Effects of Enzyme Inducers and Glutathione on the Embryotoxicity of Cyclophosphamide in Cultured Rat Embryos (효소유도제 및 glutathione이 전배자배양된 랫드태자에서 cyclophosphamide의 독성에 미치는 영향)

  • Cyclophosphamide (CP) must be enzymatically activated by cytochrome P450(CYP)-linked mixed-function oxidation pathway to be either mutagenic or teratogenic. Influences of alterations in hepatic mixed-function oxidase acitivity and glutathione (GSH) content on the embryotoxicity of CP were studied in rat whole embryo culture system. The embryotoxicity of CP was compared using rat S-9 fraction (S-9) pretreated with chemicals inducing different CYP isozymes, acetone (ACE), Aroclor 1254 (ARO), $\beta$-naphthoflavone (NAF) and phenobarbital (PHE). When 10.5 day embryos were cultured in the immediately centrifuged rat serum for 48 hrs using general gas char{ging schedule, CP$(40{\mu}g/ml)$ with S-9 induced by either NAF or PHE increased the incidence of realformations and significantly decreased embryonic growth compared with the non-induced S-9 group. ACE or ARO induced S-9 group showed no significant difference in embryonic growth. These data suggest that PB and/or NAF inducible CYP isoenzymes are mainly involved in the activation of CP. To examine the effect of GSH on the embryotoxicity of CP, 10.5 day embryos were exposed to CP and S-9 after preincubation with 10 mM of GSH for 3 hrs. In the GSH pretreated group the growth of embryos increased significantly compared with that of the untreated group, suggesting that GSH may protect embryos in culture from some toxic effects of CP.

  • 사람에게 행동의 개인차가 있듯이 rat이나 mouse에 있어서도 행동의 차이를 발견할 수 있다. Rat의 행동성향에 따른 (1)학습 및 기억 능력의 차이, (2)기억과 해마의 관계, (3)치매유발단백질의 하나로 알려진 아밀로이드 베타($A{\beta}$ )및 수종의 항 치매 약물효과를 알아보는 것이 본 실험의 목적이다. Rat의 행동관찰을 통해 두 가지 행동패턴을 관찰할 수 있었는데, 이러한 rat의 행동 특성은 심리학자 Jung이 심리유형으로 설명하고 있는 extraversion, introversion의 행동성향과 유사할 것이라는 가정 하에 실험을 계획, 실시하였다. Rat에 water maze test를 실시하여 공간 기억의 단기, 장기 기억을 분석하였는데 그 결과 두 가지 행동 성향을 가진 rat은 서로 다른 학습 및 기억 능력의 특성을 보였다. 즉, extraversion은 단기 기억의 향상을 보인 반면에, introversion은 장기 기억의 향상을 보였다. Rat을 대상으로 water maze test 외에 Y-maze, passive avoidance test를 실시하여 공간 기억(spatial memory), 작동 기억(working memory), passive avoidance memory, 그리고 단기, 장기 기억의 관계를 종합적으로 분석해 보았다. 그 결과 두 가지 행동성향에 따라 서로 영향을 미치는 기억의 종류 및 관계에 차이가 있음을 발견할 수 있었다. 또한 두 가지 행동성향을 가진 rat에 약물을 투여했을 때, 서로 다른 약물 효과를 보였으며, $A{\beta}$ 를 주입했을 때, 기억(memory) 및 해마(hippocampus) 세포 사멸(cell death)에 서로 상반된 결과를 보여주었다. 이러한 연구 결과는 개체의 행동성향에 따라 학습 및 기억의 효과가 다를 수 있음을 보여주는 결과라 할 수 있고, 개인의 적성과 소질의 인식 및 개발의 중요성에 시사하는 바가 크다. 또한 개개인의 행동과 학습 및 기억 능력의 차이를 두뇌과학적으로 이해하여, 두뇌의 장점은 살리고 단점을 보완할 수 있는 이론적 토대를 세우는데 이러한 동물실험이 그 기초를 제공해 줄 수 있을 것이다. 또한 행동성향 및 기억의 종류에 따른 약물효과의 차이는 기억과 관련된 질병인 알츠하이머 환자에 있어 개개인에게 맞는 적절한 특징적인 치료약물이 존재할 것이라는 가능성을 제공해줄 뿐만 아니라 학습과 기억력 증진 효과를 기대해 볼 수 있을 것이라고 생각된다.

Molecular Evidence for the Presence of Polymorphism in the Gene of S-100 Beta Protein Expressed in Rat Brain (쥐 뇌에서 발현되는 S-100 Beta유전자의 Polymorphism에 대한 분자생물학적 증거)

  • We examined mRNAs, isolated from the rat brain, to ascertain if there is any polymorphism for S-100 beta protein gene. As templates for polymerase chain reaction (PCR) the reverse-transcribed cDNA from the rat brain or phage DNAs isolated from the rat brain cDNA libraries were used. Although PCR products turned out to be exactly same as the expected size based on the previously reported mRNA sequence a single base substitution (CAT to CAC) was identified at nucleotide level. This change was considered as polymorphism since it did not cause any change of the primary structure for S-100 beta protein. This result should facilitate the understanding of the overall structure of the gene for S-100 beta protein.

