Study on the Gas Chromatography of Samul-tang and Sakunja-tang, each Prepared by the Different Boiling Methods (사물탕(四物湯)과 사군자탕(四君子湯)의 전탕(煎湯)방법의 차이에 관한 GC 분석 실험)

  • The GC pictures were taken to investigate the difference of the Samul-tang and Sakunja-tang, each prepared by the different boiling methods, which were the traditional method(using cover), using the condenser, and using no cover. For Samul-tang, there was not much difference in the GC pictures, but the one pick(rt about 7 min.) of the picture by the method using the condenser had the higher intensity than those in the pictures by other methods. For Sakunja-tang, the GC pictures of the method using the condenser had shown a lot of the peaks and the very high intensity, which were much better than those of the picture by the traditional method. However, the GC pictures of the method by using no cover had shown almost nothing.

  • The GC pictures were taken to investigate the difference of Samul-tang and Sakunja-tang from their distillate each. For Samul-tang, it's distillate contained many peaks which had been shown very small peaks in the GC picture of the original solution, but the one pick(rt about 7 min.) of the picture didn't been found in the GC picture of the distillate. For Sakunja-tang, it's distillate contained all peaks which had shown in the GC picture of the original solution. After distillation, there was almost no peak in the rest of the solution.

Effect of Preservation Conditions on the Stability of Samul-tang Decoctions (사물탕 전탕액의 보관 조건에 따른 안정성 분석)

  • Consumer interest in the stability of medicinal herb extracts during storage has increased. Although the advent of new technologies has improved preservation conditions, increasing the storage time, there are few studies on the preservation of herb extracts. The purpose of this study was to perform microscopic observations of Samul-tang decoctions under various preservation conditions. The storage temperature (a high temperature, room temperature, with or without light, refrigeration, or cryopreservation) and storage time (0, 15, 30, 90, and 180 days) were given to each condition Macroscopic morphology, pH, UV absorption, HPLC, and bacteriological studies were performed to determine microscopic changes in Samul-tang decoctions. The biological activity (tyrosinase inhibition) of the Samul-tang decoctions was also examined. There were no major changes in the indicated observation items when the extracts were stored in each condition. However, at higher storage temperatures and longer storage times, microscopic changes increased, although no bacteria were detected. Furthermore, the higher the storage temperature was and the longer the storage time was, the bigger the change was, despite of minor microscopic changes. Therefore, to maintain the stability of herbal extracts during storage, it is recommended to keep the Samul-tang decoction in the preservation condition of refrigeration and cryopreservation or without light rather than high temperature and room temperature as possible.

Effects of Samul-tang and Constituent Herbs on a Contracted Artery of Rabbit (사물탕(四物湯) 및 구성(構成) 약물(藥物)이 가토(家兎)의 수축혈관(收縮血管)에 미치는 영향(影響))

  • Objectives : To analyze the effects of Samul-Tang and combinations of constituent herbs on arterial contraction. Method : In order to investigate the effects, Angelicae gigantis Radix, Cnidii Rhizoma, Rehmanniae Radix, Paeoniae Radix, in which one of them, three of them, and all of them, were used to exam. Results : Samul-Tang significantly inhibited the contraction of artery induced by PE(phenylephrine), accordingly as the concentration of Samul-Tang increased and inhibited in both with intact and removed endothelium. Among the constituent herbs of Samul-Tang, Paeoniae Radix and Paeoniae Radix-Cnidii Rhizoma combination inhibited the PE-induced contraction of artery the most. Conclusions : Samul-Tang and constituent herbs of Samul-Tang inhibit the contraction of artery.

Effects of Samul-Tang Extract on Vascular Endothelial Cells from Hydrogen Peroxide-induced Injury (사물탕(四物湯)이 혈관내피세포(血管內皮細胞)에 미치는 영향(影響))

  • This study is designed to investigate the effects of Samul-Tang extract on the response of lactic dehydrogenase(LDH) release, cellular activity, lipid peroxidation, DNA synthesis and the changes of total protein of bovine pulmonary artery endothelial cells(PAEC) from hydrogen peroxide$(H_2O_2)$-induced injury. The results are as follows : 1. Samul-Tang significantly decreased $H_2O_2$-induced release of LDH from injured bovine PAEC. 2. Samul-Tang significantly repressed $H_2O_2$-induced cellular activity from injured bovine PAEC. 3. Samul-Tang significantly repressed $H_2O_2$-induced lipid peroxidation from injured bovine PAEC. 4. Samul-Tang significantly stimulated DNA synthesis in bovine PAEC. 5. Samul-Tang significantly repressed $H_2O_2$-induced changes of total protein volume from injured bovine PAEC. Above results suggest that Samul-Tang can protect bovine PAEC from $H_2O_2$-induced injury. These results can be effectively applied to the prevention and cure of cardiovascular and cerebrovascular diseases.

Subacute Toxicity Study on Samul-tang in SD Rats (SD 랫트를 이용하여 사물탕(四物湯)의 아급성 독성에 관한 연구)

  • Samul-tang(Siwu-tang) has been traditionally prescribed as a restorative. The present study was undertaken to determine the possible toxic effects of Samul-tang on SD rats. In this study, we investigated the subacute toxicity of water-extracted Samul-tang(Siwu-tang) on SD rats. Twenty rats were orally adiministered Samul-tang for 28 days at a dose of 0 mg/kg(control group) or 1500 mg/kg(treated group), respectively. All of subjects was survived. No significant difference in abnormal clinical signs, related to hematological values, serum biochemical values, water and feed intake, coagulation time, autopsy analysis, organ weight, tissue microscopically, funduscopy, urine intake and urinalysis, was detected. Compared with the control group, we could not find any subacute toxic alteration in treated group (1500 mg/kg) for 28 days. This result suggests that Samul-tang(Siwu-tang), a herbal medicine prescription, is a safe prescription to body.

