Altered Complexin Expression in Psychiatric and Neurological Disorders: Cause or Consequence?

  • Complexins play a critical role in the control of fast synchronous neurotransmitter release. They operate by binding to trimeric SNARE complexes consisting of the vesicle protein Synaptobrevin and the plasma membrane proteins Syntaxin and SNAP-25, which are key executors of membrane fusion reactions. SNARE complex binding by Complexins is thought to stabilize and clamp the SNARE complex in a highly fusogenic state, thereby providing a pool of readily releasable synaptic vesicles that can be released quickly and synchronously in response to an action potential and the concomitant increase in intra-synaptic $Ca^{2+}$ levels. Genetic elimination of Complexins from mammalian neurons causes a strong reduction in evoked neurotransmitter release, and altered Complexin expression levels with consequent deficits in synaptic transmission were suggested to contribute to the etiology or pathogenesis of schizophrenia, Huntington's disease, depression, bipolar disorder, Parkinson's disease, Alzheimer's disease, traumatic brain injury, Wernicke's encephalopathy, and fetal alcohol syndrome. In the present review I provide a summary of available data on the role of altered Complexin expression in brain diseases. On aggregate, the available information indicates that altered Complexin expression levels are unlikely to have a causal role in the etiology of the disorders that they have been implicated in, but that they may contribute to the corresponding symptoms.

메타버스 디지털 플랫폼의 메이크업 기능 제안 - 제페토를 중심으로 - (Proposal of Makeup's Function on the Metaverse Digital Platform - Focusing on Zepeto -)

  • With the popularization of 5G networks and the development of AI (artificial intelligence) technology, Metaverse, which creates production capacity by combining virtual space and reality, is attracting attention. In this study, we searched for makeup applications with more than 100 million downloads from October 11, 2020 to November 3, 2020 through the Google Play Store. As a result of the search, four applications were found: YouCam Makeup, YouCam Perfect, Beauty Plus, and Sweet Snap. Based on the functions provided by the four applications, we attempted to suggest makeup functions applicable to Zepeto's avatar. Functions for the eyes (eyeliner, eyelashes, mascara, eye shadow, eye shape, eyebrow shape, lenses, double eyelids), functions for the nose (nose shape), functions for the mouth (lipstick, lip shape, smile function) ) Functions corresponding to the facial contour (contour, skin foundation, blusher, shading, highlighter, face painting, theme makeup) and functions corresponding to the body (body adjustment) were proposed. This study is the first in the beauty field to propose a method of applying the functions of the Metaverse platform as the importance of digital platforms is highlighted, and is the first to propose a makeup function applied to the Metaverse so that it can be used as important basic data in the future.

  • With the development of technology, many changes have emerged in how consumers acquire information without restrictions on their location and time. As changes and interest in digital platforms increase, beauty industry applications are being developed in order to attract customers. Therefore, this study has provided the empirical data necessary for the development and advancement of beauty applications in the future by analyzing preferences for makeup applications according to age. The method of this study involved searching for makeup applications that had been downloaded more than 100 million times through the Google Play Store between October 11, 2020 to November 3, 2020. Four applications were identified: YouCma Makeup, YouCam Perfect, Beauty Plus, and Sweet Snap. The surveyed functions were tested on 100 people in their teens, 20s, 30s, and 40s, with a similar profile of 100 people in their teens, 20s, 30s, and 40s surveyed from January 3, 2022 to April 23, 2022. The functional preferences of the application were investigated, and the results were analyzed through frequency analysis. The results are as follows: the application that received the greatest preference for features by age was YCM, which was found to have subdivided functions that provide users a wider range of choices and more detailed work when compared to other applications which enable photo retouching.

