Objectives : This study was designed to evaluate the effect of Hasuogamibang (HGB) on hyperlipidemia and antioxidant activity. Methods : For this study, we divided four groups of rats (normal WKY group, normal SHR group, high cholesterol diet and saline-treated SHR group, high cholesterol diet and HGB-treated SHR group), and observed the change of body weight, weight of liver, cholesterol, triglyceride, glucose, albumin, histologic change of liver and aorta, lipid peroxidation and antioxidant activity of liver tissue, and cholesterol gene revelation control efficiency. Results : Total-cholesterol, LDL-cholesterol, triglycerides were decreased significantly by HGB. However, HDL-cholesterol increased significantly. The tissue of liver and aorta were controlled defect by HGB on histologic study. Lipid peroxidation and SOD of liver tissue was decreased significantly by HGB. Gene revelation of ACAT and HMG CoA reductase in hepatic tissue was decreased significantly by HGB. Conclusion : This study suggests that HOB is significantly effective on hyperlipidemia and antioxidant activity.
Our previous study showed that in vivo treatment of spontaneously hypertensive rats (SHR) with protopanaxatriol ginsenosides (PPT) reduces the blood pressure and inhibits the con- tractions induced by endothelium-derived contracting factor (prostaglandin endoperoxide ($PGH_2$) and superoxide anion) in aorta isolated from SHR. The aim of the present study is to examine whether PPT improves endothelial functions in the isolated thoracic aorta of SHR in vitro. Treatments of aortic rings with PPT, purified ginsenoside $Rg_1$ ($Rg_1$) or indomethacin normalized endotheliuln-dependent relaxation to acetylcholine, but not with protopanaxadiol ginsenosides (PPD) and purified ginsenoside Rb1 (Rb1). The effects of PPT were dose-dependent. PGH,- and oxygen free radical-inducted contractions in rat aorta without endothelium were inhibited by PPT or $Rg_1$, but not by PPD or $Rb_1$. Contractions induced by PGF2$\alpha$, U-46619, a stable thromboxane A2 agonist or KCI (60 mM) were not inhibited by PPT, $Rg_1$ or $Rb_1$. These findings demonstrate that PPT but not PPD scavenges the oxygen-derived free radicals and/or antagonize the effects of $PGH_2$ in the vascular smooth muscle and may explain the hypotensive effect of ginseng in the SHR.
The characteristics of voltage-dependent ^{45}Calcium$ uptake and norepinephrine release as factors controlling neural activities in the hypothalamus which is an important regulatory site for cardiovascular function wre studied. Two groups of animals : male spontaneously hyperterisive rat (SHR) and age-matched nomotensive wistar rat (NW) were used in this study. Animals at 4, 6 and 16 weeks of age were sacrificed by decapitiation and the hypothalamus was dissected out. Voltage-dependent calcium uptake and norepinephrine release were determined from hypothalamic synaptosomes either in low potassium (5 mM) or high potassium (41 mM) stimulatory conditions by using ^{45}Ca$ isotope and HPLC-ECD techniques. Degrees of voltage-dependent ^{45}Calcium$ uptake and norepinephrine release evoked by calcium uptake in the hypothalamus of prehypertensive phase (4 weeks old) of SHR were significantly smaller than those in NW of the same age. However, in the developmental phase (6 weeks old) and the established phase (16 weeks old) of hyperrtension in SHR, degrees of voltage-dependent ^{45}Calcium$ uptake and norepinephrine release were similar to those of age-matched normotensive wistae eats. These data imply that the deficit in hypothalamic norepinephrine release might be an important underlying factor for the development of hypertension in SHR.
