S100A4 Expression is Closely Linked to Genesis and Progression of Glioma by Regulating Proliferation, Apoptosis, Migration and Invasion

  • Background: The calcium-binding S100A4 protein is involved in epithelial to mesenchymal transition, oncogenic transformation, angiogenesis, cytoskeletal integrity, mobility and metastasis of cancer cells. This study aimed to clarify the roles of S100A4 in genesis and progression of glioma. Materials and Methods: S100A4 expression was examined by real-time RT-CPR and Western blot in glioma and paired normal brain tissue (n=69), and compared with clinicopathological parameters of tumors. In addition, glioma U251 cells transfected with an S100A4-expressing plasmid were examined for proliferation by MTT, apoptosis by Annexin V-FITC, and migration and invasion with Transwell chambers. Results: Increased S100A4 mRNA expression was found in gliomas, compared with paired non-tumor tissue (p<0.001). Gradual elevation of overexpression of S100A4 was observed with increasing glioma grade (p<0.001). Astrocytoma showed lower S100A4 mRNA expression than oligodendrogliomas, with glioblastomas having highest values (p<0.001). Similar results were obtained for S100A4 protein, a positive link being found between mRNA and protein expression in gliomas (p<0.001). There was higher growth, lower apoptosis, stronger migration and invasion of S100A4 transfectants than control and mock transfected cells (p<0.001). Conclusions: These findings indicate that up-regulated S100A4 expression is positively linked to pathogenesis, progression and histogenesis of glioma by modulating proliferation, apoptosis, migration and invasion.

  • The male and female muscle of the 16 major bands in the protein patterns of the tissues of the mud-snail, Cipangopaludina chinensis malleata, were almost similar, and the bands E (Mr. 41,500), H (Mr. 52,100), L (Mr. 71,700), N(Mr. 98,500), O (Mr. 107,900) and P (Mr. 112,900) had the same molecular weights which appear in the muscles, testis and ovary. But it showed that the testis and the ovary had their characteristic patterns, and it was also observed that, besides the bands common to all tissues, the bands which appear both in the testis and the ovary are about five: the bands d (Mr. 15,600), k (Mr. 37,100), p (Mr. 57,000), s (Mr. 80,300), and v (Mr. 105,400). The result of the amino acid analysis showed that the quantity and composition of amino acid appeared, as the protein pattersn did, in the order of ovary, muscles and testis. This reveals that in the ovary a great quantity of protein which appears in the whole body was investigated, while the testis contained only little protein due to the phenomenon of morphogenesis in which sperms were developed through the condensation of the nuclei.

Immunohistochemical study on the stomach of the duck, Anas platyrhynchos platyrhyncos Linne (청둥오리 위 내분비세포에 대한 면역조직화학적 연구)

  • The relative frequency and distribution of bovine chromogranin(BCG)-, serotonin-, somatostatin(SOM)-, and S-100 protein-immunoreactive cells in the proventriculus, gizzard and proventriculus-gizzard junction(P-G junction) of the duck(Anas platyrhynchos platyrhyncos, Linne) were studied using immunohistochemical methods. BCG-, and SOM-immunoreactive cells were detected in all ages of this study in the proventriculus and the gizzard, and BCG-immunoreactive cells were also detected in the P-G junction. SOM-immunoreactive cells were detected rarely on occasions only in 1 week, 2 weeks, 3 weeks and 9 weeks after hatching. But no S-100 protein-immunoreactive cells were detected in this study.

Immunohistochemical studies in the intestine of the duck, Anas platyrhynchos platyrhyncos Linne, with ages (연령에 따른 청둥오리 장관 내분비세포에 대한 면역조직화학적 연구)

  • The relative frequency and distribution of bovine chromogranin(BCG)-, serotonin-, somatostatin(SOM)- and S-100 protein-immunoreactive cells in the intestinal tract of the duck (Anas platyrhynchos platyrhyncos, Linne) with ages were studied using immunohistochemical methods. BCG-immunoreactive cells were detected in duodenum on 23 days of incubation. Thereafter these cells were occurred throughout the intestine. SOM-immunoreactive cells were detected in the small intestine from hatching to 9 weeks. Thereafter they were also observed in colon. Serotonin-immunoreactive cells were detected throughout the intestinal tracts after hatching. These immunoreactive cells were increased with ages except that BCG-immunoreactive cells were decreased with ages from 6 weeks after hatching. BCG- and serotonin-immunoreactive cells were most frequently detected in colon regions and SOM-immunoreactive cells in duodenum regions, but no S-100 protein-immunoreactive cells were detected in this study.

