Efficacy of Brucella abortus strain RB51 vaccine in Korean mongrel dogs against virulent strains of B. abortus biotype 1 and B. canis

  • This study was performed to test the hypothesis that Brucella abortus strain RB51 (SRB51) might protect Korean indigenous mongrel dog against challenge with either virulent B. abortus biotype 1 or B. canis. A total of 12 Korean mongrel dogs were divided into four groups (Group A, B, C and D). Dogs belonging to Group A and C were inoculated subcutaneously with $1{\times}10^9$ CFU of SRB51 in 1ml of sterile phosphate buffered saline (PBS). Dogs of Group B and D were inoculated subcutaneously with 1ml of sterile PBS as control. At 12 weeks post vaccination, dogs of Group A and B were challenged by oral inoculation of virulent strain of B. canis ($5.0{\times}10^9$ CFU) and dogs of Group C and D were challenged by oral inoculation of virulent strain of B. abortus biotype 1 ($4.4{\times}10^{10}$ CFU). The serum antibodies titers in all dogs were monitored at regular interval for eight weeks after challenge (AC) by standard tube agglutination test, plate agglutination test, rose bengal test, 2-mercaptoethanol rapid slide agglutination test and 2-mercaptoethanol tube agglutination test. No antibody titers in Group A and C was detected. Also, the challenge strains were not found from blood of all dogs of Group A and C from 1 week AC till the end of the experiment by culture and modified AMOS-PCR, whereas B. canis and B. abortus challenge strains were detected from blood of Group B and D, respectively. In addition, neither of two challenge bacteria was recovered from liver, spleen, kidneys, lymph nodes and reproductive tracts of Group A and C dogs after postmortem. However, B. canis and B. abortus challenge strains were isolated from these tissues of Group B and D, respectively. These data suggest that SRB51 could be a promising vaccine candidate for immunizing dogs to control canine brucellosis caused by B. canis or B. abortus.

Effect of Conidial Number and Nutrition on the Germination of Conidia in Septoria glycines (분생포자수 및 영양상태가 대두갈색무늬병균의 분생포자 발아에 미치는 영향)

  • Conidial germination of Septoria glycines Hemmi, brown spot fungus of soybean, was studied by slide germination test. Poor conidial germination of S. glycines was observed on sterile distilled water, but potato dextrose agar(PDA) and distilled water floated with soybean leaf disc furnished a satisfactory medium for conidial germination. Exogenous supply of carbon source was essential for conidial germination, while phosphorous and potassium were not evident as that for carbon. Soluble starch was the most suitable as a carbon source for conidial germination and followed by D-glucose, D-galactose and lactose in that order. Maximum germination was attained in the $5\times10^{-2}mol$. concentration of glucose. Germination was decreased with increment of conidial concentration and was almost completely suppressed in the density of 10,000 conidia per $mm^2$. It suggested existing a self-inhibitor(s). Non-washed conidia germinated more than washed conidia and this was obvious when the conidia density was over $2\times10^3$ conidia per $mm^2$ on the dry agar block.

  • PTCR(Positive Temperature Coefficient of Resistivity) thermistor in thin film BaTiO$_{3}$ system was prepared by using radio frequency(13.56 MHz) and DC magnetron sputter equipment. Polycrystalline, surface structure, and R-T(Resistivity-Temperature) characteristics of the specimens were measured by X-ray diffraction(D-Max3, Rigaku, Japan), SEM(Scanning Electron Microscopy: M.JSM84 01, Japan), and insulation resistance measuring system (Keithley 719), respectively. Thin films characteristics of the thermistor showed different properties depending on the substrate even with the same sputtering condition. The thin film formed on the A1$_{2}$O$_{3}$ substrate showed a good crystalline and a low resistivity at below curie point. However, the thin films prepared on slide glass and Si wafer were amorphous. The thicknesses of the three samples prepared under the same process conditions were 700[.angs.], 637.75[.angs.], and 715[.angs.], respectively.

