Antibiotic Resistance and R Plasmids in Edwardsiella tarda (양만장 사육조에서 분리한 Edwardsiella tarda의 약제 내성과 R Plasmid)

  • A total 103 strains of Edwardsiella tarda was isolated from eel culture-ponds and examined for drug resistance, distribution and transferabilities of R plasmids. The drugs used were lincomycin(LM), penicillin(PM), sulfamethazine(SF), sulfadimethoxine(SD), cephalosprin(CP), chloramphenicol(CH), streptomycin(SM), oxytetracycline(OT), ampicillin(AP), oxolinic acid(OA), kanamycin(KM), amikacin(AK), gentamicin(GM) and enrofloxacin(EF). Two strains were resistant to the all drugs used, and all isolates were multiply resistant to drugs(at least 8 among 14 drugs), mostly restricted to LM(103 strains), PM(103 strains), SD(103 strains), SF(103 strains), CP(102 strains), CM(101 strains), SM(100 strains), OT(94 strains), AM(92 strains), OA(80 strains), KM(60 strains), AK(30 strains), GM(19 strains) and EF(14 strains), in combination at high degree showing 34 different drug resistant patterns. The most frequently encountered patterns were LM PM SD SF CP CH SM OT AP OA KM(22 strains, 22.4%) followed by LM PM SD SF CP CH SM OT AP OT(12 strains, 11.7%). LM PM SD SF CP CH SM OT AP(7 strains, 6.8%), LM PM SD SF CP CH SM OT AP OA KM AK GM(6 strains, 5.8%) and LM PM SD SF CP CH SM OT AM OX KM AK GM(6 strains, 5.8%). Transfer experiment of drug resistance showed that of 103 resistant strains, 100 strains(97%) transferred part or all resistance to all drugs, indicating that most isolates carried conjugally transferrable R plasmids determining multiple drugs. The most frequently observed transferarble R plasmids were LM PM SD SF CP CH SM OT AP OA(10 strains), LM PM SD SF CP CH SM OT AP OX KM(7 strains) and LM PM SD SF CP CH AP OA(7 strains). These results sugested that eel culture-ponds were highly contaminated with different strains of Edwardsiella tarda, and that contaminated bacteria might be highly multiple resistant strains to drugs, carring transferable R plasmids.

Comparison of the Marginal and Internal Gap of Metal Coping according to Processing Method of Dental CAD/CAM System (치과 캐드캠 시스템의 가공 방식에 따른 금속 코핑의 적합도 비교)

  • The purpose of this study was to evaluate the marginal and internal gap of metal coping fabricated using additive manufacturing (AM) group and subtractive manufacturing (SM) group by dental computer-aided design (CAD)/computer-aided manufacturing (CAM) systems. Twenty same cases of stone models of abutment teeth 16 by the universal numbering system were manufactured and scanned. Ten metal copings of control group were fabricated using SM and ten metal coping of experimental group were fabricated using AM. Marginal and internal gap of copings were measured using the silicone replica technique and digital microscope (${\times}140$). The data were analyzed using IBM SPSS 21.0 Statistical Software for independent samples t-test (${\alpha}=0.05$). Mean${\pm}$ standard deviation (SD) of marginal and internal gap total size of SM group was $101.00{\pm}40.33{\mu}m$ of AM group was $83.61{\pm}40.37{\mu}m$. Mean${\pm}$SD of marginal and internal gap total size of SM group was significantly greater than that of AM group (p<0.05). This study showed that AM metal copings had a better marginal and internal gap than SM metal copings.

Transfer of R plasmids of Bacterial Isolates and Their Cloned R Genes in Natural Wastewater Environments (I) -Cloning of $Km^rCm^r$Gene- (하폐수의 자연환경에서 R plasmid와 재조합 유전자의 전이특성( I ) -$Km^rCm^r$유전자의 클로닝-)