The phospholipase C (PLC)-mediated intracellular signal transduction pathway is considered to be involved in the regulation of blood pressure. However, little information is available concerning the distributional and functional significance of PLC in the genetic hypertensive rats. As the first step of knowing the role of PLC on hypertension, we investigated the distribution of 6 PLC isozymes $(PLC-{\beta}1,\;-{\beta}3,\;-{\beta}4,\;-{\gamma}1,\;-{\gamma}2\;and\;-{\delta}1)$ in the heart and brain, which are concerned with hypertension, in the normotensive Wistar-Kyoto rat (WKY) and spontaneously hypertensive rat (SHR) using the western blotting and immunocytochemistry. The immunoreactivities of PLC isozymes in brain were detected, but there were no distributional and quantitative differences between the WKY and SHR. In the heart, but the immunoreactivities to $PLC-{\beta}1$ and $-{\gamma}2$ in the SHR were higher than those in WKY. In immunocytochemistry to confirm these western blotting data, $PLC-{\beta}1$ and $-{\gamma}2$ were localized in cardiac myocytes and the intensities of immunoreactivity in SHR were stronger than that in WKY. These results suggest that $PLC-{\beta}1$ and $-{\gamma}2$ would have possibility to concern with the establishment of spontaneous hypertension.

We have examined distribution and expression of caveolin-3 (cav-3), one of three caveolin isoforms from 16-wks-old spontaneously hypertensive rats (SHR) compared with age-matched control wistar-kyoto (WKY) rats. The expression of cav-3 was increased, whereas expression of PKB/Akt and calcineurin (Cn) was not changed in cardiac tissues of SHR compared to WKY rats. Interestingly, expression of cav-3, PKB/Akt and Cn were decreased in plasma membrane fraction in SHR compared to WKY rats. In H9c2 cardiomyoblast cells treated with phenylephrine ($50{\mu}M$, 48hr) or isoproterenol ($10{\mu}M$, 48hr), the expression of cav-3 was markedly enhanced compared to nontreated cells. Upon immunofluorescence analysis, cav-3 was localized in plasma membrane of control H9c2 cells. However phenylephrine or isoproterenol treatment caused translocation of cav-3 to perinuclear region. These results suggest that cav-3 plays as positive regulators in the development of hypertrophy in cardiac tissues of SHR rats.

To investigate the effect of ginseng on blood pressure (B.P.) in spontaneously hypertensive rat (SHR) and essential hypertension ginseng extract was given per se daily in 58 SHR and 35 essential hypertensive patients. SHR were divided into 5 groups according to the dosage of ginseng. In essential hypertension 1,000mg of ginseng extract was given. The B.P. in SHR was measured by tail cuff method. In essential hypertension side effect and changes in various laboratory examinations were evaluated. In SHR ginseng appeared to have hypertensive effect when it is given in small amount(10mg/kg). However, when it is given 60mg/kg/day or more difinite B.P. lowering effect was observed. The hypotensive effect was dosedependant and it lasted for 37days of observation. In essential hypertension in 12 $(80\%)$ among 15 patients hypotensive effect was seen with ginseng administration along and the effect lasted for 12 weeks. In the rest of hypertensive patients it is required addition of diuretics of other antihypertensive drugs to decrease B.P., no appreciable side effect was seen. In laboratory examinations no significant changes were seen except for serum cholesterol, ${\alpha}-and\;{\beta}-lipoprotein,$ and hematocrit. There was some evidence of relationship between plasma renin activity (PRA) and ginseng in hypotensive action.
The antihypertensive effect of $\alpha-Methyldopa$ administered concurrently with timolol or labetalol were studied with spontaneously hypertensive rats (SHR). Every drugs were administered orally once a day after prechecking the systolic blood pressure and heart rate of SHR. The blood pressure and heart rate of SHR were significantly decreased in groups of combination while those of non-combination groups were fallen slightly. Compared with control group, the significant changes of blood pressure was obseved in group of $\alpha-methyldopa$ with timolol(100mg+2mg/kg)and $\alpha-methyldopa$ with labetalol (100mg+12.5mg/kg). The group of $\alpha-methyldopa$ with timolo 1 (100mg + 1.0mg/kg) reduced blood Pressure in similar degree as manifested in group of $\alpha-methyldopa$ with labetalol (100mg + 50mg/kg). In the group that $\alpha-methyldopa$ was administered concurrently with timolol, the maximum antihypertensive effect and heart rate decreasing effect were appeared after 3hr and 1hr of administration respectively and those effects in group of $\alpha-methyldopa$ with labetalol were appeared after 6hr of administration. The acute oral toxicity test was performed using albino mice with $\alpha-methyldopa$ alone, $\alpha-methyldopa$ with timolol (100:1) and $\alpha-methyldopa$ with labetatol (4:1), and was found that the $LD_{50}$ of $\alpha-methyldopa$ alone was 1104 mg/kg, $\alpha-methyldopa$ with timolol (100:1) was 1115 mg/kg and $\alpha-methyldopa$ with labetalol (4:1) was 354mg/kg.

