홍삼 투여가 방사선에 조사된 생쥐 신장의 Superoxide Dismutase에 미치는 영향 (The Effect of Red Ginseng Extract on Superoxide Dismutase Activity in the Kidney of Gamma-ray Irradiated Mice)

  • 감마선에 조사된 흰쥐의 신장 내의 효소활성에 홍삼 추출물이 어떠한 영향을 미치는지를 연구하고자 생후 5주령의 생쥐를 4군으로 분류하여 감마선 조사 및 홍삼 추출물 투여 혹은 생리적 식염수를 투여하고 그로부터 1일, 2일, 3일, 4일 및 5일 등 5회에 걸쳐 도살하여 신장을 취하고 신장조직 내의 SOD, peroxidase 및 catalase 등의 활성도를 측정하였다. 그 결과 홍삼 추출물 투여군은 대조군에 비하여 SOD나 catalase 및 peroxidase의 활성이 유의성있게 증가하다가 회복하였으며 감마선 조사 후 생리적 식염수 투여군은 대조군에 비하여 SOD, peroxidase 및 catalase의 활성이 증가하다가 감소하는 경향이었다. 감마선 조사후 홍삼 추출물 투여군은 SOD, peroxidase 및 catalase의 활성도가 감마선 조사 후 생리적 식염수 투여군에 비하여 보다 빠르게 회복되는 경향이었다. 그러므로 홍삼 추출물이 이온화 방사선 조사 후 장해로 일어나는 SOD, peroxidase 및 catalase 활성도의 변화에 회복효과가 있음이 나타났다.

The Effect of Metallothionein on the Activity of Enzymes Invelved in Remival of Reactive Oxygen Species

  • To show the effects of metallothionein (MT) on the activity of enzymes involved in the removal of reactive oxygen species, MT has been added to the assay systems of superoxide dismutase (SOD), catalase and peroxidase. We have used assay systems of SOD based on NADPH oxidation and nitrite formation from hydroxylammonium chloride as an assay of superoxide breakdown rate. The two assay systems showed different results at the high concentration of MT. MT showed the scavenging of superoxide in the SOD assay system in the presence and absence of SOD. MT added to the SOD assay system behaved as an activator of SOD, but apo-MT behaved as an inhibitor. When MT was added to the assay system in the presence of a fixed amount of SOD, the breakdown rate of superoxide increased. The effects of MT on the decomposition of hydrogen peroxide and the activity of catalase and peroxidase decomposing hydrogen peroxide were evaluated. MT decreased the activities of catalase and peroxidase. We have concluded that the function of MT as an antioxidant might effect the level of superoxide scavenging and not the level of hydrogen peroxide.

흰쥐에서 Aminotriazole과 Diethyldithiocarbamate가 Paraquat의 독성에 미치는 영향 (Effects of 3-Amino-1,2,4 Triazole and Diethyldithiocarbamate on Paraquat Toxicity in Rats)

  • The effects of superoxide dismutase(SOD) and catalase on the toxicity of paraquat(PQ) were studied using diethyldithiocarbamate(DDC), 3-amino-1,2,4-triazole(AT) which are inhibitors of Cu, Zn-SOD and catalase in rats. Sprague Dawley rats were divide into 6 groups: control, DDC, PQ, AT, DDC+PQ, and AT+PQ group. The PQ (50 mg/kg body weight(BW); about half dose of $LD_{50}$) was administered with orally, otherwise AT(1.0g/kg BW) and DDC(1.0g/kg BW) were administered by intrperitoneal(iP) injection. The survival rate of rats in PQ+AT group was significantly decreased compared with PQ group while the difference of survival rate between DDC group and DDC+PQ group was not significant. The SOD activity after administration of DDC was decreased in liver, lung and kidney, but catalase activity was not changed. The catalase activity in liver, lung and kidney of AT treated rats was decreased, while SOD activity was not changed in this group. The effects of DDC and AT to the PQ toxicity was also observed in primary cultured rat Skin fibroblasts. The viable cells that was measured with MTT method, was decreased in AT+PQ treated group compared to PQ treated group, but the difference of cell viability between DDC treat group and DDC+PQ treated group was not observed. This result, AT potentlate PQ toxicity while DDC were not affect, suggested that the decreased catalase activity lead to elevation of hydrogen peroxide levels and PQ toxicity may be correlate with the hydrogen peroxide rather than the superoxides.