  • To fabricate low field magnetic sensors using magnetoresistance(MR) effects, we deposited thin layers of $600{\AA}$ in thickness of Ni-Co(0.7Ni-0.3Co) alloy on slide glasses. In the layers we ordered 4 arms of the fullbridge sensors in the shape of grid structure to be inclined at an angle of $45^{\circ}$ to main axis and made the areal rate increase to 67%. While the response characteristics of the fabricated sensors had good linearity in the magnetic field of ${\pm}0.5mT$ ranges, the white noise was 0.2 nV and the voltage sensitivity was 7.6 $nV/{\mu}T$.

An Investigation of the Control of Two-Spotted Spider Mites (Tetranychus urticae Koch) Resistant to Organo-Phosphates (유기인제저항성 점박이응애(Tetranychus urticae Koch)에 관한 연구)

  • The study involved determination of resistance levels of spider mites ta argano-phosphates using topical application and slide dip techniques; laboratory serening tests of alternative acaricides using an O/P resistant strain and a field trial of the screened materials. 1. Strains of Tetranychus were from Timaru(TR), Havelock Narth (HNR), Lincaln (LN). Germany (GR, GN). Comparisons of the resistant strains and normal strains at the LD50 and LC50 levels were as follows : (a) Using the topical application tochnique; with Parathian. resistant levels of the GR. TR and HNR strains of T. urticae were respeativuly, 1035. 484 and 452 times as resistant' as the LN strain. (b) Using the slide dip technique; with Phosdrin, resistant of GR, TR and HNR strains of T. urticae were 635, 274 and 266 times greater respeativuly, than the GN strain. 2. The laboratory sereaning tests were carried out far their contact plus stomach and residual effect to assess the toxicities of eleven alternative materials which would be used far control of O/P resistant strain of T. urticae. The acaricide groups represented were 3 organo-chlorines (Spidex, Kelthane and C 8514), 2 nitrophenyls (UC 19786 and Morocide), 2 cyclic carbonates(Eradex and Morestan). I carbamate (UC2004 7A), 1 mixture of carbamate and orano-chlorine and 2 other chemicals (C 8677 and M2527). From all acaricide tested. Kelthane and Morocide were the most effective, folowed by Spidex and M2527. Morestan, C8514. C8677 and RS 143 were intermediate, but Eradex, UC 19786 and UC 20046A were poor. 3, The number of sapmles required for estimation of the population in the field evaluation of acaricidal effects was one giving the highest practical precision. It was decided, after preliminary sampling trials. to use samples of 30 leaves per replicate which gave a $5.7\%$ standard error. 4. In the field trials, Morocide applied at the $0.05\%\;and\; 0.04\%$ a. i. conc. to black currant trees gave excellent control of O/P resistant population of T. urticae for about 12 days, but Morocide 0.025 and Kel thane $0.02\%$ a. i. cone. gave efficient control for about 6 days. In other words. first applications of Kel thane ane Moroeide gave very high degrees of control of O/P resistant population of the two-spotted spider mite. However, the results indicate that secondary application would sometimes be necessary. There was no foliage damage of black Currants and strawberries by either acaricides at the concentrations used. The Effects of Slide-Covered of Slide-Covered Contemporary Charging Automated External Defibrillator on Rapidity and Convenience of Defibrillation (슬라이드 커버 동시충전 자동심장충격기가 제세동의 신속성과 편의성에 미치는 효과)

  • This study compares the rapidity and subjective convenience of T-AED and SC-AED for health care providers and the general public. Subjects were randomly allocated to T-AED (n=77) and SC-AED (n=79) groups. Each group conducted defibrillation, with subsequent measurement of the rapidity of defibrillation in peri-shock pause, pre-shock pause, hesitation pause, and post-shock pause. Defibrillation and chest compression delay times for both equipment were analyzed by t-test. On conclusion of the experiment, subjects answered a questionnaire on the subjective convenience of defibrillation, as measured for confidence, convenience, and clear decision. Comparisons of subjective convenience analyzed by t-test revealed significantly shortened peri-shock pause (11.22s), pre-shock pause (11.04s), and hesitation pause (2.15s) in the SC-AED group, as compared to the T-AED group (p<0.001). However, no significant differences were observed for post-shock pause values. Comparing subjective convenience, confidence (T-AED: 7.62±1.25VAS vs. SC-AED: 7.80±0.98VAS, p=0.358) was not significant, whereas convenience (T-AED: 7.05±1.36VAS vs. SC-AED: 8.95±0.89VAS, p<0.001) and clear decision (T-AED: 6.58±1.73VAS vs. SC-AED: 9.08±0.98VAS, p=0.001) showed statistically significant differences. Our results indicate that compared to T-AED, SC-AED has significantly shortened pauses. Moreover, it is more convenient for the user, and significantly aids in clear decisions.