The effects of the salts fortified with seaweed functional components on blood pressure, serum minerals, and hematochemicals in spontaneously hypertensive rats (SHR/NCrj) and normotensive rats (WKY/NCrj) were investigated. SHR and WKY rats were assigned to four groups, with 8 and 6 rats in each group: laver salt (A), fucoidan+laver high salt (B), fucoidan + laver low salt (C) and refined salt as a control (D). The final blood pressure (BP) of SHR and WKY species in contrast with reference BP were low in groups A and C as compared with control group. In terms of serum mineral content, $Na^+$ levels were similar in SHR and WKY, but $K^+$ levels were higher in the group B in SHR. Serum triglyceride levels were lower in groups A and C, but the levels of HDL- and LDL- cholesterol were significantly higher and lower in group A than those of controls, respectively (p<0.05). These results demonstrated that the salts fortified with laver might suppress blood pressure in rats, and also may improve mineral and lipid metabolism.

The present study was designed to examine the effect of dietary fish oil on blood pressure and lipid status of serum. Weanling SHRs and normotensive Wistars were fed a diet containing 5%(w/w) mackerel oil(MO), soybean oil(SO) or beef tallow(BT) for 8 weeks. Growth rate was not significantly different among three dietary groups, but that of SHRs was silightly lower than that of Wistars. SHRs showed higher systolic blood pressure than Wistar rats from the beginning and become hypertensive (over 150mmHg) after 6 week s of feeding period. The MO group of SHRs showed the lowest blood pressure at the 8th week of feeding period but that of Wistars showed similar values with other groups. Tissue weights of liver, heart and kidney were not different amongdietary aroups in Wistars and SHRs. However, heart and kidney weights of SHRs were significantly higher than those of Wistars. Microscopic examination revealed that endomysium of heart tissue and urinary space of kidney were narrowed in SHRs. Serum total and HDL-cholesterol showed similar values among three different dietary fat groups but triglyceride levels were significantly low in MO groups. HDL-cholesterol levels of SHRs were lower than those of Wistars, as well as the fractions of total HDL, the sum of HDL and $HDL_{2+3}$, while VLDL fractions were higher in SHRs. MO groups had the lower values of $HDL_1,\;HDL_{2+3}$ratio than SO and BT groups. Major dietary fatty acids were more or less incorporated into serum phospholipid and triglyceride, resulting in the characteristic fatty acid profile of each dietary group. Incorporation of $C_{18:2}({\omega}_6)$ in SO groups were pronounced, but the degree of incorporation was lower in SHRs. In Mo groups, $C_{22:6}({\omega}_3)$ levels were inreased in triglyceride. It is suggested that these changes in serum lipid fatty acid composition are related to the different patterns of serum lipid by alteration of dietary fats.
This study was performed to investigate the effects of different fat diets on the serum lipid composition and on the morphology of liver tissue of the spontaneously hypertensive rats(SHR), Rats were fed for 17 weeks one of four fat diets, control(corn oil : beef tallow=1:1), beef tallow, grapeseed oil and soybean oil. Blood was withdrawn by heart puncture from each SHR, and used to determine the serum levels of total cholesterol, HDL-cholesterol, trigly ceride and phospholipid. No significant difference in body weight was observed in all diet groups. the total serum cholesterol was significantly lower in SHR fed grapeseed oil than that of other groups. The cholesterol lowering effect of grapeseed oil seemed to results from its high contents of phytosterol and linoleic acid. Also, serum HDL-cholesterol, triglyceride and phospholipid contents were low in all the groups. photochemical microscopic observations of the liver tissue revealed the congestion of sinosoid which is regarded as the characteristics of SHR, but no significant difference was observed among the groups.
