A xylanase (xylanase I) was purified 11.9-fold from the culture filtrate of Trichoderma koningii ATCC 26113 by the column chromatography on Sephadex G-75, SP-Sephadex C-50, DEAE-Sephadex A-50 and Sephadex G-50 with an overall yield of 8.2%. The molecular mass determined by gel filtration and sodium dodecyl sulfate polyacrylamide gel electrophoresis was found to be a monomeric polypeptide of ca. 35 kDa. The isoelectric point of the enzyme was estimated to be 9.3. The optimal reaction pH and temperature are 5.8 and 55.deg.C, respectively. The enzyme is stable up to 60.deg.C, while 78% of its activity is lost after the incubation for 10 min at 70.deg.C. The enzyme hydrolyzes sylan with relatively high activity, as well as carboxymethyl cellulose and avicel. The $K_{m}$ values of the enzyme for oat-spelf sylan, larchwood xylan and Avicel were 3.5, 1.6 and 10. 1 mg/ml, respectively. The enzyme hydrolyzed oat-spelt sylan to sylose, sylobiose, sylotriose and arabionoxylobiose, while it degraded larchwood xylan to xylose, xylobiose, xylotriose and arabionoxylobiose, while it degraded larchwood xylan to xylose, xylobiose and xylotriose as the major products. The hydrolysis patterns indicate that xylanase I is endo-enzyme.e.
본 연구는 터널표면의 퇴색 및 변색을 초래할 것으로 생각되는 주요 균사형진균을 결정하고, 그 진균을 방제하기 위해 부근 장소에서 분리된 세균들이 항진균 길항능을 가지는 동시에 탄산칼슘 형성을 확인하였다. 이 균주를 이용하여 모르타르에 적용했을 때 항진균 및 압축강도 증진효과를 가진 세균자원을 확보하는데 그 목적이 있다. 터널내의 오염된 지역에서 시료를 취하여 다양한 배지를 이용하여 곰팡이, 효모 및 세균을 분리하였고, 분리된 진균의 ITS-5.8S rRNA gene sequene와 세균의 16s rDNA sequence를 이용해 부분동정을 실시했다. 분리된 미생물의 터널 내 분포를 결정하였으며, 분리된 세균 5종의 탄산칼슘 형성능력을 확인하였다. 터널내 오염지역에서 가장 널리 분포하는 곰팡이인 C. sphaerospermum KNUC253 과 감수성 시험에 널리 이용되는 공시균인 A. niger KCTC6906을 대상으로 항진균 시험을 실시하였다. 터널 분리세균 5종 모두 urea-$CaCl_2$ 고체배지에서 배양했을 때 콜로니 주변부 에서 종 특이적으로 다양한 크기와 형태의 탄산칼슘을 형성함을 확인하였다. 그 중 B. aryabhatti KNUC205는 감수성 곰팡이를 대상으로 뛰어난 항진균 길항능을 보였다.

The composition changes of litters were investigated to figure out the effects of the decomposition of Humulus japonicus on nutrient circulation and decomposition process in the riverine wetlands: Tan stream and Amsa-dong. Litterbags (mesh size 1 mm and 5 mm) were installed to monitor mass and nutrient changes of 5 types of litters: H. japanicus only, Miscanthus sacchariflarus only, Phragmites communis only, mixed litters including H. japonicus, and mixed litters without H. japonicus for 7 months. It was shown that k (decay rate) of the H. japanicus ($2.68{\sim}3.12$) was higher than that of M. sacchariflorus ($1.83{\sim}2.16$) and P. communis ($0.02{\sim}1.18$). The mass and organic remainings of the mixed litters including H. japonicus at Tan stream were $47.0{\sim}55.1%\;and\;47.0{\sim}54.9%$ and those of the litterbags without H. japanicus were $49.2{\sim}65.4%\;and\;47.1{\sim}57.5%$, respectively. This result indicated that the nutrient circulation was faster at H. japanicus community than others. Ca, Na, Mg, K, P, C, N and H contents reduced to around $40{\sim}80%$ of original. However, Na concentration increased up to $407{\sim}584%$ at 100 days and decreased to $248{\sim}498%$ at the end of the experiment. Decomposition rates were similar between 1 mm and 5mm mesh size litterbags and this implies that plant litters in studied areas decomposed mainly by microbes rather than small animals. This study revealed that the fast growth of H. japonicus was resulted from fast decomposition in part: positive feedback of nutrient cycling.
