남자대학생의 흡연 및 식사습관에 따른 인체 임파구 SCE 빈도 수의 변화 (The Variations of the SCE Frequency of Human Lymphocytes by Smoking Habits and Dietary Factors in College Students)

  • Sister chromatid exchange(SCE) has recently become a common cytogenic assay system for detecting exposure to chemical mutagens and carcinogens. One application of SCE is the monitoring of populations believed to be exposed to such agents. A cross-sectional study of SCE frequency in peripheral blood lymphocytes from 40 college students aged 18 to 26 years was conducted. The effects of cigarette smoking alcohol and coffee consumption, dietary and environmental factors on SCE were assessed. A mean spontaneous SCE per cell for the smokers(4.88$\pm$0.17). The SCE levels of the smokers were associated with the personal smoking amount ; the observed increase in the SCE frequency correlated with the number of cigarettes smoked per day (P<0.05). There was no effect of age on SCE. There were positive linear relationship between SCE and food frequency score of meat and fish group (P<0.05) or instant food group(P<0.01) in non-smokers. But in smokers, a significant inverse association between SCE and food frequency score of green and yellow vegetable group(P<0.05). Alcohol intake produced a significant increase(P<0.01) of SCE in comparison with the mean SCE for those not drinking alcohol in combine subjects. Other dietary parameters, including coffee intake, use of artificial sweetners and processed foods, did not show any increase in SCE. SCEs were inversely related to blood glucose and serum cholesterol levels of the combine subjects. No significant correlations were found between SCE frequencise and any other hematological parameters of the subjects.

노인의 흡연상태와 나이가 SCE 빈도수로 본 임파구 DNA 손상에 미치는 영향 (Effects of Smoking and Age on SCE Frequency Reflecting DNA Damage of Human Lymphocytes in Elderly Koreans)

  • Sister chromatid exchange (SCE) has recently become a common cytogenic assay system for detecting exposure to chemical mutagens and carcinogens. One application of SCE is the monitoring of populations believed to have been exposed to such agents. A cross-sectional study of SCE frequency in peripheral blood lymphocytes from 45 Koreans aged 61 to 84 years was conducted. The effect of cigarette smoking and age on SCE was assessed by different degrees of smoking status such as smokers (n = 14), ex-smokers (n = 16) and non-smokers (n = 15). Mean spontaneous SCE per cell for the smokers (11.5 $\pm$ 1.1) was significantly higher (p < 0.05) than that for the non-smokers (8.8 $\pm$ 0.3). However, mean SCE frequencies per cell for the ex-smokers (10.3 $\pm$ 0.6) were not significantly different from those of the smokers or the non-smokers. The smokers showed an increased number of high SCE frequency cells (HFCs) when compared to the ex-smokers and non-smokers (p < 0.05). The mean SCE frequencies of the non-smokers showed a statistically significant increase (p < 0.05) with the subject's age. These results show that age and smoking habits contribute a great deal in setting a higher degree of basal DNA damage in elderly Koreans, and smoking appeared to be a more significant damaging factor than age.

식이성요인이 SCE 빈도수로 본 흡연자의 DNA 손상에 미치는 영향 (Influence of Dietary Factors of Smokers on Smoking-Induced DNA Damage as Reflected by Sister Chromatid Exchanges(SCE))

  • Sister chromatid exchanges(SCE) in peripheral lymphocytes is recently used as a biomarker for increased cytogenetic damage in smokers. The purpose of the investigation was to determine if there were any relationships between dietary factors and their DNA damage as measured by SCE test in a group of 62 male cigarette smokers and 36 non-smokers. As expected, smokers as compared with non-smokers had high SCE levels (10.59$\pm$0.21 versus 9.23$\pm$0.17 SCE/lymphocytes ; p<0.05). No significant relationships were observed between SCEs and age in smokers and non-smokers. In smokers, SCEs were negatively correlated with egg frequency score(r=-0.336) and total food frequency scores(r=-0.283). In non-smokers, SCEs were positively correlated with white vegetable frequency score(r=0.333) and instant food frequency score(r=0.382). There was a positive association between SCEs and the history of coffee intake of smokers(r=0.318). SCE frequency was not influenced by any other dietary factors considered ; dietary diversity and quality scores, alcohol consumption, use of processed foods and intake of burned food. No significant relationships were found between SCEs and serum cholesterol or other hematological parameters of the subjects. These results indicate that increased egg frequency score, total food frequency score which reflects dietary quality, and decreased coffee intake may reduce cancer risk by preventing smoking-induced DNA damage as reflected by sister chromatid exchanges in human lymphocytes.

