Inheritance of Resistance to Flacherie Virus in the Silkworm, Bombyx mori L. (가잠의 바이러스성 연화병에 대한 저항성 유전양식)

  • Flacherie virus (FV) is an important pathogen in the silkworm, which often gives serious damage to farmers for cocoon production. The inbred parents and F$_1$'s from an eight-parent diallel were examined to determine the inheritance of resistance to flacherie virus in the silkworm. Three resistant (R), two intermediate (M) and three susceptible (S) inbreds were used in the diallel with no reciprocals. Mean resistance was measured by survival rates of larvae which were fed on mulberry leaves sprayed with diluted mid-gut homogenate of FV infected larvae. Broad-sense heritability was obtained according to inbreds and F$_1$ family performance. Estimation of general (GCA) and specific combining ability (SCA) was made according to Griffing's Model 1, Method 2. Mean FV resistance of F$_1$ family displayed additive effect of the major gene, while heterotic effect was not significant. Considerable variation in FV resistance within F$_1$ groups of R$\times$S and S$\times$S indicated that action of minor genes for FV resistance may have been involved. FV resistance of inbreds perse and predominant effect of the major gene over minor gene(s) satisfactorily predicted the FV resistance of the hybrids. Broadsense heritability value of FV resistance on the basis of F$_1$ family performance averaged 93%, which suggested that environmental effects might have not been important in this experiment. GCA was highly significant for FV resistant among inbreds. GCA effect of 13.1 in Jam 108 was highest and -17.7 in Gyeongchu lowest. Effective selection for high FV resistance would be possible, using inbreds with high GCA effect and low GCA variance. SCA was significant among hybrids. High SCA effect in the hybrid of Geumho$\times$Mudeung (13.7) and Hansaeng #4$\times$Jam 115 (11.6) indicated that the interaction effect of minor genes for resistance to FV in the silkworm could be exploited by standard silkworm breeding procedures.

Estimation of Greenhouse Damaged Area by Heavy Snowfall using GIS and Remote Sensing Technique (논문 - GIS/RS를 이용한 비닐하우스 폭설 피해지역 추출 기법 연구)

  • This study is to estimate the possible damage area of greenhouse by heavy snowfall event using terra MODIS snow cover area (SCA) and the ground measured snowfall data (GMSD). For the 4 heavy snowfall events of January 2001, March 2004, December 2005 and January 2010, the areas exceeding the design criteria of snowfall depth for greenhouse breaking were extracted by coupling the MODIS SCA and GMSD. The main damaged regions were estimated as Gangwon province in 2001, Chungbuk and part of Gyeongbuk province in 2004, Jeonbuk and Jeonnam province in 2005, and Gangwon and part of Gyeonggi province in 2010 respectively. Comparing with the investigated number of greenhouse damaged data, the estimated areas reflected the statistical data except 2001. The 2001 greenhouse damages were caused by the high wind speed (35.7m/sec) together with snowfall. The results of this study can be improved if the design criteria of wind speed is added.

  • Six orient varieties of N.tabacum and 15 $F_1$ hybrids, not including reciprocal crosses, were evaluated for the degrees of heterosis and the combining abilities of 6 characteristics in Suweon, Korea, 1981. The values 9f general combining abilities tiers greater than those of specific combining abilities in all traits but significant SCA was also observed for Plant height, days to flowering and yield. In general, CCA effect was large in Samsun, Kavala and Izmir varieties. Large SCA values were observed for number of leaves in Samsun x Izmir, days to flowering in Samsun x Serves, leaf shape in Basma x Xantlli, and yield in Basma x Kavala and Xanthi x Izmir. Heterosis of favorable directions in $F_1$ was generally observed for all traits. Especially the $F_1$ of Samsun x Kavala flowered earlier than its midparents. Large heterosis effects were manifested for the number of leaves in Samsun x Izmir and yield in Samsun x Izmir as well as Xanthix Izmir.

  • The present study was carried out to obtain some basic informations of the breeding of tobacco varieties. Genetically divergent 8 varieties, 3 flue-cured, 2 burley and 3 sun-cured tobaccos, were used in half diallel cross. In order to analyze the heterosis, combining abilities, modes of inheritance and correlations for some agronomic and chemical characters, 8 parents and 28 $F_2$ were tested. The results obtained were summarized as follows: The percentages of heterosis for yield, stem diameter, internode length and total sugar content in $F_2$ hybrid were positive , where-as those for the other characters were negative. Yield had significant positive genotypic and phenotypic correlations with plant height, days to flower, leaves per plant, leaf length, leaf width and leaf shape index (Leaf length/leaf width). General (GCA) and specific combining abilities (SCA) for all characters of $F_2$ hybrid were significant. In the effect of GCA, yield was greater in combinations of Bulgaria and Xanthi, days to flower and leaves per plant were those of Burley 21 and Va 528, leaf length and width were those of Coker 139, respectively. In the effect of SCA, the major part of characters were greater than those of others in the combinations of Burley 21, Va 528, Hicks.

