  • Different methodologies in hair cortisol extraction may alter the final output. Thus finding the standard methodology according to a laboratory facilities is pivotal. This study was carried out to validate the feasibility of two methods of grinding hair for cortisol extraction in Korean native (Hanwoo) cattle. Hair from nine cattle including mature cows, heifers, and calves were assigned to one of the following methods for grinding hair; 1) using bead beater (BB) and 2) using surgical scissors (SS). Hair samples (> 1 g) were harvested from forehead of each individual twice (first and second measurement) to validate the results. To improve the accuracy of the obtained data, each sample was duplicated into two wells during enzyme immunoassay (EIA) analysis. Overall comparison of hair cortisol concentration (HCC) showed that the data within the range (out of the range) of standards provided by the EIA kit were 88.9% (11.1%) and 66.7% (33.3%) for BB compared with SS, respectively. In the first measurement, application of BB was tended to show higher (p = 0.056) amount of HCC compared with SS. In the second measurement application of BB showed higher (p = 0.0028) amount of HCC compared with SS. Among the cattle, calves showed higher HCC using BB compared with SS (p < 0.05). Application of BB in hair grinding methodology for Hanwoo cattle may improve cortisol extraction in comparison to application of SS method, with more consistency. Thus, it would be the preferable method to use.

  • Investigated the processing characteristics of the pollutants and runoff due to storm events in the actual application of the road fields and a Non-Point Sources (NPS) pilot scale equipment. This phenomenon has occurred in the influent bypass the blockage occurs after 90 min the expended polymeric media was filled with filtered column. When entering a treatment tank SS 200 mg/L or more high concentration of effluent treatment efficiency was reduced from the reaction time 60 min. Influent concentration less then SS 180 mg/L was stable handling. The $COD_{Cr}/SS$ ratio were analyzed with 0.67, median value. Showed 92.1% and 82.3% respectively with an average removal rate of the SS and the $COD_{Cr}$. If the influent concentration of TP is the 0.5 mg/L or less, the quality of the treated water is 0.1 mg/L levels were expressed in a stable process. And when entering the 1.0 mg/L or more of the treated water, had a greater than average 0.2 mg/L. If the influent concentration of TN is 4~10 mg/L, the treatment water quality level was kept a 1.5~3.0 mg/L. The average removal efficiency of TP and TN respectively 73.9%, 50.4%.

  • Environmental polluting materials from road surface drainage are a significant nonpoint source influenced to the eutrophication of lake and ecosystems with a transport development in recent years. To elucidate the discharge characteristics, the changing patterns in concentrations of polluting materials such as suspended solid (SS), chemical oxygen demand (COD), nitrogenous and phosphorus nutrients in drainage waters, were investigated during rainfall. Load variation of COD concentration in drainage water samples was closely related to that of SS concentration. This indicates that SS contained a greater part of organic matter. A quite difference between the past pavement and the new well-drainage pavement system was observed in the concentrations of SS and COD in drainage waters. Appreciable concentrations of nitrite and nitrate were determined in drainage waters. The present results indicate that the drainage water from road surfaces is a significant nonpoint source, and that the well-drainage pavement system introduced to skid prevention has an effect on the decreases of pollutants.

  • Corrosion inhibition of 304 stainless steel (SS) in 1 M HCl by aqueous extract of coriander seeds was studied using weight loss, potentiodynamic polarization, electrochemical impedance spectroscopy (EIS) and electrochemical frequency modulation (EFM) techniques. Values of inhibition efficiency obtained are dependent upon the concentration of extract and temperature. Generally, inhibition was found to increase with inhibitor concentration, but decrease with temperature. Physical adsorption mechanism has been proposed for the inhibition with Langmuir adsorption isotherm obeyed. Values of activation energy of the inhibited corrosion reaction of 304 SS are greater than the value obtained for the blank. Thermodynamic consideration reveals that adsorption of aqueous extract of coriander seeds 304 SS surface is spontaneous.

  • There are two types of rectangular secondary clarifier at biological nutrient removal (BNR) facility to settle MLSS; conventional activated sludge secondary clarifier and Gould Type I clarifier. In this study, the performances of two types at respective biological nutrient removal facility are compared using weekly operational data. Surface Overflow Rate (SOR), Surface Loading Rate (SLR), Sludge Volume Index (SVI), secondary effluent SS concentration are studied. It has found that Gould Type I has 3.5 times less average secondary effluent SS concentration that is 2.4 mg/L than that of conventional activated sludge secondary clarifier. Both SOR and SLR have shown little effect on secondary effluent SS concentrations at Gould Type I clarifier in contrary that SOR affects the secondary effluent SS concentrations at conventional activated sludge rectangular secondary clarifier. From this study, it is recommended that Gould Type I must be considered for secondary clarifier when BNR plant is designed.

  • Researchers developed Sediment Trap Drain Channel (STDC) as a solution of the reduction of soil erosion and muddy runoff from a alpine field. The STDC is the one that can take a role of grit chamber by installing the shield made of woods in the concrete channel. The study was conducted 8 kinds of stages according to the amount of soil loss and the inflow. Evaluation factors were ss concentration, turbidity and reduced soil. The results of study showed lessness of ss concentration and turbidity from the lower spot than the upper spot. The average reduction rate of ss concentration was 74 % and the average reduction rate of turbidity was 62 %. It was turned out that the performance related soil loss and muddy runoff of the STDC is effective. The governance was needed to expect the effectiveness of the STDC.

