  • Seral changes in floristic composition during abandoned field succession after shifting cultivation was investigated in eastern Kangwon-Do, Korea. According to the DCA ordination based on the floristic composition, sere of the dominant species was shown as follows: Digitaria sanguinalis, Persicaria spp., Commelina communis etc. in the annual stage (0∼1 years); Erigeron spp., Artemisia spp., Rubus crataegus etc. in the Erigeron-Artemisia stage (2∼6 years); Pinus densiflora, Salix spp., Miscanthus sinensis, Arundinella hirta etc. in the shrub - earlier tree stage (10∼25 years); P. densiflora, Quercus mongolica, Spodiopogon sibiricus etc. in the mid-tree stage (25∼50 years); Q. mongolica, Tripterigium regelii, Lespedeza maximowiczii, Carex siderosticta etc. in the latter tree stage (50∼80 years). In mid-tree stage, size-frequency distribution of P. densiflora was plotted as a platycurtic curve and that of Q. mongolica as a reverse J-shaped curve, which meant P. densiflora was thinned through interspecific competition and Q. mongolica was regenerated by itself in the later tree stage.

Detection of Escherichia coli O157:H7, Listeria monocytogenes, Salmonella spp. and Staphylococcus aureus using duplex real-time PCR assay with melting curve analysis on fresh lettuce

  • In this study, two duplex real-time PCR approach with melting curve analysis is presented for the detection of Escherichia coli O157:H7, Listeria monocytogenes, Salmonella spp. and Staphylococcus aureus, which are important food-borne bacterial pathogens usually present in fresh and/or minimally processed vegetables. Reaction conditions were adjusted for the simultaneous amplification and detection of specific fragments in the ${\beta}$-glucuronidase (uidA, E. coli), thermonuclease (nuc, S. aureus), hemolycin (hly, L. monocytogenes) and tetrathionate reductase (ttr, Salmonella spp.) genes. Melting curve analysis using a SYBR Green I real-time PCR approach showed characteristic $T_m$ values demonstrating the specific and efficient amplification of the four pathogens; $80.6{\pm}0.9^{\circ}C$, $86.9{\pm}0.5^{\circ}C$, $80.4{\pm}0.6^{\circ}C$ and $88.1{\pm}0.11^{\circ}C$ for S. aureus, E. coli O157:H7, L. monocytogenes and Salmonella spp., respectively. For all the pathogens, the two duplex, real-time PCR was equally sensitive to uniplex real-time PCR, using same amounts of purified DNA, and allowed detection of 10 genome equivalents. When our established duplex real-time PCR assay was applied to artificially inoculated fresh lettuce, the detection limit was $10^3$ CFU/g for each of these pathogens without enrichment. The results from this study showed that the developed duplex real-time PCR with melting curve analysis is promising as a rapid and cost-effective test method for improving food safety.

  • In this paper, we propose an improved path planning method and obstacle avoidance algorithm for two-wheel mobile robots, which can be effectively applied in an environment where obstacles can be represented by circles. Firstly, we briefly introduce the rapidly exploring random tree (RRT) and single polar polynomial (SPP) algorithm. Secondly, we present additional two methods for applying our proposed method. Thirdly, we propose a global path planning, smoothing and obstacle avoidance method that combines the RRT and SPP algorithms. Finally, we present a simulation using our proposed method and check the feasibility. This shows that proposed method is better than existing methods in terms of the optimality of the trajectory and the satisfaction of the kinematic constraints.

Rapid Detection of Salmonella spp. in Fresh-Cut Cabbage by Real-Time PCR (Real-Time PCR을 이용한 신선편이 양배추에서 Salmonella spp.의 신속검출)

  • This study was conducted to find out the minimal time needed for detection of Salmonella spp. which exist at very low concentration in foods by using real-time PCR. The sal-F and sal-R sequences were used as primers and sal-P was used as a probe. The detection limit of Salmonella spp. was $3.77{\times}10^2\;cfu/mL$ in buffered peptone water (BPW). Microbial growth was monitored after artificially inoculated Salmonella spp. into BPW. The obtained growth curve was well fitted with the equation, y=$0.0127x^2$+0.5927x-0.4317 ($R^2$=0.99), if assuming that 1 cell exists in 25 g sample (0.04 cfu/mL). The microbial concentration will be reduced to 10 fold by adding BPW during sample treatment, so actual initial concentration at the starting point of enrichment is 0.004 cfu/mL. At this condition, real-time PCR detection would be possible only when microbial concentration increase occurs to exceed the detection limit (377 cfu/mL). The time needed for microbial increase was calculated from the growth curve equation as 7 hours and 20 minutes. Therefore the total time required for detection was less than 10 hours including the PCR operating time.

