김치로부터 분리한 유산균과 효모 혼합 발효액의 제빵 최적화 (Optimization of Bread Fermentation with Lactic Acid Bactria & Yeast Isolated from Kimchi)

  • 김치로부터 유산균과 효모를 분리하여 분리균주의 혼합 배양액을 이용하여 제빵 공정의 최적화에 관하여 연구하였다. 김치로부터 젖산박테리아(lactic acid bactria)를 분리하여 제빵 환경에서 생육 조건이 좋은 것으로 나타난 저온 숙성 김치의 젖산균은 Leuconostoc mesenteroides와 Lactobacillus brevis의 2종이 선별되었고, yeast는 Saccharomyces fermentati, Saccharomyces cerevisiae 2종이 선별되었다. 분리된 4종의 균주를 적절하게 혼합배양시 co-work system을 형성하여 발효촉진의 효과를 보여주었다.

  • The cuclodextrin glucanotransferase (CGTase) gene of Bacillus macerans was subcloned at the downstream of yeast ADH1 promoter, and then the resulting plasmid pVT-CGTM(9.15 kb) was introduced into the yeast host strain, Saccharomyces cerevisias 2805. The transformed yeast, S. cerevisiae 2805/pVT-CGTM, showed the starch-hydrolyzing activity on the starch-azure plate. The optimal conditions for the CGTase expression were found to be 2% dextrose, initial pH5.5, 3$0^{\circ}C$, and 48hr cultivation. Under this condition, the extracellular CGTase activity reached at 0.53 U/mL, whereas the intracellular activity was about 0.03U/mL. This result indicates that the signal peptide of Bacillus CGTase functioned well in S. cerevisiae.

  • The effect of ginseng extract and ginseng saponins on alcoholic fermentation was studied 1. Alcoholic fermentation on gllicose medium at 30$^{\circ}C$. by Saccharomyces coveanus and Saccharomyces uvavum was stimulated when the media contained 5% and 10 % of ginseng extract, respectively. But that of Saccharomyces cerevisiae was inhibites by the addition of 10% of ginseng extract. 2. Saponin did not stimulate the alcoholic fermentation by Saccharomyces uvarum. 3. The yeast cell counts was increased remarkably by addition of ginseng extract while that of ginseng saponins was increased slightly. Dried cell weight of the broth which Contained 5% of ginseng extract was 3 times than that of control.

  • In order to investigate the characteristics of alcohol fermentation by Saccharomyces cerevisiae, compositions and free sugar content of dried riped pumpkin powder were determined. The proximate compositions of ripe pumpkin powder were as follows: moisture 11.98%, total sugar 62.18%(dried weight basis 70.64%), protein 13.38%, lipid 0.85%, fiber 7.07% and ash 4.54%. The compositions of free sugar in well ripe pumpkin were: glucese 85.36mg/g, fructose 40.68mg/g, sucrose 68.25mg/g, lactose 18.60mg/g and maltose 3.82mg/g. The optimum conditions for alcohol fermentation by S. cerevisiae were as follows; incubation temperature of 3$0^{\circ}C$, initial pH of 6.0, ripe pumpkin powder concentration of 10% and cells inoculation of 1.3$\times$$10^{6}$ cells/ml liquid medium. Ethanol production under the optimum conditions was 5.95g/100g in liquid medium containing 10% ripe pumpkin powder after 4 days incubation.

Saccharomyces cerevisiae에서 PNP의 부분 정제와 특성 (Partial Purification and Characterization of Purine Nucleoside Phosphorylase in Saccharomyces cerevisiae)

  • Intracellular purine nucleoside phosphorylase (PNP) from Saccharomyces cerevisiae was partially purified using ammonium sulfate fractionation, heat treatment, a DEAE-Sephadex A-50 anion exchange chromatography and a Sephadex G-100 gel filtration chromatography. The enzyme was purified 20 fold with 3% recovery. The stability of enzyme was kept by addition of inosine and dithiothreitol. The pH optimum was found to be from 6.3 to 7.3 PNP was sensitive to 10mM of $Hg^{2+}$ , $Cu^{2+}$ , and was inactivated completely by 2 mM of p-chloromercuribenzoate and 5,5'-dithiobis (2-nitrobenzoate). The enzyme was capable of catalyzing the phosphorolysis of inosine, deoxyinosine, guanosine, deoxyguanosine and adenosine.

