Antidiabetic activity of Argyreia speciosa (sweet) (Burm.f.)Boj. in normoglycemic and Streptozotocin-induced diabetic rats

  • Effect of ethanol (ASE) and water (ASW) extracts of Argyreia speciosa on blood glucose and lipid profile was investigated in normoglycemic and Streptozotocin (STZ)-induced diabetic animals. In oral glucose and sucrose tolerance test, treatment with ASE and ASW (100 and 200 mg/kg) and Glidenclamide (10 mg/kg) significantly improved the glucose and sucrose tolerance in normal animals. In addition, respective treatment for fifteen-day resulted in significant percentage reduction in serum glucose (SG) ie., 30.39% (lower dose of ASE) and 33.21% (higher dose of ASW). In standardized STZ (50 mg/kg, iv)-induced diabetic rats, a single dose of ASE and ASW treatment exhibited reduction in SG levels at different time intervals compared to basal levels. Administration of both the doses of ASE and ASW for fifteen-day days exhibited greater percentage reduction in glycemia (24.6%, 24.7%, 23.9% and 21.9% respectively) and also ameliorated restored to near normal value of all tested lipid parameters. Further, treatment also exhibited significantly improved glucose tolerance over the period of 120 min compared to diabetic control group. Eventhough treatment failed to increase serum insulin levels significantly but peripheral utilization of insulin was increased as evident by insulin tolerance test. Taken together, present study supports the traditional usage of title plant in the treatment of diabetes mellitus.

Cilostazol ameliorates diabetic nephropathy by inhibiting high-glucose-induced apoptosis

  • Diabetic nephropathy (DN) is a hyperglycemia-induced progressive development of renal insufficiency. Excessive glucose can increase mitochondrial reactive oxygen species (ROS) and induce cell damage, causing mitochondrial dysfunction. Our previous study indicated that cilostazol (CTZ) can reduce ROS levels and decelerate DN progression in streptozotocin (STZ)-induced type 1 diabetes. This study investigated the potential mechanisms of CTZ in rats with DN and in high glucose-treated mesangial cells. Male Sprague-Dawley rats were fed 5 mg/kg/day of CTZ after developing STZ-induced diabetes mellitus. Electron microscopy revealed that CTZ reduced the thickness of the glomerular basement membrane and improved mitochondrial morphology in mesangial cells of diabetic kidney. CTZ treatment reduced excessive kidney mitochondrial DNA copy numbers induced by hyperglycemia and interacted with the intrinsic pathway for regulating cell apoptosis as an antiapoptotic mechanism. In high-glucose-treated mesangial cells, CTZ reduced ROS production, altered the apoptotic status, and down-regulated transforming growth factor beta (TGF-β) and nuclear factor kappa light chain enhancer of activated B cells (NF-κB). Base on the results of our previous and current studies, CTZ deceleration of hyperglycemia-induced DN is attributable to ROS reduction and thereby maintenance of the mitochondrial function and reduction in TGF-β and NF-κB levels.

Effect of Dietary Vitamin E on the Microsomal Mixed Function Oxidase System of Liver and Lung in Streptozotocin-induced Diabetic Rats (식이 Vitamin E가 Streptozotocin 유발 당뇨쥐의 간 및 폐조직에서의 Microsomal Mixed Function Oxidase System에 미치는 영향)

  • The purpose of this study was to investigate the effect of dietary vitamin E on microsomal mixed function oxidase system of liver and lung in streptozotocin(STZ) induced diabetic rats. Sprague-Dawley male rats weighing 140 $\pm$ 10mg were randomly assigned to one control and three STZ-diabetic groups. Diabetic groups were divided into DM-0E(vitamin E free diet), DM-40E(40mg vitamin E kg/diet) and DM-400E(400mg vitamin E kg/diet) according to the level of vitamin E supplementation. Diabetes was experimentally induced by intravenous administration of 55mg/kg b.w of STZ in citrate buffer(pH 4.3) after 4 week feeding of three experimental diets. Animals were sacrificed at the 6th day of diabetic state. The contents of cytochrome P$_{450}$ in DM-0E, DM-40E and DM-400E groups of liver were increased by 162%, 150% and 56%, respectively, compared with that of control. Also the contents of cytochrome P$_{450}$ in lung were similar to liver. The activities of cytochrome bs in DM-0E and DM-40E groups of the liver were increased by 70% and 53%, respectively, compared with that of control, but not in DM-400E group. The activities of bs in DM-0E, DM-40E and DM-400E groups of lung were signficantly increased. Activity of cytochrome P$_{450}$ reductase in DM-0E, DM-40E of liver and lung were higher than that of control group, but the activity of DM-400E group was not different from that of control. The lipid peroxide values of DM-0E, DM-40E and DM-400E groups were 143%, 95% and 31% higher than those of control. It was concluded that dietary vitamin E had protective effects on lipid peroxidation accompanied with increased mixed function of oxidase activity in diabetic rats.

