사료 내 단백질과 지방 수준이 참돔(Pagrus major) 치어의 성장, 사료효율 및 비특이적 면역력에 미치는 영향 (Effects of Dietary Protein and Lipid Levels on the Growth Performance, Feed Utilization and Innate Immunity of Juvenile Red Seabream Pagrus major)

  • A $3{\times}3$ factorial study was conducted to investigate the effects of dietary protein and lipid levels on the growth, feed utilization and innate immunity of red seabream Pagrus major. Nine diets consisting of three protein levels (42%, 46% and 50% crude protein) and three lipid levels (10%, 14% and 18% crude lipid) were formulated. Triplicate groups of red seabream were fed the experimental diets to apparent satiation (5-6 times a day, from 08:00 to 18:00 h at 2-h intervals) for 10 weeks. At the end of the feeding trial, the weight gain and specific growth rate of fish fed P46L14 (46% protein and 14% lipid), P50L10 (50% protein and 10% lipid) and P50L14 (50% protein and 14% lipid) were significantly (P<0.05) higher than those of fish fed P42L18 (42% protein and 18% lipid). The feed conversion ratios (FCR) of the fish were affected by dietary lipid levels (P<0.039), but not dietary protein levels. The FCR tended to increase with increasing dietary lipid levels from 10% to 18% with the 46% and 50% protein levels. The weight gain, protein efficiency ratio, specific growth rate, feed intake and survival of fish were not affected by either dietary protein or lipid levels. Myeloperoxidase activity in the group fed P50L14 (50% protein and 14% lipid) was significantly higher than that in the group fed P42L10 (42% protein and 10% lipid) or P50L18 (50% protein and 18% lipid). However, the myeloperoxidase activity of fish was not affected by either dietary protein or lipid level. The fish fed P46L14 (46% protein and 14% lipid) and P46L18 (46% protein and 18% lipid) showed significantly higher superoxide dismutase activity than did the fish fed P46L10 (46% protein and 10% lipid), P50L10 (50% protein and 10% lipid) of P50L18 (50% protein and 18% lipid). In conclusion, the optimum protein and lipid levels for the growth and feed utilization of juvenile red seabream were 46% and 14%, respectively, and the optimum dietary protein to energy ratio was 27.4 g/MJ.

Identification and Characterization of Two New S-Adenosylmethionine-Dependent Methyltransferase Encoding Genes Suggested Their Involvement in Stipe Elongation of Flammulina velutipes

  • Two new SAM-dependent methyltransferase encoding genes (fvsmt1 and fvsmt2) were identified from the genome of Flammulina velutipes. In order to make a comprehensive characterization of both genes, we performed in silico analysis of both genes and used qRT-PCR to reveal their expression patterns during the development of F. velutipes. There are 4 and 6 exons with total length of 693 and 978 bp in fvsmt2 and fvsmt1, respectively. The deduced proteins, i.e., FVSMT1 and FVSMT2 contained 325 and 230 amino acids with molecular weight 36297 and 24894 Da, respectively. Both proteins contained a SAM-dependent catalytic domain with signature motifs (I, p-I, II, and III) defining the SAM fold. SAM-dependent catalytic domain is located either in the middle or at the N-terminal of FVSMT2 and FVSMT1, respectively. Alignment and phylogenic analysis showed that FVSMT1 is a homolog to a protein-arginine omega-N-methyltransferase, while FVSMT2 is of cinnamoyl CoA O-methyltransferase type and predicted subcellular locations of these proteins are mitochondria and cytoplasm, respectively. qRT-PCR showed that fvsmt1 and fvsmt2 expression was regulated in different developmental stages. The maximum expression levels of fvsmt1 and fvsmt2 were observed in stipe elongation, while no difference was found in mycelium and pileus. These results positively demonstrate that both the methyltransferase encoding genes are involved in the stipe elongation of F. velutipes.