Mathematical Description of Seedling Emergence of Rice and Echinochloa species as Influenced by Soil burial depth

  • A pot experiment was conducted to investigate the effects of soil burial depth on seedling emergences of rice (Oryza sativa) and Echinochloa spp. and to model such effects for mathematical prediction of seedling emergences. When the Gompertz curve was fitted at each soil depth, the parameter C decreased in a logistic form with increasing soil depth, while the parameter M increased in an exponential form and the parameter B appeared to be constant. The Gompertz curve was combined by incorporating the logistic model for the parameter C, the exponential model for the parameter M, and the constant for the parameter B. This combined model well described seedling emergence of rice and Echinochloa species as influenced by soil burial depth and predicted seedling emergence at a given time after sowing and a soil burial depth. Thus, the combined model can be used to simulate seedling emergence of crop sown in different soil depths and weeds present in various soil depths.

A study on effective disinfection methods of medical ultrasound probe resident floras (의료용 초음파 프로브 상제균의 효과적인 소독법에 관한 연구)

  • In the probe of a medical ultrasound device, three parts were selected randomly by the examiner and the bacteria in the probe were detected by the blood examiners. In addition, the degree of death of the pathogenic bacteria after each disinfection of the detected pathogens, disinfecting ethanol, and disinfecting tissue of the detected pathogens were analyzed quantitatively. The following was detected: S Aureus (32.3 %), Bacillus spp. (26.5 %), Micrococcus spp. (21.5 %), and CNS (20 %). With the conventional probe, S. aureus (26.2 %), a playback curve (24.2 %), and a micron (19.5 %), Micrococcus spp. (15.5 %), and CNS (14.6 %) were observed. In the fan probe, S. aureus (24.7 %), Enterococcus (24.7 %), Enterococcus (17.7 %), and CNS (13.8 %) were detected. The disinfection of the three pathogens detected revealed sterilization of most of the pathogens, and most cases contained at least 91.3 % of the total sterilizing effect (P>0.05). In addition, for the disinfection of Propolis extract and disinfecting tissue, the disinfection effect was lower than that of disinfecting ethanol, but the difference was not statistically significant (P>0.05). The results revealed bacteria on most of the ultrasound probes. Antiseptic disinfection of surgical instruments using an extract of propolis works with results similar to those of ethanol. A blood test along with disinfection can help prevent infection if an ultrasound probe is applied to food.

Prevalence of Bacillus cereus from Fried Rice Dishes and Monitoring Guidelines for Risk Management (볶음밥의 Bacillus cereus 위해 수준 및 위해 관리를 위한 모니터링 기준 설정)

  • Contamination levels of aerobic colony counts, coliforms and pathogenic bacteria were tested in fried rice dishes to monitor quality for risk management. The prevalence of Bacillus cereus in dishes from 8 Chinese-style restaurants and 2 institutional foodservices was 10%, and the bacteria's contamination levels was 3.47 log CFU/g. Echerichia coli, Staphylococcus aureus, and Salmonella were not detected in any of the 10 samples. However, for their aerobic colony counts and numbers of isolated coliforms, the samples were 30% and 70% over the microbial criteria, respectively, for ready-to-eat foods presented in the Korean Food Code. This suggests that fried rice dishes, although cooked with oil at high temperature, require special care. For the prediction of the growth curve of B. cereus spp. in the fried rice samples, an experiment design of 3 storage temperatures ($7^{\circ}C$, $35^{\circ}C$, $57^{\circ}C$) x 5 storage times (0 h, 2 h, 4 h, 6 h, 24 h) was applied. The sample exposed to $35^{\circ}C$ showed no B. cereus spp. at 0 h; however, there was a tendency of slow growth (1.0 log CFU/g) after 4 hours of storage and then faster growth at 6 h (3.7 log CFU/g) and 12 h (4.7 log CFU/g), showing a growth rate of 0.56 log CFU/g/hr. These results indicate that fried rice, despite being heat-treated, can become heavily contaminated with B. cereus spp. when held over 2 hours at room temperature. However, the samples stored at $7^{\circ}C$ and $57^{\circ}C$ over 24 hours were not contaminated with B. cereus. Based on these results, management guidelines for controlling B. cereus are suggested.