Antioxidant Effects of the Mixture of Mulberry Leaves and Silkworm Powder on the Plasma and Liver in Streptozotocin-Induced Diabetic Rats

  • This study was carried out to examine the antioxidant effects of a mixture of mulberry leaves and silkworm powder in plasma and liver of streptozotocin-induced diabetic rats. Sprague-Dawley male rats weighing 100$\pm$10 g were used and their diets were supplemented with $0.4\%$ (4 g/kg) of the mixtures. Experimental groups were diabetic rats without supplements (DM group) or with a combination of the supplements: $100\%$ mulberry leaves (M group), $25\%$ silkworm powder mixed with mulberry leaves (25SM group), $50\%$ silkworm powder mixed with mulberry leaves (50SM group), $75\%$ silkworm powder mixed with mulberry leaves (75SM group) or $100\%$ silkworm powder (100S group). The rats were fed experimental diets and water ad libitum. All animals were injected with streptozotocin at the $3^{rd}$ week for inducing diabetes and were sacrificed on $9^{th}$ day thereafter. Hepatic xanthine oxidase (XOD) activity significantly decreased in the mixture supplemented groups compared to the DM group. Hepatic superoxide dismutase (SOD) activity was not significantly different among any of the experimental groups, but glutathione peroxidase (GSH-px) activity increased in the mixture supplemented groups compared to the DM group. In particular, it was the highest in the 50SM group. The hepatic TBARS values were lower in all the mixture supplemented groups than in the DM group, and it was as lowest when ratio of mulberry leaves to silkworm powder was highest. Hepatic lipofuscin contents were similar with the TBARS value. In conclusion, the mixtures containing silkworm powder reduced oxidative damage by strengtbening the antioxidative system and suppressing oxidative stress in the STZ-induced diabetic rat. The 1:1 blend of silkworm powder and mulberry leaves was the most effective combination for antioxidant activity.

Microsomal Mixed Function Oxidase and Lipid Peroxidation in Liver and Lung of Sterptozotocin-Induced Diabetic Rats (Streptozotocin 유발 당뇨쥐의 간장 및 폐조직에서의 Microsomal Mixed Function Oxidase System과 과산화지질 생성)

  • 본 연구는 streptozotocin 유발 당뇨쥐에서의 MFO계활성 변화 및 이에 지질과산화를 관찰하고자 체중이 140kg 내외의 Sprague-Dawley 종 흰쥐 숫컷을 대조군과 당뇨 유발 시험군으로 나누어 4주간 사육하였다. 당뇨군은 STZ로 당뇨를 유발시켰으며 당뇨 유발 6일후 쥐를 희생기켜 간조직 및 폐조직 microsome 중의 cytochrome $P_{450}$ 및 cytochrome $b_[5}$ 함량과 NADPH-cytochrome $P_{450}$ reductase 활성도를 측정하고 아울러 microsome내의 지질과산화물을 측정하여 다음과 같은 결과를 얻었다. 실험군간에 식이 섭취량은 차이가 없었으나, 체중 증가량과 식이 효율은 당뇨군이 STZ군에 비해 현저하게 감소하였다. 간장 및 폐조직의 무게는 대조군과 당뇨군간에 유의적인 차이는 없었다. 간조직 및 GP조직 중의 cytochrome $P_{450}$ 함량은 대조군에 비해 당뇨군이 150%, 175%씩 증가하였다. 간조직 및 폐조직 중의 cytochrome $b_{5}$은 대조군에 비해 당뇨군이 53%,116%씩 각각 증가하였다. 간조직에서의 NADPH-cytochrome $P_{450}$ reductase 활성은 당뇨군이 대조군에 비해 46% 증가하였으며, 폐조직에서는 75%증가하였다. 지질과산화물가는 당뇨군이 대조군에 비해 간족에서는 약 95% 높았으며 폐조직에서는 73%높았다. 이상의 결과에서 STZ 유발 당뇨쥐에서는 MFO system의 활성도가 대조군에 비해 현저하게 증가되고 지질과산화반응이 같으 srudgid이 촉진되었으며 폐조직에서도 간조직에서와 비슷한 경향이었다. 이러한 것은 당뇨군에서는 MFO system의 활성증가로 free radical 생성이 증가되고 그 결과 지질과산화가 촉진되었다고 볼 수 있다.