  • The whole proteins from 10 different Saccharomyces species were separated by isoelectric focusing, which was carried out in pH gradient polyacrylamide gels with the carrier ampholytes of various pH ranges. About 25 protein bands were found in the gel using pH 3.0-10.0 carrier ampholytes. In gel using pH 4.0-7.0 carrier ampholytes, the protein band of pI 6.3 was found in Sacch. cerevisiae NCYC 478, ATCC 26787, Sacch. rosei and Sacch. uvarum, but it was absent in Sacch. cerevisiae ATCC 24903, ATCC 42949, ATCC 36029, Sacch. steineri var hara, Sacch. bayanus, and Sacch. diastaticus.

  • The cyclodextrin glucanotransferase(CGTase) gene of Bacillus macerans was expressed in Saccharomyces cerevisiae and the recombinant CGTase was partially purified from the yeast culture supernatant. The optimal pH and temperature of the CGTase were found to be 6.0 and 5$0^{\circ}C$, respectively. The pH and temperature stabilities of the recombinant enzyme were significantly enhanced and the half life at 55$^{\circ}C$ was about 60 hr. When the recombinant CGTase was reacted with 5% soluble starch, the conversion yield of total cyclodextrin (CD) from starch was estimated to be 41% at 48 hr, whereas the wild type enzyme showed the yield of 12%. This improvement of conversion yield and thermal stability of CGTase may be useful for the development of low-cost CD production process.

김치 Starter 용으로 분리한 효모의 동정

  • The eleven strains, which could be used lactic and acetic acids as carbon sources, were isolated from kimchi and identified; the strains were facultative microorganisms which could be grown at low temperature (10$\circ$C) and around pH 3.2. As results of morphological, biochemical and physiological tests, 5 species of 3 genera were identified as Debaryomyces coudertii, Pichia edia, Pichia chambardii, Pichia haplophilia and Saccharomyces fermentati. Each strain was grown in basal media. In acidic resistance and acid utilization test, Saccharomyces sp. YK- 17 and Saccharomyces fermentati YK-19 were grown well in basal and YM media containing 0.3% lactic acid. And two strains were grown in basal and YM media containing O.3% lactic acid and 0.6% acetic acid. Since strain YK-19 was grown better at 10$\circ$C than that in 25$\circ$C, strain YK-19 was known to be a psychrophilic strain.

  • This study was conducted to investigate the effects of Korean ginseng extracts and their fractions on the growth of Saccharomyces cerevsiae and Saccharomyces uvamm, their glucose consumption and alcohol production. The growth of both yeasts were stimulated by ginseng extracts and their water soluble fractions, but were supressed by ether extracts and an n-butanol extracts. Their growth were enhanced considerably by low molecular weight fractions (< 1,000) in water solubles. Similar results were also obtained with glucose consumption by yeasts. Substances increasing the growth and glucose consumption by yeasts proved to be a low molecular weight fractions (<1,000) in water solubles not saponins. The production of n-propyl alcohol by yeast was enhanced by adding ginseng extracts into the media, but that of ism-butyl alcohol was suppressed at same condition.

  • 50개의 폴리프로필렌 실관과 3개의 테르폰 실관으로 구성된 실관반응기에서 Saccharomyces cerevisiae효모를 이용하여 알콜의 연속 생산을 연구하였다. 생산된$CO_2$는 테프론 실관으로 내어 보냈고 과잉효모 세포는 shell-side를 통하여 제거하였다. Shell-side 부피를 기준으로 세포농도는 266g/L였고 알콜 생산성은 205g/L를 얻었다. 질소 결핍배지를 사용했을 경우 생산성이 너무 낮아 실제 응용할 가치는 없었다.