진돗개의 심장사상충 감염 실태 조사 (Seroprevalence of Dirofilaria immitis infections in Jindo dogs)

  • A survey on the seroprevalence of Dirofilaria immitis infection among 2,197 Jindo dogs (371 males, 1,826 females) was conducted from 2011 to 2015 using an antigen test (SNAP 4Dx plus, IDEXX, USA). The overall seroprevalence of D. immitis infection in Jindo dogs was 21.8% (479/2,197), and an increase was observed in the seroprevalence of infection from 19.4% in 2011 to 25.8% in 2015. The infection rates were higher among male dogs (32.1%, 119/371) than female dogs (19.7%, 360/1,826)(P<0.01). The infection rates of D. immitis in Jindo dogs at the age of under 1 years-old, 1 to 2 years-old, 2 to 3 years-old and over 3 years old were 10.7% (76/710), 20.0% (190/950), 36.4% (156/428) and 52.3% (57/109), respectively. The older dogs had higher infection rates than the younger dogs (P<0.01). This study indicates that the prevalence rate of D. immitis in Jindo dogs is higher than previously reported in studies which utilized an antigen test.

Genes expression monitoring using cDNA microarray: Protocol and Application

  • The major issue in the post genome sequencing era is determination of gene expression patterns in variety of biological systems. A microarray system is a powerful technology for analyzing the expression profile of thousands of genes at one experiment. In this study, we constructed cDNA microarray which carries 2,304 cDNAS derived from oligo-capped mouse cDNA library. Using this hand-made microarray we determined gene expression in various biological systems. To determine tissue specific genes, we compared Nine genes were highly-expressed in adult mouse brain compared to kidney, liver, and skeletal muscle. Tissue distribution analysis using DNA microarray extracted 9 genes that were predominantly expressed in the brain. A database search showed that five of the 9 genes, MBP, SC1, HiAT3, S100 protein-beta, and SNAP25, were previously known to be expressed at high level in the brain and in the nervous system. One gene was highly sequence similar to rat S-Rex-s/human NSP-C, suggesting that the gene is a mouse homologue. The remaining three genes did not match to known genes in the GenBank/EMBL database, indicating that these are novel genes highly-expressed in the brain. Our DNA microarray was also used to detect differentiation specific genes, hormone dependent genes, and transcription-factor-induced genes. We conclude that DNA microarray is an excellent tool for identifying differentially expressed genes.

The Effects of Lead Exposure on Glutamatergic Nervous System in Rat Cerebellar Cells

  • Changes in glutamate release and uptake on cerebellar cells after the chronic exposure to lead were investigated. Rats were received 0.25% lead acetate in drinking water from the beginning of the pregnancy. The control group was given 0.125% sodium acetate in drinking water. The cerebellar cells from 7 or 8 day-old pups were cultured. Amino acid release from cerebellar granule cells and the glutamate uptake into cerebellar glial cells were measured using HPLC-ECD. Basal glutamate release and NMDA-induced glutamate release didn't show significant difference. However, the other amino acids in the granule cells obtained from lead exposed pups were less released than the control after the stimulation by NMDA (50$\mu$M). SNAP-induced (50$\mu$M) glutamate release was significantly reduced in granule cells prepared from lead exposed pups. The basal glutamate uptake in glial cells didn't show any difference. However, the uptake in glial cells prepared from lead exposed pups was significantly less blocked by PDC (24$\mu$M) compared to the control group. These results indicate that lead exposure to the mother might affect the Excitatory amino acid system during the development of the offspring.

국내 신문 인쇄의 색재현 특성에 관한 연구 (A Study on The Color Reproduction Properties of Domestic Newspaper Prints)

  • Web offset newspaper prints is the most widely used printing process in domestic printing industry, but there are no industry-wide specifications for press control to assure consistent quality across newspaper printing plants. As newspaper printing becomes more of a commodity and less of an art, it is necessary to develop the print quality specification standard suitable for Korea web offset newspaper printing field. There are a few commonly employed printing standards such as, SNAP (Specification for Non-Heatset Publication) of the US, IFRA(INCA-FIEJ Research Association), J-color (Japan printing machinery association). And there exists KS for domestic standards, however, since it has been directly imported from ISO standards, there are discrepancies in printing materials and media. This paper aims to examine the color reproduction quality of domestic web offset newspaper prints by comparing with foreign standards. Printing experiments on this study have been performed at one of the domestic newspaper printing companies. The color reproduction properties of the web offset newspaper prints are measured by means of densitometry and colorimetry. These measures are evaluated comapratively with the international standards by means of analyzing, the major factors that determine the color reproduction quality of web offset newspaper prints.

