  • nif (nitrogen fixation)-H.D, K genes of Frankia sp. SNU 014201. a symbiotic strain isolated from root nodule of Alnus hirsura, were found to be located in the genome on 13.5 kb of EcoRI, 18.0 kb of BamHI, 10.5 kb of BglII and 4.5 kb of KpnI fragments. Using EMBL-3 BamHI arms of bacteriophage lambda. the genomic library was constructed. from which fourteen recombinant phage nif-clones were selected. Among them, Ahnif-I2 had insert DNA of 18 kb, in which 7.9 kb of BamHl fragment contained nif-H, D, K and 3.6 kb of HindlIl/KpnI had nif-H and partial -D. Therefore, the 7.9 kb and 3.6 kb fragments were subcloned and partial restriction maps were constructed. As the results, nif-F1, D.K genes were found to be located continuously on the 6.5 kb of HindII/BamHI and 5.2 kb of SalIIBamHI fragment in the genome of Frankia sp. SNU 014201.

Conservation Environmental Assessment and Microbial Distribution of the Songsan-ri Ancient Tombs, Gongju, Korea (공주 송산리 고분 내 미생물 분포 및 보존환경 연구)

  • After occurrence of Cyanobacteria in 1997, Songsan-ri tombs located in Gonju have been investigated to monitor for biological damage. The room temperature of Tomb No.6 was $18.6{\sim}19.8^{\circ}C$ and the relative humidity was 94.3~99.9%. The temperature of Royal Tomb of King Muryeong was $17.3{\sim}18.53^{\circ}C$ and the relative humidity was 73.2~96.45%. The variation of relative humidity increased after setting up air vents. If the outside temperature increases, dew condensation occurs on the floor and the north side. When conditioning equipment operates, the maximum temperature differences between walls is $2.8^{\circ}C$. Bacteria from the air of the tomb and on the surface of the walls outnumbered fungi. 20 species of fungi including Alternaria sp., Aspergillus sp., Penicillium sp., and 19 species of bacteria including Pseudomonas sp., Arthrobacter sp., are identified. Microbes in the tombs may damage cultural heritage. The growth possibility of microbes should be estimated because the microbes in the tombs may damage mural painting. The interrelation between microenvironmental condition and biological damage of mural painting should be researched to come up with an long-term conservation method.

Effects of Deep Seawater on the Growth of a Green Alga, Ulva sp.(Ulvophyceae, Chlorophyta)

  • In order to examine the effects of deep seawater (mesopelagic water in the broad sense) on the growth of macroalgae, the growth and nutrient uptake (nitrate and phosphate) of Ulva sp. (Ulvophyceae, Chlorophyta) were investigated by cultivation in deep seawater (taken from 687 m depth at Yaizu, central Japan, in August 2001), surface seawater (taken from 24 m depth), and a combination of the two. Culture experiments were carried out in a continuous water supply system and an intermittent water supply system, in which aerated 500-mL flasks with 4 discs of Ulva sp. (cut sections of ca. 2 $cm_2$) were cultured at 20$^{\circ}C$ water temperature, 100 $\mu$mol photons $m^{-2}{\cdot}s^{-1}$ light intensity, and a 14:10 light:dark cycle. Nutrient uptake by Ulva sp. was high in all seawater media in both culture systems. The frond area, dry weight, chlorophyll a content, dry weight per unit area, and chlorophyll a content per unit area of Ulva sp. at the end of the experimental period were the highest in deep seawater and the lowest in surface seawater in both culture systems. These values, except for dry weight per unit area and chlorophyll a content per unit area, for each seawater media in the intermittent water supply system were higher than those in the continuous water supply system. We conclude that not only deep seawater as the culture medium but also the seawater supply system is important for effective cultivation of macroalgae.

