by 85-90 %. We also experimented with the polarized Th1 cells/ Th2 cells and their production of IFN-γ and IL-4, respectively. There also were inhibitory effects on the polarized cells like there was on neutral cells, they were not as strong on the polarized cells. Under Th1 polarized condition, SCT acted dose-dependently, while in Th2 cells, the IL-4 production was inversely proportional to the doses of SCT. From the current study, it can be concluded that SCT exerts inhibitory effects on cytokine production without interfering with immune cells' activity. The result that SCT inhibits IFN-γ and IL-4 confirms that it does have a effect on immunomodulation.

Isolation, Identification, and Activity of Rosmarinic Acid, a Potent Antioxidant Extracted from Korean Agastache rugosa (한국산 방아잎(배초향, Agastache rugosa O. Kuntze)에서 항산화물질 로즈마린산의 분리, 동정 및 활성)

  • Thirty plants were screened for their antioxidative activity. The extracts of Agastache rugosa, Schizonepeta tenuifolia and Lycopus lucidus had high free radical(2,2-diphenyl-1-picrylhydrazyl) scavenging activities. Methanol extract of Agastache rugosa was fractionated with hexane, chloroform, ethyl acetate, and water. The ethyl acetate fraction showed the highest antioxidant activity in the DPPH test. The ethyl acetate fraction was applied to Sephadex LH-20 column, and the fractions showing antioxidative activity were collected and used for identification of the substance. The purified substance was applied to mass, IR, UV and NMR spectroscopy. The spectra of mass, IR, UV and NMR implied that the substance is a rosmarinic acids as a kind of phenolic compound. The rosmarinic acid has more antioxidative effect than those of BHT and ${\alpha}-tocopherol$ in the Rancimat test.

  • In the present study, we investigated the glutathione S-transferase (GST) induction of medicinal plants using a cultured PC12 cells. The methanol extracts of Dendrobium nobile Lindley, Schizonepeta tenuifolia Briquet, Glycyrrhiza uralensis Fischer, Paenoia lactiflora Pall were tested. As the result, exposure of PC12 cells to G. uralensis resulted in the significant induction of GST. On the continuous experiment, G. uralensis was extracted with methanol, ethanol, and acetone. Among these extracts, methanol extracts showed the highest GST induction. The methanol extracts were further fractionated with hexane, diethyl ether, ethyl acetate, and water layer according to the degree of polarity. The diethyl ether layer showed the highest exhibited GST induction on PC12 cells. Based on these results suggest that the extracts of G. uralensis can be applicable for the development of a new antioxidant agent.

The Inhibitory Effects of GHJ on Allergic Inflammatory Response in Human Mast Cells (HMC-1) (고삼, 형개, 자초 혼합물(GHJ)의 인간비만세포에서의 항염증 효과)

  • Objectives : This study aimed to evaluate inhibitory effects of GHJ on allergic inflammatory response in human mast cells (HMC-1). Methods : To investigate the inhibitory effect of GHJ (62.5, 125, 250, 500, $1000{\mu}g/mL$), HMC-1 cells were stimulated with phorbol 12-myristate 13-acetate plus calcium ionophore A23187 (PMACI). Enzyme-linked immunosorbent assays (ELISAs), RT-PCR and Western blot analysis were investigated using GHJ extract. Results : GHJ inhibited levels of $TNF-{\alpha}$ and IL-6 of $1000{\mu}g/mL$ concentration in ELISA and mRNA expression. GHJ had inhibitory effects in level of MAPKs, $p-I{\kappa}B-{\alpha}$ and p-NF-kB also. GHJ attenuated Compound 48/80-stimulated histamine release. In addition, GHJ inhibited PCA reaction in vivo. Conclusion : This study indicated that GHJ extract can inhibit allergic responses in HMC-1 cell.

Effects of Medicinal Herb Extracts on Osteoblast Differentiation and Osteoclast Formation (한약재 추출물의 조골세포 분화 및 파골세포 형성에 미치는 영향)

  • Bone is continuously remodeled by osteoblasts and osteoclasts. We investigated the effects of medicinal herbs, which act on bone metabolism. Fifteen kinds of medicinal herb extracts were screened for bone formation activity with osteoblastic cells, and MC3T3-E1 and bone resorption were screened with osteoclasts derived from mouse bone marrow macrophages. Among these samples, Actinidia polygama, Eucommia ulmoides Oliv., Schizonepeta tenuifolia, Sorbus commixta, and Zingiber officinale Rosc. extracts showed strong bone-forming activity accompanied with osteoblast proliferation and alkaline phosphatase activity. In addition, these extracts decreased tartrate-resistant acid phosphatase activity against osteoclast differentiation. The results indicate that these medicinal herb extracts can potentially prevent bone-related diseases such as osteoporosis by increasing osteoblast differentiation and reducing osteoclast activity.