Arabidopsis thaliana의 Ethylene Triple Response Mutant에서 에틸렌 생합성 과정의 생리 생화학적 특성

  • Arabidopsis thaliana의 에틸렌 관련 돌연변이체인 에틸렌 과다 생성 돌연변이체(eto1-1)와 에틸렌에 대한 반응이 둔감한 돌연변이체(etr1-3과 ein2-1)의 생리·생화학적 특성을 분석하였다. 2∼3주된 Arabidopsis 식물로부터 얻은 성숙한 잎을 재료로 이용하였다. 에틸렌 생성량은 eto1-1은 야생형의 2배, etr1-3은 야생형의 4배, 그리고 ein2-1은 야생형의 4.5배 더 많았다. eto1-1에서의 ACC synthase와 ACC oxidase 활성은 야생형과 비슷하였으나 ACC 함량은 야생형보다 4.5배 더 많았다. ACC-N-malo-nyltransferase의 활성은 eto1-1이 야생형보다 3배 더 높았으며 SAM synthetase의 활성은 야생형보다 1.5배 더 높았다. 이들 결과로부터 eto1-1로의 변형은 SAM에서 ACC로 전환하는 과정 이전에 있음을 추측할 수 있다. etr1-3과 ein2-1에서 ACC synthase의 활성은 야생형보다 높았으나 ACC oxidase의 활성은 야생형보다 낮았다. 그러나 SAM synthetase 활성은 etr1-3에서는 야생형과 비슷하였고 ein2-1에서는 야생형보다 1.7배 높았다. 이것은 etr1-3과 ein2-1이 에틸렌에 대한 반응에 결함이 있기 때문에 그것으로 인하여 자가 조절이 되지 않았기 때문으로 추정된다. etr1-3의 ACC 함량은 야생형보다 2.3배 더 많았으며 ACC N-malonyltransferase의 활성은 야생형보다 3.9배 더 높았다. 그리고 ein2-1의 ACC 함량은 야생형보다 1.7배 더 많았으며 ACC N-malonyltransferase의 활성이 촉진된 것으로 추정된다. In vitro kinase assay를 한 결과 eto1-1과 ein2-1에서 36 kDa 단백질의 인산화를 관찰할 수 있었다.

Purification and Characterization of Arylphorin of the Chinese Oak Silkmoth, Antheraea pernyi

  • The arylphorin was purified from the pupal haemolymph of the Chinese oak silkmoth, Antheraea pernyi, and characterized physiologically and biochemically, The protein was purified by a simple preparative polyacrylamide gel electrophoresis (PAGE) and subsequent diffusive elution. The preparation was shown to be homogeneous by 7.5% native-PAGE. The native molecular weight of arylphorin was 450 kDa with a 80 kDa single subunit, suggesting hexamer, The protein contained high amounts (18.3%) of aromatic amino acids, phenylalanine (9.7%) and tyrosine (8.6%). Therefore, the protein was identified as a kind of a storage protein referred to as an arylphorin. The protein was stained by Schiff's reagent, suggesting a glycoprotein. The protein contained 4.9% (w/w) sugar and mannose and N-acetylglucosamine were major components. Also, degradation of the protein was begun by heat treatment at 90 for 20 minutes. These results showed that the A. pernyi arylphorin in the study is hexamer associated with the six subunits consisting of a 80kDa single subunit, and is different from that of Kajiura et al. (1998) in the subunit composition.

Analysis of Diapause-Associated Changes in Silkworm Egg Proteins

  • The patterns of diapause-associated proteins of silkworm eggs were analyzed by two-dimensional (2-D) gel electrophoresis. Among the hundreds of spots on the 2-D gels, at least two proteins were considered to be associated with diapause. A protein, spot 4, with an approximate molecular weight of 38 kDa and pI 6.1 was observed in the HCI-treated, cold-treated, and diapause eggs, respectively. Spot 4 was undetectable in unfertilized eggs and non-diapause eggs at two days after oviposition, suggesting that this protein may be associated with the entrance to diapause. A protein, spot 11, with an approximate molecular weight of 21 kDa and pI of 61 was detected in the unfertilized, HCl-treated, and cold-treated eggs, respectively, after oviposition by normal moths. In diapausing eggs, a protein corresponding to spot 11 was observed in 3-, 5-, and 30-day-old eggs, while the protein was not detected one-day-old eggs. The protein corresponding to spot 11 was not detected in unfertilized and non-diapause eggs obtained from subesophabeal ganglion (SG)-extirpated moths either. Spot 11 was also considered to be a diapause specific protein, which occurred at only early embryonic stage under the control of diapause-downregulated gene.

무균성수막염과 뇌염환자 사이의 인구학과 검사소견의 비교: 나이와 C-반응단백질의 중요성 (Comparison of the Demographic and Laboratory Profiles of Patients with Aseptic Meningitis and Encephalitis: Significance of Age and C-reactive Protein)

  • Background: Viruses can cause either meningitis or encephalitis. It is unclear why some people suffer from aseptic meningitis, and others acquire aseptic encephalitis when infected with the same viral pathogens. The aim of this study was to compare demographic and laboratory factors between patients with aseptic meningitis and encephalitis. Methods: The demographic and laboratory differences were analyzed according to age, sex, diabetes, hypertension, C-reactive protein in the blood, white blood cell and protein in the cerebrospinal fluid, and glucose ratio (cerebrospinal fluid/blood). Additionally, we analyzed the nation-wide differencesin age between the patients with aseptic meningitis and those with encephalitis in Korea. Results: The patients with aseptic encephalitis were older, more likely to have hypertension, and had higher levels of C-reactive protein than did the patients with aseptic meningitis. However, the numbers of white blood cells in the cerebrospinal fluid were significantly higher in the patients with meningitis than in the patients with encephalitis. Multivariable analysis revealed that age >49 years, hypertension and a C-reactive protein level >5.81 mg/dL were independent and significant variables in the prediction of aseptic encephalitis. Additionally, the patients with aseptic encephalitis were older than those with aseptic meningitis in the nation-wide Korean database. Conclusions: Older age, hypertension, and higher levels of C-reactive protein are useful factors for the prediction of aseptic encephalitis.