인삼의 섭취가 흡연성인의 인체임파구 SCE 빈도수에 미치는 영향 (The Effect of Ginseng Intake on the Frequency of Sister Chromatid Exchanges of Human Lymphocyte of Adult Smokers)

  • This study was intended to investigate the anticarcinogenic effect of ginseng previously elucidated by other researches in animal studies. The sister chromatid exchange(SCE) method of human lymphocytes was used as a biomarker. Based on the literature search and the results of our laboratory, smoking was used as a parameter elevating the SCE frequency of general human population. To evaluate the smoking and ginseng effect on SCE frequency, 98 male healthy factory workers aged 23 to 58 years were divided into 4 groups : smoker with ginseng (SG), smoker control(SC), non-smoker with ginseng(NSG), and non-smoker control(NSC) groups, according to their smoking habits and ginseng intake. The mean sponteneous SCE per cell for the SG(10.8$\pm$0.3) and SC(10.4$\pm$0.3) groups were significantly higher than the NSG(9.1$\pm$0.2) and NSC(9.3$\pm$0.3) groups(p<0.05). High frequency cells (HFCs, cells with 15 SCEs) in SG and SC groups were also greater than those in NSG and NSC groups. However, the SCE levels of the SG and SC groups were not associated with the personal smoking history and the number of cigaretts smoked per day. Ginseng intake did not show any effect on the increased SCE caused by smoking. There were no correlations of the elevated SCE among smoking and ginseng types, history of ginseng intake, and consumption frequencies of ginseng intake. These results does not support the findings of other researchers that ginseng could be a protective agent to DNA damage.

식이 요인이 SCE 빈도수로 본 흡연노인 임파구 DNA손상에 미치는 영향 (Effects of Dietary Factors on Lymphocyte DNA Damage in Smoking Elderly People in Korea)

  • 영양소의 섭취를 포함한 여러 식이성요인에 따라 일부 지역 남자 노인의 인체 임파구 SCE 빈도수가 어떤 영향을 받는지를 조사해 보고자하여 60세 이상의 노인 45명을 대상으로 설문조사를 실시한 후 채혈하여 임파구 SCE 시험을 실시하였다. 조사 대상자의 식이성 요인이 SCE 빈도수에 미치는 영향에 대해 분석한 결과는 다음과 같다. 1. 채혈 대상자를 흡연여부에 따라 흡연군(n=14), 담배를 피우다가 현재는 끊은 금연군(n=16), 비흡연군(n=15)으로 나눈 후 SCE 빈도수를 비교한 결과 흡연군의 SCE빈도수가 비흡연군에 비해 유의적으로 높았으며 노인의 나이가 많을수록 DNA 손상정도가 증가하였다. 대상 노인의 인구학적 요인 중 신장, 혹은 체중이 전 대상자군에서 SCE 빈도수와 각각 역의 상관관계를 보였으며 흡연군에서는 신장과 SCE빈도수가 역의 상관관계, 그리고 pack years로 본 흡연력과 SCE 빈도수가 정의 상관관계 를 보였다. 2. 전 대상자군에서는 24시간 회상법으로부터 구한 영양소의 섭취와 SCE빈도수 간에 상관관계가 없었으나 흡연군의 경우는 식이섬유질 섭취량, 금연군의 경우는 비타민 C 섭취량과 SCE 빈도수가 각각 정의 상관관계를 보였다. 그러나 비 흡연군에서는 지방, 인 혹은 비타민 A 섭취량과 SCE빈도수 간에 역의 상관관계를 보였다. 3. 흡연군에서 육어류 섭취빈도와 SCE빈도수 간에 정의 상관관계를 보였고, 비 흡연군에서 식 사균형도가 SCE 빈도수와 역의 상관관계를 보였다. 세 군을 모두 합한 전대상자군의 경우, 혈당수준과 SCE 빈도수 사이에 정의 상관관계가 나타났다. 금연군에서 감미료를 섭취하는 군의 SCE 빈도수가 섭취하지 않는 군보다 유의적으로 높았다. 나머지 식이성 요인들은 SCE 빈도수와 상관성을 보이지 않았다. 본 연구 결과는 앞으로 DNA손상도가 높은 노인 대상으로 이의 회복을 위하여 식이성 요인을 이용한 영양중재실험을 계획할 때, 혹은 노인 대상의 영양교육 시에 중요한 기초자료로 활용될 수 있을 것이다.