Unique epithelial expression of S100A calcium binding protein A7A in the endometrium at conceptus implantation in pigs

  • Objective: S100A7A, a member of the S100 protein family, is involved in various biological processes, including innate immunity, antimicrobial function, and epithelial tumorigenesis. However, the expression and function of S100A7A in the endometrium during the estrous cycle and pregnancy are not well understood in pigs. Therefore, this study determined the expression and regulation of S100A7A at the maternal-conceptus interface in pigs. Methods: We obtained endometrial tissues from pigs throughout the estrous cycle and pregnancy, conceptus tissues during early pregnancy, and chorioallantoic tissues during midto late pregnancy and analyzed the expression of S100A7A in these tissues. We also determined the effects of steroid hormones, estradiol-$17{\beta}$ ($E_2$) and progesterone, and interleukin-$1{\beta}$ (IL1B) on S100A7A expression in endometrial tissues. Results: We found that S100A7A was expressed in the endometrium during the estrous cycle and pregnancy in a pregnancy status- and stage-dependent manner and was localized to endometrial luminal epithelial (LE) and superficial glandular epithelial cells with strong intensity in LE cells on day 12 of pregnancy. Early stage conceptuses and chorioallantoic tissues from day 30 to term pregnancy also expressed S100A7A. The expression of S100A7A was increased by $E_2$ and IL1B in endometrial tissues. Conclusion: S100A7A was expressed at the maternal-conceptus interface at the initiation of implantation in response to conceptus-derived estrogen and IL1B and could be a unique endometrial epithelial marker for conceptus implantation in pigs. These findings provide an important insight into the understanding of conceptus-endometrial interactions for the successful establishment of pregnancy in pigs.

Effect of Temperature on Some Quality Characteristics of Aqueous Extracts of Sea Mustard (미역 추출액의 품질 특성에 미치는 온도의 영향)

  • Effect of temperature on some quality characteristics of aqueous extracts of sea mustard was investigated. The concentration of total solids and protein and their yields in the extracts were increased as the extraction temperature raised from $50^{\circ}C$ to $100^{\circ}C$ during initial period of extraction. However the lower values of the percent of supernatant was obtained at $50^{\circ}C$ and $100^{\circ}C$. The percent of yields of solids and protein for 1 hour extraction at $100^{\circ}C$ were 21.56% and 4.7%, respectively. The highest viscosity, which was $2.5{\sim}4.0$ times of higher values of the other extracts, was obtained by 2 hours extraction at $100^{\circ}C$. The turbidity was gradually decreased after 1 hour extraction. The activation energy calculated for initial increase rate of solid and protein concentration were 1.18 and 3.90 Cal/mole, respectively.

Changes in Specific Gravity, Total Solid and Protein Contents of Human Milk During the Course of Lactation in Korean Women (한국인 모유의 수유기간별 비중, 충고형분 및 단백질 함량의 변화)

  • The lonitudinal determination of specific gravity, total solid and protein contents of human milk were carried out in 27 Korean women. Human milk samples from the subjects were collected at 15, 30, 60, 90, 120, 150th days of lactation. 1) The average values of specific gravity of the milk was 1, 0281 (S.D. 0.0018, n=12), with a range of 1.0200-1.0383. 2) The average values of total soild of the milk was 12.07g/100ml(S.D. 0.38, n=112), with a range of 9.36-15.88g/100ml. 3) The average values of protein content of the milk was 1.20g/100ml (S.D. 0.14, n=112), with a range of 1.09-1.46g/100ml. A slight decrease of specific gravity and protein content in human milk was found during the course of lactation, but significant decrease was not found in total solid content.

Improved recovery of active GST-fusion proteins from insoluble aggregates: solubilization and purification conditions using PKM2 and HtrA2 as model proteins

  • The glutathione S-transferase (GST) system is useful for increasing protein solubility and purifying soluble GST fusion proteins. However, purifying half of the GST fusion proteins is still difficult, because they are virtually insoluble under non-denaturing conditions. To optimize a simple and rapid purification condition for GST-pyruvate kinase muscle 2 (GST-PKM2) protein, we used 1% sarkosyl for lysis and a 1 : 200 ratio of sarkosyl to Triton X-100 (S-T) for purification. We purified the GST-PKM2 protein with a high yield, approximately 5 mg/L culture, which was 33 times higher than that prepared using a conventional method. Notably, the GST-high-temperature requirement A2 (GST-HtrA2) protein, used as a model protein for functional activity, fully maintained its proteolytic activity, even when purified under our S-T condition. This method may be useful to apply to other biologically important proteins that become highly insoluble in the prokaryotic expression system.

  • Ox-RBC로부터 Moddy法에 依하여 Ox-RCM抗原을 抽出하여 몇가지 分析을 實施하였다. Lyophilized 한 Ox-RCM protein의 化學的 分析 結果 total protein은 $55\\pm 1.3%$, total nitrogen은 9.2%, carbohydrate 8.5% 그리고 total acid soluble phosphorus는 $0.96\\pm 0.1%$였다. Polyacrylamide gel electrophoresis한 結果 Ox-RCM은 12 $\\sim$ 15個, Pre-VRS fraction은 8個 및 PHR 分劃은 9個의 pattern을 나타냈다. Amino acid의 分析 結果로 cystine이 4.93 $\\sim$ 3.72 moles/100 moles 이고 glycine이 最高로 26.7 $\\sim$ 13.5 moles/100 moles였고 最低로 methionine 이었으나 試料의 抽出方法에 따라 그 値의 變動을 볼 수 있다.
