The mycelial growth of P. coccineus strain was good in PDA and YMA, but mycelial growth was low in MEA. Light irradiation during the incubation period affected the pigment formation and density of mycelia. Mushroom of P. coccineus strain was able to produce fruiting bodies in both bottle and bag cultivation, and oak sawdust was found to be the most suitable substrate for spawn culture and cultivation. In artificial cultivation using sawdust medium, fruiting body was grown to the extent that visual observation was possible from the 15th day, and it formed about 5 days fast in the treatment group with low relative humidity. From 40 to 45 days of mushroom development, mature fruiting bodies could be harvested, and the lower relative humidity of the growing room favored mushroom development and growth. Antioxidant activity of fruiting bodies harvested from artificial cultivation showed that ABTS radical scavenging activity of bottle-cultivated and wild fruit bodies were shown at 505㎍/㎖ and 515㎍/㎖, respectively. However, fruiting bodies harvested in bag cultivation were high at 910㎍/㎖. As a result of DPPH radical scavenging activity, all extracts were found to be inactive, exhibiting IC50 value of more than 2,000㎍/㎖ concentration. The ethyl acetate extract of mushrooms obtained from bottle cultivation showed the highest activity with 1,550㎍/㎖ IC50 value. Methanol extract of wild fruit bodies had the highest ABTS radical scavenging activity at the same concentration (10mg/㎖).
Sparassis crispa(Cauliflower mushroom) was an edible mushroom that shows remarkably high contents of ${\beta}$-glucan compared to other edible mushroom. In this study, we aimed to select suitable conditions and materials for mycelial growth and fruit body production. The longer saccharification time of barely result in higher sugar content for eight hours. The optimal sugar content of media for mycelial growth was showed at level of 6 $^{\circ}Brix$. Optimum temperature and pH for mycelial growth in liquid spawn were $25^{\circ}C$ and pH 5.0~6.0, respectively. In case of sawdust was used Larix kaempferi as main material, the fruit body yield and cultivation period of supplemented with 10% wheat flour and 20% corn flour were highest and fastest, respectively.
Culture maturity assessment can be used to control fruiting body flush timing. Culture maturity of sawdust-based substrate was evaluated by using oak mushroom, (Lentinula edodes (Berk.) Pegler). The influence of substrate water potential (${\psi}$) on the growth and fruiting of three genotypes of L. edodes was also investigated. Glucosamine content revealed a peak at the fruiting body senescent stage. Glucosamine increased steadily to the sporophore senescent stage, and sharply declined at crop treatment. Lipid phosphate and ergosterol contents peaked at pinning and button break stages, respectively. Therefore lipid phosphate and ergosterol contents would be considered as the convenient measurement for judging culture maturity and fruiting potentials. The substrate pH values before inoculation and on the fruiting stage were varied from 6.3 to 4.0. This pH changes were detected as changes in color from bluish purple to yellow by direct bromphenol blue(BPB) spraying, and shown a good correlation with fruit body yield of the 1 st flush. Concerning water potential of the cultures, a slight reduction of water potential, -0.5MPa, stimulated mycelial and colony growths on liquid, agar and sawdust-based substrates. The water potential of well-colonized matured substrate was -0.7MPa and -4.0MPa, before and after the fruiting, respectively. Excellent water providing capacity (higher ${\psi}$) is expected to well-matured cultures with a high density of mycelial colonization. Also, the substrate water potential significantly affected by the interaction between genotypes and spawn run time.
The effects of punching treatment on mycelial culture and fruiting body productivity were investigated in a new Lentinula edodes cultivar, "Jadam", in sawdust medium for the stable production of oak mushroom. As the punching volume and number increased, the weight loss rate and color difference increased and the L value decreased. After spawn inoculation, the sawdust medium temperature and CO2 concentration reached their highest values at 33 and 19 days of incubation, respectively. The O2 concentration showed the lowest value on the 14th day of incubation, which was the opposite pattern to the CO2 concentration. As the punching volume and the number increased, the medium temperature and O2 concentration increased, and the CO2 concentration decreased. Higher punching volumes and numbers resulted in higher temperatures and lower CO2 concentrations. The best fruiting body yield was 5 × 70 mm - 30 (punching diameter × depth - number), and the total yield after three cycles was 644.7 g.
The process of cultivation and production of oak mushroom (Lentinula edodes (Berk.) Pegler) on sawdust surface beds were investigated. Sawdust surface bed cultivation is the method by which oak mushrooms are cultured and produced on sterilized sawdust surface bed without using bags. The bed was made by inoculating with 3 to 1 ratio of bed sawdust to oak mushroom mycelial inoculum. The sawdust bed medium with 65% water content was pasteurized at $65^{\circ}C$, inoculated with sawdust spawn and spread on the surface on vinyl film in cultivation shed. During 78 days of cultivation period, water content in the medium varied from 61 to 72%, its pH decreased from 5.6 to 3.9~4.6 and ergosterol concentration increased to $0.33{\sim}0.59{\mu}g/g$. $CO_2$ concentration in the medium rapidly increased to 8.06% in two weeks. In seven weeks the medium surface started browning and $CO_2$ concentration increased to about 5.63%. Until 11th week the $CO_2$ concentration was maintained at 6~7%. After removing the plastic cover on the bed for ventilation in 12 weeks, $CO_2$ within the bed reduced dramatically to 1.5%. In the cultivation shed the internal temperature was $7.1{\sim}29^{\circ}C$ and humidity was 27.3 to 100%, while bed temperature ranged $11.6{\sim}30^{\circ}C$. Oak mushroom fruiting started from late July, in 120 days after bed establishment in late March and continued for approximately 100 days until early December with eight cycles of irrigation treatment. The mushroom yield of the eight cycles were 288~352 kg during the 1st (7/29~8/4) to 3rd cycle (9/3~9/7), 800 kg at the 4th cycle (9/19~9/24), 1,296~1,853 kg during 5th (10/3~10/8) to 7th cycle (4.11~11/9) and 990 kg at 8th cycle (11/23~12/7). Total production was approximately 7.4 tons from 33.0 tons of oak sawdust medium, thus harvest efficiency of the mushroom production was approximately 22.4%.
