이 연구는 AP-PCR 기법을 이용하여 Salmonella 균주의 유전적 다양성과 근연관계를 비교하고자 수행하였다. 18종의 Salmonella 균주에 대해 8종류의 primer를 이용하여 각 균주에 대한 DNA 밴드를 검출한 결과 총 AP-PCR 단편의 수는 39에서 52개의 범위였으며 평균 43.6개가 검출되었다. 총 349개의 표지인자가운데 다형성을 나타내는 단편들은 185개로서 53.0%의 다형성 수준을 보여주었다. 살모넬라균주들은 GEN 703과 GEN 708 primer에서 각각 0.682와 0.676의 높은 다형성 수준을 보여주었으나 GEN 603, GEN 604, 및 GEN 607 primer에서는 각각 0.404, 0.460 및 0.472의 비교적 낮은 수준의 다형성을 나타냄으로써 살모넬라균주간 상동성 비율이 높음을 시사해 주고 있다. 따라서, 이들 primer들은 Salmonella 균주들의 AP-PCR 분석에 대단히 효율적임을 확인할 수 있었다. S typhimirium CU 2001의 다형성 수준은 77%로 S typhimirium CU 2002와 가장 근연관계가 가까운 것으로 나타났으며 다음이 S typhimirium CU 2003으로 63%, S typhimirium ATCC 14028이 50%, 그리고 S typhimirium CU 2004가 43%의 근연관계를 나타내었다. S enteritidis ATCC 13076의 다형성 수준은 83%로 S enteritidis CU 2005와 근연관계가 가장 가깝게 나타내었으며 다음이 S enteritidis CU 2006으로 63%, S enteritidis CU 2007이 58%의 근연관계를 나타내었다. S choleraesuis CU 2009의 다형성 수준은 67%로 S choleraesuis CU 2010과 가장 가까운 근연관계를 나타내었으며 S choleraesuis CU 2008은 53%의 근연관계를 나타내었다. S gallinarum CU 2011과 S gallimarum CU 2012의 다형성 수준은 70%로 근연관계가 가장 가깝게 나타났으며 S gallinarum ATCC 9184는 60%의 근연관계를 나타내었다. 마지막으로 S pullorum CU 2013과 S pullorum CU 2014의 다형성 수준은 80%로 매우 높은 근연관계를 나타낸 반면 S pullorum No 11은 53%로 근연관계가 먼 것으로 나타났다. 따라서, AP-PCR 분석은 Salmonella 균주의 유전적 다양성 및 근연관계를 추정하기 위한 강력한 도구로서 이용할 수 있을 것이다.
가금티프스는 닭, 오리, 칠면조, 꿩 등 가금류에 Salmonella gallinarum이 원인균이 되어 발병하는 질병이다. 본 연구는 가금티프스를 방제하기 위한 생균제 개발을 위한 기초조사의 목적으로 가금티프스 원인균인 5. gallinarum 의 생육을 저해시킬 수 있는 길항균을 야생꿩에서 분리, 선발했다. 최종적으로 3종의 우수 길항균주를 선발하였으며 선발, 분리된 길항균을 동정한 결과 Ll9, L33균주는 similarity가 100%인 Sphinromonas sanguis로 동정 되었고 L50균주는 similarity가 96.2%로써 Sphinromonas sanguis의 근연종으로 동정되었다. 선발된 길항균주가 생산한 길항물질의 특성을 확인해본 결과, S. gallinarum을 저해하는 물질이 고분자성 물질이고 내열성임을 추정 할 수가 있었다. S. sanguis Ll9의 경우 배양 20시간 이후부터 길항물질이 생산되었으며 탄소원, 질소원을 각각 maltose, $(NH_4)_2SO_4$를 첨가하였을 때 가장 높은 길항력을 나타내었다. S. sanguis L33의 경우 배양 22시간 이후 부터 길항물질이 생산되었으며 탄소원, 질소원을 각각 galactose, urea를 첨가하였을 때 가장 높은 길항력을 나타내었다. S. sanguis L50은 배양 10시간 이후부터 길항물질을 생산하였으며 탄소원, 질소원을 각각 saccharose, $(NH_4)_2S_2O_8$음로 하였을 때 가장 높은 길항력을 나타내었다.
주요 가금티푸스 유발균인 Salmonella gallinarum에 대항하는 항체를 계란으로부터 효율적으로 얻기 위하여 사료 첨가제, 산란계에 백신 주사 시 adjuvant 종류, target균 외의 첨가균의 여부에 따른 계란 내의 면역 단백질 생산을 비교 측정하였다. 참숯의 급여에 의한 면역처리구의 specific IgY 함량을 보면 0.5% 참숯을 급여한 처리구가 가장 높은 함량을 나타내었으며, specific IgY의 생산에 참숯의 급여가 큰 영향을 미치는 것으로 나타났다. Adjuvant 처리에 따른 total IgY 함량의 변화는 거의 없는 것으로 나타났으나 specific IgY는 Freund's adjuvant를 사용하여 백신 처리한 처리구에서 높게 나타났다. Adjuvant 사용 시 결핵균을 같이 첨가한 경우 더 많은 specific IgY가 생성되었으나, 수산화 알미늄 처리구에서는 결핵균 첨가에 의한 영향이 나타나지 않았다. 15주령에 면역처리를 해준 경우 21주령에 면역처리를 해준 경우 보다 산란율이 높았으며, 사료에 참숯을 첨가한 경우 일반사료 급여군에 비해 산란율저하가 완화되었다.
