Small Molecules Targeting for ESX-Sur2 Proteins' Interaction

  • It's been known that overexpression of the oncoprotein Her2 (eu/ErbB2), transmembrane receptor protein, occurs in human breast cancer. Her2-positive breast cancer patients who have Her2 overexpression show less therapeutic efficacy with enhanced metathesis and increased resistance to chemotherapy. So far, a humanized monoclonal antibody against Her2 protein called Herceptin is the only drug approved by Food and Drug Administration for treatment of Her2-overexpressing breast tumors. However, antibody therapy of Herceptin may not be ideal method for therapeutic intervention of Her2 protein expression. The therapeutic intervention of Her2 protein expression may be more efficiently achieved by inhibiting the expression of Her2 gene rather than by down-regulating the Her2 protein already overexpressed. Here, we found that the interaction of two proteins of ESX (an epithelial-restricted transcription factor) and DRIP130/CRSP130/Sur2 (a Ras-linked subunit of human mediator complexes) mediates the expression of Her2 gene. The association of ESX with Sur2 is mediated by a small hydrophobic face of 8-amino acid helix in ESX, suggesting that the ESX-Sur2 interaction can be a new novel target for Her2-positive cancer. The process to develop potent ESX-Sur2 interaction inhibitors targeting for Her2-positive cancer therapeutics will be discussed.

A Study on Clinical Diagnosis of Temporomandibular Joint Disorders Using Bone Scan (골스캔을 이용한 측두하악관절장애의 임상진단에 관한 연구)

  • The author examined the clinical signs and symptoms , routine radiographs, laboratory test and bone scan in 73 subjects with TMJ disorders and studied the responses of clinical test on patients, the distribution of signs and symptoms of joints, the simple uptake rate(SUR) of affected joints , the SUR of subclassified groups of TMJ disorders, active joints of subclassified groups of TMJ disorders and the SUR of joints with noises. The obtained results were as follows : 1. The percentage of joint pain on palpation, joint noises joint pain on function, mandibular dysfunction and active finding of bone scan in aptients with TMJ disorders were higher 2. The SUR was higher in joints affected by joint pain on function, joint pain on palpation, mandibular dysfunction and creptius. 3. The SUR of osteoarthrosis was the lowest. 4. The percentage of active joints were the highest in joints with discdisplacement without reduction, and followed by osteoarthritis, and disc displacement with reduction. 5. The SUR of TMJ showing joint noises only was lower.

The Evaluation of Bone Scan and SPECT Before and After Treatment of TMD Patients (골관절염을 가진 측두하악장애 환자의 치료 전, 후 골스캔과 SPECT의 평가)

  • The aim of this study was to evaluate bone scan(SUR) and SPECT(hot spot) in patients with TMJ osteoarthritis. 99mTc-MDP bone scan and SPECT were done in 27 patients, total 54 temporomandibular joints were examined with clinical examination, plain radiography. 42 TMJs were osteoarthritis and 12 TMJs were normal case, clinically diagnosed. We compared osteoarthritis group and control group of bone scan(SUR) and SPECT(hot spot). thus we compared before and after treatment of bone scan(SUR) and bone SPECT(hot spot) according to history, contributing factor, symptom degree, treatment, treatment period. The obtained results were as follows. 1. Bone scan(SUR) and SPECT(hot spot) activity of osteoarthritis group were higher than control group(P<0.05, P<0.01). 2. Bone scan(SUR) and SPECT(hot spot) activity were decreased after treatment at TMJ with osteoarthritis(P<0.01). 3. Bone scan(SUR) and SPECT(hot spot) activity were decreased at treatment group with splint(P<0.01, P<0.05). 4. Bone scan(SUR) and SPECT(hot spot) activity were decreased at Chronic group(P<0.01, P<0.05). 5. Bone scan(SUR) and SPECT(hot spot) activity were decreased at low noise NAS(<6) group(P<0.01,P<0.05). 6. Bone scan(SUR) and SPECT(hot spot) activity were decreased at TMD patient with parafunctional habit and without trauma history, psychosocial factor. 7. Bone scan(SUR) and SPECT(hot spot) activity were higher at before treatment required more treatment period.

Expression of ATP-sensitive Potassium Channel and Sulfonylurea Receptor in Neonate and Adult Rat Tissues

  • The ATP-sensitive potassium $(K_{ATP})$) channel is a member of inward rectifier potassium channel (Kir) that is inhibited by intracellular ATP and functions in close relation to sulfonylurea receptors (SUR). Although the molecular mechanism and physiological function of $K_{ATP}$ channels are well understood, the expression pattern during development or treatment with the channel modulators such as glybenclamide is little known. In this work, we determined mRNA levels of a $K_{ATP}$ channel (Kir6.2) and a sulfonylurea receptor (SUR2) in rat tissues by RNase protection assay. Levels of Kir6.2 and SUR2 mRNA in the rat brain and skeletal muscle were higher in adult $(90{\sim}120\;days)$ than in neonate $(2{\sim}8\;days),$ whereas those in the heart were not much different between neonate $(2{\sim}8\;days)$ and adult $(90{\sim}120\;days).$ In addition, none of $K_{ATP}$ channel modulators (opener, pinacidil and nicorandil; blocker, glybenclamide) affected the Kir6.2 mRNA levels in the heart, brain and skeletal muscle. The results indicate that the expression of Kir and SUR genes can vary age-dependently, but the expression of Kir is not dependent on the long-term treatment of channel modulators. The effect of the channel modulators on mRNA level of SUR is remained to be studied further.