Thirty strains of genus Agrocybe which had been kept in National Institute of Horticultural and Herbal Science, were tested for the formation of fruiting body. Nineteen strains of collected genus Agrocybe were formed fruiting body at bottle culture of sawdust medium. Five strains (including ASI 19003) and thirteen strains (including ASI 19007) could be grouped as A. cylindracea and A. chaxingu. Although ASI 19008 showed the formation of fruiting body, morphological characteristics were significantly different from the two groups. Cultural period of ASI 19003 strain at sawdust substrates was 29 days and ASI 19007 train was 30 days in liquid spawn inoculation. The yield and quality of ASI 19003 strain was excellent in the spring, fall, and winter, whereas ASI 19007 strain was excellent in the summer. Accordingly, these cultivar might be contribute to farmers' income by stable year-round production if using the season-oriented strain.
To replace the sawdust in the growth medium of oyster mushroom to utilize its by-product spent mushroom substrates (SMS) as feed for ruminant, we performed cultivation test using cotton seed hull pellet (CSHP), corn stalk pellet (CSP), corncob (CC), and analyzed the feed chemical properties of those SMS. As a result of cultivation test, CC and CSHP treatment took 27 days for spawn run, 4 days for primordium formation, and 3 days for development fruiting bodies, resulting in a total cultivation period of 34 days. The yield per bottle was 134 g for CC treatment, similar to 130 g for control, while CSHP treatment (112 g) and CSP treatment (68 g) were lower than that of control. The highest biological efficiency (BE) was shown in CC treatment as 80.1%, which was 11.4% higher than 68.7% of control. The SMS of CC treatment had a relatively low content of neutral detergent fiber and acid detergent fiber, and in particular, lignin content was the lowest and crude protein content was the highest among other treatments. Therefore, CC as a substitute material for sawdust was capable of stable mushroom production and excellent nutritional value as a feed for its by-products.
'Baekseung', a new variety of Flammulina velutipes, was bred by mating two monokaryotic strains isolated from KMCC 4210 and KMCC 4216 in the Mushroom Research Division, Baekseung ARES in 2016. The Baekseung and Uri1ho strains showed fast mycelial growth and mycelial density on malt extract agar media after 7 days of incubation. The spawn running period on the sawdust substrate required a cultivation period and temperature of 30 days and $25^{\circ}C$, respectively, for primordia formation where in fruiting body development occurred from $11{\pm}1days$ at $14^{\circ}C$ and $14{\pm}1days$ at $7^{\circ}C$. The length of the pilei and stipes of the Baekseung harvested in optimal stag were $11.3{\pm}0.4$ and $89.2{\pm}7.1mm$, respectively, whereas the values for Uri1ho were $10.7{\pm}1.0$ and $91.3{\pm}20.8mm$, respectively. The yield of the Baekseung and Uri1ho strain grown on the sawdust substrate was $153.7{\pm}12.5$ and $139.8{\pm}17.8g$, respectively, per 850 ml in bottle cultivation. The inferred tree exhibited a phylogenetic relationship between the Korean white fruiting body strains of Baekseung, Uri1ho and Fv-14-a-38, Fv-14-a-51, and the Japanese white fruiting body-forming strains of KMCC 4226, and these were confirmed to be genetically related.
The study was carried out on the artificial cultivation of the abalone mushroom, Pleurotus cystidiosus O.K.Miller. The pine sawdust substrates with 20% rise bran were good for mycelial growth and high quantity of P. cystidiosus in the bottle cultivation. Moreover, the proper volume for bottle cultivation was 850 ml and the removal of spawn and surface layer of the medium before pin-heading was more efficient. The yields of P. cystidiosus were higher in sawdust substrates added calcium carbonate than those not added calcium carbonate. The volume of 3 kg polypropylene bag is good for yield and biological efficiency in bag cultivation of P. cystidiosus. Cotton wastes were proper substrates for bag cultivation. In the effect of different cultivation temperature, $28{\pm}2^{\circ}C$ cultivation temperature was good for for primordial formation after inoculation.
A new oak mushroom cultivar 'Hwadam' was bred from monokaryotic strains of 'GMLE36062' and 'LE15401'. The optimum temperature for mycelial growth of 'Hwadam' on potato dextrose agar was 25℃. The cultivation period of 'Hwadam' lasted for 129 days until the first harvest of the fruiting bodies, including 35 days for spawn run, 84 days for browning, and 10 days for development of fruiting bodies. The fruiting bodies of 'Hwadam' had a hemispherical, brighter pileus and thicker stipe than the control cultivar (Sanjo 701ho). The three-cycle yield of 'Hwadam' was 554 g, which was higher than that of control cultivar (405 g). In addition, 'Hwadam' contained 79.4 mg/g of beta-glucan and 0.93 mg GAEs/g of phenolic compounds, which was similar to that of the control cultivar but higher than that of 'L808' and 'Chujae 2ho'.