  • In order to study the transfer of antibiotics resistance genes of the genetically cloned bacteria in water environments, DK1 strain, which is resistant to kanamycin (Km), chloramphenicol (Cm), streptomycin (Sm), and sulfadiazine (Su), was selected from the Gram-negative bacterial isolates from wastewater. One of 4 plasmids harboured in the DK1 strain was found to possess Km$^{${\gamma}$}$Cm$^{${\gamma}$}$ gene and be about 68 kb in size, and it was designated as pDK101. The plasmid of pDK101 was also found to have 16, 32, and 6 restriction sites for EcoRI. .PstI, and SalI, respectively. From the digestion fragments of pDK101 plasmid and pKT230 used as a vector by EcoRI restriction endonuclease, pDT309 and pDT529 were constructed as chimeric plasmids which possess Km$^{${\gamma}$}$Cm$^{${\gamma}$}$ gene and are 30.9 and 52.9 kb in size, respectively. When the chimeric plasmids were trasformed into E. coli C600 or E. coli HB101, transformants of DKC601, DKC602, DKH102, and DKH103 were obtained as cloned bacterial cells. The Km$^{${\gamma}$}$Cm$^{${\gamma}$}$ genes were well expressed in those cloned cells and the chimeric plasmids were clearly detected in the cloned cells of DKC601 and DKH103.

Soil Salinity and Salt Spray Drift Tolerance of Native Trees on the Coastal Windbreaks in the South-Sea, Korea (한국 남해안방풍림 자생수종의 내염성 및 내조성 수종 선발)

  • This study was carried out to investigate the soil salinity and salt spray drift of the indigenous windbreak trees, and its main purpose was to provide basic data for the selection of salt-tolerant trees in the saline coastal region in the South Sea of Korea. The soil salinity($EC_{1:5}$)was $0.18dSm^{-1}$, which was an average degree of the whole areas of investigation whose salinity degree ranged from $0.05dSm^{-1}$ to $0.58dSm^{-1}$. The level of soil salinity gradually decreased as it moved farther inland, except the belt I. The level of decreasing soil salinity was found to be in the following order: belt II, belt III, belt I, belt IV. The degree of soil salinity was $EC_{1:5}$ $0.22dSm_{1:5}$, $0.22dSm_{1:5}$ $0.19dSm^{-1}$ and $0.13dSm^{-1}$ respectively. The total 110 taxa, which consisted of 45 families, 74 genus, 101 species, and 9 varieties, were found to be tolerant to both soil salinity and salt spray drift. The trees that grow in the highest degree of salinity($EC_{1:5}$ $0.50dSm^{-1}$)were Parthenocissus tricuspidata(Siebold & Zucc.), Planch and Lonicera japonica Thunb. The next group of trees that grow in the high degree of salinity ranging from $EC_{1:5}$ 0.41 to $0.50dSm^{-1}$ was Cudrania tricuspidata(Carr.) Bureau ex Lavall$\acute{e}$e, Rubus parvifolius L., Zanthoxylum schinifolium(Siebold & Zucc.), Hedera rhombea(Miq.) Bean., Robinia pseudoacacia L., Quercus serrata Thunb., Callicarpa dichotoma(Lour.) K. Koch, and so on. The woody species which grew in the entire belts were Pueraria lobata(Willd.) Ohwi and Vitis flexuosa Thunb., and Vitex rotundifolia L. f. which was known to be highly tolerant to salt spray drift was found only in belt I. The woody species with high important value(IV) were Zelkova serrata(Thunb.) Makino., Celtis sinensis Pers., Koelreuteria paniculata Laxmann, Mallotusjaponicus(Thunb.) Muell. Arg., Trachelospermum asiaticum(Siebold & Zucc.) NAKAI, and Pueraria lobata(Willd.) Ohwi. These species were classified as native windbreak trees that are comparatively more tolerant to salt spray drift than other kinds.

Construction of the Intelligence Stress Predictor for Compression Strength Evaluation (압축강도 평가를 위한 지능형 응력예측기 구축)

  • This work is concerned with construction of the intelligence stress predictor far compression strength evaluation using neural network-ultrasonic waves. The contact pressure in jointed plates was measured by using ultrasonic technique. Neural network is used to evaluate and predict contact pressure from the results of the calibration curves. The organized neural system was leaned with the accuracy of 99%, as a result of learning the ultrasonic echo ratio to the contact pressure measurement between SM45C and STS410 materials. And it could be evaluated and predicted with the accuracy of 90% in the evaluation of ultrasonic echo ratio difference in the same surface roughness and contact pressure, and 85% in the prediction of virtual ultrasonic echo ratio. Thus the proposed stress predictor is very useful for the evaluation and prediction of the contact pressure between SM45C and STS410 materials.