Genetic Analyses of Heading and Maturing Dates and Their Relationship to Freezing Resistance in Barley (보리 출수기와 성숙기의 유전분석 및 내동성과의 관계)

  • The combination of early heading time, maturing time and short grain-filling period is very important to develop early varieties in winter barley. The 4 parental half diallel crosses (parents, $F_1$s, $F_2$s) were cultivated at the field. The heading date was from April 3 to 26, maturing date from May 15 to 27 and grain-filling period from 31 days to 42 days, showing that the varietal differences about the 3 traits were remarkable. According to half diallel cross analyses, Dongbori 1 for heading time (late heading) was dominant, but Oweolbori (early heading) was recessive, showing partial dominance with high additive component of genetic variance. Dongbori 1 for maturing time was dominant, but Oweolbori was recessive, showing partial dominance with high additive variance. Reno for grain-filling period (short grain-filling period) was dominant, but Oweolbori (long grain-filling period) was recessive with additive, and partial dominance. There were highly significant mean squares for both GCA and SCA effects on the heading and maturing times, and GCA/SCA ratios for all traits were high, showing the additive gene effects more important. Sacheon 6 and Oweolbori had greater GCA effects for early heading and maturing times, and Dongbori 1 and Reno had greater GCA effects for late times. GCA effects were highly significant in $F_1$ and $F_2$ generations, showing high GCA/SCA ratios (7.02). The heading and maturing times in field were positively correlated with antifreeze proteins concentrations, accumulation, resistance to photoinhibition and winter survival, respectively) but the grain-filling period did negatively correlated with the trails.

Studies on the Combining Ability for Silage Yield and Major Agronomic Characters of Corn (사료용 옥수수 선발을 위한 수량 및 주요형질의 조합능력 분석)

  • This experiment was conducted to identify the superior corn hybrids for silage production and to determine the combining ability of their parental inbred lines. A 7-entry diallel cross was evaluated for grain and stover yields, and agronomic characters under 3 different seasons. Hybrids Hi34 x Tx601, Hi26 x Hi34, and Hi29 x Hi34 showed high total dry matter yields. In general, late maturing hybrids had higher grain and silage yields than early maturing hybrids. Leaf area index was correlated with grain and stover yields. Rust rating was negatively correlated with yields and all characters, indicating that rust (Puccinia sorghi) was one of the major factors affecting yields. Diallel analysis showed that inbred Hi34 which was late in maturity and resistant to rust was the best combiner for both grain and silage yields. General combining ability (GCA) and specific com-bining ability (SCA) effects for yields and other characters were significant, suggesting that both additive and non-additive gene effects are involved for those characters. Ratio of GCA/SCA mean squares showed that GCA effects were more important than SCA effects for yields and other characters. GCA x season and SCA x season interactions were significant for grain and stover yields, revealing that gene effects were not stable for those yields under dissimilar environments.

  • Objectives: Many neurological diseases are known to be caused by expansion of trinucleotide repeats (TNRs). It is hard to diagnose the alteration of TNRs with single cell level for preimplantation genetic diagnosis (PGD). In this study, we describe methods optimized for PGD of TNRs related diseases such as Huntington's disease (HD), spinocerebellar ataxia 3 (SCA3) and fragile X syndrome (FXS). Methods: We performed the preclinical assays with heterozygous patient's lymphocytes by single cell PCR strategy. Fluorescent semi-nested PCR and fragment analysis using automatic genetic analyzer were applied for HD and SCA 3. Whole genome amplification with multiple displacement amplification (MDA) method and fluorescent PCR were carried out for FXS. Amplification and allele drop-out (ADO) rate were evaluated in each case. Results: The fluorescent semi-nested PCR of single lymphocyte showed 100.0% of amplification and 14.0% of ADO rate in HD, and 94.7% of amplification and 5.6% of ADO rate in SCA3, respectively. We could not detect the PCR product of CGG repeats in FXS using the fluorescent semi-nested PCR alone. After applying the MDA method in FXS, 84.2% of amplification and 31.3% of ADO rate were achieved. Conclusions: Fluorescent semi-nested PCR is a reliable method for PGD of HD and SCA3. The advanced MDA method overcomes the problem of amplification failure in CGG repeats of FXS case. Optimization of methods for single cell analysis could improve the sensitivity and reliability of PGD for complicated single gene disorders of TNRs.