  • This experimental study was done to research effects of a number of extract on the anti-allergic effect. The results were obtained as follows: 1. In effect of herb-extract on compound $\frac{48}{80}$-mediated histamine release from Evans blue skin assay, Isatis Japonica Miquel, Dictamnus dasycarpus Turcz, Spirodela polyrhiza, Cimicifuga heracleifolia, Bupleurum chinense, Magnolia liliflora, Forsythia koreana, Aster tataricus L., Xanthium strumarium L.(MtOH), Trichosanthes kirilowii, Phellodendron amurense Rupr, Schizonepeta tenuifolia Var, Betula platyphylla show considerable visible anti-allergic effect. In the result of quantification of histamine induced compound $\frac{48}{80}$, Spirodela polyrhiza, Isatis Japonica Miquel, Trichosanthes kirilowii, Bupleurum chinense, Forsythia koreana inhibit histamine release effectively. 2. In effect of Herb-Extract on compound $\frac{48}{80}$-mediated histamine release from RPMC, Spirodcla polyrhiza, Cimicifuga heracleifolia inhibit histamine release effectively. 3. In effect of Herb-Extract on anti-DNP IgE-mediated histamine release from Evansblue skin assay. Spirodela polyrhiza, Cimicifuga heracleifolia(0.1mg/ml). Forsythia koreana, Aster tataricus L., Xanthium strumarium L.(0.1mg/ml), Trichosanthes kirilowii(0.1mg/ml) show considerable visible anti-allergic effect. In the result of quantification of histamine induced anti-DNP IgE, Spiradela polyrhiza, Isatis Japonica Miquel, Trichosanthes kirilowii, Bupleurum chinense, Forsythia koreana inhibit histamine release effectively 4. In the result of genetic manifestative inhibition about the human mast cell-1(HMC-1), Cimicifuga heracleifolia has considerable effect in IL-4 in IL-5, and Tussilage farfara L. has in IL-4. According to the above results, it is suggested that several Herb-Extract have anti-allergic effect.

  • Objectives This paper investigates the origin, the progressive changes and the constructive principles of Jeoryoungchajeonja-tang(猪苓車前子湯).Methods Jeoryoungchajeonja-tang and other related prescriptions were analyzed in terms of their pathological indications, based on previous literature including 『Sanghannon(傷寒論)』, 『Keumkuaeyoryak(金匱要略)』, 『Donguibogam(東醫寶鑑)』, 『Donguisusebowon‧Sasangchobongwon(東醫壽世保元‧四象草本卷)』, 『Donguisusebowon‧Gabogubon(東醫壽世保元‧甲午舊本)』, 『Donguisusebowon‧Sinchukbon(東醫壽世保元‧辛丑本)』Results and Conclusions 1) The origin of Jeoryoungchajeonja-tang is discovered in the prescriptions for Lee-su(利水), that is, Jeoryoung-tang(猪苓湯). 2) Ohryoung-san(五苓散) introduced in 『Donguisusebowon‧Sasangchobongwon(東醫壽世保元‧四象草本卷)』progressively transformed into Shihosaryoung-san(柴胡四苓散)(『Donguisusebowon‧Gabogubon(東醫壽世保元 ‧甲午舊本)』) and ultimately into Jeoryoungchajeonja-tang(『Donguisusebowon‧Sinchukbon(東醫壽世保元‧辛丑本)』), a prescription appropriate for usage in the Mangeum symptomatology(亡陰證). 3) The Jeoryoungchajeonja-tang is composed of 10 herbs. Of these, Notopterygium incisum Ting(羌活), Aralia cordata Thunb.(獨活), Schizonepeta tenuifolia Briq.(荊芥), Saposhnikovia divaricata Schischk.(防風) make the Pyoeum(表陰) drop, and Anemarrhena asphodeloides Bge.(知母), Gypsum fibrosum(石膏) scatter the uihwa(胃火), and Poria cocos Wolf.(茯苓), Alisma canaliculatum(澤瀉), Polyporus umbellatus Fr.(猪苓), Plantago asiatica L.(車前子) facilitate urination.

A Study on the Meaning of Soyangin Gangpyoeum(降表陰) Treatment and Application of Gangpyoeum(降表陰) Medicine (소양인 강표음 치법의 의미와 강표음약 용약법에 대한 고찰)

  • Objectives : Not many studies have been done on the mechanism of the Gangeum or Gangpyoeum method which treats the Soyang Exterior Pattern, nor on which collective or individual roles the Gangeum medicinals play. Therefore in this study, four medicinals categorized as Gangpyoeum which are Jingjie, Fangfeng, Qianghuo, Duhuo, were analyzed in terms of their individual and collective effects. Methods : Content analysis based on the verses in the Donguisusebowon, and medicinal analysis on contents from the Donguibogam, Bencaogangmu, and Benjingshuzheng were carried out followed by a medicinal combination analysis using the InSAm 1.1 program. Results : The four medicinals Jingjie, Fangfeng, Qianghuo, Duhuo had the common effect of normalizing the Wei qi, allowing for unobstructed Qi flow to prevent production of dampness and phlegm, which ultimately lowers Yin[降陰, gangeum] and helps to preserve the Kidney Yin Qi. Jingjie works at the highest position of the upper body; Fangfeng, the most basic medicinal of the Soyangin pattern, works mostly at the upper body although it defends the entire body from wind-dampness. Qianghuo and Duhuo are more or less similar. The difference is that Qianghuo mostly pulls Yang qi upwards while eliminating pathological byproducts, and Duhuo stabilizes the lower body that pulls Wei qi upwards. Conclusions : Analysis on the relationship between Gangpyoeum of the Soyangin pattern and Wei qi, the individual and collective effects of the four Gangpyoeum medicinals were carried out. Further research on other major Soyangin medicinals, along with those of other constitution patterns is anticipated.
