Biphasic Study to Characterize Agricultural Biogas Plants by High-Throughput 16S rRNA Gene Amplicon Sequencing and Microscopic Analysis

  • Process surveillance within agricultural biogas plants (BGPs) was concurrently studied by high-throughput 16S rRNA gene amplicon sequencing and an optimized quantitative microscopic fingerprinting (QMF) technique. In contrast to 16S rRNA gene amplicons, digitalized microscopy is a rapid and cost-effective method that facilitates enumeration and morphological differentiation of the most significant groups of methanogens regarding their shape and characteristic autofluorescent factor 420. Moreover, the fluorescence signal mirrors cell vitality. In this study, four different BGPs were investigated. The results indicated stable process performance in the mesophilic BGPs and in the thermophilic reactor. Bacterial subcommunity characterization revealed significant differences between the four BGPs. Most remarkably, the genera Defluviitoga and Halocella dominated the thermophilic bacterial subcommunity, whereas members of another taxon, Syntrophaceticus, were found to be abundant in the mesophilic BGP. The domain Archaea was dominated by the genus Methanoculleus in all four BGPs, followed by Methanosaeta in BGP1 and BGP3. In contrast, Methanothermobacter members were highly abundant in the thermophilic BGP4. Furthermore, a high consistency between the sequencing approach and the QMF method was shown, especially for the thermophilic BGP. The differences elucidated that using this biphasic approach for mesophilic BGPs provided novel insights regarding disaggregated single cells of Methanosarcina and Methanosaeta species. Both dominated the archaeal subcommunity and replaced coccoid Methanoculleus members belonging to the same group of Methanomicrobiales that have been frequently observed in similar BGPs. This work demonstrates that combining QMF and 16S rRNA gene amplicon sequencing is a complementary strategy to describe archaeal community structures within biogas processes.

  • 최근 들어 신시간 멀티미디어를 지원하는 응용들이 증가됨에 따라, 하나의 도메인 내에서의 서비스 품질보장을 지원하기 위한 멀티캐스트 프로토콜에 대한 연구가 증가하고 있다. 그러나 진정한 서비스 품질 보장을 위해서는 도메인간 환경에서의 서비스 품질보장이 이루어져야 한다. 본 논문에서는 도메인간 환경에서 서비스 품질보장 멀티캐스트 트리 구축을 위한 새로움 메커니즘, COINQoS 알고리즘을 제안하였다. 또한 서비스 품질보장을 위한 네트워크 토폴로지 정보 전송을 위하여 기존의 BGP4를 확장한 서비스 품질보장 BGP4를 추가로 제안하였다. COINQoS는 멤버 도메인으로의 경로 설정을 위하여 각 AS들이 자신들이 제공하는 서비스를 몇 개의 클래스로 구분하여 라우팅 정보를 관리하도록 하였고, 통과 도메인에서의 흐름상태 정보의 양을 줄이기 위하여 AS의 출구 노드와 QoS 요구사항을 기반으로 흐름 집단화를 수행하였다.

QoS Support Mechanisms in Mobile MPLS VPN (이동 MPLS VPN에서의 QoS 지원 방안)

  • Network based VPN(Virtual Private Network) using MPLS(Multiprotocol Label Switching) technology, called PE(Provider Edge router) based MPLS VPN, is regarded as a good solution for intranets or ext3nets because of the low cost and the flexibility of the service provision. In this paper, we describe a mechanism that allows the VPN users to move from one site to another site of the VPN network based on the BGP-E MPLS technology. This mechanism is designed for PE(Provider Edge) routers of the backbone network. PE routers connected to the VPN sites establish a new MPLS path to the mobile node after they detect movement of the mobile VPN node. The new location may belong to the same VPN or to different VPN. We desisted VPN management and control functions of the PE routers in order to interface with the Mobile IP protocol and support the QoS mechanism. The pilot implementation and performance measurement were carried out on a simulation using COVERS tool.

  • 본 고에서는 Peer모델 PPVPN 기술인 BGP/MPLS를 비롯하여, 현재 IETF의 PPVPN WG에서 논의되고 있는 IP기반의 여러 가지 PPVPN 프로토콜들의 기술적 특징 및 장단점을 비교하고, 이들의 연구동향 및 발전방향에 대하여 분석한다. 3계층의 IPSec VPN은 확장성이 부족한 단점을 가지며, 2계층 MPLS VPN의 확장성을 개선한 BGP/MPLS는 보안성 및 범용성에 문제가 있다. 이러한 PPVPN 프로토콜의 기술적 특성을 강화하기 위하여 각 기술들을 융합하거나 기존 기술을 개량하는 등의 다양한 제안 및 논의가 이루어지고 있다. VPN 사용자의 다양한 욕구를 만족시켜 줄 필요성과 네트워크 운용의 효율화를 통한 사업자의 수익증대 측면에서 기존의 IP PPVPN 기술들은 향후에 지속적인 개선과 발전이 예상되며, 이는 본 고에서 논의하는 바와 같이 확장성, 보안성 등의 특성을 강화하는 방향으로 진행될 것이다.

  • VPN(Virtual Private Network)은 물리적인 장비나 회선을 논리적으로 구분하여 기존 전용회선을 기반으로 하는 사설 네트워크에 비해 상대적으로 적은 구축 비용, 융통성 있는 확장성, 저렴한 운용비용의 장점을 보유한 별도의 사설 네트워크를 구성하는 기술이다. 다양한 VPN 기술 중에서도 MPLS VPN 은 확장성, QoS 제공을 용이하게 해주는 장점을 가지고 있어 차세대 네트워크 기술로 부각되고 있다. 일반적으로 MPLS VPN 은 ISP 에서 제공하는 서비스로 인식되나 규모가 크고 다양한 사설 네트워크를 필요로 하는 기업의 네트워크 모델로도 적합하다. 또한 Virtual Router VPN 은 Customer Edge(CE) 장비로서 활용성이 큰 Switch 장비에 적용하여, CE 를 MPLS VPN 과 쉽게 연동하게 한다. 본 논문에서는 MPLS VPN 및 Virtual Router VPN 을 이용하여 기업 네트워크를 효과적으로 구성하는 방안을 기술한다.

