Antimicrobial Activity of Turmeric(Curcuma aromatica Salab.) Extracts Against Various Pathogens and Spoilage Bacteria Isolated from Tofu (강황추출물이 두부 부패미생물과 병원성 미생물에 미치는 항균활성)

  • The antimicrobial activity of Curcuma aromatica Salab. was investigated. The Curcuma aromatica Salab. extract showed antimicrobial activity against six pathogens tested. These were Listeria monocytogenes ATCC 19115, Pseudomonas fluorescens ATCC 21541,Staphylococcus aureus ATCC 29273, Salmonella typhimurium ATCC 21541, vibrio parahaemolyticus ATCC 17802, and Aeromonas hydrophila KCTC 2358. Antimicrobial activity was alsonoted when the extract was tested against four isolates of Bacillus sp. purified from spoiled tofu. The growth of various pathogens was significantly inhibited (100 10,000-fold) upon growth in tryptic soy broth containing 0.05 0.2%(w/v) Curcuma aromatica Salab. extract(CE), after incubation for 12hr at $37^{\circ}C$. The growth of the four Bacillus isolates was also significantly inhibited in nutrient broth containing 0.05 0.2% CE after incubation for 24hr at $37^{\circ}C$. Although the antimicrobial activity of CE was decreased by heat treatment at temperatures above $80^{\circ}C$, the activity remained relatively high after heat treatment at $121^{\circ}C$ for 15min. The minimum inhibitory concentrations of CE were 0.1 0.3%(v/v culture) for the six pathogens, and 0.2 0.25% for the Bacillus isolates, respectively.

Prevalence of Antibiotic-Resistant Strains among Bacteria Isolated from Bovine Mastitis, Swine Diarrhea, and Swine Pneumonia. (소 유방염, 돼지설사, 돼지 폐렴의 원인균 분리 동정과 항생제 내성균 분포)

  • Microorganisms causing bovine mastitis, swine diarrhea and swine pneumonia were isolated from farms in Chungbuk and Kyunggi Province and the isolates were identified using microbial identification system(MIS). The most common isolates from bovine mastitis were Staphylococcus sp.,Streptococcus sp., and Corynebacterium sp., those from swine diarrhea were Escherichia coli, Salmonella sp., and Clostridium perfringens, those from swine pneumonia were Bordetella bronchiseptica and Pasteurella multocida. Examination of 16 antibiotics against these pathogens revealed that the incidence of antibiotic-resistant microoganisms were very high and that many of these isolates had multiple resistance to various commercially available antibiotics such as penicillin, ampicillin, erythromycin, streptomycin, norfloxacin, and tetracycline. These results suggest that the use of antibiotics in a farm should be controlled in order to decrease the number of antibiotic-resistant microorganisms were very high and that many of these isolates had multiple resistance to various commercially available antibiotics such as penicillin, ampicillin, erythromycin, streptomycin, norfloxacin, and tetracycline. Antimicrobial Activities of Scutellaria baicalensis Georgi Against Various Pathogens and Spoilage Bacteria Isolated from Tofu (두부 부패 미생물과 병원성 미생물에 대한 황금의 항균효과)

  • Antibacterial activities of Scutellaria baicalensis Georgi (SBG) extract were examined against spoilage bacteria isolated from commercial tofu and various pathogens such as Listeria monocytogenes ATCC 19115, Pseudomonas fluorescens ATCC 21541. Staphylococcus aureus ATCC 29737, Salmonella Typhimurium ATCC 11806, Vibrio parahaemolyticus ATCC 17802, and Aeromonas hydrophila KCTC 2358. Four kinds of spore forming organisms were isolated from commercial tofu and identified as Bacillus sp. KN-4, Bacillus sp. KN-6, Bacillus sp. KN-10 and Bacillus sp. KN-20 using API CHB kit. The SBG extract showed high antibacterial activities and significantly inhibited the growth of the isolated spoilage bacteria and pathogens. The inhibitory effects against the organisms increased as the concentration of the SBG extract increased. The antimicrobial activities of the SBG extract were maintained markedly after heat treatments $(80^{\circ}C/30\;min,\;100^{\circ}C/30\;min\;and\;121^{\circ}C/15min)$. The minimum inhibitory concentrations (MIC) of the SBG extract against the organisms ranged from 1,000 ppm to 5,000 ppm.