십전대보탕의 항산화작용에 관한 연구 (Study on Antioxidant Aaction of Sibjeondaebo-tang)

  • In order to examine the antioxidant activities of SJDBT(Sibjeondaebo-tang), the study was done through measurement of parameters such as LPO(lipidperoxidation), GSH(glutathione), SOD(superoxidation dismutase), catalase, GOT, GPT, ALP. The results were obtained as follows: For the weight changes, the kidney and testis of group given SJDBT showed decrease in the weight compared to the control group, and the left cerebrum, right cerebrum, cerebellum and liver of group given SJDBT showed increase. But the changes were not significant. The left cerebrum and right cerebrum of group given SJDBT showed significant decrease in the content of LPO compared to the control group, and in the activity of SOD and catalase showed significant increase. The cerebellum of group given SJDBT showed significant decrease in the content of LPO and GSH compared to the control group, and in the activity of SOD and catalase showed significant increase. For the changes of LPO, GSH in the liver, the group given SJDBT showed significant decrease in the content of LPO and GSH compared to the control group, and in the activity of catalase showed significant increase. For the changes of LPO in the kidney, the group given SJDBT showed significant decrease in the content of LPO compared to the control group. For the changes of LPO, GSH and the activity of SOD, catalase in the testis, the group given SJDBT showed significant decrease in the content of LPO and GSH compared to the control group, and in the activity of SOD and catalase showed significant increase. From above results, the antioxidant action of SJDBT is effective. And it is expected to be necessary to the study of the mechanism in the antioxidant of SJDBT.

Rhodobacter sphaeroides D230이 생성하는 산소 유도체에 작용하는 효소의 특성 (Characterization of Enzymes Against Oxygen Derivatives Produced by Rhodobacter sphaeroides D-230)

  • 산소의 존재 유ㆍ무 등과 같은 배양 환경의 변화에 따라 통성 혐기성 광합성 세균인 Rhodobacter sphaerodes B-230이 만들어내는 산소 유도체에 작용하는 효소의 특성을 조사한 결과 세포내 SOD는 호기적 배양에서는 초기 배양액의 pH가 7일 때, 혐기적 배양에서는pH 8일 때 활성이 높은 반면 세포외 방출 SOD는 두 배양조건에서 모두 약산성인 pH 6에서 활성이 높았다. Catalase는 두 조건 모두 중성 부근에서 최고의 활성을 보였으며, 산성 pH 부위에서는 급격히 활성이 낮아졌다. Mn-SOD의 활성 유도제인 methyl viologen을 첨가했을 때 두 조건 모두에서 성장의 저해를 보였으며, 배지에 철 이온을 첨가하여 배양 하였을 때 호기적 조건에서만 두 배 이상 활성이 증가되었다. 혐기적 조건에서는 전체적인 활성이 낮아 금속이온의 추가적인 첨가에도 더 이상 활성이 유도되지 않았다. Mn-SOD 활성 저해제인 $NaN_3$와 CuZn-SOD활성 저해제인 NaCN를 배양액에 첨가했을 때 NaCN은 두 가지 배양 조건에서 생성되는 SOD 모두를 저해하지 않았으며, $NaN_3$는 혐기적 배양조건에서만 0.3 mM 이상에서 급격한 SOD활성의 저해를 가져왔다. 따라서 Rhodobacter sphaeroides D-230도 혐기적 배양 조건에서 Mn-SOD가 생성되는 것을 확인할 수 있었으며,호기적 조건에서는 Fe-SOD가 생성되는 것을 확인할 수 있었다. Catalase의 활성도 두 가지 배양조건 모두에서 methy1 viologen에 의해 활성이 유도되었으며, NaCN와 $NaN_3$에 의해서 급격히 저해되었다.

  • 녹차카테킨(GTC)의 항산화 작용을 알아보고자 in vitro와 in vivo에서 지질과산화와 superoxide dismutase(SOD), catalase 활성에 미치는 영향에 대한 실험을 행하였다. In vitro 시험계에서의 항산화활성 실험결과, GTC는 peroxide value와 과산화지질 생성을 유의성 있게 억제시켰고, SOD 활성이 매우 높았다. 또한 GTC를 rat에 경구투여 한 후 항산화활성실험 결과, GTC는 $CCl_4$로 유도된 rat의 간 microsome의 지질과산화를 유의성 있게 억제시켰으며, SOD와 catalase 활성을 유의성 있게 증가시켰다. 따라서 GTC는 암과 노화의 예방과 관련이 되는 항산화 활성이 있는 것을 알 수 있었다.

치주염 환자의 혈장과 적혈구내 S.O.D와 Catalase 활성도에 관한 연구 (SUPEROXIDE DISMUTASE - AND CATALASE - ACTIVITY IN BLOOD PLASMA AND RED BLOOD CELLS IN PERIODONTITIS)

  • It has been believed that the increased release of free oxygen radicals and their tissue damaging potency might be a contributing factor in the pathogenesis of periodontal disease. Antioxidant enzymes such as superoxide dismutase(SOD) and catalase can protect the tissue damage from the free oxygen radicals($O_2^-,H_2O_2$, and $OH^-$). In order to investigate the SOD- and catalase - activity in the blood plasma and red blood cells(RBCs) of the patients with perodontitis, 19 male periodontitis patients($25{\sim}35$ years old) who had good general health, more than 10 teeth with severely inflamed gingiva, attachment loss more than 6mm and bone loss were selected as periodontitis group, and 13 male volunteers($22{\sim}29$ years old) with good general and periodontal health were selected as normal group. After blood plasma and RBC were separated from peripheral blood of 2ml collected from antecubital vein of each subject, SOD- activity in blood plasma and RBCs was measured by the same method that Paoletti et al. did, and catalase - activity in RBC was measured by the same method that Beers et al, did. The difference of SOD- and catalase - activity between the normal and the periodontitis groups was statistically analyzed by Student t-test with SPSS/PC program.The results were as follows : 1. SOD activity in blood plasma was significantly lower in the periodontitis group($1.986{\pm}0.893$) than in the normal group($3.324{\pm}1.044$)(p<0.05). 2. In vivo Antioxidant Effects of Aralia elata Seemann Ethanol Extract Administered with Benzo($\alpha$) pyrene

