  • The purpose of this study was to examine effects of treadmill running training on superoxide product of spontaneously hypertensive rats. 20 males aged five week old rats were used for the experiments. They were randomly selected into experimental group (10 rats) and control group (10 rats) after being housed in laboratory for two weeks. The rats of experimental group performed treadmill exercise from 15 minutes for the first time to 35 minutes for the last one in order to familiar with experimental protocol ($VO_{2}max\;65{\sim}70%$, 0.9km/h, 5 times/week, 35 minutes/session). They began to perform treadmill running at the age of eight weeks for eight weeks. Superoxide production and NADPH oxidase activities were measured on the aortal from end of experiment. All statistical analyses and description methods were computed by SPSS Version 10.0. Differences in the values between experimental group and control group were calculated using t-tests. The results of this study were: there wewe no significant differences in superoxide production, NADPH oxidase-drivened superoxide activity, NADPH oxidase activity between two groups (p>.05). Spontaneously hypertensive rats of experimental group were characterized by higher superoxide production and lower NADPH oxidase-drivened superoxide activity and NADPH oxidase activity in comparison with the control group. One possible explanation of this finding was that NADPH oxidase activity of experimental group was lower than the control group. It can be concluded that treadmill running for eight weeks was associated positively with superoxide product of spontaneously hypertensive rats.

  • ABSTRACT - To examine antihypertensive components of Ganoderma lucidum in Korea, two kinds of fruiting bodies (J and K) were used for extraction with water and the extracts were purified by ethanol precipitation and dialysis. Three fractions, i.e., the aqueous total extract(A), the ethanol supernatant(B} and the purified precipitate(C), were compared for antihypertensive activity in anesthetized spontaneously hypertensive rats (SUR). Although fractions A and B showed the activity, fraction C did not. Particularly, fraction B of sample K produced 44.3% reduction in diastolic blood pressure and 30.6% reduction in heart rate after i.v. administration of a dose of 10 mg/kg. Direct effects of this fraction B to the heart were observed in the isolated blood perfused heart preparation of the dog. It induced positive chronotropic and inotropic responses dose-dependently in the case of sample J. In the case of sample K, marked chronotropic and inotropic effects on atrial muscle but not on ventricular muscle were induced. In both samples, coronary blood flow (CBF) was dose-relatedly increased.reased.

  • Background: Panax ginseng has distinct and impressive health benefits, such as improved blood pressure and immune system functioning. Rg3-enriched Korean Red Ginseng (REKRG) isolated from Korean Red Ginseng contains a high percentage of Rg3. Methods: In this study, we examined the effects of REKRG on endothelial cell nitric oxide synthase (eNOS) activation and adhesion molecules in endothelial cells and vascular function in rats. Results: REKRG dose-dependently increased eNOS phosphorylation and nitric oxide (NO) production in endothelial cells. In addition, REKRG markedly inhibited the tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$)-mediated induction of intercellular adhesion molecule (ICAM)-1 and cyclooxygenase (COX)-2 expressions in endothelial cells. REKRG improved endothelium-dependent vasorelaxation in the Wistar-Kyoto (WKY) rat and spontaneously hypertensive rats (SHRs) compared with controls. Furthermore, REKRG treatment for 6 weeks increased serum NO levels and reduced the mean aortic intima-media thickness compared with controls. Conclusion: Taken together, these results suggest that REKRG increased vascular function and improved immune system functioning. Therefore, REKRG is a very useful food for preventing or improving various cardiovascular diseases.