중국의 명수법은 기록은 구어체를 사용하고, 계산은 산대를 사용하는 이중 구조를 가지고 있었다. 또 산서는 실생활의 문제만 다루는 과정에서 수학적 구조를 나타내는 방법을 택하여 계산 과정을 제외하면 이들에서 취급한 수는 모두 명수(名數)들이어서 순수한 수론의 발전을 이룰 수 없었다. 송대에 0의 도입과 함께, 천원술의 표현에서 나타나는 계수를 산대로 표시하는 방법을 통하여, 산대가 계산 도구와 함께 추상수의 기수법(記數法)이 되는 과정을 밝힌다. 수량의 단위를 사용한 소수의 표현도 이 과정에서 산대 표현으로 대치되었다. 그러나 명대에 산대 계산이 주산으로 대치되고 천원술이 잊히게 되어 추상수의 개념도 함께 잊히게 되었다. 청대의 산학자 심사계(沈士桂)가 저서 간첩이명산법(簡捷易明算法)에서 분수의 소수표시와 계산을 하는 과정에서 순환소수를 인지하고 이들의 계산법을 확립한 것도 보인다.

The scientific information between microbial community and chemical properties of reclaimed tidal soil is not enough to understand the land reclamation process. This study was conducted to investigate the relation between chemical properties and microbial activities of soils from reclaimed tidal lands located at south-western coastal area (42 samples from Goheuong, Samsan, Bojun, Kunnae, Hwaong and Yeongsangang sites). Most of the reclaimed soils showed chemical characteristics as salinity soil based on EC. Only $Na^+$ in exchangeable cation was dependent on EC of reclaimed soil, whereas other cations such as $K^+$, $Ca^{2+}$, and $Mg^{2+}$ were independent on EC. The mesophilic bacteria decreased with an increase in EC of soil. Microbial population increased with soil organic content in the range of $0{\sim}10g\;kg^{-1}$ and dehydrogenase activity less than $100{\mu}g-TPF\;g^{-1}h^{-1}$. Microbial population of soils from reclaimed tidal lands was closely related to the microbial community containing hydrolytic enzyme activities of cellulase, amylase, protease, and lipase.

Purpose: This study is to conduct usability evaluations from the perspective of developing comfort-shoes for the middle-aged and elderly to identify key factors and derive implications for optimal comfort-shoes production. Research design, data, and methodology: A total of 10 middle-aged and elderly women in their 50s and 60s were selected as eligible for the rescue. For data collection, the study was conducted in a Gang Survey, where pre-explanations, shoes test, and interviews were conducted. The collected data were analyzed in a total of four stages. In step 1, the contents obtained through interviews with the subjects were recorded in text, organized and analyzed systematically, and in step 2, unnecessary vocabulary, sentences, and overlapping opinions were eliminated. In step 3, we classified areas around key functions and carried out categorization tasks. Finally, in Step 4, the results and implications of the study were derived by classifying each usability evaluation shoe as positive and negative text around categorized data. Results: There are a total of seven factors for comfort-shoes usability evaluation, which are categorized as cushion, fitting, stability, flexibility, lightweight, comfort, and pressure. Positive/negative factors for the derived usability evaluation factors were shown in the form of a positive-centered, negative-centered, and positive-mixed mix for each of the four products. Positive-focused products are VA products, which are seven times more positive than negative factors. Negative-centered products are CL and SA products, which are five times more negative than positive factors. Positive mixing was a CA product with a ratio of 1:1. Text-based usability evaluations allow us to proceed with analysis based on more scientific data rather than simply listening to opinions and judging by comments. Conclusions: The study discussed implications of developing comfort-shoes for middle-aged consumers and future directions were discussed.
Objectives : Cnidii Fructus is prescribed as the fruit of Cnidium monnieri (L.) Cusson or Torilis japonica (Houtt.) DC. in Korea pharmacopoeia. Although there are differences in the composition of useful components, two species have been used without distinction. In order to discriminate them, DNA sequencing and taste pattern analysis were used in this study. Methods : Primers ITS 1 and ITS 4 were used to amplify the intergenic transcribed spacer(ITS) region of nuclear ribosomal DNA from seven T. japonica and six C. monnieri samples. Taste pattern of samples were measured by using taste-sensing system SA402B equipped with five foodstuff sensors(CT0, C00, AAE, CA0, and AE1). The five initial taste(sourness, bitterness, astringency, umami, and saltiness) and three aftertaste(aftertaste of bitterness, astringency, and umami) of two species were compared. Results : According to the results of ITS region sequence analysis, two species showed 94 base pairs differences. The similarity of two sequences was 85%. From the taste pattern analysis, sourness, bitterness, aftertaste of bitterness(aftertaste-B), and umami showed a different pattern. Especially, bitterness and aftertaste-B of C. monnieri were significantly higher than T. japonica. In addition, two species were shown to have two markedly different clustering by these two flavors. Conclusion : T. japonica and C. monnieri were effectively discriminated using DNA sequencing and taste pattern analysis. These methods can be used to identify the origin of traditional medicine in order to maintain therapeutic efficacy.
