Adherence of Salivary Proteins to Various Orthodontic Brackets (다양한 교정용 브라켓 표면에 부착하는 타액단백질에 관한 연구)

  • The principal aims of this study were to identify the composition of salivary pellicles formed on various orthodontic brackets and to obtain a detailed information about the protein adsorption profiles from whole saliva and two major glandular salivas. Four different types of orthodontic brackets were used. All were upper bicuspid brackets with a $022{\times}028$ slot Roth prescription; stainless steel metal, monocrystalline sapphire, polycrystalline alumina, and plastic brackets. Bracket pelicles were formed by the incubation of orthodontic brackets with whole saliva, submandibular-sublingual saliva, and parotid saliva for 2 hours. The bracket pellicles were extracted and confirmed by employing sodium dodecyl sulfatepolyacrylamide gel electrophoresis, Western transfer methods, and immunodetection. The results showed that low-molecular weight salivary mucin, ${\alpha}-amylase$, secretory IgA (sIgA), acidic proline-rich proteins, and cystatins were attached to all of these brackets regardless of the bracket types. High-molecular weight mucin, which promotes the adhesion of Streptococcus mutans, did not adhere to uy orthodontic brackets. Though the same components were detected in all bracket pellicles, however, the gel profiles showed qualitatively and quantitatively different pellicles, according to the origins of saliva and the bracket types. In particular, the binding of sIgA was more prominent in the pellicles from parotid saliva and the binding of cystatins was prominent in the pellicles from the form plastic brackets. This study indicates that numerous salivary proteins adhere to the orthodontic brackets and these salivary proteins adhere selectively according to bracket types and the types of the saliva.

A study on the correlations between salivary levels of lysozyme, lactoferrin and secretory Immunoglobulin A to Streptococcus mutans and caries susceptibility (치아우식감수성과 타액내 Lysozyme, Lactoferrin 및 Streptococcus mutans에 대한 secretory IgA 수준과의 상관관계에 관한 연구)

  • Saliva plays an important role in modulating the oral microbial ecology. And it is suggested to influence the initiation and progression of the dental caries. To evaluate the correlations between the salivary antimicrobial agents and the caries susceptibility, the 51 subjects were divided into 3 groups according to caries experience ; caries resistant group, medium caries susceptible group, and high caries susceptible group. Stimulated whole saliva was collected, and the salivary levels were measured for lysozyme, lactoferrin, and secretory-IgA to Streptococcus mutans. The lysozyme level was estimated using Micrococcus diffusion plate, lactoferrin level was determined with a non-competitive avidin-biotin enzyme immunoassay, and the titer of secretory IgA to Streptococcus mutans was assayed with ELISA. The results were as follows: 1. Lysozyme levels of each group showed no significant difference statistically (p>0.05). 2. The caries resistant group and the medium caries susceptible group had significantly higher levels of lactoferrin than the high caries susceptible group (p<0.05). But no clear difference was observed between the caries resistant group and the medium caries susceptible group(p>0.05). 3. The caries resistant group and the medium caries susceptible group showed relatively higher levels of the secretory IgA to Streptococcus mutans than the pigh caries susceptible group, but no significant difference was observed statistically (p>0.05).

COMPARISON OF THE DENTAL CARIES, CARIOGENIC MICROORGANISMS AND LEVELS OF SALIVARY IMMUNOGLOBULIN BETWEEN NORMAL PERSON AND MENTAL RETARDED PERSON (정상인과 정신지체인의 치아우식증과 치아우식 원인균, 타액내 면역항체의 비교)

  • The aims of this study were to compare the status of dental caries between mental retarded persons(MR) and normal persons and investigated the association among dental caries and oral factors(plaque index, S. mutans, Lactobacillus, buffering capacity, salivary total IgA and anti-S. mutans IgA titers). DMFS index and plaque index were significantly greater in the MR person group than in the normal person group. The concentration of S. mutans-specific IgA was significantly greater in the normal person group than in the MR person group, but that of salivary total IgA was not show the significant difference. Salivary buffering capacity was significantly greater in the normal person group than in the MR person group, but the counts of salivary S. mutans and Lactobacillus were not significantly different. By age group(I: 9-11Y, II: 12-14Y, III: 15-18Y), DMFS index and plaque index were significantly greater in the MR person group than in the normal person group at III. The S. mutans counts and Lactobacillus counts were significantly greater in the normal person group than in the MR person group at I, but those were contrary at II, III. There was a high correlation among caries index and buffering capacity, level of S. mutans and Lactobacillus, plaque index at III.

Effect of Aromatherapy according to the type of Sasang constitution (사상체질별(四象體質別) 향요법(香療法)의 효과(效果))

  • The purpose of this quasi-experimental study was to explore the differences in the effect of aromatherapy according to the type of Sasang constitution. The subjects were fifty-three nursing students experiencing clinical practice from March 10th. 2000 to April 22nd, 2000. For experiment, researcher used inhalation of 2% grape seed oil mixed with Rosemary oil for 2 minutes and massage with same oil for 20 minutes on the back of subjects in the speed of 20 strokes per minutes. The instruments used for this study were Questionnaire of Sasang Constitution Class II program for personal computer developed by Kim Sun Ho et al., (1996), and Mood Questionnaire developed by Ryman. Biersner, & Larocco (1974). The concentration of IgA within saliva was analyzed by immunoturbidimeter assay. The data were analyzed using SPSS and hypotheses were examined with paired t-test and ANCOVA. The results were as follows : 1) The mean score of mood was increased significantly after aromatherapy. 2) The mean concentration of salivary IgA was increased significantly after aroma therapy. 3) There was no significant difference in the mean score of mood after aromatherapy according to the type of Sasang constitution, 4) There was no significant difference in the mean concentration of salivary IgA after aromatherapy according to the type of Sasang constitution. In conclusion, the results suggest that aromatherapy with Rosemary oil have effect on the improvement of mood and IgA within saliva in the nursing students under stress caused by clinical practice regardless of the type of Sasang constitution.

