알코올 투여후 마우스 간 크산틴 산화효소 활성에 미치는 인삼의 영향 (Effect of Ginseng Saponins on Xanthine Oxidase Activity after Ethanol Treatment in Mouse Liver)

  • A dose, 1g/kg of ethanol produced experimental hyperuricemia in mouse. Ginseng saponins were tested for their ability to alter the hepatic xanthine oxidase activity and the blood level of uric acid in the ethanol-treated mouse. Intraperitoneal injection of ginseng saponin 4mg/kg markedly decreased the xanthine oxidase activity in the ethanol-treated mouse liver. It was also observed that ginseng saponin reduced the blood concentration of uric acid in experimentally induced hyperuricemia by alcohol treatment. In vitro, it was found that a low concentration of ginseng saponin in the reaction mixture incresed the hepatic xanthine oxidase activity, while a high concentration inhibited both enzyme preparations of normal and ethanol treated mice. In contrast with the xanthine oxidase, uricase activity was not influenced by ginseng saponin as well as in vivo. These results suggest there is a possibility that ginseng saponin may have some therapeutic effect on gout and other hyperuricemia syndrome.

  • Two novel steroidal saponins designated as spicatosides A(1) and B(2) were isolated from the tubers of Liriope spicata and their structures were elucidated as 25(S)-rus-cogenin -1-0- fJ - D-glucopyranosyl (1$\rightarrow$2)- [$\beta$ - D-xylopyranosyl (1$\rightarrow$3)] -$\beta$- D- fucopyranoside (1) and 26-0-$\beta$-D-glucopyranosyI25(S)-22-0-methyl-furost-5-en-l$\beta$, 3$\beta$, 26-trioll-0-fJ -D¬glucopyranosyl (1$\rightarrow$2)- [$\beta$- D-xylopyranosyl (1$\rightarrow$3)] - $\beta$- D- fucopyranoside (2), respectively.

홍삼의 알콜 농도별 추출 엑스의 수율과 사포닌 함량 (Extract Yields and Saponin Contents of Red Ginseng Extracts prepared with various Concentrations of Ethanol)

  • Red ginseng extracts were prepared with various concentrations of ethanol. Extract yields were examined and saponins in the extracts were identified and determined by TLC and HPLC, respectively. Yields of the extracts, $19.7{\sim]50.3%$, were the highest in water extract and showed significant decrease with the increase of ethanol concentration used for extraction. Contrary to the extract yields, saponin yields from red ginseng were conspicuously increased with the increase of ethanol concentration and were $3.47{\sim}5.13%$ of crude saponins and $1.28{\sim}1.93%$ of six major ginsenosides. Saponin contents in the red ginseng extracts were $6.9{\sim}24.2%$, of crude saponin and $2.57{\sim}9.22%$, of six major ginsenosides.

  • Two triterpenoid saponine were isolated from the stems of Acer ginnala Maxim. The structures of triterpenoid saponins were established as ilexoside O, $3-O-{\alpha}-L- rhamnopyranosyl(1{\rightarrow}2)-{\beta}-D-glucopyranosyl(1{\rightarrow}2)-{\beta}-D-xylopyranosyl-pubescenolic$ acid 28-{\beta}-D-glucopyranosyl$ ester(1) and ilexoside K, $3-O-{\beta}-D-glucopyranosyl(1{\rightarrow}2)-{\beta}-D-xylopyranosyl-pubes-cenolic$ acid $28-{\beta}-D-glucopyranosyl$ ester(2). Their chemical structures have been elucidated on the basis of spectral methods.

  • After rats were exposed to 5,000 ppm carbon monoxide for 30 minutes, the amounts of catecholamine neurotransmitters in stratum were measured using high performance liquid chromatograph equipped with electrochemical detector. The concentration of dopamine in stratum was significantly decreased after carbon monoxide intoxification, but those of dihydroxyphenylacetic acid, norepinephrine, and epinephrine was not changed. However the pretreatments of Ginseng total saponin and panaxatriol saponin increased the concentrations of dopamine and its acidic metabolites (DOPAC and HVA). Ginseng total saponin also increased the concentrations of norepinephrine and epinephrine. Similar results were obtained from aged rats.

  • The effect of ginseng extracts and ginseng saponins on alcoholic fermentation and production of higher alcohols in malt wort by Sacch. uvarnm were studied The results otained were as follows. 1. Alcoholic fermentation of the wort contained 1-5% of ginseng extracts was inhibited slightly, but the wort contained 0.1-0.5% of ginseng extracts were same as the control. 2. 0.02-0.2% of saponin stimulated alcoholic fermentation. 3. Higher alcohol contents were decreased when the wort contained 0.1-0.5% of ginseng extracts. 4. Higher alcohol content were increased when the wort contained 0.02-0.2% of ginseng saponin. Iso-amylalcohol content of fermented wort which contained ginseng saponins were higher 18 -35mg/1 than those of control.

  • The effect of ginseng extract and ginseng saponins on alcoholic fermentation was studied 1. Alcoholic fermentation on gllicose medium at 30$^{\circ}C$. by Saccharomyces coveanus and Saccharomyces uvavum was stimulated when the media contained 5% and 10 % of ginseng extract, respectively. But that of Saccharomyces cerevisiae was inhibites by the addition of 10% of ginseng extract. 2. Saponin did not stimulate the alcoholic fermentation by Saccharomyces uvarum. 3. The yeast cell counts was increased remarkably by addition of ginseng extract while that of ginseng saponins was increased slightly. Dried cell weight of the broth which Contained 5% of ginseng extract was 3 times than that of control.

  • Glucosidic bonds at the C20 position of the sapogenins were hydrolyzed easily in the lower pH, higher temperatures and longer times to give prosapogenins and sugars. The glucosidic bond of saponin at the C3 of ginsenoside-Rb1, which is secondary carbon, was relatively stable due to the low electron density of -0.2. But the bond of saponin at the C20 position, which is tertiary carbon with the relatively high electron density of -0.3, was liable to be hydrolyzed even in weakly acidic solution by the increase of heating time. On the other hand, red ginseng contained 13.34 mg/g of citric acid, 8.78 mg/g of malonic acid, 3.70 mg/g of oxalic acid, 2.13 mg/g of malic acid and 0.44 mg/g of succinct acid. Ginseng saponins were very stable in ginseng extract neutralized with sodium carbonate or sodium bicarbonate corresponding to the equivalent amount of the total organic acid in the red ginseng.

  • Ten saponins named phytolaccoside $A{\sim}I$ and O were separated and shown to contain only $_D-glucose$ as sugar except phytolaccoside A which contains only $_D-xylose$. Phytolaccagenic acid was found to be a genin for phytolaccosides A and D, phytolaccagenin for B, E and F, jaligonic acid for G, and esculentic acid for C, H, I and O, respectively. The total saponin fraction exhibited strong anti-inflammatory activity.

  • In oriental drug therapy it has been considered by experience that the intoxication of alkaloids may be controlled by some drugs, and this was not based on scientific evidence. These drugs would contain saponins, as saponins are widely distributed in many vegetable drugs and, therefore, the present experiment was undertaken to find out if effects of saponin on the extracting process of aconite alkaloids could decrease the amount of the alkaloids. The data obtained in the extractions using various concentrations of saponin solution, show a decrease in alkaloid extract, suggesting that saponin forms a less soluble complex with the alkaloids or acts as a detergent.