Primary Culture of Human Hepatocytes from Small Size Sample

  • Human and rat hepatocytes were isolated by nonperfusion method and cultured for longer than 5 days. Human liver biopsy sample and rat liver were used as hepatocyte source. Several physical and chemical factors which were influencing on hepatocyte isolation procedure were examined and a batch isolation procedure was established for small size sample of rat liver. Isolated hepatocytes showed normal morphlologica characteristics in microscopy and electron microscopical examinations and a morphologica response to phalloidin. Isolated cells were cultured as a monolayer and proven to have intact morphological characteristics for longer than 15 days. Because human liver sample is harder and tighter compared with rat liver, a standard procedure for rat hepatocytes was slightly modified to reduce mechanical damage. Similarly with rat hepatocytes, isolated human hepatocytes showed a normal morphological characteristics and could be cultured for longer than 15days. Human and rat hepatocytes were examined on their functional integrities including cytochrome-P450 related enzyme activity and it's inducibility, hormonal inducibility of AIB uptake and TAT activity, albumin synthesis, DNA synthesis, cellular protein maintenance. In all parameters used in the present study, human and rat hepatocytes showed normal functional characteristics.

  • Endotoxin from the cell wall of marine V. vujnificus was .extracted using the hot phenol-water method, injected endotoxin into rat, and tested the toxic effect of endotoxin on the blood component In rat blood. The results showed that blood glucose, blood urea nitrogen, white blood cell, and reticulocyte were Increased and red blood cell was the same as the number of control group(normal blood), but platelet was decreased. Above results suggested that endotoxin induced a malfunction of liver and that the Increase of white blood cell was for the removal of foreign toxic substance.

Functional Recovery Following the Transplantation of Olfactory Ensheathing Cells in Rat Spinal Cord Injury Model

  • Study Design: Olfactory ensheathing cells (OECs) from rat olfactory mucosa were cultured, characterized, and transplanted into a rat model of spinal cord injury (SCI). Purpose: To evaluate different doses of OECs in a rat model of SCI. Overview of Literature: SCI causes permanent functional deficit because the central nervous system lacks the ability to perform spontaneous repair. Cell therapy strategies are being explored globally. The clinical use of human embryonic stem cell is hampered by ethical controversies. Alternatively, OECs are a promising cell source for neurotransplantation. This study aimed to evaluate the efficacy of different doses of allogenic OEC transplantation in a rat model of SCI. Methods: OECs were cultured from the olfactory mucosa of Albino Wistar rats; these cells were characterized using immunohistochemistry and flow cytometry. Rats were divided into five groups (n=6 rats each). In each group, different dosage ($2{\times}10^5$, $5{\times}10^5$, $10{\times}10^5$, and >$10{\times}10^5$) of cultured cells were transplanted into experimentally injured spinal cords of rat models. However, in the SCI group, only DMEM (Dulbecco's modified Eagle's medium) was injected. Rats were followed up upto 8 weeks post-transplantation. The outcome of transplantation was assessed using the Basso, Beattie, Bresnahan (BBB) scale; motor-evoked potential studies; and histological examination. Results: Cultured cells expressed 41% of p75NTR, a marker for OEC, and 35% of anti-fibronectin, a marker for olfactory nerve fibroblast. These cells also expressed $S100{\beta}$ and glial fibrillary acid protein of approximately 75% and 83%, respectively. All the transplanted groups showed promising BBB scores for hind-limb motor recovery compared with the SCI group (p<0.05). A motor-evoked potential study showed increased amplitude in all the treated groups compared with the SCI. Green fluorescent protein-labeled cells survived in the injured cord, suggesting their role in the transplantation-mediated repair. Transplantation of $5{\times}10^5$ cells showed the best motor outcomes among all the doses. Conclusions: OECs demonstrated a therapeutic effect in rat models with the potential for future clinical applications.

Effects of Commelina communis L. on the Blood Glucose Level in Alloxan Induced Diabetic Rat and the Biochemical Properties of Glucose-6-Phosphate Dehydrogenase from the Rat Livers (당뇨유발쥐에서 닭의장풀의 혈당감소효과와 간조직내의 Glucose-6-Phosphate Dehydrogenase의 효소활성에 미치는 효과)

