(E)-3-(3-methoxyphenyl)-1-(2-pyrrolyl)-2-propenone (MPP) is an aldol condensation product resulting from pyrrole-2-carbaldehyde and m- and p- substituted acetophenones. However, its biological activity has not yet been evaluated. Since it has been reported that some propenone-type compounds display anti-inflammatory activity, we investigated whether MPP could negatively modulate inflammatory responses. To do this, we employed lipopolysaccharide (LPS)-stimulated macrophage-like RAW264.7 cells and examined the inhibitory levels of nitric oxide (NO) production and transcriptional activation, as well as the target proteins involved in the inflammatory signaling cascade. Interestingly, MPP was found to reduce the production of NO in LPS-treated RAW264.7 cells, without causing cytotoxicity. Moreover, this compound suppressed the mRNA levels of inflammatory genes, such as inducible NO synthase (iNOS) and tumor necrosis factor (TNF)-${\alpha}$. Using luciferase reporter gene assays performed in HEK293 cells and immunoblotting analysis with nuclear protein fractions, we determined that MPP reduced the transcriptional activation of nuclear factor (NF)-${\kappa}B$. Furthermore, the activation of a series of upstream signals for NF-${\kappa}B$ activation, composed of Src, Syk, Akt, and $I{\kappa}B{\alpha}$, were also blocked by this compound. It was confirmed that MPP was able to suppress autophosphorylation of overexpressed Src and Syk in HEK293 cells. Therefore, these results suggest that MPP can function as an anti-inflammatory drug with NF-${\kappa}B$ inhibitory properties via the suppression of Src and Syk.
In this paper, an analytical approach hwas been conductsed to clarify the relationships between the axial force and the deformation capacity of steel- encased reinforced- concrete beam-columns. The analytical model was defined as a cantilever. Several parameters influencing the inelastic performance of the beam-columns were selected, as follows: including encased steel area ratios, and sectional shapes of the encased steel, material strengths, and shear-span- to-depth ratios. The Analytical results of the analysis showed that the axial force had to have a maximum limit to ensure the stable behavior of a steel- encased reinforced- concrete beam-column when it was subjected to both axial and repeated lateral loading under a constant rotation angle amplitude. The maximum axial force of the beam-column to be resisted under cyclic lateral loading was defined as the stable-limit axial force to ensure the required rotation angle amplitude. The Analytical results of the analysis indicate that the stable-limit axial load ratio increases as the steel strength increases or as the compressive strength of the concrete decreases. The stable-limit axial load ratio decreases as the encased steel ' s sectional area increases in the case of a 1-shaped sections and it is almost not influenced by the steel sectional area in the case of a cross-shaped section.
Allulose, a monosaccharide isomer of fructose, was evaluated as a sucrose replacer for healthy cookie production with benefits such as low glycemic impact and low calorie content. Sucrose (as a reference), fructose, glucose, and allulose were used to explore the effects of sugar-replacer type on solvent retention capacity (SRC), differential scanning calorimetry (DSC), rapid visco-analyzer (RVA), and wire-cut cookie baking. SRC results indicated the lowest swelling of solvent-accessible arabinoxylans in allulose compared to that in other sugar solutions. DSC and RVA results showed retardation of starch gelatinization and onset of starch pasting, respectively, in the following order: water < allulose < fructose < glucose < sucrose. Among sugars, wire-cut cookies formulated with glucose showed the least desirable attributes with respect to cookie diameter and thickness. Although the baking response of allulose was slightly inferior to that of the sucrose control, the sugar exhibited a superior baking response to that of fructose, suggesting it could be used successfully as a fructose alternative or sucrose alternative for producing wire-cut cookies with reduced calorie content and low glycemic impact.
