Comparison of Bacterial Diversity in the Water Columns of Goseong Deep Seawaters (고성 심해에서 수심에 따른 해양미생물의 다양성 비교)

  • Microbial diversities in the 300 m and 500 m deep seawaters near Goseong, Gangwon Province (South Korea), were investigated. Pyrosequencing of 16S rRNA genes of marine microbes resulted in 19,474 reads from the 300 m deep seawaters, which consisted of Alphaproteobacteria (57.41%) and Gammaproteobacteria (38.85%), and 82,806 reads from the 500 m deep seawaters, which consisted of Gammaproteobacteria (99.64%) mostly. Rhodobacterales (57.31%) were dominant in the 300 m deep seawaters, but Alteromonadales (45.65%) and Oceanospirillales (34.61%) were dominant in the 500 m deep seawaters. On the bases of operational taxonomic units and diversity indexes (Shannon and Simpson), biodiversity of marine bacteria in the 500 m deep seawaters was shown to be higher than that in the 300 m deep seawaters.

Effect of Salt Accumulation on the Germination and Growth of Lettuce(Lactuca Sativa, L.) (염류집적(鹽類集積)이 상추의 발아(發芽) 및 생육(生育)에 미치는 영향(影響))

  • Study on germination ratio and growth of lettuce affected by accumulated salt in vinyl house cultivation soils was investigated by pot test with EC 1.65. 3.50, 5.75. 7.15. 9.50 and 13.57 dS/m. (Germination rate of lettuce in different electric conductivity of 0, 2, 4, 6, 8 and 10 dS/m controlled with KCl were 96.7, 96.7, 87.8, 82.2, 52.2, and 27.8 % respectively. Standing ratio of lettuce in soil below 6 dS/m was more than 60% and in soils of 7.15, 9.50 and 13.57 dS/m they were 45, 32 and 31%, respectively. Growth and fresh weight of lettuces increased significantly in a low EC content soil. The fresh weight of lettuces in the soil of EC 3.50 dS/m was higher than that of the soil EC 1.65 dS/m by 22%, while another soils(EC: 5.75, 7.15, 9.50 and 13.57 dS/m) were decreased 3, 15, 60 and 62%, respectively. Relationship between soil EC and standing ratio of lettuce showed high correlation coefficient($r=-0.9057^{**}$). Therefore, in the field of vinyl houses concentrated salt, standing ratio of lettuce can be foreseen by soil EC [Y = -4.313x+ 82.95 (Y:standing ratio, x:soil EC)], also standing ratio and fresh weight of lettuce showed high correlation coefficient($r=0.8396^{**}$).

A Study on Development of Machine-readable Platform for S-100 ECDIS

  • Logistics movement using ships has been used for many centuries, but maritime accidents are still difficult to predict and they occur on a large scale. The International Maritime Organization (IMO) made it mandatory to install Electronic Chart Display and Information System (ECDIS) for safe seas, but S-57 standards applied to equipment were not updated over 30 years. In addition, it is difficult to keep the equipment up-to-date and revise standards as ships move around the world. In consideration of the limitations of S-57, the IHO developed the new standard as S-100 rather than an update of the S-57. In this study, the previously developed S-100 tool and S/W were configured as one platform and applied to the S-100 ECDIS. The platform conducts as the S-100 cycle and ecosystem from the creation of standards to release and practical use in ships. The hydrographic information standard is machine-readable as defined in S-100, and it has been validated that the latest standard can be applied using the Plug and Play (PnP) function without software system updates. It is expected that international organization such as the International Hydrographic Office (IHO) and the International Association of Marine Aids to Navigation and Lighthouse Authorities (IALA) shall develop and release standards and mariners can easily apply the latest standards to equipment through the machine-readable platform.

  • 1996년에 3판이 발행된 S-57 표준은 디지틀 수로데이터의 국제적인 교환 표준형식이며 ENC는 S-57을 기본형식으로 하고 특정 상품을 위한 생산품 규약에 의해 만들어진 것이다. ECDIS에 사용되기 위해 만들어진 디지틀 해도 데이터 상품이지만 앞으로 예상되는 ENC 데이터의 일반적 목적을 갖고 있는 사용자들이 모두 S-57과 ENC 생산품 규약을 이해하고 데이터를 읽기 위해 시간과 노력을 투자하는 것은 무척 소비적인 일이 될 것이다. 즉 사용자의 요구는 데이터의 형식이 아니라 데이터의 사용에 집중된다. 그러므로 사용자가 데이터의 내부형식을 알지 못하더라도 사용하고자 하는 의도에 맞게 데이터의 내용을 보여주고 필요한 형식으로 재가공하여 사용할 수 있도록 데이터를 제공하는 프로그램이 필요하며 이는 ENC의 보급 및 활용도를 높이는데 매우 중요하다고 할 수 있다. 본 논문에서는 Data Loader 개발의 기초 단계로 ENC 데이터를 텍스트 형태로 전환하여 ENC에 담긴 내용을 파악할 수 있도록 하는 ENC Data Loader의 기능과 프로그램 개발과정에 대하여 기술하고 텍스트가 아닌 그림으로 보여주며 일반적인 데이터 교환형식으로 변환하는 둥의 새로운 모듈 개발과정을 설명한다. 이를 위해 ENC 데이터의 형식과 내용을 분석하고 GIS 측면에서의 데이터 해석을 시도한다. 또한 이러한 프로그램을 이용한 향후 ENC 데이터의 활용방안에 대하여 기술한다.

  • This project defines Tiled-SENC format for S-57 ENC data usage of the mobile platforms and low-end terminals which are not included in ECDIS carriage requirements of SOLAS Chapter V. Also, the architecture modeling and converting technology of Tiled-SENC have been developed and the performance has been compared with ENC Format.

Cloning and characterization of a gene encoding ABP57, a soluble auxin-binding protein

  • Auxin-binding protein 57 ($ABP_{57}$), a soluble auxin-binding protein, acts as a receptor to activate plasma membrane (PM) $H^+-ATPase$. Here, we report the cloning of abp57 and the biochemical characterization of its protein expressed in E. coli. The analysis of internal amino acid sequences of $ABP_{57}$ purified from rice shoots enabled us to search for the corresponding gene in protein DB of NCBI. Further BLAST analysis showed that rice has four abp57-like genes and maize has at least one homolog. Interestingly, Arabidopsis seems to have no homolog. Recombinant $ABP_{57}$ expressed in E. coli caused the activation of PM $H^+-ATPase$ regardless of the existence of IAA. Scatchard analysis showed that the recombinant protein has relatively low affinity to IAA as compared to natural $ABP_{57}$. These results collectively support the notion that the cloned gene is responsible for $ABP_{57}$.
