The objective of this study was to optimize the selection of sperm sources, optimal culture systems and vitrification method depends on sperm sources. The oocytes were inseminated with either KPN 105, 114, 191, SNU 101, 102, 103 or epididymis and then embryos inseminated were cultured in oviductal cell co-culture or HECM-6 as defined me dium. The blastocysts produced were pooled according to sperm sources as KPN, SNU or epididymis and then vitrified by OPP vitrification method. The results obtained were as follows: 1. The cleavage(86.2 or 84.7%) and development rates to blastocyst (30.6 or 32.0%) were not significantly different between oviductal cell co-culture or HECM-6 culture systems(P<0.05). 2. To determine the optimal sperm sources for using IVF in this system, cleavage rates in KPN 191 and SNU 101 (74.2, 55.8%) were significantly lower rather than those in KPN 105, 114, SNU 102, 103 or epididymis (86.7, 85.1, 89.8, 85.5 or 81.2%), but development rates to blastocyst in KPN 114, SNU 103 or epididymis sperm (30.0, 33.0 or 28.6%) were significantly higher rater than those in KPN 105, 191, SNU 101, 102(21.4, 15.4, 14.9 or 25.4%), respectively (P<0.05). 3. The blastocysts produced were pooled according to sperm sources as KPN, SNU or epididymis and then vitrified by OPP vitrification method. The survival rates were not significantly different among sperm sources (89.6%: 43/48 ; 90.1%: 46/51 ; 83.3% : 20/24). These results obtained indicate that the defined medium, HECM-6, could be use to produce of IVP bovine embryos. Since the frozen semen must be required to maintain of unvariation data in IVP embryo production system, KPN 114 and SNU 103 produced in our laboratory were useful for this purpose. The blastocysts produced by different sperm sources as KPN, SNU or epididymis were vitrified by OPP vitrification method and survived very high rates. The OPP vitrification method could be susceptibility to use of IVP bovine blastocyst embryos.
A cytotoxic compound, MCHS-4, was isolated from the mycelium of Streptomyces T70 and identified as manumycin A. The compound showed significant cytotoxicity against human gastric carcinoma cell line, SNU-1 and human hepatocellular cell line, SNU-354.

Objectives: Many efforts have shown multi-oncologic roles of galectin-3 for cell proliferation, angiogenesis, and apoptosis. However, the mechanisms by which galectin-3 is involved in cell proliferation are not yet fully understood, especially in human colon cancer cells. Methods: To cluster genes showing positively or negatively correlated expression with galectin-3, we employed human colon cancer cell lines, SNU-61, SNU-81, SNU-769B, SNU-C4 and SNU-C5 in high-throughput gene expression profiling. Gene and protein expression levels were determined by using real-time quantitative polymerase chain reaction (PCR) and western blot analysis, respectively. The proliferation rate of human colon cancer cells was measured by using a 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay. Results: Expression of ${\gamma}$-aminobutyric acid B receptor 1 (GABABR1) showed a positive correlation with galectin-3 at both the transcriptional and the translational levels. Down-regulation of galectin-3 decreased not only GABABR1 expression but also the proliferation rate of human colon cancer cells. However, Korean herbal extract, HangAmDan-B (HAD-B), decreased expression of GABABR1 without any expressional change of galectin-3, and offset ${\gamma}$-aminobutyric acid (GABA)-enhanced human colon cancer cell proliferation. Conclusions: Our present study confirmed that GABABR1 expression was regulated by galectin-3. HAD-B induced galectin-3-independent down-regulation of GABABR1, which resulted in a decreased proliferation of human colon cancer cells. The therapeutic effect of HAD-B for the treatment of human colon cancer needs to be further validated.
