Effect of Ca and BSA on Hydrogen Ion Concentration in Bovine Sperm Washed Solution (Ca과 BSA가 소 정자세척액내 수소이온농도에 미치는 영향)

  • This study was carried out to investigate the effects of Ca and BSA on hydrogen ion concentration in sperm washed solution. The results obtained were as follows : 1. The hydrogen concentration in 1st and 2nd sperm washed solutions was signifcinatly(p<0.01) higher when sperm was washed with SHPsolution containing 2mM Ca than when sperm washed with SHP solution or SHP solution containing 10mM Ca. 2. The hydrogen ion concentration in sperm washed solution was significnatly(p<0.05) higher when seprm was washed with SHP solution containing BSA-FAF than when sperm was washed with SHP solution or SHP solution containing BSA-V.

  • This paper presents the methodology to analyze flow duration characteristics and performance prediction technique for small hydro power(SHP) Plants and its application. The flow duration curve can be decided by using monthly rainfall data at the most of the SHP sites with no useful hydrological data. It was proved that the monthly rainfall data can be characterized by using the cumulative density function of Weibull distribution and Thiessen method were adopted to decide flow duration curve at SHP plants. And, the performance prediction technique has been studied and development. One SHP plant was selected and performance characteristics was analyzed by using the developed technique, Primary design specfications such as design flowrate, plant capacity, operational rate and annual electricity production for the SHP plant were estimated, It was found that the methodology developed in this study can be a useful tool to predict the performance of SHP plants and candidate sites in Korea.

  • The correlation between hydrologic performance design parameters of small hydro power(SHP) sites and rainfall condition have been analyzed for major river systems. The model, which can predict flow duration characteristic of stream, was developed to estimate the inflow caused from rainfall. And another model to predict hydrologic performance for SHP plants is established. Based on the models developed in this study, the hydrologic performance characteristics for SHP sites have been analyzed. The results show that the hydrologic performance characteristics of SHP sites have some difference between the river systems. Especially, the specific design flowrate and specific output of SHP sites located on North Han river and Nakdong river systems have large difference compared with other river systems. It was found that the hydrologic performance design parameters such as specific design flowrate and specific output were affected by rainfall condition in basin area of SHP sites.

  • Objective: We inversigated Small Heterodimer Partner (SHP) gene mutation in Korean Polycystic Ovarian Syndrome (PCOS) patients. SHP protein regulates the activity of nuclear receptors which regulate the cellular development and differentiation. Recently, the mutation of SHP gene was found in the obesity and diabetes patients in Japanese group, and suggested that its mutation may involved in pathogenic mechanism of PCOS. Methods: This study was performed in 20 PCOS patients and 20 normal women. The DNAs were extracted from the peripheral bloods, and amplified at each exon (1 and 2) of SHP gene by PCR method. Subsequently, each PCR product was digested with the restriction enzyme indicated below for studying restriction fragment length polymorphism (RFLP). After enzyme digestion, the results of RFLP were compared PCOS patients with control women to find any sequence variation. Results: We examined 9 regions of exon 1 with Msp I, Pvu II, Dde I and 3 regions of exon 2 with Pst I, Dde I. There is no heterozygous or homozygous mutation in patients and control women at these restriction sites. Conclusion: The genetic analysis at our restriction sites in the SHP gene did not show any genetic variation in Korean PCOS patients. Our PCR-RFLP analysis was not covered the entire SHP gene (68 bp/1,006 bp), we need to further analysis of the entire SHP gene.

Analysis of the SHP-2 Binding Site of Helicobacter pylori CagA Protein in Korean (한국인에서 획득한 Helicobacter pylori의 CagA에 존재하는 SHP-2 binding site의 분석)

  • Recently the pathological actions of CagA of Helicobacter pylori (H. pylori) on gastric epithelial cells have been reported. CagA is directly injected into the host cytoplasm and undergoes tyrosine phosphorylation in the cells. In addition, translocated CagA forms a physical complex with SHP-2. There are two major CagA subtypes according to the amino acid sequence in the 3'region of CagA; i) the East Asian type (A-B-D of EPIYA motifs) and ii) the Western type (A-B-C of EPIYA motifs). Repeated EPIYA motifs in the 3'region of CagA are involved in the interaction with SHP-2. The East Asian type conferred stronger SHP-2 binding activity than the Westrrn type of CagA. Here we analyzed the amino acid sequences of the SHP-2 binding site of cagA gene in H. pyzori, and investigated whether there is my relationship between the diversities of cagA and the disease out-come in Korea. Most of Korean H. pylori strains showed A-B-D motifs(the East Asian type), and only one strain showed A-B-B-D motifs. In Korea, the incidence of atrophic gastritis and gastric cancer is significantly high compared with Western countries. The high frequency of the East Asian type CagA among Korean H. pylori strains would be involved in increasing the risk of gastric cancer in Korean populations.

