어성초의 추출물에 따른 항산화력 연구 (Antioxidant Properties in Water and 70% Ethanol Extracts of Houttuynia Cordata Thunb)

  • 본 연구는 여러 생리활성 성분이 다량 함유되어 있다고 알려진 전통 약용 식물인 어성초의 추출 조건에 따른 항산화력을 관찰하였는데, 추출조건으로는 식용으로 활용가능한 물 추출과 70% 에탄올 추출을 통해 항산화력을 비교하였다. 항산화력은 총 페놀 함량, DPPH 라디칼 소거능, ABS 양이온 소거능, FRAP 방법으로 측정하였다. 결과적으로 총 페놀 함량은 어성초 물추출물이나 70% 에탄올 추출물 모두 77.8 mM Tannic acid/g extract로 나타나 유의적인 차이는 없었고 DPPH 라디칼 소거능은 물 추출물 2 mg/mL 농도에서 78.4%, 70% 에탄올 추출물은 91.8%로 유의적으로 소거능이 70% 에탄올 추출 시에 우수한 것으로 나타났다. 또한 ABTS 양이온 소거능에서도 물 추출물은 2 mg/mL 농도에서 47.7%, 70% 에탄올 추출물에서는 54.8%로 유의적으로 70% 에탄올 추출물이 저해능이 우수한 것으로 나타났으며 FRAP 방법으로 통한 환원 능도 70% 에탄올 추출물이 우수한 것으로 관찰되었다. 이와 같은 결과로 어성초 추출물 조건중 70% 에탄올 추출 시 항산화력을 높힐 수 있는 생리활성 성분이 추출되는 것으로 사료되며 식용 가능한 기능성 식품소재로써 어성초 에탄올 추출물을 사용할 수 있을 것으로 사료된다.

Effects of Drying Temperature on Antioxidant Activities of Tomato Powder and Storage Stability of Pork Patties

  • This study was performed to evaluate the antioxidant activity of oven-dried tomato powder (OTP) as affected by drying temperature and the effect of OTP on the product quality of pork patties. Three OTP products were obtained by drying of fresh tomato at 60, 80 and 100℃ oven until constant weight was obtained. Total phenolic content of three kinds of OTPs ranged from 1.95 to 5.94 g/100 g. The highest amount of total phenolic compound was observed in OTP dried at 100℃. Antioxidant activity of three kinds of OTPs was measured by 1,1-diphenyl-2-pycrylhydrazyl (DPPH)-radical scavenging activity, iron chelating ability, reducing power and measurement of lipid peroxide in linoleic acid emulsion system. In all parameters, OTP at 100℃ showed the higher antioxidant activity than other temperatures (p<0.05). Based on the model study, the physicochemical properties, and antioxidant and antimicrobial activities of pork patties containing 1% OTP were measured. Redness of pork patties were increased with the addition of OTPs (p<0.05). Thiobarbituric acid reactive substances (TBARS) values of raw pork patties containing OTPs were lower than those of control (CTL) until 7 d of storage, regardless of drying temperatures (p<0.05). Peroxide values of pork patties made with OTP (1%) were lower than those of CTL until the end of storage time (p<0.05). However, no antimicrobial activities were observed among the treatments (p>0.05). Therefore, OTPs could be used as a natural antioxidant in meat products.

아몬드 첨가 설기떡의 품질 특성 및 항산화성 (Quality Characteristics and Antioxidant Activities of Sulgidduk Added with Almond Powder)