Analysis of the Results of Blood Cultures, 1984~1987 at Yeungnam University Hospital (형랙배양검사 성적의 분석 -1984년에서 1987년까지 -)

  • Reviewing the results of the blood cultures performed at Yeungnam University Hospital during 4-year-period through January. 1, 1984 to December 31, 1987, the following results were obtained. 1) Out of 808:3 blood specimens cultured microorganisms grew in 582 specimens with positivity rate of 7.20%. Polymicrobial bacteremia was found in 16 patients. 2) Among 582 positive specimens, Gram-positive cocci grew in 189 specimens, and Gram-negative bacilli, in 393 specimens. Clinically significant microorganisms consisted of 82 Staphylococcus aureus, and 20 Strptococcus species in Gram-positive cocci group, 80 Salmonella typhi, 72 Escherichia coli, 72 Salmonella paratyphi A in Enterobacteriaceae, and 46 Pseudomonas cepacia, and 16 Pseudomonas aeruginosa in glucose non-fermentating microorganisms. 3) Increasing incidence of Serratia, Acinetobacter and Pseudomonas species as major nosocomial infection source is noteworthy. They showed increased tendency from 6.3% of 1984 to 17.7% of 1987 of total positive blood cultures. 4) High isolation rate of Pseudomonas species and Aeromonas hydrophilia was noted in summer, while Salmonella typhi showed high prevalence from May to September and in January. 5) In susceptibility tests of isolated organisms, staphylococcus aureus was sensitive to basic antimicrobial agents except for ampicillin. The glucose non-fermentating microorganisms showed high resistance to basic antimicrobial agents in 32.2%. In conclusion, considering the relatively higher incidence of growth of Staphylococcus epidermidis than ideal level indicates that sampling technique should be improved. Secondly, all the hospital staffs in cooperation with Hospital Infection Committee are desirable to pay efforts to decrease the nosocomial infection.

  • To evaluate nutritional values and to detect pharmacologically active and antibacterial components of wild edible plants in Korea, 20 species were examined. 1) The essential free amino acid compositions of these plants were determined by TLC. The plants contained threonine, leucine, valine and methionine. 2) In antibacterial tests of 17 species the plants Hemerocallis sp. and Plantago asiatica showed an antibacterial activity against Staphylococcus, Bacillus, Eschericia, and Salmonella species. 4) Rutin. quercitrin, $k\ddot{a}mpferol$, chlorogenic acid and caffeic acid were also identified.

  • Microorganisms capale of utilizing linear alkylbenzene sulfonates(LAS) as sole carbon source were isolated from industrial effluent by using LAS agar plates. The isolated strains were identified as Salmonella sp(BC-2) and Escherichia sp.(BC-3) from the results of morphological, cultural and biochemical tests. The optimal condition for the growth and biodegradation of LAS was the initial pH 7.0 and LAS concentration 0.1%. The isolated BC-2 and BC-3 strains harbored plasmid and LAS-degrading activity was lost when the plasmids were cured by mitomycin C. The plasmids were transformed into E. coli and transformants have the LAS-degrading activity. Isolated strains were examined for primary biodegradation rate of LAS in the medium by methylene blueactive substance(MBAS) method. Of these isolates, BC-2 and BC-3 strains degradated LAS upto 60% and high resistant to CdCl$_{2}$ and HgCl$_{2}$. Isolated strains were sensitive to chloramphenicol, kanamycin, rifampicin, streptomycin and tetracycline but resistant to ampicillin and lincomycin.] Its minimal inhibitory concentration(MIC) for ampicillin was more than 1500 $\mu $g/ml.

Diversity and Biological Activities of Endophytic Fungi of $Emblica$ $officinalis$, an Ethnomedicinal Plant of India

  • In the present study, an attempt to evaluate the antimicrobial and antioxidant activity of fungal endophytes inhabiting $Emblica$ $officinalis$ has been made keeping in view the medicinal importance of the selected host plant in Indian traditional practices. A total of four endophytic fungi belonging to Phylum Ascomycetes were isolated from different parts of the plant which were characterized morphologically and by using rDNA-internal transcribed spacer. The most frequently isolated endophyte was $Phomopsis$ sp. The antioxidant activity by 2, 2-diphenyl-1-picrylhydrazyl (DPPH) and reducing power assay, and total phenol were evaluated using ethanolic extract of endophytic fungi. DPPH activities in all the ethanolic extract increased with the increase in concentrations. Endophytes, $Phomopsis$ sp. and $Xylaria$ sp. showed highest antioxidant activity and also had the higher levels of phenolics. Antimicrobial activity of fungal extract were tested against four bacteria namely, $Escherichia$ $coli$ MTCC730, $Enteroccocus$ $faecalis$ MTCC2729, $Salmonella$ $enterica$ ser. $paratyphi$ MTCC735 and $Streptococcus$ $pyogenes$ MTCC1925, and the fungus $Candida$ $albicans$ MTCC183. In general, the fungal extracts inhibited the growth of test organisms except $E.$ $coli$.