Provider Provisioned based Mobile VPN using Dynamic VPN Site Configuration (동적 VPN 사이트 구성을 이용한 Provider Provisioned 기반 모바일 VPN)

  • Increase in the wireless mobile network users brings the issue of mobility management into the Virtual Private Network (VPN) services. We propose a provider edge (PE)-based provider provisioned mobile VPN mechanism, which enables efficient communication between a mobile VPN user and one or more correspondents located in different VPN sites. The proposed mechanism not only reduces the IPSec tunnel overhead at the mobile user node to the minimum, but also enables the traffic to be delivered through optimized paths among the (mobile) VPN users without incurring significant extra IPSec tunnel overhead regardless of the user's locations. The proposed architecture and protocols are based on the BGP/MPLS VPN technology that is defined in RFC24547. A service provider platform entity named PPVPN Network Server (PNS) is defined in order to extend the BGP/MPLS VPN service to the mobile users. Compared to the user- and CE-based mobile VPN mechanisms, the proposed mechanism requires less overhead with respect to the IPSec tunnel management. The simulation results also show that it outperforms the existing mobile VPN mechanisms with respect to the handoff latency and/or the end-to-end packet delay.

Whitening and inhibiting NF-κB-mediated inflammation properties of the biotransformed green ginseng berry of new cultivar K1, ginsenoside Rg2 enriched, on B16 and LPS-stimulated RAW 264.7 cells

  • Background: Main bioactive constituents and pharmacological functions of ripened red ginseng berry (Panax ginseng Meyer) have been frequently reported. Yet, the research gap targeting the beneficial activities of transformed green ginseng berries has not reported elsewhere. Methods: Ginsenosides of new green berry cultivar K-1 (GK-1) were identified by HPLC-QTOF/MS. Ginsenosides bioconversion in GK-1 by bgp1 enzyme was confirmed with HPLC and TLC. Then, mechanisms of GK-1 and β-glucosidase (bgp1) biotransformed GK-1 (BGK-1) were determined by Quantitative Reverse Transcription-Polymerase Chain Reaction and Western blot. Results: GK-1 possesses highest ginsenosides especially ginsenoside-Re amongst seven ginseng cultivars including (Chunpoong, Huangsuk, Kumpoong, K-1, Honkaejong, Gopoong, and Yunpoong). Ginseng root's biomass is not affected with the harvest of GK-1 at 3 weeks after flowering period. Then, Re is bioconverted into a promising pharmaceutical effect of Rg2 via bgp1. According to the results of cell assays, BGK-1 shows decrease of tyrosinase and melanin content in α-melanocyte-stimulating hormone challenged-murine melanoma B16 cells. BGK-1 which is comparatively more effective than GK-1 extract shows significant suppression of the nuclear factor (NF)-κB activation and inflammatory target genes, in LPS-stimulated RAW 264.7 cells. Conclusion: These results reported effective whitening and anti-inflammatory of BGK-1 as compared to GK-1.

Detection of the Portent of Distributed DoS Attacks on the Internet AS-level Topology (인터넷 AS 레벨 토폴로지에서 분산서비스거부 공격 징후 탐지)

  • Despite lots of efforts to obtain an accurate picture of structure at the level of individual ASes, there is a few application works using the AS-level Internet topology. In this paper, we show that the power-law fits the number of down-stream customer ASes very well and also present the distributions of AS links with the "public view" from UCLA IRL laboratory. Moreover, we obtain the distributions of source-destination pairs of routing hops for two sites in Korea and the United States, and then we propose a new method to decide the randomness of Internet traffic using the obtained distributions and the BGP valley-free routing policy. The randomness of traffic must be a portent of outbreak of the distributed denial-of-service attacks.

Gram-Scale Production of Ginsenoside F1 Using a Recombinant Bacterial β-Glucosidase

  • Naturally occurring ginsenoside F1 (20-O-${\beta}$-$\text\tiny{D}$-glucopyranosyl-20(S)-protopanaxatriol) is rare. Here, we produced gram-scale quantities of ginsenoside F1 from a crude protopanaxatriol saponin mixture comprised mainly of Re and Rg1 through enzyme-mediated biotransformation using recombinant ${\beta}$-glucosidase (BgpA) cloned from a soil bacterium, Terrabacter ginsenosidimutans Gsoil $3082^T$. In a systematic step-by-step process, the concentrations of substrate, enzyme, and NaCl were determined for maximal production of F1. At an optimized NaCl concentration of 200 mM, the protopanaxatriol saponin mixture (25 mg/ml) was incubated with recombinant BgpA (20 mg/ml) for 3 days in a 2.4 L reaction. Following octadecylsilyl silica gel column chromatography, 9.6 g of F1 was obtained from 60 g of substrate mixture at 95% purity, as assessed by chromatography. These results represent the first report of gram-scale F1 production via recombinant enzyme-mediated biotransformation.

  • In paper, we present design and implementation of MPLS based VPN with QoS capabilities. We propose a design architecture of an edge router for MPLS based VPN with QoS capabilities in backbone network. We design functional blocks, interface specifications, detail routing and forwarding information based structure, and packet forwarding engine for the purpose of implementation of an edge router, We extend bgp-4 software for implementation differentiated service packet forwarding schemes in MPLS based VPN.