  • 본 연구는 설기떡에 쌀에 부족한 단백질 등 영양이 풍부한 아몬드를 첨가하여 설기떡의 건강 기능성을 향상하고자 아몬드를 6, 12, 15% 첨가한 설기떡을 제조하였다. 가용성 고형물 함량은 아몬드 첨가량이 증가할수록 유의적으로 감소하였고 환원당은 아몬드 첨가량이 증가할수록 유의적으로 감소하였다. pH와 산도는 아몬드 첨가량에 따른 변화가 없었다. 색도는 아몬드의 첨가량이 증가할수록 명도는 유의적으로 감소하였고, 적색도와 황색도는 유의적으로 증가하였다(P<0.05). 항산화능은 DPPH 라디칼 소거능 및 hydroxyl라디칼 소거능 모두 아몬드의 첨가량이 증가함에 따라 $IC_{50}$ 값이 감소하여 아몬드의 첨가량이 증가함에 따라 항산화능이 증가하였다. 이는 총페놀 함량이 아몬드의 첨가량이 증가함에 따라 농도 의존적으로 증가하는 결과에 기인하는 것이다. 관능검사 결과 강도 특성으로는 아몬드를 첨가할수록 색과 아몬드 향, 아몬드 맛에 대한 점수가 높게 나타났으며, 짠맛은 아몬드 첨가량이 증가할수록 높아졌다. 기호도에서 외관, 조직감, 전체적인 기호도는 아몬드 12% 첨가군에서 가장 높은 점수를 받았다(P<0.05). 이상의 결과로부터 설기떡에 아몬드 첨가 시 설기떡의 품질 특성 및 항산화능이 개선되었으며, 설기떡에 첨가하는 아몬드의 양은 12%가 적당한 것으로 생각되었다.

Antioxidant and Hepatoprotective Effects of Tomato Extracts

  • The objective of present study was to investigate the anti oxidative and hepatoprotective effects of tomato extracts. Total antioxidant capacity and total antioxidant response were 5.5 and $19.8{\mu}g$ Trolox equivalent per mg of tomato extract, respectively. DPPH radical scavenging activity of tomato extracts ($10mg\;ml^{-1}$) was 70% as compared to 100% by pyrogallol solution as a reference. The effect of the tomato extracts on lipid peroxidation was examined using rat liver mitochondria induced by iron/ascorbate. Tomato extracts at the concentration of $0.5mg\;ml^{-1}$ significantly decreased TBARS concentration. Tomato extracts prevented lipid peroxidation in a dose-dependent manner. The effect of the tomato extracts on reactive oxygen species (ROS) generation was examined using cell-free system induced by $H_2O_2/FeSO_4$. Addition of $1mg\;ml^{-1}$ of tomato extracts significantly reduced dichlorofluorescein (DCF) fluorescence. Tomato extracts caused concentration-dependent attenuation of the increase in DCF fluorescence, indicating that tomato extracts significantly prevented ROS generation in vitro. The effect of tomato extracts on cell viability and proliferation was examined using hepatocyte culture. Primary cultures of rat hepatocytes were incubated with 1mM tert-butyl hydroperoxide (t-BHP) for 90 min in the presence or absence of tomato extracts. MTT values by addition of tomato extracts at the concentration of 2, 10, and $20mg\;ml^{-1}$ in the presence of t-BHP were 13, 33 and 48%, respectively, compared to 100% as control. Tomato extracts increased cell viability in a dose-dependent manner. These results demonstrate that tomato extracts suppressed lipid peroxidation and t-BHP-induced hepatotoxicity and scavenged ROS generation. Thus antioxidant and hepatoprotective effects of tomato extracts seem to be due to, at least in part, the prevention from free radicals-induced oxidation, followed by inhibition of lipid peroxidation.

크랜베리 농축액을 첨가한 젤리의 품질 및 항산화 활성 (Quality and Antioxidant Properties of Gelatin Jelly Incorporated with Cranberry Concentrate)

