Bacillus sp. $\beta$-mannanase was purified by DEAE-sephadex ion exchange column chromatography. The specific activity of the purified enzyme was 21.57 units/$m\ell$ protein, representing an 95.33-folds purification of the original crude extract. The final preparation thus obtained showed a single band on SDS-polyacrylamide gel electrophoresis. The molecular weight was determined to be 38.9 kDa. Guar gum galactomannan was hydrolyzed by the purified $\beta$-mannanase, and then the hydrolysates was separated by activated carbon column chromatography and Sephadex G-25 gel filtration. The main hydrolysates were composed of D.P. (Degree of Polymerization) 5 and 7 galactomannooligosaccharides. To investigate the effects of guar gum galactomannooligosaccharides on in vitro growth of Bifidobacterium longum, B. bifidum, B. infantis, B. adolescentis, B. animalis, and B. breve, Bifidobacterium spp. were cultivated individually on the modified-MRS medium containing carbon source such as D.P. 5 and D.P. 7 galactomannooligosaccharides, respectively B. longum and B. bifidum grew up l0-fold and 9.8-fold more effectively by the treatment of D.P. 5 galactomannooligosaccharides, compared to those of standard MRS medium. Especially, D.P. 5 was more effective than D.P. 7 galactomannooligosaccharide on the growth of Bifidobacterium spp.
Bacillus thuringiensis is a well-known species of entomophathogenic bacteria that is widely used as a biopesticide against many insect pests. It produces parasporal crystals ($\delta$-endotoxin) and endospores during sporulation. In this report, the $\delta$-endotoxin produced by Bacillus thuringiensis BT-1 and BT-2 were characterized by Scanning Electron Microscope(SEM), Transmission Electron Microscope(TEM), SDS-PACE, and solubilization activity by alkaline solution. BT-1, BT-2 were cultured in the GBY medium, and the $\delta$-endotoxin of them was purified with discontinuous sucrose density gradient centrifugation. Their $\delta$-endotoxin was observed by SEM and TEM. Morphologically, the $\delta$-endotoxin of BT-1 was a square and flat type, whose size was $1.73{\mu}m{\times}0.7{\mu}m$, and the $\delta$-endotoxin of the BT-2 was spherical form whose size was $1.1{\mu}m{\times}0.9{\mu}m$ determined by SEM and TEM. The $\delta$-endotoxin of the BT-1 was composed of 28 kDa and 21 kDa, however, it of the BT-2 was composed of 50 kDa, 35 kDa, and 22 kDa bands determined by SDS-PACE. The purified crystals of BT-1 and BT-2 were dissolved gradually in alkaline solution as time goes by, and it was perfectly dissolved after 3 hours. It is supposed that the $\delta$-endotoxin of crystal was converted to a state of activation in the course of time in the intestines of insect.
Random amplified polymorphic DNA (RAPD) markers were used to identify the genetic diversities among and within varieties and landraces of Rehmannia glutinosa. Polymorphic and reproducible bands were produced by 10 primers out of total 20 primers used in the experiment. In RAPD analysis of the 11 genotypes, 64 fragments out of 73 amplified genomic DNA fragments were polymorphic which represented an average 6.4 polymorphic fragments per primer. Number of amplified fragments with random primers ranged from 2 (OPA-1) to 13 (OPA-11) and varied in size from 200 bp to 1,400 bp. Especially, OPA-10, OPA-11 and OPA-19 primers showed specific bands for varieties of Korea Jiwhang and Jiwhang il ho, which could be useful for discriminating from other varieties and landraces of R. glutinosa. Percentage polymorphism ranged from a minimum of 50% (OPA-1) to a maximum of 100% (OPA-11), with an average of 87.7%. Similarity coefficients were higher in the genotypes of Korea Jiwhang and Jiwhang il ho than in other populations. In cluster analysis, genotypes of Korea Jiwhang, Jiwhang il ho, and Japanese accession were separated from those of other varieties and landraces. Average of genetic diversity within the population $(H_S)$ was 0.110, while average of total genetic diversity $(H_T)$ was 0.229. Across all RAPD makers the $G_{ST}$ value was 0.517, indicating that about 52% of the total genetic variation could be explained by RAPDs differences while the remaining 48% might be attributable to differences among samples. Consequently, RAPD analysis was useful method to discriminate different populations such as domestic varieties and other landraces. The results of the present study will be used to understand the population and evolutionary genetics of R. gllutinosa.

