Sauchinone, a Lignan from Saururus chinensis, Suppresses iNOS Expression through the Inhibition of Transactivation Activity of RelA of NF-kB

  • Sauchinone, a known lignan, was isolated from the root of Saururus chinensis as an active principle responsible for inhibiting the production of NO in LPS-stimulated RAW264.7 cells by activity-guided fractionation. Sauchinone dose-dependently inhibited not only the production of NO, but also the expression of iNOS mRNA and protein in LPS-stimulated RAW 264.7 cells. Furthermore, sauchinone prevented LPS-induced NF-kB activation, which is known to play a critical role in iNOS expression, assessed by a reporter assay under the control of NF-kB. (omitted)

Suppression of RelA/p65 Transactivation Activity by a Lignoid Manassantin isolated from Saururus chinensis

  • In our search for NF-kB inhibitors from natural resources, we have previously identified two structurally related dilignans, manassantin A and B as specific inhibitors of NF-kB activation from Saururus chinensis. However, their molecular mechanism of action remains unclear. We here demonstrate that manassantin A and B are potent inhibitors of NF-kB activation by the suppression of transciptional activity of RelA/p65 subunit of NF-kB. These compounds significantly inhibited the induced expression of NF-kB reporter gene by LPS or TNF-a in a dose-dependent manner. (omitted)

  • In the course of our search for intercellular adhesion molecule-1 (ICAM-1)/leukocyte function-associated antigen-1 (LFA-1) mediated cell adhesion inhibitors from natural sources, new type of cell adhesion inhibitors were isolated from the MeOH extract of Saururus chinensis roots. On the basis of spectral evidence, the structures of the active compounds were identified as manassantin A and B. Manassantin A and B inhibited phorbol 12-myristate 13-acetate (PMA)-induced homotypic aggregation of the human promyelocytic leukemia HL-60 cells without cytotoxicity with MIC value of 1.0 and 5.5 nM, respectively.   • This study was performed to determine the cytotoxic effect of methanol extract from medicinal plants. The cell viability was determined by the MTT method. Their cytotoxic activities against three cancer cell lines such as A549, MDA-MB-231 and SNU-C4 cell line were tested. Among them, The methanol extract of Saururus Chinensis Bail showed the strongest cytotoxic effect against SNU-C4 cells. These results suggest that the methanol extract of Saururus Chinensis Bail possessed a potential antitumorous agent

Renoprotective and antioxidant effects of Saururus chinensis Baill in rats fed a high-fructose diet

  • This study investigated the preventive effect of Saururus chinensis Baill against renal damage induced by a high-fructose diet in rats. The rats (n = 30) were fed either a cornstarch-based (65%), high-fructose (65%), or high-fructose (64.5%) diet with 0.5% S. chinensis Baill extract for 10 weeks. Twenty-four hour urine collections were obtained and the animals were sacrificed after an overnight fast. Serum urea and creatinine and urine albumin were measured using colorimetric methods, and creatinine clearance was determined. In addition, thiobarbituric acid reactive substances (TBARS), reduced glutathione (GSH), and the activity of superoxide dismutase (SOD) in the kidney were determined. Kidney samples were also examined histologically. The fructose-fed rats showed renal dysfunction, indicated by decreased creatinine clearance, increased albumin in the urine, and increased urea and creatinine in the serum. These renal function parameters were comparable to control levels in rats that consumed S. chinensis Baill. Fructose consumption increased renal TBARS and reduced GSH and SOD activity, whereas these levels were near-normal in the rats consuming S. chinensis Baill The kidneys of fructose-fed rats showed glomerular basement membrane thickening, mesangial matrix expansion, and tubule dilation. These pathological changes were not seen in the rats that consumed S. chinensis Baill. Therefore, S. chinensis Baill effectively alleviated fructose-induced renal damage in these rats, at least partially due to antioxidant activity.

삼백초 건근을 첨가한 국수의 품질 특성 (Quality Characteristics of Noodle Added with Dried Saururus chinensis Baill. Root Powder)

  • 본 연구에서는 삼백초 건근 분말을 새로운 식품소재로 활용하고자 하는 연구의 일환으로 밀가루에 삼백초 건근 분말 첨가량을 달리하여 국수를 제조한 후 품질특성을 분석하였다. 삼백초 첨가량이 증가할수록 색도의 L값과 b값은 감소하였으며, a값은 증가하였다. 조리특성에 있어서 무게와 부피가 증가하였으며, 또한 국물의 탁도도 증가하여 조리중 고형분의 손실량이 증가함을 알 수 있었다. 조직감은 생면의 경우 전반적으로 증가하는 경향을 보이는 반면, 조리면에 있어서는 경도를 제외하고는 큰 차이가 나지 않았다. 관능검사 결과 외관 및 향은 유의적인 차이가 없었으나, 맛, 조직감은 삼백초 건근 분말의 첨가량이 증가함에 따라 감소하는 경향을 나타내었고 종합적 기호도에서 삼백초 건근 분말을 3%까지 첨가할 경우 대조군과 유의적인 차이가 나지 않았다. 따라서 삼백초 건근 분말을 첨가하여 국수를 제조할 경우 3% 첨가가 적절한 함량으로 판단된다.