  • 크랜베리 농축액의 첨가량을 0~15%로 달리하여 젤리를 제조한 후 물리화학적 품질, 항산화 특성 및 소비자 기호도를 비교하였다. 크랜베리 농축액의 첨가 비율이 증가함에 따라 젤리의 pH와 경도는 감소하였고, 가용성 고형분 함량은 증가하는 경향을 보였다(P<0.05). 밝기를 나타내는 $L^*$값은 농축액의 첨가량이 증가할수록 유의적으로 감소하였고, 적색도를 나타내는 $a^*$값은 현저하게 증가하였다(P<0.05). 황색도를 나타내는 $b^*$값은 농축액 첨가에 따라 현저하게 감소하였으나, 첨가 농도와 직접적인 연관관계는 발견되지 않았다. 한편 농축액의 첨가량이 증가함에 따라 항산화 활성을 나타내는 DPPH와 ABTS 라디칼 소거능은 유의적 차이를 나타내며 증가하였다(P<0.05). 소비자 기호도 검사 결과 6% 첨가군이 대부분의 평가항목에서 유의적으로 높은 점수를 얻었고, 특히 전체적인 기호도 측면에서도 유의적으로 가장 높게 평가되어 젤리의 관능품질과 건강기능성 효과 등을 고려할 때 최적 첨가 농도로 6%가 가장 적합한 것으로 판단된다.

The Effect of Ground Cherry Extract on the Activity of Hepatic Aniline Hydroxylase in Mice

  • To evaluate the effect of ground cherry extract on the activity of aniline hydroxylase, we gave ground cherry extract in doses of 100, 200 or 400 mg/kg i.p to mice for 1, 2 or 4 days. The aniline hydroxylase activity in the group treated with ground cherry extract increased in a dose dependant manner in all experimental groups compared with the control group, and was significantly higher in the group treated with ground cherry extract at a dose of 200 mg/kg, which also exhibited a time dependant increase over 4 days. Enzyzme kinetic analysis was performed for hepatic aniline hydroxylase activity in the group treated with 200 mg/kg for 4 days. There was no change of the Km values for aniline hydroxylase between the experimental group and the control group, but the Vmax values for aniline hydroxylase was 21% lower in the experimental group compared with the control. The experimental group also showed lower lipid peroxide and reduced glutathione content, and there were no significant difference in serum alanine aminotransferase activity between the experimental group and the control. Aniline was injected into both the experimental group mice treated with ground cherry extract at a dose of 200 mg/kg for 4 days and the control group, and then the level of blood aniline was assayed at 1hr. The level of blood aniline was lower in the experimental than the control group. This study suggests that ground cherry extract induces hepatic aniline hydroxylase activity and might accelerate the scavenging system of reactive oxygen species. It is likely that ground cherry extract influences the metabolism of xenobiotics by activating AH activity substituted for CYP2E1.

Effect of p-Phenylenediamine Application to Rat Skin on the Liver Oxygen Free Radical Systems

  • To evaluate the influence of hepatic oxygen free radical systems on liver injury by topical p-phenylenediamine (PPD) application on rat skin, PPD (25 mg/16.5 $\textrm{cm}^2$) was topically applied to the abdominal region 5 times every other day and sacrificed. By PPD treatment, increasing rate of liver weight/body weight (%), serum activities of alanine aminotransferase and aspartate aminotransferase and decreasing rate of microsomal glucose-6-phosphatase activity were higher in the rats fed tungstate supplemented diet than those fed a standard diet. These findings indicate that group fed tungstate supplemented diet have more severe liver injury compared with group fed standard diet on topical PPD application. However, the activities of oxygen free radical generating enzymes such as xanthine oxidase (XO) and cytochrome P450 dependent aniline hydroxylase and those of oxygen free radical scavenging enzymes were not found to be different between these two animal groups. In the present study, a novel monitoring method to detect the generating of oxygen free radicals in liver extract was devised. Throughout this method, the oxidized PPD produced by oxygen free radicals was determined colorimetrically. The increasing rate of PPD oxidation by liver homogenate was higher in tungstate fed animals than in standard diet fed ones. Among the fractionations of liver extract, the mitochondrial and postmitochondrial fractions in the liver extract of tungstate fed animals led to a higher availability of PPD oxidation by PPD treatment compared with standard diet fed ones. In conclusion, these results suggest that an enhanced liver injury in tungstate fed animals treated with PPD may be due to oxygen free radicals produced in other systems except oxygen free radicals generating from cytosolic XO system. Especially, oxidative availability by PPD can be used for oxygen free radical detection in some tissue.