  • Two opioid peptides, YPLDL and YPLDLF, were isolated from enzymatic digests of spinach ribulose-1, 5-bisphosphate carboxylase/oxygenase (RuBisCO) and named rubiscolin-5 and -6, respectively. These peptides were selective for delta-receptor and the latter was about 3 times more potent than the former. After oral administration in mice at the dose of 100 mg/kg, rubiscolin-6 showed analgesic activity in tail pinch test. It also stimutated learning performance at the same dose in passive avoidance experiment using step-through apparatus. An immunostimulating peptide, MITLAIPVNKPGR, was isolated from a trypsin digest of soybean protein and named soymetide. Immunostimulating activy of soymetide was mediated by fMLP receptor. Interestingly, after oral administration in rats at a dose of 300 mg/kg (po.), soymetide-4 (MITL) protected alopecia (hair-loss) induced by etoposide, a cancer chemotherapy agent. Stimulation of IL-1 release by the peptide was involved in the mechanism. Ovokinin(2-7), RADHPF, is a vasorelaxing peptide released from ovalbumin by the action of chymotrypsin. It lowered blood pressure of spontaneously hypersensive rats (SHR) after oral administration at a dose of 10 mg/kg. RPLKPW, which was designed by replacing 4 amino acid residues in ovokinin(2-7), exhibited hypotensive activity at a dose of 0.1 mg/kg (po.). This peptides was introduced into 3 homologous sites in soybean beta-conglycinin alpha' subunit by site-directed mutagenesis of the cDNA and expressed in E. coli. The minimum effective dose for hypotensive activity of the genetically modified beta-conglycinin alpha' subunit was 10 mg/kg (po.), which is about 1/200 that of ovalbumin.

After 11days by fed with SRT, the cholesterol had no significant in all treated group compared with the control group. 4. After 11days by fed with SRT, the triglyceride had no significant in all treated group compared with the control group. 5. The cholesterol of hypercholesterolemic rat induced by feeding with cholesterol was decreased significantly in all treated group compared with the control group. 6. The triglyceride of hypercholesterolemic rat induced by feeding with cholesterol was decreased significantly in all treated group compared with the control group. 7. The HDL-cholesterol of hypercholesterolemic rat induced by feeding with cholesterol was decreased significantly in all treated group compared with the control group. 8. The phospholipid of hypercholesterolemic rat induced by feeding with cholesterol was decreased significantly in Sample B of the treated group. 9. The total lipid of hypercholesterolemic rat induced by feeding with cholesterol was decreased significantly in Sample B of the treated group. 10. The total protein of hypercholesterolemic rat induced by feeding with cholesterol was decreased significantly in Sample B of the treated group. 11. The albumin of hypercholesterolemic rat induced by feeding with cholesterol was decreased significantly in all treated group compared with the control group. With the above result, it is thought that Shiryungtang can be applied effectively to the hypertension and hyperlipidemia.

  • The present study was designed to investigate the effects of various electroacupuncture(EA) stimulation on NADPH-diaphorase and Neuropeptide Y(NPY) in the cerebral cortex of Sprague-Dawley Rats. We evaluated the changes of NADPH-d-positive neurons using a histochemical method and the changes of NPY-positive neurons using a double labeling immunohistochemical method. The staining intensities of NADPH-d-positive neuron and NPY-positive neurons were assessed in a quantitative fashion using a microdensitometrical method based on optical density by means of an image analyzer. As to frequency, the optical density of NADPH-d-positive neurons of 2Hz-1 time EA group was significantly higher than that of 100Hz-1 time EA group in M1, Cg, and that of 100Hz-3 times EA group was significantly higher than that of 2Hz-3 times EA group in Vi, Au, Cg and Ins. As to treatment-time, the optical density of NADPH-d-positive neurons of 2Hz-1 time EA group was higher than 2Hz-3 times EA group in Vi, Au, Cg and Ins. And in Vi 100Hz-1 time EA group and in M1 100Hz-3 times EA group was more efficient. As to frequency, the optical density of NPY-positive neurons of 2Hz-1 time was significantly higher in Vi, and that of 100Hz-1 EA group was higher in M1. And that of 2Hz-3 times EA group was higher than 100Hz-3 times EA group in Cg. As to treatment-time, the optical density of NPY-positive neurons of 2Hz-1 time EA group was significantly higer than 2Hz-3 times EA in S1, Vi, Au. And that of 100Hz-1 time EA group was significantly higher than that of 100Hz-3 time EA in Cg. The present results demonstrated that EA changes the activity in NO and NPY system in the cerebral cortex of SHR and EA stimulation, like frequency and treatment-time, is of importance for this effect.