New Insights into Autonomic Nerve Preservation in High Ligation of the Inferior Mesenteric Artery in Laparoscopic Surgery for Colorectal Cancer

  • Aim: To take a deeper insight into the relationship between the root of the inferior mesenteric artery (IMA) and the autonomic nerve plexuses around it by cadaveric anatomy and explore anatomical evidence of autonomic nerve preservation in high ligation of the IMA in laparoscopic surgery for colorectal cancer. Methods: Anatomical dissection was performed on 11 formalin-fixed cadavers and 12 fresh cadavers. Anatomical evidence-based autonomic nerve preservation in high ligation of the IMA was performed in 22 laparoscopic curative resections of colorectal cancer. Results: As the upward continuation of the presacral nerves, the bilateral trunks of SHP had close but different relationships with the root of the IMA. The right trunk of SHP ran relatively far away from the root of IMA. When the apical lymph nodes were dissected close to the root of the IMA along the fascia space in front of the anterior renal fascia, the right trunk of SHP could be kept in suit under the anterior renal fascia. The left descending branches to SHP constituted a natural and constant anatomical landmark of the relationship between the root of IMA and the left autonomic nerves. Proximal to this, the left autonomic nerves surrounded the root of the IMA. Distally, the left trunk of the SHP departed from the root of IMA under the anterior renal fascia. When high ligation of the IMA was performed distal to it, the left trunk of SHP could be preserved. The distance between the left descending branches to SHP and the origin of IMA varied widely from 1.3 cm to 2.3 cm. Conclusions: The divergences of the bilateral autonomic nerve preservation around the root of the IMA may contribute to provide anatomical evidence for more precise evaluation of the optimal position of high ligation of the IMA in the future.

  • Objective : The present study was carried out to investigate the preventive effect of Several Herb - Combined Prescription (SHP) on streptozotocin (STZ) - induced diabetes mellitus. Methods : SHP was given to mice with the combination of oral administration and herbal-acupuncture stimulation. Experimental diabetes was induced by the injection of STZ(50mg/kg) to the rat via the peritoneum The effect of SHP on STZ-induced diabetes was observed by measuring the serum level of insulin, glucose, triglyceride, total cholesterol and lipid peroxides. Hepatic activities of catalase and reduced glutathione were examined. Results : SHP treatment protected them from the hyperglycemia. STZ induced increase of serum triglyceride lowered by SHP treatment. Conclusions : The SHP treatment showed protective effect on diabetic mouse model, and action mechanism of the effect was thought to be concerned with anti-oxidative stress.

Assessment of the Effect of a Disinfectant to Control Vibrio Strain from White Leg Shrimps Cultured in Korea (국내산 양식 흰다리새우 유래의 비브리오균에 대한 소독제의 방제 효과 검증)

  • White leg shrimps which were cultured in a private aquaculture farm showed abnormal swimming behavior and appetite reduction in July 2023. Then, gradual mortality was observed in the aquaculture farm. During the diagnosis, bacterial strain KNUAFVaSHP08147 was isolated from the hepatopancreas of the dead shrimps. Based on the sequence of 16S rRNA gene, KNUAFVa-SHP08147 was proved to be Vibrio alginolyticus, showing 99.71 % nucleotide identity with that of V. alginolyticus MG54 and GS MYPK1. According to the result of the growth of KNUAFVa-SHP08147, the treatment a commercial disinfectant, VirkonTMS was not proved to be effective to prohibit the growth of Vibrio.

Hypoxic repression of CYP7A1 through a HIF-1α- and SHP-independent mechanism

  • Liver cells experience hypoxic stress when drug-metabolizing enzymes excessively consume O2 for hydroxylation. Hypoxic stress changes the transcription of several genes by activating a heterodimeric transcription factor called hypoxia-inducible factor-1α/β (HIF-1α/β). We found that hypoxic stress (0.1% O2) decreased the expression of cytochrome P450 7A1 (CYP7A1), a rate-limiting enzyme involved in bile acid biosynthesis. Chenodeoxycholic acid (CDCA), a major component of bile acids, represses CYP7A1 by activating a transcriptional repressor named small heterodimer partner (SHP). We observed that hypoxia decreased the levels of both CDCA and SHP, suggesting that hypoxia repressed CYP7A1 without inducing SHP. The finding that overexpression of HIF-1α increased the activity of the CYP7A1 promoter suggested that hypoxia decreased the expression of CYP7A1 in a HIF-1-independent manner. Thus, the results of this study suggested that hypoxia decreased the activity of CYP7A1 by limiting its substrate O2, and by decreasing the transcription of CYP7A1.

E-Band Wideband MMIC Receiver Using 0.1 ${\mu}m$ GaAs pHEMT Process

  • In this paper, the implementations of a $0.1{\mu}m$ gallium arsenide (GaAs) pseudomorphic high electron mobility transistor process for a low noise amplifier (LNA), a subharmonically pumped (SHP) mixer, and a single-chip receiver for 70/80 GHz point-to-point communications are presented. To obtain high-gain performance and good flatness for a 15 GHz (71 GHz to 86 GHz) wideband LNA, a five-stage input/output port transmission line matching method is used. To decrease the package loss and cost, 2nd and 4th SHP mixers were designed. From the measured results, the five-stage LNA shows a gain of 23 dB and a noise figure of 4.5 dB. The 2nd and 4th SHP mixers show conversion losses of 12 dB and 17 dB and input P1dB of -1.5 dBm to 1.5 dBm. Finally, a single-chip receiver based on the 4th SHP mixer shows a gain of 6 dB, a noise figure of 6 dB, and an input P1dB of -21 dBm.