Effects of SP3 and GV6 Acupuncture on the STZ-treated Rats for Induction of Diabetes (척중 및 태백의 침자가 Streptozotocin(STZ)으로 유발된 고혈당 생쥐에 미치는 영향)

  • This study has been carried out to understand the effect of SP3 and GV6 acupuncture on the hyperglycemic rat induced with Streptozotocin (STZ). Diabetes was induced in experimental groups by inraperitoneal injection of STZ (50mg/kg of body weight) twice by 24 h interval and the additional 100mg/kg 3 days after the earlier treatment. Control group was treated with tail-non acupoint, and experimental groups were treated SP3, GV6 and SP3+GV6 after hyperglycemic induction for 6 weeks. The body weight of control was lower than the experimental groups. The blood glucose was decreased significantly in the experimental groups. Glucose tolerance in acupuncture treated groups was improved. Blood cholesterol level and transaminase activites were lower in the experimental groups than in the control. In the SP3 or GV6 treated rats, hepatocyte degeneration were apparently decreased and the organelles were properly arranged. Furthermore, decrease in liver IGF-I mRNA expression was improved by the acupuncture in STZ-induced diabetic rats. In conclusion, our observation indicate that SP3 or GV6 acupoints treatment can exert beneficial effects in diabetes, with preservation of ${\beta}-cell$ function and liver function.

The Effects of Chungkookjang Biopolymer on Blood Glucose and Serum Lipid Lowering in Streptozotocin-Induced Diabetic Rats (청국장 점질성 중합체의 첨가 식이가 Streptozotocin 유발 당뇨쥐의 혈당 및 혈청지질 감소에 미치는 영향)

  • The purpose of this study was to investigate the effects of Chungkookjang viscous biopolymer supplementation on blood glucose and serum lipid-lowering in Streptozotocin (STZ)-induced diabetic rats. The rats were divided into three experimental groups; a normal group (N), diabetic control group (D), and a diabetic group with the supplementation of Chungkookjang viscous biopolymer (DCB). The groups were given experimental diets for four weeks. The normal group (C) was fed a casein-based diet, and the Chungkookjang viscous biopolymer group (DCB) received 3% biopolymer added to the casein-based diet. In the diabetic group (D), food intake increased significantly, but weight gain decreased significantly. The food efficiency ratio was significantly lower in the diabetic group. Liver weight increased significantly in the D group as compared to the N group. However, the DCB group showed a significant decrease in liver weight when compared to the D group. Blood glucose decreased significantly in the DCB group after receiving the experimental diet for four weeks, as compared to the diabetic control (p<0.05). Serum triglyceride levels were significantly lower in the DCB group than in the D group, whereas HDL-cholesterol levels were significantly higher (p<0.05). Total cholesterol was decreased in DCB group, but there were no significant differences. Also, LDL-cholesterol level was not significantly different among the experimental groups. Hepatic triglyceride and cholesterol were lower in the DCB group with no significant differences among groups. These results indicate that dietary supplementation of Chungkookjang viscous biopolymer improved glucose lowering and lipid metabolism in diabetic rats.