밀리타리스 동충하초(Cordyceps militaris)의 면역 활성에 미치는 영향 (Effects of Cordyceps militaris on Immune Activity)

  • In order to determine the functional benefits of Cordyceps militaris in the immune system, we examined the immunomodulatory activities of C. militaris using an immunocompromised C57BL/6 mice, mouse spleen cells, RAW 264.7 macrophage cells, and A549 lung carcinoma cells. Mice were injected intraperitioneally with an immunosuppressive drug, cyclophosphamide, and then administered orally with 30, 100 and 300 mg/kg of 50% ethanol extract of C. militaris (CME 30, CME 100 and CME 300) for 14 days. CME increased splenocyte proliferation and natural killer (NK) cell activity compared to 3% hydroxypropyl methylcellulose-treated control mice. CME also increased the production of Th1 cytokines, IL-2 and TNF-${\alpha}$ in spleen cells isolated from CME-injected mice and in vitro, which suggested the enhanced cellular immunity in response to CME. CME also increased splenocyte proliferation, NK cell activity, and IL-2 and TNF-${\alpha}$ production compared to 1 ${\mu}M$ methotrexate-treated spleen cells in vitro. We examined whether C. militaris regulates the production of inflammatory mediators in LPS-stimulated RAW 264.7 cells. CME inhibited LPS-induced NO production and iNOS expression in a dose dependent manner, while COX-2 expression was remained unchanged. In addition, CME also has free radical scavenging activity, indicating its antioxidant activity. These results indicate that C. militaris enhances immune activity by promoting immune cell proliferation and cytokine production.

  • Radio frequency (RF) energy harvesting technology have become a reliable and promising alternative to extend the lifetime of power-constrained wireless networks by eliminating the need for batteries. This emerging technology enables the low-power wireless devices to be self-sustaining and eco-friendly by scavenging RF energy from ambient environment or dedicated energy sources. These attributes make RF energy harvesting technology feasible and attractive to an extended range of applications. However, despite being the most reliable energy harvesting technology, there are several challenges (especially power conversion efficiency, output DC voltage and sensitivity) poised for the implementation of RF energy harvesting systems. In this article, a detailed literature on RF energy harvesting technology has been surveyed to provide guidance for RF energy harvesters design. Since signal strength of the received RF power is limited and weak, high efficiency state-of-the-art RF energy harvesters are required to design for providing sufficient DC supply voltage to wireless networks. Therefore, various designs and their trade-offs with comprehensive analysis for RF energy harvesters have been discussed. This paper can serve as a good reference for the researchers to catch new research topics in the field of RF energy harvesting.

Analysis of the Phenolic Content and Antioxidant Activities of Soybean Extracts from Different Regions and Cultivars

  • Phenolic compounds such as isoflavones contribute to the antioxidant properties of soybean. This study investigated differences in the phenolic content and antioxidant activity of ten soybean cultivars grown in two different locations in Korea. We observed a wide range of variation in total isoflavone and phenolic content by location and cultivar. The total isoflavone content of cultivars grown at Pyeongchang (sub-highland) was significantly higher than that in cultivars grown at Gangneung (lowland). In contrast, the total phenolic content of cultivars grown at Gangneung was greater than that of cultivars grown at Pyeongchang. The radical scavenging activity of DPPH was similar to that of the total phenolic content rather than that of the total isoflavone content. These results suggest that antioxidant activity of soybean was associated with phenolic compounds rather than isoflavones. To identify the individual antioxidant components, we used an on-line HPLC-$ABTS^+$-based assay system, ESI/MS, and NMR. The results showed that the strongest antioxidant activity was linked to epicatechin.