  • BACKGROUND: The Doam watershed in Korea has been managed for the reduction and the prevention of non-point source pollution since 2007. Especially, the water quality of the Doam reservoir is a primary issue related to the Doam dam reoperation. We have carried out the modeling to evaluate the water quality based on suspended solids (SS) of the Doam watershed and the Doam reservoir. Two powerful hydrological and water quality models (HSPF and CE-QUAL-W2) were employed to simulate the combined processes of water quantity and quality both in the upland watershed of the Doam reservoir and the downstream waterbody. METHODS AND RESULTS: The HSPF model was calibrated and validated for streamflow and SS. The CE-QUAL-W2 was calibrated for water level, water temperature, and SS and was validated for the only water level owing to data lack. With the parameters obtained through the appropriate calibration, SS concentrations of inflow into and in the Doam reservoir were simulated for three years (2008, 2004 and 1998) of the minimum, the average, and the maximum of total annual precipitation during recent 30 years. The annual average SS concentrations of the inflow for 2008, 2004, and 1998 were 8.6, 10.9, and 18.4 mg/L, respectively and those in the Doam reservoir were 9.2, 13.8, and 21.5 mg/L. CONCLOUSION(s): The results showed that more intense and frequent precipitation would cause higher SS concentration and longer SS's retention in the reservoir. The HSPF and the CE-QUAL-W2 models could represent reasonably the SS from the Doam watershed and in the Doam reservoir.

  • Saponins are glycosidic compounds present in many plant foods. They are characterized by their ability to lyse cell membranes due to their surface-active properties. Saponins are believed to interact primarily with cholesterol in the cell membrane. In this study, the interaction of soybean(SS) with cell membrane was investigated using erythrocytes as a model. Mechanisms of interaction was also investigated by measuring their binding capacity with different membrane lipid fractions. Throughout the study, gypsophilla saponin(GS) and quillaja saponin(QS) were used to evaluate the membranolytic activity of soybean saponins. All saponins released hemoglobin in a concentration-dependent manner. SS induced 40% hemolysis at the concentration of 400 ppm, however there was no increase in hemoglobin release above 400ppm concentration. 5ppm of GS and 8 ppm of QS hemolyzed 100% of erythrocytes. Isolation of SS fractions by thin layer chromatography revealed that only one non-polar saponin possesses strong hemolytic activity. When saponins were incubated decreased the release of cholesterol. When the hemolytic activity of saponins was measured in the presence of other major membrane lipid components, sphingomyelin significantly reduced the hemolytic activity of SS, while cholesterol reduced the activity of QS. GS showed high affinity to other component(s) in the incubation media as well as lipids. These results suggest that the membranolytic activity of saponins are related to their specific chemical structure, which determines the interaction behavior between saponins and different membrane components, and thereby influence the biological activity.

  • We studied runoff characteristics of combined sewer overflows in a city while it was raining. The event mean concentration (EMC) of biochemical oxygen demand (BOD), chemical oxygen demand ($COD_{Cr}$), suspended solids (SS), total nitrogen (TN), and total phosphorus (TP) in one of the combined sewer sites in Chuncheon was 63.5-211.6 mg/L, 114.9-523.8 mg/L, 70.3-436.4 mg/L, 6.4-33.0 mg/L, and 1.09-6.81 mg/L, respectively. In another combined sewer, the EMC of BOD, COD, SS, TN, and TP was 42.1-131.4 mg/L, 107.7-256.5 mg/L, 33.7-221.1 mg/L, 7.9-26.4 mg/L, and 1.16-3.91 mg/L, respectively. The ratio of the cumulative pollutant mass and the cumulative discharged volume determined using all parameters (BOD, $COD_{Cr}$, SS, TN, and TP) was over 1.0, which shows the first flush effect. Relationships between flow and loadings of BOD, $COD_{Cr}$, SS, TN, and TP were 0.90, 0.89, 0.88, 0.89, 0.92, respectively. Although the size of two areas was almost same, pollutant concentration and loading were different because of the amount of rainfall, rainfall intensity and basin area.

  • The effects of activated carbon originated Ballasted Flocculant (BF) on the settleability of activated sludge and the recovery of BF by Hydro-cyclone (HC) were analyzed experimentally. Two kinds of BF (M-I: 125-250 ㎛, M-II: 250-425 ㎛ in dia.) and three kinds of activated sludges with different SS concentration (2,300-7,100 mg/L) were applied for this study. With the dosage variation of BF from 0.14 to 1.3 g-BF/g-SS, we could obtain 24-31% improvement in SV30 (Sludge Volume after 30min sedimentation) for the lowest SS concentration sludge (2,300 mg/L). Whereas the SV30 improvement was much higher as 44-48% for the highest SS concentration sludge (7,100 mg/L). The settling characteristics of the sludge with BF followed Vesilind model the best among three models (Vesilind, Takacs and Cho model). HC could effectively separate BF with the separation efficiency of 70-90% and over 95% separation efficiency could be obtained when the HC was applied twice.