This paper presents an experimental study of three two-dimensional (2D/planar) steel reinforced concrete (SRC) T-shaped column-RC beam hybrid joints and six 3D SRC T-shaped column-steel beam hybrid joints under low cyclic reversed loads. Considering different categories of steel configuration types in column cross section and horizontal loading angles for the specimens were selected, and a reliable structural testing system for the spatial loading was employed in the tests. The load-displacement curves, carrying capacity, energy dissipation capacity, ductility and deformation characteristics of the test subassemblies were analyzed. Especially, the seismic performance discrepancies between planar hybrid joints and 3D hybrid joints were intensively compared. The failure modes for planar loading and spatial loading observed in the tests showed that the shear-diagonal compressive failure was the dominating failure mode for all the specimens. In addition, the 3D hybrid joints illustrated plumper hysteretic loops for the columns configured with solid-web steel, but a little more pinched hysteretic loops for the columns configured with T-shaped steel or channel-shaped steel, better energy dissipation capacity & ductility, and larger interlayer deformation capacity than those of the planar hybrid joints. Furthermore, it was revealed that the hysteretic loops for the specimens under $45^{\circ}$ loading angle are generally plumper than those for the specimens under $30^{\circ}$ loading angle. Finally, the effects of steel configuration type and loading angle on the seismic damage for the specimens were analyzed by means of the Park-Ang model.
Purpose: To identify the potential therapeutic role of postoperative radiotherapy (RT) in patients with locally advanced (stage II and stage III) gastric signet ring cell carcinoma (SRC). Materials and methods: Patients with locally advanced gastric SRC from the Surveillance, Epidemiology, and End Results program database between 2004 and 2012 were included in our study. Univariate and multivariate Cox proportional models were performed, and survival curves were generated to evaluate the prognostic effect of postoperative RT and surgery alone on SRC patients. Propensity score matching (PSM) was used to avoid selection bias among the study cohorts. Results: We found that patients with postoperative RT had better probability of survival compared with those who did not receive RT (overall survival [OS], P<0.001; cancer-specific survival [CSS], P<0.001). After PSM, analysis of both overall and CSS showed that patients who underwent postoperative RT had better prognosis than those receiving surgery alone in the matched cohort (OS, P=0.00079; CSS, P=0.0036). Multivariate Cox proportional model indicated that postoperative RT had better effect on prognosis compared with surgery alone with respect to both overall (hazard ratio [HR], 0.716; 95% confidence interval [95% CI], 0.590-0.87; P=0.001) and CSS (HR, 0.713; 95% CI, 0.570-0.890; P=0.003). Conclusions: Postoperative RT had better prognosis compared with surgery alone for both overall and CSS for patients with locally advanced gastric SRC.
This paper presents an investigation on the dynamic behavior of SRC columns with built-in cross-shaped steels under impact load. Seven 1/2 scaled SRC specimens were subjected to low-speed impact by a gravity drop hammer test system. Three main parameters, including the lateral impact height, the axial compression ratios and the stirrup spacing, were considered in the response analysis of the specimens. The failure mode, deformation, the absorbed energy of columns, as well as impact loads are discussed. The results are mainly characterized by bending-shear failure, meanwhile specimens can maintain an acceptable integrity. More than 33% of the input impact energy is dissipated, which demonstrates its excellent impact resistance. As the impact height increases, the flexural cracks and shear cracks observed on the surface of specimens were denser and wider. The recorded time-history of impact force and mid-span displacement confirmed the three stages of relative movement between the hammer and the column. Additionally, the displacements had a notable delay compared to the rapid changes observed in the measured impact load. The deflection of the mid-span did not exceed 5.90mm while the impact load reached peak value. The impact resistance of the specimen can be improved by proper design for stirrup ratios and increasing the axial load. However, the cracking and spalling of the concrete cover at the impact point was obvious with the increasing in stiffness.

Estrogen has diverse effects on cardiovascular function, including regulation of the contractile response to vasoactive substances such as serotonin. The serotonin system recently emerged as an important player in the regulation of vascular tone in humans. However, hyperreactivity to serotonin appears to be a critical factor for the pathophysiology of hypertension. In this study, we examined the modulatory mechanisms of estrogen in serotonin-induced vasoconstriction by using a combinatory approach of isometric tension measurements, molecular biology, and patch-clamp techniques. $17{\beta}$-Estradiol (E2) elicited a significant and concentration-dependent relaxation of serotonin-induced contraction in deendothelialized aortic strips isolated from male rats. E2 triggered a relaxation of serotonin-induced contraction even in the presence of tamoxifen, an estrogen receptor antagonist, suggesting that E2-induced changes are not mediated by estrogen receptor. Patch-clamp studies in rat arterial myocytes showed that E2 prevented Kv channel inhibition induced by serotonin. Serotonin increased Src activation in arterial smooth muscle required for contraction, which was significantly inhibited by E2. The estrogen receptor-independent inhibition of Src by E2 was confirmed in HEK293T cells that do not express estrogen receptor. Taken together, these results suggest that estrogen exerts vasodilatory effects on serotonin-precontracted arteries via Src, implying a critical role for estrogen in the prevention of vascular hyperreactivity to serotonin.