PEROXIDASE ACTIVITY ASSOCIATED WITH Puccinia sorghi INFECTION IN MAIZE (옥수수에 있어서 녹병감염에 관한 Peroxidase의 활성)

  • Five parents and their 10 $F_1$ crosses of maize (Zea mays L.) were tested by means of diallel analysis for the inheritance of peroxidase activity following Puccinia sorghi rust infection. Peroxidase activity was measured at day zero and at 2 days and 8 days after inoculation. Peroxidase activity was increased significantly by P. sorghi infection in all 15 genotypes after 8 days, but not after 2 days. Highly significant differences in peroxidase activity were detected among the 15 genotypes. The highly genera] resistant inbred, CM105, and its hybrids showed exceptionally high peroxidase activity in both healthy and infected plants. However, another highly resistant inbred, Oh545, showed exceptionally low peroxidase activity. Significant general combining ability (GCA) and specific combining ability (SCA) means quares were detected for peroxidase activity independent of disease. GCA mean spuares, however, were consistently a major contiribution to the inheritance of peroxidase activity in the infected plants whereas SCA men square contributions were minor. Rust resistant maize plants controlling mongenic dominant $Rp_1^d$gene showed stronger peroxidase reroxidase responses than their susceptible counterparts in the gel electrophoresis and densiometric tracings. The increased peroxidase activity occurred in both major leaf peroxidases, $Px_3\;and\; Px_7$.

Studies on Genetic Analysis by the Diallel Crosses in $F_2$ Generation of Cowpea(Vigna sinensis savi.) (동부 Diallel Cross$ F_2$세대의 유전분석에 관한 연구)

  • Genetic studies on the $F_2$ generation of a set of half diallel crosses involving six cowpea varieties were conducted. by the randomized block design with three replications to determine combining ability, gene action and the relationships between parents and their $F_2$ hybrids. The 12 agronomic characters namely, days to flowering, days from flowering to maturity, days to maturity, diameter of stem, length of internode, number of branches per plant, length of pod, number of pods per plant, number of grains per pod, number of grains per plant, 100 grain weight and grain weight per plot were observed, and the $F_2$ generation of this diallel set of crosses was analysed for each character according to the method by Jinks and Hayman. The results obtained are summarized as follows: 1. Vr-Wr graphical analyses; The following seven characters, days to flowering, number of branches per plant, length of pod, number of pods per plant, number of grains per plant, 100 grain weight and grain weight per plot appeared to be partially dominant, and over dominance was found for days from flowering to maturity, days to maturity, length of internode and number of grains per pod. But diameter of stem indicated partial dominance near complete dominance. 2. Estimates of genetic variance components; In the degree of dominance,. eight characters, that is, days to flowering, days from flowering to maturity, days to maturity, length of internode, number of pods per plant, number of grains per pod, number of grains per plant and grain weight per plot were expressed larger than 1. And the characters, days from flowering to maturity, number of branches per plant and number of grains per plant as the degree of mean dominance ($H_1$/D) were found to be negative value over other characters. On the other hand, apprent asymmetry of dominance-recessive allele ($H_2$ /$4H_1$) produced comparatively estimates with lower value on days from flowering to maturity, length of internode, number of branches per plant and number of grains per pod. 3. Analyses of combining ability; Mean square value of GCA(general combining ability) appeared to be more important than those of SCA (specific combining ability) for most characters, and among them, grain weight per plot showed the highest mean square value in GCA and SCA. 4. Effect of combining ability; Variety 178 was expressed as the highest GCA effects in five characters (days to flowering days to maturity, number of pods per plant, number of grains per plant and grain weight per plot). SCA effects were differed from parents, characters and crosses, but crosses between TVu 1857 $\times$ TVu 2885 and TVu 2702 $\times$ J78 were shown to be highly with SCA effects on yield.

Correlation and Combining Ability related to the Growth Characteristics in F1 Hybrids by Diallel Cross of Viola tricolor (팬지의 일대 잡종에서 생육관련 형질의 상관관계 및 조합능력)

  • This study was conducted to determine correlations and combining abilities of leaf lengths, leaf widths, petiole lengths, number of branches, number of leaves, leaf areas, fresh weights, dry weights and number of flowers in F1 crosses made with the partial seven-parent diallel cross in Viola tricolor. Leaf lengths and leaf widths showed highly positive correlation with petiole length and negative correlation with number of branches, number of leaves and number of flowers. There was positive correlation between the number of leaves and flowers as well as between leaf area and fresh weight. Mean squares of general combining ability(GCA) and specific combining ability(SCA) were highly significant for all the parameters. Variance component values of SCA were greater than those of GCA for all the parameters except leaf length, implying preponderance of non-additive gene actions for these characters. The lines C and G for leaf lengths and widths, the lines A and F for number of leaves, the lines A, B and G for leaf areas, and the lines B and F for number of flowers showed relatively high GCA effects. The crosses of A×B and B×D exhibited high SCA effects on increasing leaf lengths, leaf widths, number of leaves, fresh weights and number of flowers. And also the crosses of B×E and D×G exhibited high SCA effects on decreasing leaf lengths, leaf areas and fresh weights as well as increasing number of flowers. The broad sense heritabilities of most characters were high compared with the narrow sense one. Those of leaf length, leaf width, petiole length and number of leaves were high in both the broad and narrow sense heritabilities.