Effects of Opuntia ficus-indica Complexes B(OCB) on Blood Glucose and Lipid Metabolism in Streptozotocin-induced Diabetic Rats (손바닥선인장 복합물이 당뇨 쥐의 혈당 및 지질대사에 미치는 영향)

  • This study was conducted to examine the effects of Opuntia ficus-indica complexes(OCB) on the intake of water and food, body weight, blood glucose levels, glucose tolerance and lipid metabolism in streptozotocin(STZ)-induced diabetic rats. Thirty-two male Sprague-Dawley rats were assigned to four different groups; non-diabetic control(NC), diabetic control (DC), diabetic OCB of 2%(OCB-2), and diabetic OCB of 5%(OCB-5). The animals were fed on each experimental diet for 3 weeks. The DC, OCB-2 and OCB-5 groups showed a higher intake of water and food than the NC group. The fasting blood glucose levels were 100 $ mg/d{\ell}$ and 416 $ mg/d{\ell}$ for the NC and DC groups, respectively. The OCB-5 group presented a significantly low fasting glucose level of 21%(P<0.05), while OCB-2 group had a decrease of 13% compared to the DC group. As for the results of the glucose tolerance test, the highest blood glucose level was observed for all the groups at 30 minutes after the glucose injections as well as higher plasma insulin levels in the OCB-5 group. Plasma total cholesterol, triglyceride, non-esterified fatty acids(NEFA) and LDL-cholesterol concentrations were also lower in the OCB-2 and OCB-5 groups. The experimental diet did not affect the HDL-cholesterol levels. The overall results suggest that the higher intake of food by the OCB-2 and OCB-5 groups improved the blood glucose levels and lipid metabolism in STZ-induced diabetic rats.

The Immunological Study on Expression of the Clusterin in Sterin in Streptozotocin-Induced Diabetic Rats (당뇨시 백서 타액선 조직내이 clusterin변화에 관한 면역학적 연구)

  • Clusterin is a highly glycosylated protein composed of two disulfide linked subunits. Although its biolobical action is not clearly defined, clusterin seems to be involved not only in remodeling of damaged tissue, but also in production of halitosis, the present study was designated to elucidate the expression of clusterin in the salivary gland of diabetic rats. For this study, 24 Sprague-Dawley rats were used for the experiment and divided into 2groups: control and experimental. The experimental group was composed of 18 rats and the control goup was 6 rats. After nduction of diabetes by STZ injection, the animals were sacrificed at 1,3,5,7,10,14 days. The parotid and submamndibular glands were observed histologically and the transcriptional expression of clusterin in the glands by Northern blot. The finding were as follows : 1. In experimental group, the salivary glands were observed at day 3 and then a seven destructive pattern was found in the glands at day 5. Howere, regeneration of gland tissue occured at day 14. 2. In experimental goup, destructive change was examined in the septal connective tissue after 7 days, and gradually more serious. 3. In experimental group, clusterin was expressed in the submandibular glands after 5 days, but in parotid glands to a lesser extent after 10 days. These results suggest that clusterin seems to be closely associated with histologic changes in the mucous glands rather serous glands.

Antihyperglycemia Effect of Medicinal Plants Mixture in Streptozotocin-Induced Diabetic Rats (Streptozotocin 유발 당뇨쥐에서 항당뇨 생약 복합물의 혈당강하 효과)

  • This study was performed to investigate the hypoglycemic effect of single and repeated oral administration of medicinal herbal mixture (AD) in streptozotocin (STZ) induced diabetic rats. Angelica decursiva, Lycium chinense and Adenophora triphylla var. japonica Hara were selected by oral glucose tolerance test (OGTT) and mixed for AD mixture. In an oral glucose tolerance test, the AD inhibited the increase in blood glucose levels at 1 hr and 2 hr and decreased incremental glycemic response area under the curve. In a single administration of AD1 (100 mg/kg) and AD2 (500 mg/kg), significant reductions by 5.3% and 12.3% were observed in fasting blood glucose level for 4 hours. During the 1 month of the experimental period, AD1 and AD2 was given to the STZ induced diabetic rats. At 4th week, the fasting blood glucose levels of AD1 and AD2 caused a fall of 25.5% and 37.9%, respectively. In addition, the body weights were decreased by 7.7% (AD1) and 1.7% (AD2), respectively, compared with diabetic control (DC, decreasing of 10.2%). This study suggests that AD could be potentially useful for fasting and post-prandial hyperglycemia treatment and all these effects concluded to the use of this plant extract to manage diabetes mellitus.