Effects of Minor Arginyl tRNA and Isoleucyl tRNA on the Expression of Clostridium botulinum Neurotoxin Light Chain in Escherichia coli

  • Botulinum neurotoxin type A (BONT/A) is an extremely potent toxin, which is produced by Clostridium botulinum. The light chain of this protein (BONT/A LC), which is known as a zinc endopeptidase, cleaves SNAP-25 involved in the exocytosis process. In this work, the expression of recombinant BoNT/A LC in E. coli is described. The BONT/A LC gene of C. botulinum contains a high frequency of the arginine AGA and isoleucine ATA codons that are rarely used in genes of E. coli, hampering the translation of recombinant protein. The argD and ilex tRNA genes were cloned into pACYC184 vector, resulting in pAAD131X plasmid. The translational stress of the toxin gene related to codon bias was reversed by fupplernentation of the AGA arginyl tRNA of T4 phage and AUA isoleucyl tRNA of E. coli. This system may be applicable for the expression of a variety of AT-rich heterologous genes in E. coli.

Prion Protein Does Not Interfere with SNARE Complex Formation and Membrane Fusion

  • In prion disease, spongiform neurodegeneration is preceded by earlier synaptic dysfunction. There is evidence that soluble N-ethylmaleimide sensitive factor attachment receptor (SNARE) complex formation is reduced in scrapie-infected in vivo models, which might explain this synaptic dysfunction because SNARE complex plays a crucial role in neuroexocytosis. In the present study, however, it is shown that prion protein (PrP) does not interfere with SNARE complex formation of 3 SNARE proteins: syntaxin 1a, SNAP-25, and synaptobrevin. Sodium dodecyl sulfate-resistant complex formation, SNAREdriven membrane fusion, and neuroexocytosis of PC12 cells were not altered by PrP. Thus, PrP does not alter synaptic function by directly interfering with SNARE complex formation.

  • Background: Nitric oxide (NO), a cytotoxic molecule is produced in various tissues including tumor cells during interleukin-2 (IL-2) therapy . Lymphokine-activated killer (LAK) cells are induced during IL-2 therapy, and have cytotoxic activity against tumor cells. The current study investigated the effects of NO synthesized in target cells or exposure of target cells to NO on the sensitivity of target cells to LAK cell cytotoxicity. Methods: Cytotoxicity was measured using 4 h chromium release assays. LAK cells which were induced by a 4 day incubation of BALB/c mouse splenocytes with IL-2 (6,000 IU/mL) were employed as effector cells. RD-995 skin tumor cells originated from a C3H/HeN mouse were employed as target cells. NO synthesis in target cells was induced by a 24 h incubation of RD-995 cells with $IFN{\gamma}$ (25 U/mL), TNF (50 U/mL) and IL-1 (20 U/mL). S-nitrosyl acetylpenicillamine (SNAP), an NO donor, was used to expose target cells to NO. $N^G$-monomethyl-L-arginine (MLA) and carboxy-PTIO were added during cytotoxicity assays to inhibit NO synthesis, and to scavenge NO produced by target cells, respectively. Results: Sensitivity of NO-producing RD-995 cells to LAK cell cytotoxicity was decreased by addition of MLA and carboxy-PTIO during cytotoxicity assays. However, the two reagents had no effect on the sensitivity of non-NO-producing RD-995 cells. Pretreatment of RD-995 target cells with SNAP increased the sensitivity in comparison with untreated cells. Conclusions: Sensitivity of target cells to LAK cell cytotoxicity is increased by target cell NO synthesis or exposure to NO. Further studies are needed to evaluate whether these in vitro results have relevance to in vivo phenomena.