만성외상성뇌병증(Chronic traumatic encephalopathy, CTE)은 운동선수와 매우 밀접하게 관련되어 있으며 장기간에 걸쳐 반복적인 외상성뇌손상(traumatic brain injury, TBI)로 인한 퇴행성뇌질환이다. 신경영양인자(neurotrophic factor)는 여러 종류가 알려져 있으며 이들은 뇌와 척수의 물리적 손상시에 신경보호효과가 있다. 따라서, 신경영양인자의 혼합물인 cebrolysin을 이용하여 CTE질환에 가장 적합하다고 여겨지는 repetitive mild TBI (rmTBI) 모델에서 cerebrolysin의 신경보호효과를 알아보고자 하였다. 실험군은 5군(groups 1 and 2: rmTBI for 4 weeks following cerebrolysin injection for 4 weeks; groups 3 and 4: rmTBI for 8 weeks with or without cerebrolysin injection for 4 weeks; group 5: control)으로 나누어 진행하였다. CTE의 가장 대표적 표시인자인 tau 단백질의 인산화를 조직학적으로 조사한 결과, 대뇌겉질과 해마내 CA3 영역에서 phospho-tau단백질의 발현이 증가되었으며 cerebrolysin ($10{\mu}l$ of 1 mg/ml)를 미정맥으로 투여시 p-tau발현이 감소되었다. CTE의 병인으로 알려진 별아교세포와 미세아교세포의 활성을 각각의 표시인인 GFAP, iba-1을 이용하여 면역조직화학염색을 시행하였다. 별아교세포의 활성은 rmTBI에 의하여 증가하였으며 cerebrolysin에 의해 회복되었으나 미세아교세포의 활성은 관찰되지 않았다. 또한 rmTBI모델에서 체내 탐식세포(macrophage)의 뇌내유입유무를 관찰하고자 CD45 염색을 시행하였으나 유의한 차이를 관찰하지 못하였다. 이상의 결과를 종합하면, cerebrolysin이 rmTBI에 의한 tau단백질의 인산화 및 별아교세포의 활성을 조절하는 것으로 사료된다. 따라서 cerebrolysin이 CTE 환자에 대한 치료 약물의 후보가 될 수 있음을 시사한다.
이 연구는 미생물의 생체광물형성작용 중 미생물의 방해석 석출 작용(micro-biologically induced calcite precipitation, MICP)을 이용하여 환경적인 문제를 배려한 차세대 스마트 콘크리트의 개발이 목적이다. 현재 콘크리트의 개질(改質) 및 성능향상을 목적으로 미생물의 방해석 석출 작용을 이용한 기술은 대표적 미생물인 Sporosarcina pasteurii에 의해 그 가능성이 제안되어 왔다. 이 연구에서는 이러한 미생물의 방해석 석출작용을 이용하는 것으로서 선행 연구의 Sporosarcina pasteurii외에 콘크리트 구조물에서 탐색하여 16S rDNA 염기서열 분석법에 의해 동정된 4종의 신규 유용미생물자원들을 추가적으로 이용하였으며, 이렇게 확보된 방해석을 석출하는 유용미생물자원들에 대한 소개와 미생물의 방해석 석출 작용에 따른 시멘트 결정성을 평가하였다. 또한 콘크리트의 개질 및 성능 향상을 목적으로 이러한 유용미생물자원들을 우선적으로 모르타르 환경에 적용하여 양생조건별 압축강도의 특성을 평가하고, 모르타르에 인위적 균열을 만들어 미생물의 방해석 석출 작용에 따른 균열의 충전 가능성을 검토하였다. 이러한 유용미생물들의 적용에 따른 효과는 보수 기능뿐만 아니라 환경 문제를 배려한 새로운 재료로서의 개발로 이어져 향후 더욱 더 중요한 연구주제의 하나가 될 것으로 기대된다. 또한 이 연구의 큰 의미는 실제 콘크리트 구조물에 상생하고, 자연환경에서 방해석을 석출하는 미생물을 이용한다는 것이며, 긴 시간동안 자연환경에서 살아남은 이 미생물들은 환경적으로 안전할 뿐만 아니라 새로운 환경 저부하성 기능재료로서의 이용이 가능할 것으로 판단된다.
현재 고로폐광산에서 가동 중인 인회석 배수체계의 설계는 다음과 같다 ; 인회석 배수시스템은 침사조, 반응조 및 침전조로 구성되며 폐갱도 및 광미장의 침출수를 배관을 통하여 집수한 후 인회석 배수시스템으로 수리구배에 의해 운반되게 설계되었다. 2004년도에 실시된 실내실험 결과에 의하면, 인회석 38.8 톤을 매년 반응조에 새로 충진시켜 줘야 하며 침전물은 매 3개월 마다 침전조에서 제거해줘야 한다. 본 연구의 목적은 석회석 및 인회석을 이용한 실내실험을 실시하여 ARD 내의 비소저감 효율성을 평가하고 침출수 처리 체계 내에 사용되는 침전제를 재평가하는 것이다. 실내실험 결과, 석회석 및 인회석의 비소 저감율은 64.7-98.3%이며 인회석의 경우 비소 저감율은 입도에 반비례하였다. 비소화합물은 비산염인회석 그리고 칼슘비산염수화물의 형태인 것으로 추측된다. 따라서, 인회석 및 인이 포함된 석회석은 저 농도의 비소제거를 위한 침전제로 사용될 수 있는 것으로 판단된다.