We studied the effects of hot water extract of Inonotus obliquos mushroom on the proliferation and apoptosis of the human colon adenocarcinoma, HT-29 and the human stomach adenocarcinoma, SNU-484 cell. Cells were maintained with Dulbecco's modified Eagle medium/Ham's F-12 nutrient mixture supplemented with 10% fetal bovine serum at 37$^{\circ}C$ in a humidified $CO_2$. For the cell proliferation experiments, cells were seeded in 35 mm dishes, and were treated with the various concentrations of the extract for the different time course. Apoptosis was measured by caspase-3 activity. When we incubated HT-29 cells for 24, 48, 72, and 96 hours after treatments, the cell proliferation was more suppressed with more treatment time. In case of the human stomach cancer cell, SNU484, the extract significantly decreased the cell number. Thus, the treatment of 1.5 mg/$m\ell$ extract decreased almost half of the cell number. Caspase-3 activity in HT-29 was increased by the treatment of mushroom extracts. In SNU484, caspase-3 activity tended to increase in proportion to the amounts of the extracts and the treatment of Inonotus obliquos affected the activity a lot. Therefore, Inonotus obliquos is suggested for the prevention of gastro-intestinal cancer and strongly recommended for the treatment of stomach cancer. (Korean J Nutrition 36(1) : 18~23, 2003)
종양세포는 포도당섭취 및 포도당 대사가 정상세포에 비해 증가된 특징을 가진다. 포도당 유사체인 $^{18}F$ fluorodeoxyglucose(FDG)의 섭취를 이용한 PET 검사가 종양의 진단에 많이 쓰이고 있다. 이 연구에서는 유사한 성질을 가진 사람의 대장암 세포주간의 FDG 섭취량 및 섭취 속도의 차이점을 비교하고, 그 포도당 수송체의 발현의 관련성을 규명하고자 한다. 사람대장암 세포 SNU-C2A, SNU-C4, SNU-C5를 이용하여 FDG 섭취를 측정하였다. 또한 세포의 포도당 섭취에 중요 역할을 하는 포도당 수송체 1(GLUT1)의 발현을 Western blotting으로 비교하였다. $1{\times}10^6/ml$의 대장암 세포에 HEPES-buffered saline에 희석한 $1{\mu}Ci/ml$ FDG를 가하여 $37^{\circ}C$에서 1시간 배양하였을 때 SNU-C2A($16.8{\pm}1.36cpm/{\mu}g$ of protein), SNU-C4($12.3{\pm}5.55$), SNU-C5($61.7{\pm}2.17$) 섭취를 보였다. 시간당 FDG의 섭취는 SNU-C2A($0.29{\pm}0.03cpm/ min/{\mu}g$ of protein), SNU-C4($0.21{\pm}0.09$), SNU C5($1.07{\pm}0.07$)이었으며, 시간이 경과함에 따라 비례하여 증가하였다. Western blotting으로 측정한 GLUT1 은 SNU-C5의 경우 다량 발현되었으나 SNU-C2A와 SNU-C4는 소량 발현되었다. 따라서 SNU-C2A, SNU-C4, SNU-C5 세포는 이들 세포가 비록 유사한 특징을 가졌지만 FDG 섭취량과 섭취 속도 및 GLUT1의 발현이 다르고, 이들 세포의 배가시간(doubling time)은 FDG 섭취와 상관관계가 없었다. 이들 세포의 FDG 섭취와 GLUT1의 발현은 밀접한 상관관계가 있었다.