가금티푸스 유발균인 Salmonella gallinarum 대항하는 항체의 함량이 높은 계란을 생산하기 위해 면역처리 후 산란계의 난황에서 항체를 분리하여 총 IgY와 특수 IgY의 함량의 변화와 그 특성을 조사하였다. 먼저 난황으로부터 항체의 수율과 경제적인 측면을 비교하기 위하여 난황 분리된 난황 25 g을 λ-카라기난(Sigma C-3889) 0.1%, 냉수 가용성(MSC #10387) 1%, 2%, 90℃ 가용성 (MSC #10386) 1%, 2% 및 냉수 가용성(0.1%) : 90℃ 가용성(0.1%) = 1 : 1 1%, 2%로 사용하여 분리하는 하였을 때 λ-카라기난 0.1%, 냉수 가용성 1% 비교적 높은 회수율과 total IgY, specific IgY 함량을 나타내었지만 λ-카라기난 0.1%가 더 나은 것으로 나타났다. Specific IgY는 마지막 백신 접종 후 44주간까지 꾸준히 검출되고 있는 것으로 나타났다. 산도 변화에 의한 total IgY의 잔존 함량을 보면 pH 5-9 범위에서 70% 이상 잔존하며, specific IgY도 pH 4-10에서 74% 이상 존재하여 산에 대하여 대체로 안정적임을 알 수 있었다. 열처리 후 조항체 용액을 total IgY, specific IgY 역가 높게 유지되는 것을 관찰 할 수 있었다. 이로써 난황으로부터의 면역 항체 생산의 가능성이 확인되었고 이러한 IgY는 추출과 가공처리 등에 의해 비교적 안정적인 것으로 나타나 IgY를 이용한 가금티푸스의 예방 또는 치료용의 대량 생산 가능성이 높은 것으로 사료되었다.
The study was carried out to evaluate and compare the immune responses in mice experimentally infected with either wild-type or isogenic mutants of Salmonella enterica serovars Enteritidis (SE), Salmonella Typhimurium (ST) and Gallinarum (SG). The mutant strains were constructed by allelic replacement of some virulence-associated genes in the wild-type strains. Seven-week-old female BALB/c mice were orally or intraperitoneally inoculated by injecting bacterial suspension. To evaluate the immune responses, enzyme-linked immunosorbent assay (ELISA) and enzyme-linked immunospot (ELISPOT) assay were conducted with serum and fecal samples. As a result, the mice group infected orally with the SE mutant strain showed the highest level of specific IgA-secreting splenocytes, compared to the other groups. The peritoneally injected groups showed the greater levels of IgG1 than the orally injected groups, which was in a good agreement with the previous studies. In addition, the mutant infected groups had the similar secretion levels of antibodies with the wild-type infected groups. These results demonstrated that the SE mutant strain elicited humoral immune response as much as wild-type, implying that it can be useful as a delivery vehicle as well as a candidate of a live attenuated vaccine.
Protective efficacy of trivalent Salmonella inactivated vaccine containing Chlorhexidine-inactivated S. Enterltidis (SE), S. Typhimurium (ST), and S. Gallinarum (SG) strains, was evaluated in this study. A total of 70 brown nick layers were divided into 7 groups, A to G, containing 10 hens per group. All hens in groups B to D were intramuscularly immunized with approximately 7×108 cells (3×108 cells of SE+1×108 SE+1×108 cells of ST+3×108 cells of SG) of the trivalent vaccine in 0.5 mL of PBS. All chickens in groups E to G were injected with sterile PBS. All hens of groups B and E, groups C and F, and groups D and G were orally challenged with approximately 2 ×109 CFU of wild-type SE, ST, and SG, respectively. Serum IgG titers and CD3+CD4+ T-cells, and CD3+CD8+ T-cells levels of groups B to D significantly higher than those of group A. In addition, all animals in groups A to C, E and F showed no clinical symptoms and survived after the virulent challenges, whereas one chicken in group D died and all chickens in group G died following the challenge. The protection against wild-type SE and ST in liver, spleen, cecum, and cloaca of groups B and C chickens was significant effective as compared with those in groups E and F. These indicate that the trivalent inactivated vaccine can be an effective tool for prevention of Salmonella infections by inducing robustly protective immune responses and cellular immune response in chickens.

Objective: This study investigated the effects of Bacillus subtilis CSL2 (B. subtilis CSL2) administration before Salmonella challenge on the fecal microbiota and microbial functionality of Hy-line Brown (HLB) laying hens. Methods: Fecal samples were collected from control (CON), Salmonella-infected (SAL) and Salmonella-infected, probiotic-treated (PRO) groups before and after Salmonella challenge for microbiome analysis using 16S rRNA gene pyrosequencing. Results: Infection with Salmonella led to decreased microbial diversity in hen feces; diversity was recovered with Bacillus administration. In addition, Salmonella infection triggered significant alterations in the composition of the fecal microbiota. The abundance of the phylum Firmicutes decreased while that of Proteobacteria, which includes a wide variety of pathogens, increased significantly. Bacillus administration resulted in normal levels of abundance of Firmicutes and Proteobacteria. Analysis of bacterial genera showed that Salmonella challenge decreased the population of Lactobacillus, the most abundant genus, and increased populations of Pseudomonas and Flavobacterium genera by a factor of 3 to 5. On the other hand, Bacillus administration caused the abundance of the Lactobacillus genus to recover to control levels and decreased the population of Pseudomonas significantly. Further analysis of operational taxonomic units revealed a high abundance of genes associated with two-component systems and secretion systems in the SAL group, whereas the PRO group had more genes associated with ribosomes. Conclusion: The results of this study indicate that B. subtilis CSL2 administration can modulate the microbiota in HLB laying hens, potentially acting as a probiotic to protect against Salmonella Gallinarum infection.