  • Shigella spp. constitutes some of the key pathogens responsible for the global burden of diarrhoeal disease. With over 164 million reported cases per annum, shigellosis accounts for 1.1 million deaths each year. Majority of these cases occur among the children of the developing nations and the emergence of multi-drug resistance Shigella strains in clinical isolates demands the development of better/new drugs against this pathogen. The genome of Shigella flexneri was extensively analyzed and found 4,362 proteins among which the functions of 674 proteins, termed as hypothetical proteins (HPs) had not been previously elucidated. Amino acid sequences of all these 674 HPs were studied and the functions of a total of 39 HPs have been assigned with high level of confidence. Here we have utilized a combination of the latest versions of databases to assign the precise function of HPs for which no experimental information is available. These HPs were found to belong to various classes of proteins such as enzymes, binding proteins, signal transducers, lipoprotein, transporters, virulence and other proteins. Evaluation of the performance of the various computational tools conducted using receiver operating characteristic curve analysis and a resoundingly high average accuracy of 93.6% were obtained. Our comprehensive analysis will help to gain greater understanding for the development of many novel potential therapeutic interventions to defeat Shigella infection.

Developmental Characteristic of Cyanobacterial Bloom in Lake Daecheong (대청호의 남조세균 수화 발달 특성)

  • The occurrence of cyanobacterial bloom in Korean lakes of the summer is generalized. The characteristic of cyanobacterial community was explored. And the developmental stage of cyanobacterial bloom was divided into three phases, 'preparatory phase', 'bloom phase' and 'extinction phase' Cyanobacterial bloom started during the end of June at site 1, transition Bone of Lake Daecheong. The period of water bloom in normal year was about 60~70 days at site 4, lacustrine Bone, but it was unusually 11 days from July 19 in 1999. M. aerugilnosa first occurred in June, had a peak of standing crop curve from the end of August to the beginning of September in 1998 and 2002 and the end of July in 1999 and 2001. The standing crop of M. aeruginosa occupied $68.1\%$ of phytoplankton, $74.2\%$ of cyanobacteria and $88.8\%$ of genus Microcystis, Anabaena spp. first occurred in April, was above 10,000 cells $mL^{-1}$ from the end of August to about the middle of September in 1998. The effect of rainfalls on cyanobacterial bloom was different according to the phases. The rainfalls of preparatory phase assist the growth of cyanobacteria, but accelerate the decrease of cyanobacteria in extinction phase. In bloom phase, the heavy rainfalls reduce the development of the bloom, while the slight ones display only a little effects.

Clinical manifestation of Campylobacter enteritis in children

  • Purpose: Timely antibiotic therapy in selected cases of diarrhea associated with bacterial infections can reduce the duration and severity of illness and prevent complications. The availability of a predictive index before identification of causative bacteria would aid in the choice of a therapeutic agent. Methods: The study included patients admitted to the pediatrics unit at Konyang University Hospital for acute inflammatory diarrhea from August 1, 2015 to July 31, 2016 who underwent multiplex polymerase chain reaction testing. Of 248 patients, 83 had positive results. The clinical symptoms and blood test results were examined in 61 patients with Campylobacter spp. (25 patients), Salmonella spp. (18 patients), and Clostridium perfringens (18 patients) infections. The mean age of the 61 patients (male:femal=31:30) was $84.0{\pm}54.8months$, and the mean hospital stay was $4.6{\pm}1.7days$. Results: There were no statistical differences in sex, age, clinical symptoms, or signs. Patients with Campylobacter infection were significantly older (P=0.00). C-reactive protein (CRP) levels in patients with Campylobacter infection were higher than those in the other 2 groups, at $9.6{\pm}6.1mg/dL$. The results of receiver-operating characteristic curve analysis showed that the cutoff age was ${\geq}103.5months$ (sensitivity, 72%; specificity, 86%) and the CRP cutoff level was ${\geq}4.55mg/dL$ (sensitivity, 80%; specificity, 69%). Conclusion: Age (${\geq}103.5months$) and higher CRP level (${\geq}4.55mg/dL$) were good predictors of Campylobacter enterocolitis. If neither criterion was met, Campylobacter enterocolitis was unlikely (negative predictive value 97.2%). When both criteria were met, Campylobacter enterocolitis was highly likely.