  • Objectives: Chungpaesagan-tang (CPSGT), which is frequently used for treating patients of cerebrovascular disease, has not been reported by clinical doctors concerning the effect of neuronal aptosis caused by brain ischemia. To study the effect of CPSGT on focal cerebral ischemia in normal and diabetic rats and SHR, focal cerebral ischemia was induced by transient MCAO, and after onset CPSGT was administrated. Methods: Rats (Sprague-Dawley) were divided into four groups: sham-operated group, MCA-occluded group, CPSGTadministrated group after MCA occlusion, and normal group. The MCA was occluded by intraluminal method. CPSGT was administrated orally twice (l and 4 hours) after middle cerebral artery occlusion. All groups were sacrificed at 24 hours after the surgery. The brain tissue Was stained with $2\%$ triphenyl tetrazolium chloride (TTC) or $1\%$ cresyl violet solution, to examine effect of CPSGT on ischemic brain tissue. The blood samples were obtained from the heart.~. Tumor necrosis $factor-\alpha$ level and interleukin-6 level of serum was measured from sera using enzyme-linked immunoabsorbent assay (ELISA). Then changes of immunohistochemical expression of $TNF-\alpha$ in ischemic damaged areas were observed. Results: In NC+MCAO+CP and DM+MCAO+CP, CPSGT significantly (p<0.01) decreased the number of neuron cells compared to the control group. CPSGT markedly reduced (p<0.01) the infarct size of the forebrain in distance from the interaural line on cerebral ischemia in diabetic rats. CPSGT significantly reduced the $TNF-\alpha$ expression in penumbra region of damaged hemisphere in diabetic rats. Conclusions: CPSGT had a protective effect on cerebral ischemia in SD rats, especially in diabetic rats compared with normal SD rats.

  • The goal of this study was to develop a high value Korean red wine possessing antihypertension activity. The effects of some medicinal plants and grapes on the alcohol fermentation process and the angiotensin I-converting enzyme (ACE) inhibitory activity of Vitis hybrid red wine were investigated. Various Vitis hybrid red wines were vinified by the fermentation of a mixture of Vitis hybrid must and some medicinal plants and grapes at $25^{\circ}C$ for 10 days. Of these red wines, the Vitis hybrid-Vitis coignetiae red wine exhibited a high ethanol content of 12.0% and had a good level of acceptability. It also showed a high antihypertensive ACE inhibitory activity of 68.5%. After post-fermentation of 60 days, the ACE inhibitory activities of the Vitis hybrid-Vitis coignetiae red wine exhibited the highest ACE inhibitory activity of 80.7% ($IC_{50}$: 28 mg/mL) and also had the best acceptability. The $C_{18}$ solid phase extracts of the Vitis hybrid-Vitis coignetiae red wine, after 60 days post-fermentation, showed clear antihypertensive effects on spontaneously hypertensive rats. Our results reveal that the Vitis hybrid-Vitis coignetiae red wine has the potential to become a new functional red wine due to its good acceptability and high antihypertensive activity.

  • The renoprotective effects of Korean Red Ginseng were examined in STZ-induced diabetic spontaneously hypertensive rats (SHR). After 3 day administration of streptozotocin (STZ), animals were divided into four groups : Group 1, hypertensive rats (H); Group 2, hypertensive rats with diabetes (HD); Group 3, hypertensive rats with diabetes administered with 100 mg/kg of ginseng total saponin(GTS); Group 4, hypertensive rats with diabetes administered with 600 mg/kg of ginseng non-saponin (GNS). After 2 weeks oral administraions of GTS and GNS, body weight, kidney weight, plasma glucose, urinary albumin excretion, serum creatinine, urea nitrogen and blood pressure were examined. After 3,7 and 21 day of STZ administration, expressions of TGF-${\beta}$1 and fibronectin in kidney were analyzed by immunoblotting and/or immunohistochemistry. GTS and GNS treatments slightly decreased blood pressure when compared to H and HD groups. Also, GTS and GNS treatments ameliorated kidney hypertrophy without affecting plasma glucose levels. Meanwhile, GNS treatment increased Cu/Zn-SOD activity in kidney and generally showed more efficient renoprotective effects than GTS. We suggest that the renoprotective effects of ginseng partially result from downregulations of TGF-${\beta}$1, fibronectin expressions and anti-oxidative activity of ginseng non-saponin.