Evaluation of IH-1000 for Automated ABO-Rh Typing and Irregular Antibody Screening (ABO 및 RhD 혈액형 검사와 비예기항체 선별검사를 위한 자동화장비 IH-1000의 평가)

  • Background: Despite modern advances in laboratory automated medicine, work-process in the blood bank is still handled manually. Several automated immunohematological instruments have been developed and are available in the market. The IH-1000 (Bio-Rad Laboratories, Hercules, CA, USA), a fully automated instrument for immunohematology, was recently introduced. In this study, we evaluated the performance of the IH-1000 for ABO/Rh typing and irregular antibody screening. Methods: In October 2011, a total of 373 blood samples for ABO/Rh typing and 303 cases for unexpected antibody screening were collected. The IH-1000 was compared to the manual tube and slide methods for ABO/Rh typing and to the microcolumn agglutination method (DiaMed-ID system) for antibody screening. Results: For ABO/Rh typing, concordance rate was 100%. For unexpected antibody screening, positive results for both column agglutination and IH-1000 were observed in 10 cases (four cases of anti-E and c, three of anti-E, one of anti-D, one of anti-M, and one of anti-Xg) and negative results for both were observed in 289 cases. The concordance rate between IH-1000 and column agglutination was 98.7%. Sensitivity and specificity were 90.9% and 99.3%, respectively. Conclusion: The automated IH-1000 showed good correlation with the manual tube and slide methods and the microcolumn agglutination method for ABO-RhD typing and irregular antibody screening. The IH-1000 can be used for routine pre-transfusion testing in the blood bank.

Stellite bearings for liquid Zn-/Al-Systems with advanced chemical and physical properties by Mechanical Alloying and Standard-PM-Route

  • An important business-field of world-wide steel-industry is the coating of thin metal-sheets with zinc, zinc-aluminum and aluminum based materials. These products mostly go into automotive industry. in particular for the car-body. into building and construction industry as well as household appliances. Due to mass-production, the processing is done in large continuously operating plants where the mostly cold-rolled metal-strip as the substrate is handled in coils up to 40 tons unwind before and rolled up again after passing the processing plant which includes cleaning, annealing, hot-dip galvanizing / aluminizing and chemical treatment. In the liquid Zn, Zn-AI, AI-Zn and AI-Si bathes a combined action of corrosion and wear under high temperature and high stress onto the transfer components (rolls) accounts for major economic losses. Most critical here are the bearing systems of these rolls operating in the liquid system. Rolls in liquid system can not be avoided as they are needed to transfer the steel-strip into and out of the crucible. Since several years, ceramic roller bearings are tested here [1.2], however, in particular due to uncontrollable Slag-impurities within the hot bath [3], slide bearings are still expected to be of a higher potential [4]. The today's state of the art is the application of slide bearings based on Stellite\ulcorneragainst Stellite which is in general a 50-60 wt% Co-matrix with incorporated Cr- and W-carbides and other composites. Indeed Stellite is used as the bearing-material as of it's chemical properties (does not go into solution), the physical properties in particular with poor lubricating properties are not satisfying at all. To increase the Sliding behavior in the bearing system, about 0.15-0.2 wt% of lead has been added into the hot-bath in the past. Due to environmental regulations. this had to be reduced dramatically_ This together with the heavily increasing production rates expressed by increased velocity of the substrate-steel-band up to 200 m/min and increased tractate power up to 10 tons in modern plants. leads to life times of the bearings of a few up to several days only. To improve this situation. the Mechanical Alloying (MA) TeChnique [5.6.7.8] is used to prOduce advanced Stellite-based bearing materials. A lubricating phase is introduced into Stellite-powder-material by MA, the composite-powder-particles are coated by High Energy Milling (HEM) in order to produce bearing-bushes of approximately 12 kg by Sintering, Liquid Phase Sintering (LPS) and Hot Isostatic Pressing (HIP). The chemical and physical behavior of samples as well as the bearing systems in the hot galvanizing / aluminizing plant are discussed. DependenCies like lubricant material and composite, LPS-binder and composite, particle shape and PM-route with respect to achievable density. (temperature--) shock-reSistibility and corrosive-wear behavior will be described. The materials are characterized by particle size analysis (laser diffraction), scanning electron microscopy and X-ray diffraction. corrosive-wear behavior is determined using a special cylinder-in-bush apparatus (CIBA) as well as field-test in real production condition. Part I of this work describes the initial testing phase where different sample materials are produced, characterized, consolidated and tested in the CIBA under a common AI-Zn-system. The results are discussed and the material-system for the large components to be produced for the field test in real production condition is decided. Outlook: Part II of this work will describe the field test in a hot-dip-galvanizing/aluminizing plant of the mechanically alloyed bearing bushes under aluminum-rich liquid metal. Alter testing, the bushes will be characterized and obtained results with respect to wear. expected lifetime, surface roughness and infiltration will be discussed. Part III of this project will describe a second initial testing phase where the won results of part 1+11 will be transferred to the AI-Si system. Part IV of this project will describe the field test in a hot-dip-aluminizing plant of the mechanically alloyed bearing bushes under aluminum liquid metal. After testing. the bushes will be characterized and obtained results with respect to wear. expected lifetime, surface roughness and infiltration will be discussed.

