Saponins from the Aerial Parts of Aralia continentalis

  • Seven triterpenoid saponins such as oleanolic acid 28-O-{\beta}-D-glucopyranosyl$ ester, hederagenin $28-O-{\beta}-D-glucopyranosyl$ ester, chikusetsusaponin IVa, udosaponin A, salsoloside C, udosaponins F and C were isolated from the aerial parts of Aralia continentalis, among which two $28-O-{\beta}-D-glucopyranosyl$ esters of oleanolic acid and hederagenin are isolated for the first tome from this plant. These results suggested that the chemical components of Korean Dokwhal are practically identical to those of japanese Udo supporting the chemotaxonomical point of view.

  • Effects of total ginseng saponin, 20-S-protopanaxadiol saponin, 20-S-protopanaxatriol saponin and playcodon saponin on the osmotic behavior of liposomes were investigated by optical measurement. These saponins showed different activities on liposomal membrane, and cholesterol in liposomes was an important factor to this variation of saponin activities.

  • One hundred and forty-four extracts have been tested for the presence of saponins, using the method of froth formation, precipitate formation by acid hydrolysis, Liebermann-Buchard reaction on the acid hydrolysate, and hemolytic activity of the extracts. Among the samples tested, thirteen extracts showed positive saponin reaction in all the tests employed and one hundred and thirty-six showed positive in one or more tests of saponins. The results are tabulated.

  • The application of LC/MS system with FRIT-FAB LC/MS interface in both positive and negative ion modes to the analytical separation of isomeric saponins was studied. The two isomeric saponins such as rosamultin and arjunetin were well separated and analyzed FRIT, FAB LC/MS system. This is another examples to structure elucidation of natural products.

  • Two new triterpenoidal saponins B(1) and C(2) were isolated from the fructus of Kochia scoparia. On the basis of chemico-spectral evidences, the structures of 1 and 2 were elucidated as oleanolic acid 3-O-$\beta$-D-ribopyranosyl-(1$\rightarrow$2)-.betha.-D-glucuronopyranoside and 3-O-$\beta$-D-xylopyranosyl-(1$\rightarrow$3)-$\beta$-D-glucuronopyranosyl-olean-12-en-28-O-$\beta$-D-glucopyranosyl ester, respectively.

  • Total saponins extracted from Panox ginseng have no effect on the cytotoxic T-cell activity, and natural killer cell activity in mice infected with A/WSN influenza virus. The saponins, however suppressed delayed-type hypersensitivity responses to the virus and to sheep erythrocytes when administered to the animal before sensitization. Thus a prophylactic anti-inflammatory action of the total saponins of ginseng is observed, which may be related to their steroid-like structure.

  • Pontential mutagenic and antimutagenic activities of four steroidal saponins from Smilax china rhizomes were investigated. These saponins did not revealed mutagneicity in the Ames and SOS umu test. For antimutagenic activity by SOS umu test, two spirostanol glycosides, dioscin and gracillin, inhibited the activity of ${\beta}-galactosidase$ induced by AF-2, but their proto-type furostanol glycosides did not show this activity.

Extraction Yields of Ginseng Saponins in the Extracts of Crude Drug Preparations with Various Concentrations of Ethanol (생약복방제의 에탄을 농도별 엑스중 인삼 사포닌의 이행량 조사)

  • As a part of studios on the Quality control of index components in crude drug preparations, extraction yields of ginseng saponins from crude drug extracts were identified by TLC and quantified by HPLC. So-Shi-Ho-Tang(小柴胡湯), Sa-Kun-Ja-Tang(四君子湯), Yook-Kun-Ja-Tang(六君子湯) and In-sam-Tang(人蔘湯) were extracted with water, 30%-ethanol, 50%-ethanol, 80%-ethanol and absolute ethanol to analyze ginseng saponins in the crude drug extracts prepared with various concentrations of ethanol. Ginseng saponins were extracted considerably more from the extracts with higher concentrations of ethanol than those with water or lower concentrations of ethanol. Extraction yields of ginseng-side-Rb$_1$, -Rb$_2$ and -R$_c$ from four crude drug preparations were the lowest as 4.9~45.9%, 5.0~40.1, and 6.3~43.7% in water extract and the highest as 29.5~62.6%, 26.7~61.4% and 31.4~62.0% in absolute ethanol extract, compared with those of 80%-methanol extracts.

  • The present study was undertaken to determine the inhibitory effects of orally administered ginseng saponins (GS), protopanaxadiol saponins (PD), protopanaxatriol saponins (PT) and ginseng ether fraction (GE) on the development of morphine induced tolerance and physical dependence in mice and also to determine the hepatic glutathione contents. GS, PD and PT inhibited significantly the development of morphine induced tolerance and physical dependence, but GE was effective only on the inhibition of the development of morphine induced physical dependence. GS, PD, PT and GE also inhibited the hepatic glutathione level decrease induced by morphine multiple injections.

  • Strong anti-inflammatory saponins Phytolacca americana (Phytolaccaceae) wereobtained from callus mass derived from the stems and also from that derived from the roots of cultivated Phytolacca americana (which were designated as PAS and PAR, respectively). The callus were grown on Linsmair and Skoog's agar medium supplemented with 1ppm OF 2, 4-dichlorophenoxyacetic acid. Phytol accoside B and D were obtained from PAS and phytolaccoside A and B from PAR. The thin layer chromatograms of the crude saponins from PAS and PAR were similar to those of original plants. PAS contained phytolaccoside B as a major component while phytolaccoside E was a major saponin in original plants.