THE EFFECT OF GALVANIC CURRENT BETWEEN DENTAL RESTORATIONS ON HUMAN SALIVA (치아 수복재에 의한 갈바닉 전류가 인체 타액에 미치는 영향)

  • The purpose of this study was to evaluate the effect of galvanic current between different metallic restorations on human saliva. The rate of salivary flow and concentrations of IgG, IgM, sIgA and lactoferrin were measured. In this study, unstimulated whole saliva collected before restoration was regarded as control group and unstimulated whole saliva collected 10 minutes, 1 day, 1 week, and 1 month after restoration were regarded as experimental groups. Following results were obtained from this study. 1. There were some differences in values of salivary flow rate between experimental groups, but the changes in values compared to those of the control group were not statistically significant(P>0.05). 2. Measurements of major antibacterial components of saliva showed that while the concentrations of IgG and IgM decreased significantly 1 week and 1 month after restoration(P<0.05), changes in values of sIgA and lactoferrin were not statistically significant(P>0.05). 3. In vitro measurements of galvanic currents decreased sharply in the first 20 seconds and thereafter decreased gradually. Galvanic current values measured in the early stages were greatly varied, but after 2 hours, the values in all groups approximated each other.

  • Immunoglobulin G4 (IgG4)-related disease is an inflammatory condition characterized by IgG4 positive plasma cell infiltration. It can affect any organ in the body and mainly involves the pancreas, liver, biliary tracts, orbits, salivary glands and lymph nodes. It can manifest as an inflammatory pseudotumor. Pseudolymphoma as an inflammatory pseudotumor is a group of benign tumors that exhibit histological and clinical features suggestive of malignant lymphoma. Studies on IgG4-related disease are rarely reported, and no case of the disease that involved the maxillary bone and adjacent soft tissue, except for the skin, has been reported. Therefore, we report herein a case of pseudolymphomatous IgG4-related disease that involved the maxilla, with a literature review.

Effect of Prunus mume extract(PME)-containing chewing gum mastication on the change of saliva ingredients

  • Objectives: The purpose of the study is to investigate the effect of chewing gum containing Prunus mume extract(PME) on the change of saliva ingredients. On the basis of the biological background of molecules and diagnostic indices in the use of saliva, the mastication effect of chewing gum containing PME was demonstrated in terms of secretory IgA concentration and total protein concentration in stimulated saliva. Methods: This study is an experimental research on the use of a research design before and after applying a randomized control group. Participants were distributed randomly to the experimental group and the control group, respectively. The experiment group was instructed to masticate the chewing gum containing PME for 10 minutes for one month after each meal within 30 minutes. Salivary secretion was collected by the participants between 8 and 10 a.m in the morning in the research office. For the measurement of secretory IgA and total protein concentrations in the saliva, indirect enzyme-linked immunosorbent assay(ELISA) was used. Results: The salivation stimulation rate was significantly increased after four weeks of masticating chewing gum containing PME after each meal(p<0.001). Mastication of chewing gum containing PME for four weeks decreased the concentration of secretory IgA much more significantly than that after mastication for one week(p=0.003). The concentration of total protein in the saliva was decreased after four weeks in the experimental and control groups. Conclusions: Mastication of chewing gum containing PME stimulated salivary secretion and led to oral disease prevention in patients with xerostomia. Furthermore, it seems to be urgent to seek measures that can be utilized in intervention for patients with xerostomia.

Effects of Back Massage with Lavender Essence Oil on the Stress Response of the ICU Nurses (라벤더 정유를 이용한 등마사지가 중환자실 간호사의 스트레스 반응에 미치는 효과)

  • This quasi-experimental study was to explore the effects of back massage with Lavender essence oil on the stress response of the ICU nurses. Method: Twenty nurses were selected as an experimental group and twenty nurses were in the control group. Lavender essence oil mixed with Jojova carrier oil was used to massage the neck, shoulder, back, and low back. Massage was done three times a week for 20 minutes for 2 weeks by the massage therapist. The stress response was measured with a subjective stress score, Cortisol and IgA level in saliva. The cortisol level in saliva was analyzed by an radioimmunoassay before and after back massage. The level of IgA in saliva was analyzed by an immunoturbidimeter assay and subjective stress scores was measured by Visual Analogue Scale. The data was analyzed using SPSS 8.0 and the hypotheses were tested by ANCOVA and the Pearson coefficient correlation. Result: 1) Score of subjective stress decreased significantly after the use of back massage with Lavender essence oil. 2) Level of Cortisol in saliva did not change significantly after the use of back massage with Lavender essence oil. 3) Level of IgA in saliva increased significantly after the use of back massage with Lavender essence oil. 4) After the use of back massage with Lavender essence oil, there was significant negative correlation between subjective stress and level of IgA in saliva. Conclusion: Back massage with Lavender essence oil is an effective intervention to reduce the stress response in the ICU nurses.