  • The hypoglycemic and metabolic effects of Commelina communis L. extract were investigated in alloxan induced diabetic rats. The increased blood glucose level in the diabetic rats was significantly reduced and the loss of body weight was recovered with the treatment of the plant protein fractions($30{\sim}70%$ ammonium sulfate precipitates). Administration of the plant protein fractions elicited the significant increase of glucose-6-phosphate dehydrogenase (G-6-P DH) activity and liver weight which were decreased in the diabetic rat liver. G-6-P DH was partially purified from extract- or insulin-treated diabetics, diabetic control, and normal rat liver and studied for the biochemical properties. The $K_m$ value(9.002 mM) of diabetic rat liver enzyme was greatly higher than that (0.033 mM) of normal enzyme indicating the affinity of enzyme for the substrate was significantly reduced in the diabetic rat liver. This reduced affinity of enzyme for the substrate in the diabetic rat was recovered in the extract- or insulin-treated rat liver enzyme having 0.164 or 0.208 mM of their $K_m$ values, respectively. Although there was no significant difference in the optimum pH(6.0) and optimum temperature($37^{\circ}C$) of enzyme among the experimental groups, the dependence of their activities on pH appeared to be slightly resistant in the extract- or insulin-treated group compared to the diabetic group. In order to investigate the antigenicity of rat liver enzyme among experimental groups, enzyme-linked immunosorbent assay was carried out by using anti-G-6-P DH anti-serum. Absorbance(0.102) shown in the normal rat liver was reduced even below zero in the alloxan-diabetic rat liver, but increased again in the extract- or insulin-treated rat liver(0.096 or 0.118, respectively). The result of this study suggested that G-6-P DH may be used as a marker enzyme to diagnose and to indicate the progress of the diabetics, and the hypoglycemic effect of the extracts of Commelina communis L. was certainly associated with action or mode of G-6-P DH on the rat liver.

Effects of Silkworm Powder, Mulberry Leaves and Mulberry Root Bark Adminstered to Rat on Gastrointestinal Function (누에분말, 뽕잎 및 상백피 투여가 흰쥐의 장관기능에 미치는 영향)

  • This experiment was carried out to investigate the effect of silkworm powder, mulberry leaves and mulberry root bark administered to S.D. rat on gastrointestinal function. Plot containing ten percent sericultural products(silkworm powder, mulberry leaves and mulberry root bark) was supplied with S.D. rat. The food efficiency ratio(FER) of S.D. rat fed with sericultural products were 0.1486~0.1573. The transit time also were 512.5~574.0 minutes, and transit speed were 25.05~29.11 mm/min. The pH of S.D. rat's feces fed with sericultural products were 5.95~6.72. The daily amount of S.D. rat's feces were 4.12~5.42g. As above results, sericultural products was evaluated to improve the function of S.D. rat's gastrointestine.

Neuroprotective Effects of Herbal Ethanol Extracts from Gynostemma pentaphyllum on L-DOPA Therapy in 6-hydroxydopamine-lesioned Rat Model of Parkinson's Disease (돌외 에탄올 추출물 엑스가 6-hydroxydopamine-유도 파킨슨병 백서 모델에서의 L-DOPA 요법에 미치는 영향)

  • The neuroprotective effects of herbal ethanol extracts from Gynostemma pentaphyllum (GP-EX) in 6-hydroxydopamine (6-OHDA)-lesioned rat model of Parkinson's disease treated with L-DOPA were investigated. Rats were prepared for the Parkinson's disease model by 6-OHDA-lesioning for 14 days. The rats were then treated with L-DOPA (10 and 20 mg/kg) with or without the oral administration of GP-EX (30 mg/kg, daily) for 28 days. L-DOPA (20 mg/kg) treatment for 28 days enhanced dopaminergic neuronal cell death in 6-OHDA-lesioned rat groups, but L-DOPA (10 mg/kg) did not. However, the oral administration of GP-EX (30 mg/kg) for 28 days ameliorated the enhanced neurotoxic effects induced by chronic L-DOPA treatment in 6-OHDA-lesioned rat groups by increasing tyrosine hydroxylase (TH)-immunohistochemical staining and the number of TH-immunopositive cells surviving in the substantia nigra. In addition, GP-EX administration (30 mg/kg) for 28 days recovered the levels of dopamine and norepinephrine of the striatum in 6-OHDA-lesioned rat groups, which were markedly reduced by L-DOPA treatment (20 mg/kg). GP-EX (30 mg/kg) did not produce any signs of toxicity, such as weight loss, diarrhea, or vomiting in rats during the 28-day treatment period. These results suggest that GP-EX has protective functions against chronic L-DOPA-induced neurotoxic reactions in dopaminergic neurons in the 6-OHDA-lesioned rat model of Parkinson's disease. Therefore, GP-EX may be beneficial in the prevention of adverse symptoms in parkisonian patients.

  • Samples of two species of genus Ratts(black rat, Rattus rasttus Linnaeus; common rat, Rattus norvegicus Berkenhaut) in Korea were trapped and their 31 morphometric charcters were analyzed statistically in order to determine the range of geographic variation within each species and the interspecific differences. In addition, chromosomal G-bands and C-bands were compared and the fragment patterns of mtDNA resulted from the digestion with restriction enzymes were also analyzed. Samples of black rats from six localities in Korea were similar with one another in their morphometric characters: in head and body length, length of tail vertebrae, conventional karyotype and C-bands, they are comparable to Rattus rattus tanezumi in Japan. Specimens of common rats from seven localities in Korea were similar with one another in their morphometric characters: in conventional karyotype, they are comparable to Rattus norvegicus caraco in eastern Asia. Common rats differ from black rats in their morphometric characteris, chromosomal karyotypes and mtDNA. It is confirmed that correct species name of black rat in Korea is Rattus rattus tanezumi Tempminck: species name of common rat in Korea is Rattus norvegicus caraco Pallas: the common rat is a species, which is distinct from the black rat.