A Study on Mechanical Properties and Friction Weldability of SKH51 and SM45C (SKH51/SM45C의 마찰용접특성에 관한 연구)

  • The present study examined the mechanical properties of the friction welding of shaft made of SKH51 and SM45C, of which the diameter is 12mm. Friction welding was done at welding conditions of 2,000rpm, friction pressure of 104MPa, upset pressure of 134MPa, friction time of 0.5sec to 2.5sec by increasing 0.5sec, upset time of 2 seconds. Under these conditions, a tensile test, a bending test, a shear test, a hardness test and a microstructure of weld interface were studied. When the friction time was 1.0 second under the conditions, the maximum tensile strength of the friction weld observed to be 963MPa, which is 89% the tensile strength of SKH51 base metal and 101% of the tensile strength of SM45C base metal. When the friction time was 1.0 seconds under the conditions, the maximum bending strength of the friction weld happened to be 1,647MPa, which is 78% the bending strength of SKH51 base metal(2,113MPa) and 87% of the bending strength of SM45C base metal(1,889MPa). When the friction time was 1.0 seconds under conditions, the maximum shear strength of the friction weld was observed to be 755MPa, which is 92% the shear strength of SKH51 base metal and 122% of the shear strength of SM45C base metal. According to the hardness test, the hardness distribution of the weld interface varied from Hv282 to Hv327. HAZ was formed from the weld interface to 1.2mm of SKH51 and 1.6mm of SM45C. Upon examination it was found that the microstructure became finer along with increase of friction revolution radius.

The Sedative Effects of Submucosal Midazolam in Children (소아 진정 시 구강 협점막하로 투여한 Midazolam의 효과에 관한 연구)

  • Purpose: This retrospective study assessed the effectiveness of submucosal (SM) midazolam (M), oral chloral hydrate (CH), hydroxyzine (H), and nitrous oxide (N) 99 uncooperative pediatric dental patients over 182 sedation sessions. Methods: Selection criteria included good medical health (ASA I), from 2 to 6 years of age, uncooperative behavior, and the need for restorations and dental surgery. The children who was administerated with CH H, N was Group 1 (48 case). The children who was administerated with CH, H, N, M was Group 2 (51 case). All the patients received oral CH (50-70 mg/Kg), H (1 mg/Kg) and N (less than 50%). Additionally the Group 2 received SM midazolam (0.1-0.2 mg/kg). Behavior response was rated as quiet (Q), crying (C), movement (M), or struggling (S) every 5 minutes through operative procedures. Behavior response rating was assigned to Houpt scale of Overall behavior. Data was analyzed using Chi-Square Test. Results: The Group 2 had more effective (88.2%) than the Group 1 (68.8%)(P = 0.05). Also, vomiting response decreased in the Group 2 (94.4%) than those with oral administration of chloral hydrate alone without SM midazolam (77.1%)(P = 0.05). Conclusions: SM midazolam improved the quality of sedation and vomiting response.

  • the vector, pGUR19, for Rhizobium gene manipulation, was constructed by combining the replication and mobilization function of indigenous multicopy plasmid from Acacia(Robinia pseudoacacia L.) Rhizobia sp86 with E. coli cloning vehicle, pBR322. The vector could be efficiently mobilized by RP4 tra function incorporated into chromosome of E. coli named SM10 and efficiently transferred to various gram negative hosts including Rhizobium and Afrobacterium by transformation. Mobilization frequency of the constructed vector was ranged from $1.2\times 10^{-2}$ (E.coli HB 101) to $4.6\times 10^{-4}$ (A. tumefaciens 15955) and transformation frequency was ranged from $5.4\times 10^{-7}$(E. coli HB101) to $1.2\times 10^{-10}$ (A. tumefaciens 15955). The vector, pGUR19, was stably replicated and maintained in a variety of Rhizobium and Agrobacterium.

The Study of Curing Day Reduction by Step Curing of HTPB/AP Propellant (HTPB/AP계열의 고체 추진제의 Step 경화 방법을 통한 경화일(기간) 단축)

  • In this paper, step-curing, which includes the change of curing temperature on the curing process, was applied to reduce curing day of HTPB/AP based propellant. This study targets the improvement of productivity of HTPB/AP based solid rocket motor. Comparison of mechanical properties of propellant resulted in the change of normal curing condition (60℃, 5 days) to step-curing condition (60℃, 1 day / 65℃, 3 days). Post-cure test was conducted to determine the impact on the shelf life of the solid rocket motor. The aging characteristics of propellants were analyzed by measuring mechanical properties and thermal expansion factor. To step-cured propellant, accelerated aging test was performed for 12 weeks, followed by tensile test. Sm(bar) and Em(%) were higher than 8 bar and 40% each, showing excellent mechanical properties.