Epizootiologic Surveys on Canine Parvovirus Infections of Dogs Kyungbuk West Area (경북서부지역에서의 개 파보바이러스 감염상황 조사)

  • The disease syndrome characterized by the acute vomiting and diarrhea with high mortality had been greatly epidemic in Kyungbuk West Area since March 1990 and it was followed serologically for the classification of the agent. The agent present in feces of dogs associated with this syndrome had characteristic feature in agglutinating pig red blood cells that was specifically inhibited by anti-CPV reference dog serum. This also showed the serological identity with the reference CPV antigen in Hemagglutinating inhibition test. The result obtained were summarized as follows : 1. During 5 years(March. 1990∼September. 1994), 1,470 dogs were investigated on the actual condition of CPV infections. The Infection rate of CPV from dogs was 62.5% and mortality rate was 59.8%. 2. Among 24 fecal samples collected from the dogs with enteric disease, all showed the hemagglutinating activity to porcine erythrocyte ranging from 40 to 5,120 of HA titers. 3. Among 12 sera samples collected from the dogs with enteric disease, all showed the serological identity with the reference CPV antigen from 5 to 5,120 of HI titers. 4. Bacteriologic examination of fecal specimens resulted in the isolation of pathogeric bacteria such as Staphylococcus sp, Streptococcus sp, Escherichia coli and Bacillus. Cultures for salmonella sp and Clostridium remaind negative. 5. The prevalence and identification of internal parasites were determined by fecal examination using the floatation methods. From 20 fecal samples 12(60.0%) were isolated and their species were Toxacara canis, Toxascaris leonina, and coccidium.

Microbial Contamination Levels in Porphyra sp. Distributed in Korea (국내 유통 김(Porphyra sp.)의 미생물 오염도 평가)

  • Aerobic bacteria, coliforms, Escherichia coli, and pathogenic bacteria were investigated in laver Porphyra sp. samples from various regions of Korea. The mean bacterial counts were $6.9{\pm}0.87log\;CFU/g$ (range 4.0 to 7.7) log CFU/g in dried laver, $2.83{\pm}4.36log\;CFU/g$ in roasted laver, and $4.93{\pm}1.43log\;CFU/g$ in seasoned laver. Coliforms were most abundant (mean count: $2.1{\pm}1.01log\;CFU/g$) in dried laver. No pathogenic bacteria, including Staphylococcus aureus, Salmonella spp. Vibrio parahaemolyticus, or Listeria monocytogenes, were detected in any of the samples. Aerobic microorganisms were the most diverse microorganisms in dried laver. Staphylococcus spp. were predominant, but S. aureus was not detected. Standardization of laver production is necessary to ensure a hygienic product because laver products are often ingested without heating or cooking, and the production process is simple.

Characterization of the Bacteriocin Produced by a Leuconostoc mesenteroides Strain Inhibiting the Growth of Lactobacillus sakei (Lactobacillus sakei 생육저해활성 보유 Leuconostoc mesenteroides가 생산하는 Bacteriocin의 특성)

  • Lactobacillus sakei is known to be the most populous lactic acid bacteria in over-ripened kimchi. Twenty three strains of Leuconostoc species inhibiting the growth of Lb. sakei were isolated from kimchi and amongst these the Leuconostoc mesenteroides strain CK0122 exhibited the highest antagonistic activity against Lb. sakei. The culture supernatant of the strain CK0122 was fractionated by a molecular weight cutter and lyophilized. The fraction with a molecular weight of less than 3,000 Da showed antagonistic activity against Lb. sakei. The antibacterial activity of the active fraction was sensitive to proteinase K treatment, confirming its proteinaceous nature (bacteriocin). The crude bacteriocin was active in the pH range of 4 to 7 and extremely stable after 15 min of heat treatment at $121^{\circ}C$. The crude bacteriocin inhibited the growth of Bacillus cereus, Listeria monocytogenes, Staphylococcus aureus, Alcaligenes xylosoxydans, Flavobacterium sp., and Salmonella typhimurium.