  • This study has examined the effects of Aralia elata Seemann ethanol extract on antioxidant enzyme systems inrats along with benzo($\alpha$) pyrene(B(a)P) administration . The ethanol extract of Aralia elata Seemann (50mg/kg body wt.) was fed to rats for 4 weeks by stomach tubing. The extract administration increased antioxidant activities of glutathione sulfur transferase(GST) comparing to the control. also total superoxide dismutase(SOD) and Cu, Zn-SOD activities were stimulated. Catalase activities were increased by 50% with the extract feeding compared to the control . Combined administration of B($\alpha$)P and the extract increased GST activity in B($\alpha$) P group. Although total SOD acitivity was decreased , Cu, Zn-SOD was greately increased from 0.10unit to 0.18 unit and catalase activity also was increased compared to the group of B($\alpha$) P. GST activity in CLE group was 1.32 unit, increased by 33% comparing to the group CL of 0.99unit. Cu, Zn-SOD and catalase activities in thegroup fed high fat and ethanol extracts were increased by 25% and 39%, respectivley comparing to the group of high fat. In addition , total SOD was decreased but, Cu, Zn-SOD acitivity was increased from 0.09 unit to 0.18unit. Catalase activity was 76.05 unit in the group of B($\alpha$) P and extract comparing to 65.26 units in B($\alpha$)P group. Serum $\alpha$-tocopherol of rat was markedly increased by theextract. Administration of B9$\alpha$)P reduced $\alpha$-tocopherol levels in the serum, on the other hand, lard in the diet increased $\alpha$-tocopherol levels in the serum. The above results indicate that Aralia bud exerts antioxidant functions in vivo against B($\alpha$)P. Further research may be necessary for the identification fo the biologically active material.

육미지황탕의 항산화작용에 관한 연구 (Study on antioxidant action of Yukmijihwang-tang)

In the left cerebrum, the group given YMJHT showed significant increase compared to the control group on the activities of SOD, catalase. In the right cerebrum, the group given YMJHT showed significant decrease on the content of LPO and showed significant increase on the activity of catalase. In the cerebellum, the group given YMJHT showed significant decrease on the content of LPO and showed significant increase on the activities of SOD, catalase. In the liver, the group given YMJHT showed significant decrease on the content of GSH and showed significant increase on the activity of SOD. In the kidney, the group given YMJHT showed significant decrease on the contents of GSH, GSH and showed significant increase on the activities of SOD, catalase. In the testis, the group given YMJHT showed significant decrease on the contents of LPO, GSH and showed significant increase on the activity of SOD. From above results, the antioxidant action of YMJHT is effective. And it is expected to be necessary to the study of the mechanism in the antioxidant of YMJHT.

일산화탄소 폭로후 고압산소 투여가 흰쥐 신장에서의 malondialdehyde 함량과 catalase 및 superoxide dismutase 활성에 미치는 영향 (Effects of Hyperbaric Oxygen Treatment on the Malondialdehyde Level and Activities of Catalase and Superoxide Dismutase in the Kidney of the Rats Exposed to Carbon Monoxide)

  • In an attempt to define the effects of hyperbaric oxygen treatment on the lipid peroxidation and oxygen free radical reactions in rats exposed to carbon monoxide, we studied malondialdehyde (MDA) level and activities of catalase and superoxide dismutase in the kidney of the rats exposed to carbon monoxide. Male Sprague-Dawley albino rats weighing 240 to 260 gm were used. Experimental groups consist of Control group (=breathing with air), HBO group (=exposed to hyperbaric oxygen 〔HBO, 3ATA, 100%〕 after air breath), CO group (=exposed to CO〔3,970 ppm〕after air breath), CO-Air group (=exposed to CO after air breath followed by air breath) and CO-HBO group (=exposed to CO after air breath followed HBO treatment). The CO group showed significantly higher MDA level, catalase activity and SOD activity as compared to that of control group. The CO-HBO group showed significantly lower MDA level as compared to that of CO group, and did not show significantly lower catalase activity and SOD activity as compared to that of CO group. These results suggest that the excessive oxygen free radicals is an important determinant in pathogenesis of CO-induced nephrotoxicity and HBO inhibits the lipid peroxidation caused by excessive oxygen free radicals in the kidney of the rats exposed to carbon monoxide.