Emulsification Activity of Acinetobacter sp. 2-3A Isolated from Petroleum Oil-Contaminated Soil (유류오염 토양에서 분리한 Acinetobacter sp. 2-3A의 유화활성)

  • Fifty hydrocarbon-metabolizing microorganisms were isolated from soil samples polluted by the petroleum oils in Gamman-dong, Busan. Among them, strain 2-3A, showing strong emulsification activity, was selected by oil film-collapsing method. This bacterium was identified as Acinetobacter sp. and designated as Acinetobacter sp. 2-3A. The optimum temperature and pH on the growth of Acinetobacter sp. 2-3A were $25^{\circ}C$ and pH 7.0, respectively. The carbon and nitrogen sources for the most effective emulsification activity were 3.0% olive oil and 0.5% peptone, respectively. The 0.15% potassium phosphate was the most effective emulsification activity as a phosphate source. The optimum emulsification activity condition was $20^{\circ}C$, pH 7.0, and 2.0% NaCl. The optimum time for the best production of biosurfactant was 27 hrs. The emulsification stability was maintained at the temperature range from $4^{\circ}C$ to $100^{\circ}C$, pH range from 6.0 to 10.0, and NaCl range from 0% to 10%. For the oil resolvability of the biosurfactant, the residual oils were investigated by gas chromatography. As a result, it was verified that the biosurfactant decreased and decomposed crude oils from $_nC_{10}$ to $_nC_{32}$.

  • The strain which produces agar degrading enzyme was isolated from chiton(Liolophura japonica). The strain was identified as Cytophaga sp. through its morphological, physiological, and biological characteristics. For the production of agar degrading enzyme, 0.3% nutrient broth, 0.2% yeast extract and 0.5% agar was used as nitrogen and carbon source, respectively. The optimal initial pH, NaCl and temperature for the agar degrading activity of Cytophaga sp. were 7.0, 2.0% and $30{\pm}2^{\circ}C$, respectively. Agar degrading activity of enzyme obtained from Cytophaga sp. was increased until the incubation of 96hrs, but after 96hrs, the activity was decreased.

  • Self-potential (SP) is sensitive to groundwater flow and there are many causes to generate SP. Among many mechanisms of SP, pore-fluid flow in porous media can generate potential without any external current source, which is referred to as electrokinetic potential or streaming potential. When calculating SP responses on the surface due to geothermal fluid within an engineered geothermal system (EGS) reservoir, SP anomaly is usually considered to be generated by fluid injection or production within the reservoir. However, SP anomaly can also result from geothermal water fluid within EGS reservoirs experiencing temperature changes between injection and production wells. For more precise simulation of SP responses, we developed an algorithm being able to take account of SP anomalies produced by not only water injection and production but also the fluid of geothermal water, based on three-dimensional finite-element-method employing tetrahedron elements; the developed algorithm can simulate electrical potential responses by both point source and volume source. After verifying the developed algorithm, we assumed a simple geothermal reservoir model and analyzed SP responses caused by geothermal water injection and production. We are going to further analyze SP responses for geothermal water in the presence of water production and injection, considering temperature distribution and geothermal water flow in the following research.

  • A marine bacterium which was isolated from the enrichment culture for the emulsification of Bunker-C oil produced a bioemulsifier potently. The strain identified as an Achromobacter sp. M-1220. The bioemulsifier was produced during mid-logarithmic phase in hexadecane oil medium at 18$^{\circ}C$. It appeared to be a cationic peptidolipid substance and showed an active stabilizing effect on the emulsion of crude oils and a few vegetable oils.

  • Seolgiddeok a representative rice cake was prepared by the addition of silkworm powder(SP) at various concentrations, and their physical characteristics were monitored by sensory evaluation, chromaticity, and rheometric measurement. In the proximate composition of SP cake, the contents of crude protein and ash were increased as the ratio of SP increased, but the moisture content was decreased. In the sensory evaluation, 3%-SP cake showed the highest preference, old showed the highest values in color, flavor, taste. texture, and overall duality. Lightness(L) value of SP cake was decreased as the ratio of SP increased. In the rheometer test, 15%-SP cake showed the highest value in the hardness, but 3%-SP cake showed the highest value in cohesiveness, gumminess and brittleness.