Background: Platelet-rich plasma (PRP) stimulates cell proliferation and enhances matrix gene expression and synthesis. However, there have been no comparative study of the PRP effect on the normal and degenerative tenocytes. The purpose of this study was to compare the effect of PRP on tenocytes from normal and degenerative tendon. Methods: Tendon tissues were obtained from patients undergoing arthroscopic repair (n=9) and from healthy donors (n=3). Tenocytes were cultured with 10% (vol/vol) platelet-poor plasma, PRP activated with calcium, and PRP activated with calcium and thrombin. The total cell number was assessed at days 7 and 14. The expressions of type I and III collagen, decorin, tenascin-C, and scleraxis were evaluated by quantitative real-time reverse transcriptase polymerase chain reaction. The total collagen and glycosaminoglycan (GAG) synthesis was evaluated at days 7 and 14. Results: No differences were observed between the groups at day 7, but cell proliferation was remarkably increased in tenocytes from the degenerative tendon at day 14. In both tenocyte groups, the gene expressions of type I and III collagen were up-regulated. GAG synthesis was greater in the normal tendon, whereas the expressions of decorin and tenascin-C were increased in tenocytes from the degenerative tendon. Tenocytes from the degenerative tendon had higher fold-change of GAG synthesis and a lower collagen III/I ratio than normal tenocytes. Conclusions: PRP promoted the cell proliferation and enhanced the synthesis of tendon matrix in both groups. PRP has a greater positive effect on cell proliferation, matrix gene expression and synthesis in tenocytes from degenerative tendon.
Background : Anticancer effects of herbal medicine have been reported in various types of cancer, but the systematic approaches to explain molecular mechanism(s) are not established yet. Objective : To find the anticancer-effect and mechanism(s) of Water Extract of Coptis japonica (WECJ) colon cancer cell (SNU-81). Methods : We first selected 11 herbals, and anti-cancer effects of water-extracts from those herbals have been tested in human colon cancer cell line, SNU-81. Among the tested herbals, the WECJ significantly reduced proliferation of SNU-81. To establish a basis of understanding for anti-cancer mechanism, whole proteins have been obtained from SNU-81 harvested at 48 and 96 hrs after the treatment of WECJ, protein expression has been profiled by 2DE-based proteomic approach. Results : Various changes of the protein expression have been monitored, and most frequent dysregulation was found in the molecular chaperons including heat shock protein 90-alpha (Hsp90-alpha), 14-3-3 protein epsilon, T-complex protein 1 subunit alpha, protein disulfide-isomerase A3, and calreticulin. Interestingly, proliferation-associated protein 2G4 has been up-regulated, and it suggests the possible effect of Coptis japonica on ErbB3-regulated signal transduction pathway and growth control of human colon cancer cells. Conclusion : Based upon the present findings, the further study will focus on monitoring various cancer survival factors after artificial regulation of the proteins identified, and it would be the basis for the understanding of the Coptis japonica anti-cancer effect(s) at the molecular level.
Levans produced from Microbacterium laevaniformans were isolated, characterized, and fractionated by molecular weight. TLC, HPLC, and GC-MS analyses of the exopolysaccharide showed that it was a fructan-type polymer and was composed of (2,6)- and (2,1)-glycosidic linkages. $^{13}C$-NMR analysis proved that the polysaccharide was mainly a $\beta$-(2,6)-linked levan-type polysaccharide. To investigate the cytotoxicity of the acetone-precipitated levan fractions such as M1, M2, and M3, HepG2, P388D1, U937, SNU-1, and SNUC2A cell lines were screened. Among the cell lines tested, the cytotoxicity of M1- M3 fractions were detected from only SNU-1 and HepG2 cells at the dosage level of $100-800\mu\textrm{g}ml$. The M2 fraction M_r$, 80,000) at 400 $mu{g/ml}$ had the greatest cell growth inhibition (84.6%) on SNU-1, while the M1 $(M_r$, 50,000) at $800\mu\textrm{g}ml$ showed the greatest (46.32%) on HepG2. To obtain more uniform M_r$ fractions of levan, the levan was further fractionated from S1 $(M_r$ 1,000,000) to S5 $(M_r$ 10,000) using gel permeation chromatography. Again, the S1-S5 fractions had strong cytotoxicity on SNU-1 and HepG2 cell lines. The greatest inhibition effects of S4 $(M_r$ 80,000) on SNU-1 and S5 $(M_r$ 10,000) on HepG2 were shown to be 49.5% and 73.0%, respectively. The cytotoxicity of the levan fractions was more effective on SNU-1 than on HepG2. Although the relationship between the Mw and the cytotoxicity was not clear, smaller $M_r$, fractions of levan showed greater growth inhibition effect on the cancer cell lines in general. Therefore, it was indicated that a specific Mw class of levan is responsible for the effective cytotoxicity.