This article presents a comparative study of the effect of steel layouts on the seismic behavior of transition steel-concrete composite connections, both experimental and analytical investigations of concrete filled steel tube-reinforced concrete (CFST-RC) and steel reinforecd concrete-reinforced concrete (SRC-RC) structures were conducted. The steel-concrete composite connections were subjected to combined constant axial load and lateral cyclic displacements. Tests were carried out on four full-scale connections extracted from a real project engineering with different levels of axial force. The effect of steel layouts on the mechanical behavior of the transition connections was evaluated by failure modes, hysteretic behavior, backbone curves, displacement ductility, energy dissipation capacity and stiffness degradation. Test results showed that different steel layouts led to significantly different failure modes. For CFST-RC transition specimens, the circular cracks of the concrete at the RC column base was followed by steel yielding at the bottom of the CFST column. While uncoordinated deformation could be observed between SRC and RC columns in SRC-RC transition specimens, the crushing and peeling damage of unconfined concrete at the SRC column base was more serious. The existences of I-shape steel and steel tube avoided the pinching phenomenon on the hysteresis curve, which was different from the hysteresis curve of the general reinforced concrete column. The hysteresis loops were spindle-shaped, indicating excellent seismic performance for these transition composite connections. The average values of equivalent viscous damping coefficients of the four specimens are 0.123, 0.186 and 0.304 corresponding to the yielding point, peak point and ultimate point, respectively. Those values demonstrate that the transition steel-concrete composite connections have great energy dissipating capacity. Based on the experimental research, a high-fidelity ABAQUS model was established to further study the influence of concrete strength, steel grade and longitudinal reinforcement ratio on the mechanical behavior of transition composite connections.
The composite steel reinforced concrete (SRC) columns have been widely used in Structural Engineering due to their good performances. Many studies have been done on the SRC columns' performances, but they focused on the ordinary types with conventional configurations and materials. In this study, nine new types of steel reinforced lightweight aggregate concrete (SRLAC) short columns with cross-shaped (+shaped and X-shaped) steel section were tested under monotonically axial compressive load; the studied parameters included steel section ratio, steel section configuration, ties spacing, lightweight aggregate concrete (LWAC) strength, and longitudinal bars ratio. From the results, it could be found that the specimens with larger ties ratio, concrete strength, longitudinal bars ratio, and steel section ratio achieved great strength and stiffness due to the excellent interaction between the concrete and steel. The well-confined concrete core could strengthen the steel section. The ductility and toughness of the specimens were influenced by the LWAC strength, steel section ratio, and longitudinal bars ratio; in addition, larger ties ratio with smaller LWAC strength led to better ductility and toughness. The load transfer between concrete and steel section largely depends on the LWAC strength, and the ultimate strength of the new types of SRLAC short columns could be approximately predicted, referring to the codes' formulas of ordinary types of steel reinforced concrete (SRC) columns. Among the used codes, the BS-5400-05 led to the most conservative results.

In order to study the seismic response of the main plant of steel reinforced concrete (SRC) structure of the CAP1400 nuclear power plant under the influence of different high-mode vibration, the 1/7 model structure was manufactured and its dynamic characteristics was tested. Secondly, the finite element model of SRC frame-bent structure was established, the seismic response was analyzed by mode-superposition response spectrum method. Taking the combination result of the 500 vibration modes as the standard, the error of the base reactions, inter-story drift, bending moment and shear of different modes were calculated. Then, based on the results, the influence of high-mode vibration on the seismic response of the SRC frame-bent structure of the main plant was analyzed. The results show that when the 34 vibration modes were intercepted, the mass participation coefficient of the vertical and horizontal vibration mode was above 90%, which can meet the requirements of design code. There is a large error between the seismic response calculated by the 34 and 500 vibration modes, and the error decreases as the number of modes increases. When 60 modes were selected, the error can be reduced to about 1%. The error of the maximum bottom moment of the bottom column appeared in the position of the bent column. Finally, according to the characteristics of the seismic influence coefficient αj of each mode, the mode contribution coefficient γj•Xji was defined to reflect the contribution of each mode to the seismic action.