Acute Toxicity Study on Samul-tang(Siwu-tang) in Mice (ICR 마우스를 이용하여 사물탕(四物湯)의 급성 독성에 관한 연구)

  • Objectives: Samul-tang(Siwu-tang) has been traditionally prescribed a medicine as a restorative. Methods: In this study, we investigated the acute toxicity about water-extracted Samul-tang(Siwu-tang). Twenty-five mice completed 14 days of oral Samul-tang(Siwu-tang) at the respective doses of 0(control group), 2560, 3200, 4000 and 5000mg/kg. Results: We observed survival rates, general toxicity, change of body weight, and autopsy. Conclusions: Compared with the control group, we could not find any toxic alteration in all treated groups (2560, 3200, 4000 and 5000mg/kg). $LD_{50}$ of Samul-tang(Siwu-tang) was over 5000mg/kg and it is very safe to ICR mice.

Immuno stimulatory activities of Samul-tang, Sagunja-tang, Pamul-tang and Sipjeondaebo-tang in vitro (사물탕, 사군자탕, 팔물탕, 십전대보탕의 약리 활성 비교 연구)

  • Objectives : Samul-tang(SM). Sagunja-tang(SG). Pamul-tang(PM) and Sipjeondaebo-tang(SJ) was used many diseases such as sterility. menstrual disorder. general prostration. recruitment in Korea. We investigated the immune stimulatory activities of SG. SM. PM and SJ in in vitro. Methods: For comparision for effective of SM. SG. PM and SJ. this study examined anti-inflammation(NO. PGE2 assay). anti-oxidation(DPPH assay) and immune response in in vitro assay. For immune response activities. this study used NO synthesis on RAW 264.7 cells, splenocyte proliferation and cytokine assay(IL-2, IL-4) in splenocyte. Results: The results showed that SG. SM. PM and SJ were no significant effect anti-oxidation and anti-inflammatory effects. For immune response. they showed the splenocyte proliferation and macrophage proliferation. We confirmed that they synthesised NO a dose-dependent manner significantly and secreted the IL-4. Conclusions : These results suggested the SG, SM, PM and SJ had immune stimulatory activity. A efficacy of immune response in them had produced similar results.

The Effect of Samul-tanggahyangbuja on Depression and Learning on Repeated Stress in Ovariectomized Rats (사물탕가향부자(四物湯加香附子)가 난소적출 흰쥐의 우울 및 학습에 미치는 영향)

  • Objectives: In this research, the effect of samul-tanggahyangbuja on depression and learning in ovariectomized rats subjected to repetitive stress were assessed. Samul-tanggahyangbuja is the prescription consisting of Samul-tang and Cyperi Rhizoma. Methods: Ovariectomized rats were repeatedly stressed over a 2-week period. After being orally medicated with samul-tanggahyangbuja (100 or 400 mg/kg), rats performed the Morris water maze test and forced swimming test, and social exploration was assessed in a behavior test. As well, sucrose intake was measured and measurements of blood serum corticosterone and the change of interleukin-$1{\beta}$ (IL-$1{\beta}$) and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) in blood samples were made. Results: 1. In the Morris water maze test, rats medicated with 100 mg samul-tanggahyangbuja mastered the maze in a shorter time on the 4th day in comparison with the control group, while rats medicated with 400 mg samul-tanggahyangbuja mastered the maze more quickly (p<0.05 on the 3rd day ; p<0.01 on the 4th day, as compared to control). 2. Immobility time in the forced swimming test was significantly decreased in rats receiving 400 mg samul-tanggahyangbuja compared with the control group (p<0.05). 3. Sucrose intake and active social behavior of rats receiving 400 mg samul-tanggahyangbuja were markedly increased in comparison with the control group (p<0.01). 4. Blood serum corticosterone measurements revealed decreased blood serum corticosterone level after medicating with samul-tanggahyangbuja. But it was not statistically significant. 5. Treatment with either dose of samul-tanggahyangbuja significantly reduced IL-$1{\beta}$ and TNF-${\alpha}$ (p<0.05). Conclusions: These results suggest that samul-tanggahyangbuja possesses the anti-depressant and cognitive-enhancing activities related to menopause.

A Prior Study on the Effect of Samul-tang to Regeneration of Injured Peripheral Nerve Fiber (사물탕(四物湯)이 손상된 말초신경섬유 재생에 미치는 효과에 대한 사전 연구)

  • Peripheral axons in vertebrate animals can regenerate after nerve injury and accomplish its functional recovery. Numerous studies have revealed that diverse molecular factors are induced during axonal regeneration and their potential roles in axonal regeneration have been studied. Examples is N-CAM, L1, P0, nerve growth factors, GAP-43 and so forth. However, most of the studies on axonal regeneration have been primarily focused on axon fiber regrowth and elucidating molecular factors, and relatively less is known about functional recovery. Also, specific drugs or drug components used in the oriental medicine in relation to nerve fiber regeneration have not been known. And thus, in the present, a study on the effect of Samul-tang components and Samul-tang extracts to regeneration of peripheral axon fiber is underway by immunofluorescence staining. Therefore, this prior application of Samul-tang with documents consideration is reported with a plea for further investigation.