Three more unusual macrolides in addition to concnamycin B were isolated from the mycelium of Streptomyces sp. strain Bal6. These four compounds showed a potent cytotoxity to hunian cancer cell lines, SNU-1 (stomach cancer cell line), SNU-354 (liver cancer cell line), MCF- 7 (breast cancer cell line) and KB-3-1 (oral epidermoid carcinoma cell line). Interestingly, these compounds confered slight differential cytotoxity on RHEK-1, a human epidermal keratinocyte cell line immotalized by AD12-SV40 hybrid virus and RHEK-1/pSV$_{2}$ ras which was resulted from H-ras transfomation of RHEK-1. These compounds were determined to be concanamycin A, conca- namycin E and 0-methyl concanamycin B by NMR and other spectral analysis.
목적 : 사람의 두경부종양 세포주를 대상으로 방사선 조사 후에 일어나는 아포토시스를 측정하여 전체 세포사에서의 중요성 및 방사선감수성과의 관련성을 알아보고자 하였다. 대상 및 방법 : 방사선치료가 주 치료방법인 두경부종양 세포주(PCI-1, PCI-13, SNU-1066)와 정상세포 중 섬유모세포 세포주(LM217), 혈액종양 세포주 중 백혈병 세포주(CCRF-CEM)를 대상으로 하였다. 방사선 조사는 동물실험용 Cs-137 방사선조사기를 사용하였다. 전체 세포사는 집락형성능측정을 이용하였고, 아포토시스의 측정은 annexin-V와 propidium iodide를 이용하는 염색법을 사용하였다. 결과 : 2 Gy 방사선 조사 시의 생존분획인 $SF_2$는 PCI-1, PCI-13, SNU-1066, CCRF-CEM, LM217 세포주에서 각각 0.741, 0.544, 0.313, 0.302, 0.100으로, LM217 세포주가 방사선감수성이 가장 높았고 PCI-1 세포주의 방사선감수성이 가장 낮았다. 두경부암 세포주인 PCI-1, PCI-13, SNU-1066에서는 모두 72시간이 경과한 후 아포토시스지수가 최대치에 도달하였으며, LM217과 CCRF-CEM에서는 24시간 후에 최대치에 도달하였다. 방사선량의 증가에 따라서 전체세포사는 현저하게 증가하였으나 아포토시스지수의 변화는 매우 작았다. 전체 세포사에 대한 아포토시스의 분획(아포토시스분획)은 2 Gy 조사 시 PCI-1, PCI-13, SNU-1066, CCRF-CEM, LM217에서 각각 $46\%,\;48\%,\;46\%,\;24\%,\;19\%$이었고, 6 Gy 조사 시 각각 $20\%,\;33\%,\;35\%,\;17\%,\;20\%$이었다. 아포토시스의 정도와 6 Gy 조사 시의 방사선 감수성과는 일정한 관계를 보이지 않았으나, 2 Gy조사 시 방사선에 감수성이 비교적 높은 세포주가 아포토시스분획이 작았다. 결론 : 본 연구에서 사용한 두경부암세포주에서 방사선 조사 후 아포토시스가 관찰되었으며, 발생 양상이 시간적으로 정상 섬유모세포 및 백혈병세포주와 다른 것을 확인하였다. 또한 아포토시스보다는 다른 종류의 세포사인 증식사가 더 중요하게 작용하고 있음을 알 수 있었다. 아포토시스분획과 2 Gy 조사 시 방사선감수성 사이에 관련 가능성이 제시되었다.