Acute abdomen following COVID-19 vaccination: a systematic review

  • Purpose: Conduct a systematic review of case reports and case series regarding the development of acute abdomen following coronavirus disease 2019 (COVID-19) vaccination, to describe the possible association and the clinical and demographic characteristics in detail. Materials and Methods: This study included case report studies and case series that focused on the development of acute abdomen following COVID-19 vaccination. Systematic review studies, literature, letters to the editor, brief comments, and so forth were excluded. PubMed, Scopus, EMBASE, and Web of Science databases were searched until June 15, 2023. The Joanna Briggs Institute tool was used to assess the risk of bias and the quality of the study. Descriptive data were presented as frequency, median, mean, and standard deviation. Results: Seventeen clinical case studies were identified, evaluating 17 patients with acute abdomen associated with COVID-19 vaccination, which included acute appendicitis (n=3), acute pancreatitis (n=9), diverticulitis (n=1), cholecystitis (n=2), and colitis (n=2). The COVID-19 vaccine most commonly linked to acute abdomen was Pfizer-BioNTech (messenger RNA), accounting for 64.71% of cases. Acute abdomen predominantly occurred after the first vaccine dose (52.94%). All patients responded objectively to medical (88.34%) and surgical (11.76%) treatment and were discharged within a few weeks. No cases of death were reported. Conclusion: Acute abdomen is a rare complication of great interest in the medical and surgical practice of COVID-19 vaccination. Our study is based on a small sample of patients; therefore, it is recommended to conduct future observational studies to fully elucidate the underlying mechanisms of this association.

세수추계모형의 예측력 비교

  • 본 연구는 세입증가율 예측을 위해 사용되는 각종 세수추계모형의 예측능력을 상호비교하는 데 목적이 있다. 본 연구에서 고려하는 세수추계 방식은 네 가지이다. 첫째는 단순 ECM 모형으로서 오차수정모형(error correction model)을 각각의 세목에 적용하여 세수를 예측하는 것이다. 둘째는 SUR-ECM 모형으로서 단순 ECM 모형의 개별 회귀방정식을 통합하여 SUR(Seemingly Unrelated Regression) 방식으로 추정한 후 이를 이용하여 세수를 예측하는 것이다. 셋째와 넷째는 흔히 사용되는 탄성치 방식으로서, 과거의 연도별 탄성치를 5년간 또는 10년간 평균하여 이를 바탕으로 향후의 세수를 예측하는 것이다. 이러한 모형비교를 통해 얻은 결과는 다음과 같이 요약될 수 있다. 첫째, 단순 ECM 모형과 5년 평균 탄성치 모형은 예측력에 있어 큰 차이가 없다. 둘째, SUR-ECM 모형과 10년 평균 탄성치 모형은 예측력에 있어 큰 차이가 없다. 셋째, 단순 ECM 모형보다는 SUR-ECM 모형의 예측력이 높으며, 5년 평균 탄성치 모형보다는 10년 평균 탄성치 모형의 예측력이 높다. 넷째, 어느 경우에든 예측 오차가 상당히 크고 이러한 오차는 예측시계가 넓어질수록 커진다. 예를 들어, 5년 후의 세수에 대한 예측치는 평균적으로 오차의 절대값이 10% 수준에 이른다. 탄성치 모형이 단순 ECM 모형이나 SUR-ECM 모형에 비해 그리 나쁜 예측결과를 낳지 않는다는 것은 새로운 사실이다. 또한 5년 평균 탄성치보다 10년 평균 탄성치를 사용하는 것이 더 나은 예측치를 낳는다는 것은 세수예측에 있어 최근의 자료만을 사용하는 것보다는 과거 꽤 오랜 기간의 자료를 사용하는 것이 바람직하다는 점을 시사한다.

Genotoxic Effects on Gas Station Attendants in South-southeastern México due to Prolonged and Chronic Exposure to Gasoline

  • Background: Gasoline, a complex mixture of volatile organic compounds is classified as possibly carcinogenic to humans. Gasoline station attendants, consistently exposed to its hazardous components, may face genotoxic effects. This study aimed to assess the influence of varying work shift durations on DNA damage in gasoline station attendants. Methods: Ninety individuals from three locations in southern México were studied. Peripheral blood mononuclear cells (PBMCs) were isolated, and DNA damage was assessed using the comet assay. Demographic, occupational, and lifestyle data were collected. Statistical analyses included t-tests, ANOVA, and Pearson correlation. Results: Significant differences in DNA damage parameters were observed between exposed and unexposed groups. The impact of tobacco, alcohol, and exercise on DNA damage was negligible. Extended work shifts (12 and 24 hours) showed heightened DNA damage compared to 8-hour shifts and the unexposed group. A novel finding revealed a modest but significant correlation between DNA damage and job seniority. Conclusion: The study highlights the intricate relationship between occupational exposure to gasoline components, DNA damage, and work shift lengths. Extended shifts correlate with heightened genotoxic effects, emphasizing the importance of personalized safety measures. The significant correlation between DNA damage and job seniority introduces occupational longevity as a determinant in the genetic health of gasoline station attendants. This discovery has implications for implementing targeted interventions and preventive strategies to safeguard workers' genetic integrity throughout their years of service. The study calls for further exploration of unconsidered factors in understanding the multifactorial nature of DNA damage in this occupational setting.