Powdery mildew (PM) caused by Podosphaera aphanis is a major disease that can result in significant yield losses in strawberry (Fragaria ${\times}$ ananassa Duchesne). For preventing PM, pesticides are usually applied in strawberry. In this study, molecular markers were developed to increase breeding efficiency of PM-resistance cultivars by marker-assisted selection (MAS). An $F_2$ population derived from a cross between PM-resistance 'Seolhyang' and PM-susceptibility 'Akihime' was evaluated for disease resistance to PM and RAPD (random amplification of polymorphic DNA)-BSA (bulked segregant analysis). Among 200 RAPD primers tested, OPE10 primer amplified a 311bp-band present in with 331bp. Sequence alignment performed for searching polymorphisms and six single nucleotide polymorphism (SNP) were found in amplified regions. To develop polymorphic marker for distinguishing between resistant and susceptible, RAPD was converted to cleaved amplified polymorphic sequence (CAPS) marker. Among restriction enzymes associated with six SNPs, Eae I (Y/GGCCR) was successfully digested to 231bp in susceptible. The results suggest that the selected CAPS marker could be used for increasing efficiency of selecting powdery mildew resistant strawberry in breeding system.
The KT(Kauth-Thomas) and IHS(Intensity-Hue-Saturation) transformation techniques were introduced and compared to investigate fire-scarred areas with single post-fire Landsat 7 ETM+ image. This study consists of two parts. First, using only geometrically corrected imagery, it was examined whether or not the different level of fire-damaged areas could be detected by simple slicing method within the image enhanced by the IHS transform. As a result, since the spectral distribution of each class on each IHS component was overlaid, the simple slicing method did not seem appropriate for the delineation of the areas of the different level of fire severity. Second, the image rectified by both radiometrically and topographically was enhanced by the KT transformation and the IHS transformation, respectively. Then, the images were classified by the maximum likelihood method. The cross-validation was performed for the compensation of relatively small set of ground truth data. The results showed that KT transformation produced better accuracy than IHS transformation. In addition, the KT feature spaces and the spectral distribution of IHS components were analyzed on the graph. This study has shown that, as for the detection of the different level of fire severity, the KT transformation reflects the ground physical conditions better than the IHS transformation.
Alginate lyase from Streptomyces violaceoruber was purified by DEAE sephacel chromatography and SP sepharose chromatography. The specific activity of the purified enzyme was 14.6 units/mg protein, representing a 40.6-fold purification of the crude extract. The final preparation thus obtained showed a single band on Tricine-SDS polyacrylamide gel electrophoresis whose molecular weight was determined to be 23.3 kDa. The polyMG block of sodium alginate was hydrolyzed by the purified alginate lyase and then separated by activated carbon column chromatography and bio gel P-2 gel filtration. The main hydrolysates were composed of hetero type M/G-oligosaccharides with the degrees of polymerization (D.P.) being 6 and 8. To investigate the effects of hetero type M/G-oligosaccharides from the sodium alginate on the growth of some intestinal bacteria, cells were cultivated individually on the modified-MRS medium containing D.P. 6 and 8 M/G-oligosaccharides. B. longumgrew 4.25-fold and 6.44-fold more effectively by the treatment of D.P. 6 and 8 M/G-oligosaccharides compared with those of standard MRS medium. In addition, B. bifidumgrew 3.3-fold and 5.4-fold more effectively by the treatment of D.P. 6 and 8 M/G-oligosaccharides. In conclusion, D.P. 8 was more effective than D.P. 6 hetero M/G-oligosaccharides as regards the growth of Bifidobacteriumspp. and Lactobacillus spp.