혈장 총 율기 포집 능력(TRAP) 수준 및 항산화 비타민 영양상태와 인체 임파구 DNA 손상정도와의 상호관련성 연구 (The Associations between Plasma Concentrations of Total Radical-Trapping Antioxidant Potential(TRAP), Antioxidant Vitamins and DNA Damage in Human Lymphocytes)

  • The spontaneous frequency of genetic damage and the possible relationship of this damage to total radical-trapping antioxidant potential(TRAP) and antioxidant vitamins, including plasma levels of $\alpha$-carotene, $\beta$-carotene, cryptoxanthin, retinol, $\alpha$-tocopherol and ${\gamma}$-tocopherol in humans were investigated in 57 subjects using two indices of genetic damage, SCE & HFC frequency. The mean of SCE and HFC frequencies were weakly correlated with plasma TRAP(r=-0.305, p<0.1 for SCEs: r=-0.297, p<0.1 for HFCs, respectively), but those were strongly negatively correlated with plasma $\beta$-carotence(r=-0.385, p<0.01 for SCEs : r=-0.392, p<0.01 for HFCs) and cryptoxanthin(r=-0.312, p<0.05 for SCEs : r=0.335, p<0.05 for HFCs, respectively) levels in the subjects. However, those DNA damage markers including SCE and HFC did not correlate with either plasma $\alpha$-carotene, $\alpha$-tocopherol or retinol concentrations. The mean of SCE and HFC frequencies were positively correlated with plasma ${\gamma}$-tocopherol level(r=0.421, p<0.01 for SCEs : r=0.399, p<0.01 for HFCs, respectively). These findings indicate that increased cytogenetic DNA changes, as determined by SCE and HFC frequencies are possibly associated with generation of free radicals in lymphocytes and decreased plasma antioxidant vitamin($\beta$-carotene and cryptoxanthin) status in the subjects. Increased Sister Chromatid Exchange in Peripheral Blood Lymphocytes from Humans Exposed to Pesticide: Evidence Based on a Meta-analysis

  • Background: Sister chromatid exchange (SCE) in human peripheral blood lymphocytes is one of the most extensively studied biomarkers employed to evaluate genetic damage subsequent to pesticide exposure. Objective: To estimate the pooled levels of SCE in human peripheral blood lymphocytes among population exposed to pesticide. Materials and Methods: Meta-analysis on the association between SCE frequency and pesticide exposure was performed with STATA 10.0 software package and Review Manager 5.0.24 in this study. Results: The overall means of SCE were 7.88 [95% confidence intervals (95%CI): 6.71-9.04] for exposure group and 6.05 (95%CI: 5.13-6.95) for controls, respectively. There was statistically significant difference in the SCE frequency in human peripheral blood lymphocytes between pesticide-exposed groups and control groups, and the summary estimate of weighted mean difference was 1.69 (95%CI: 1.01-2.38). We also observed that pesticide-exposed population had significantly higher SCE frequency than control groups among smokers, nonsmokers, pesticide applicator, pesticide producer, other exposure population and Asian population in stratified analyses. Conclusions: Data indicate that the SCE frequency in human peripheral blood lymphocytes might be an indicator of early genetic esffects for pesticide-exposed populations.