A Study on Torch Path Generation for Laser Cutting Process (레이저 절단공정에서의 토지경로 생성에 관한 연구)

  • This paper addresses the problem of a torch path generation for the 2D laser cutting of a stock plate nested with resular or irregular parts. Under the constaint of the relative positions of parts enforced by nesting, the developed torch path algorithm generate feasible cutting path. In this paper, the basic object is a polygon( a many-slide figure) with holes. A part may be represented as a number of line segments connected end-to-end in counterclockwise order, and formed a closed contour as requied for cutting paths. The objective is to tranverse this cutting contours with a minimum path length. This paper proposes a simulated annealing based dtorch path algorithm, that is an improved version of previously suggested TSP models. Since everypiercing point of parts is not fixed in advance, the algorithm solves as relazed optimization problem for the constraint, thich is one of the main features of the proposed algorithm. For aolving the torch path optimization problem, an efficient generation mechanism of neighborhood structure and as annealing shedule were introduced. In this way, a global solution can be obtained in a reasonable time. Seveeral examples are represented to ilustrate the method.

A STUDY ON THE MAXILLARY INTERINCISAL DIASTEMA (상악중절치간(上顎中切齒間) 정중이개(正中離開)에 관(關)한 연구(硏究))

  • The purpose of this study was to investigate the interrelationship of the maxillary interincisal diastema, spacing and crowing to the type of superior labial frenum and the type of intermaxillary suture by analyzing histories, intraoral radiographs, orthopantomographs, intraoral slide films, and dental casts. The data for this study were compiled from 500 outpatients of the Department of Orthodontics, Infirmary of Dental College, Seoul National University. The following conculusions were obtained: 1. The occurrence of maxillary interincisal diastema in the permanent dentition decreased rapidly compared with that in the mixed dentition. And there was no sex difference in the occurrence of diastema in the both dentitions. 2. Frenum attachment remained relatively constant between the two dentitions but assumed a higher level in the permanent dentition than in the mixed dentition. 3. Except suture type D which was not yet fused completely, there was no significant difference in the distribution of suture type between the mixed dentition and the permanent dentition. 4. Frenum and suture type were highly related; frenum types 5 and 6 were associated with suture types III and V. 5. In the permanent dentition, frenum type 5 and 6 and suture types III and IV occurred more frequently in the spacing group than in the normal or crowding group. 6. The relationship between the diastema and frenum types 5 and 6 and sture types III and IV showed a strong correlation.