The Marine Environment Observation Information System supplies oceanographic information elements such as water temperature, chlorophyll, float, etc. based on satellite images to consumers. The data produced by the Korean marine environmental observatories are located in the coastal areas of Korea. But if the range is too far from a particular area of interest, due to distance or spatial constraints, the accuracy and up-to-dateness of the data can not be relied upon. Therefore, it is necessary to perform fusion and complex operation to solve the difference between the field observation and the marine satellite image. In this study, we develop a system that can process marine environmental information in the user's area of interest in the form of layered character (numeric) information considering the readability and light weight rather than the satellite image. In order to intuitively understand satellite image information, we characterize (quantify) the marine environmental information of the area of interest and we process the satellite image band values into layered characters to minimize the absolute amount of transmitted / received data. Also we study modular location-based interest information service method to be able to flexibly extend and connect interested items that diversify various observation fields as well as application technology to serve this.
Recently, land cover spectral libraries have been widely used in studies to classify land cover based on hyperspectral images. Overseas, various institutions have built and provided land cover spectral libraries, but in Korea, the building and provision of land cover spectral libraries is insufficient. Against this background, the purpose of this study is to suggest the possibility of using domestic and foreign spectral libraries in the classification studies of domestic land cover. Band matching is required for comparative analysis of the spectral libraries and land cover classification using the spectral libraries, and in this study, an automation logic to automatically perform this is presented. In addition, the directly constructed domestic land cover spectral library and the existing overseas land cover spectral library were comparatively analyzed. As a result, the directly constructed land cover spectral library had the highest correlation coefficient of 0.974. Finally, for the accuracy evaluation, aerial hyperspectral images of the study area were supervised and classified using the domestic and foreign land cover spectral libraries using the SAM technique. As a result of the accuracy evaluation, it is judged that Soils, Artificial Materials, and Coatings among the classification items of the foreign land cover spectral library can be sufficiently applied to classify the cover in Korea.
With the recent increase in dementia patients due to aging, measures to prevent their wandering behavior and disappearance are urgently needed. To solve this problem, various authentication methods and location detection techniques have been introduced, but the security problem of personal authentication and a system that can check indoor and outdoor overall was lacking. In order to solve this problem, various authentication methods and location detection techniques have been introduced, but it was difficult to find a system that can check the security problem of personal authentication and indoor/outdoor overall. In this study, we intend to propose a system that can identify personal authentication, basic health status, and overall location indoors and outdoors by using wristband-type wearable devices in a private blockchain environment. In this system, personal authentication uses ECG, which is difficult to forge and highly personally identifiable, Bluetooth beacon that is easy to use with low power, non-contact and automatic transmission and reception indoors, and DGPS that corrects the pseudorange error of GPS satellites outdoors. It is intended to detect wandering behavior and abnormal signs by locating the patient. Through this, it is intended to contribute to the prompt response and prevention of disappearance in case of wandering behavior and abnormal symptoms of dementia patients living at home or in nursing homes.

CaF2 single crystal has a large band gap (12 eV), and it is used for optical windows, prisms, and lenses due to its excellent transmittance in a wide wavelength range and low refractive index. Moreover, it is expected to be one of the materials for ultraviolet transmissive laser optical components. CaF2 belongs to the fluoride compounds and has a face-centered cubic (FCC) structure with three sub-lattices. The representative method for CaF2 single crystal growth is Czochralski, which method has the advantages of high production efficiency and the ability to make large crystals. In this study, X-ray diffraction (XRD), X-ray rocking curves (XRC) measurement, and chemical etching were performed to analyze the crystallinity and defect density of the CaF2 single crystals, grown by the Czochralski method. Fourier-transform infrared spectroscopy (FT-IR) and UV-VIS-NIR spectroscopy systems were used to investigate the optical properties of the CaF2 crystal. The provability of various applications, including UV application, was systematically investigated with various analysis results.