한국인 집단의 유전학적 연구 29. 약물치료를 받은 정신분열 살인환자의 자매염색분체 교환 빈도 (Genetic Studies of Korean Population 29. Sister Chromatid Exchange Frequency in Homicidal Schizophrenic Patient Treated with Antipsychotic Drugs)

  • 정신분열 살인환자로 격리 치료를 받고 있는 집단을 대상으로 이들을 치료하고자 복용시키는 Haloperidol, Perphenazine, Lithium carbonate 등과 같은 항정신질환 치료제가 자매염색분체 교환(Sister Chromatid Exchange SCE)에 미치는 영향을 조사하였다. 항정신질환 치료제를 계속적으로 복용하고 있는 환자 100명(남자: 76명, 여자: 24명)과 치료제를 전혀 복용하지 않은 남자 환자 10명을 대조군으로 하여 SCE의 빈도를 분석한 결과 항정신질환 치료제를 복용한 환자군에서의 SCE 평균빈도는 세포당 12.24$\pm$0.20으로 항정신질환 치료제를 복용하지 않은 대조군에서의 평균빈도인 세포당 8.77$\pm$0.20보다 높아 유의한 차이를 볼 수 있었다 그러나 항정신질환 치료제를 복용하지 않은 대조군에서의 평균빈도는 이미 보고된 바 있는 정상인 한국인 집단에서의 평균빈도인 세포당 8.78$\pm$0.24(Park et al. , 1992)와 별 차이가 없었다 한편 항정신질환 치료제를 장기간 복용하는데 따른 SCI 빈도의 차이가 있는지의 여부를 보기 위하여 1년 미만에서 6년 정도까지의 치료제 복용기간에 따른 SCE의 평균빈도를 비교 분석한 바 복용기간에 따른 SCE 평균빈도의 유의한 차이는 볼 수 없었다. 정신분열환자들이 복용하고 있는 항정신질환 치료제가 SCE에 영향을 줄 수 있다는 결과를 얼었다.

Age-related Increase of Sister Chromatid Exchange Frequency in Bone Marrow Cells of Senescence Accelerated Mouse and Its Inhibition by Chronic Treatment of Ginseng

  • Age-related change in the frequency of spontaneous sister chromatid exchange (SCE) and chromosornal aberrations were investigated in bone marrow cells of accelerated senescence-resistant mice (SAM R1) and senescence accelerated ones (SAM P1). And the effect of chronic treatment of ginseng extract (Panax ginseng C.A. Meyer) on these chromosomal abnormalities was tested in SAM P1. SCE frequency in the cells was progressively increased with age in both mice, but it was consistently higher in SAM P1 than in SAM R1 at all corresponding age. Chromosomal aberrations were, however, not significantly changed with age except that it was slightly increased in only aged SAM P1. Interestingly, the rate of these genetic instabilities in SAM P1 was remarkably retarded by long-term administration of ginseng water extract (0.05% in drinking water). These results suggest that frequency of spontaneous SCE in bone marrow cells increase in parallel with senescence of the mice, and SAM P1 is in the condition of being more exposed than SAM R1 to DNA damaging factors. These also indicate that long-term treatment of ginseng may reduce the genetic damage.

Di-2-EthylHexyl Phthalate, 2-EthylHexanoic Acid 및 Di-2-Ethyl Phthalate의 유전독성 평가 (Genotoxicity of Di-2-Ethylhexyl phthalate, 2-EthylHexanoic Acid and Di-2-Ethyl Phthalate in Human Lymphocytes in vitro)

  • DEHP is one of well known endocrine disrupter and it is used as additives for the production of PVC. There has been contradictional result on the genotoxicity of DEHP. In order to examine genotoxicity of a endocrine disruptors, DEHP (Di-2-EthylHexyl Phthalate) and it's metabolites, EHA (2-EthylHexanoic Acid) and DEP (Di-2-Ethyl Phthalate), chromosome aberration (CA), sister chromatid exchange (SCE), micronuclei (MN) and single cell gel electrophoresis were analysised. No increase of the frequency of CA was observed by DEHP and its two metabolites. DEHPincreased the frequency of SCE and MN whereas EHA only increased the frequency of SCE. DEP increased the frequency of SCE but the increase was not statistically significant. DEHP and DEP, also induced DNA damage. It is suggested that combination of different methods were recomended to find the genotoxicity of DEHP and its metabolites.
