Efficient Regeneration of Plants Independent of Exogeneous Growth Regulators Using Tissues in Pepper (Capsicum annuum L.) (고추의 조직을 이용한 생장조절제 처리가 식물체 재분화 효율에 미치는 영향)

  • The study was carried out to develop efficiency of transformantion in pepper plants. We used cotyledons of two native varieties, 'Subicho' and 'Kumtap' to establish some conditions of plant regeneration. Differentiation rate of shoot was higher in 2 to 4 mg/l zeatin and 0.05 mg/l NAA than in other treatments. Meanwhile, differential rate of roots was the highest in 0.5 mg/l NAA.

Nitrogen Removal using Zeolite at On-site Wastewater Treatment System (소규모 오수처리 시스템에서의 제올라이트에 의한 질소 제거)

  • Recently, absorbent biofilters, which are inexpensive and easy to manacle, have been supplied to the rural areas, but have limitations in removing the nutrients effectively. Accordingly, as an alternative plan. natural zeolites were arranged in front or at the rear of the absorbent biofilters, and their removal efficiency for nitrogen and, ultimately, their applicability to the on-site wastewater treatment system were studied. Furthermore, the same experiments were carried out on artificial zeolites, made from coal ashes at National Honam Agricultural Experiment Station, to compare natural zeolites with artificial ones. Treated wastewater through the Absorbent Biofilter showed 22.6% nitrogen removal efficiency, while 64.6% was attained when natural Zeolites were placed in front of the absorbent biofilters (Zeolite-Aerobic process). As an addition, phosphorus was also efficiently removed. On the other hand, Aerobic-Zeolite process, which arranged natural zeolites at the rear of the biofilters, did not have significantly higher nitrogen removal as compared to the treatment using only the absorbent biofilters. Furthermore, upon regeneration of the natural zeolite, the ion exchange rate was fecund to increase over 10% as compared to before regeneration. Our results show that natural zeolites, applied to the on-site wastewater treatment system through the Zeolite-Aerobic process, not only increase the removal efficiency of nutrients, but, by choosing the appropriate regeneration time, can also be cast-effective. Artificial zeolites, on the other hand, though more efficient in removing nutrients, cannot be regenerated and, therefore, are not cost-effective.

New embryogenesis from atypical bodies and plant regeneration from long-term subcultured embryogenic callus in rose (장기간 계대배양 된 장미 배발생 캘러스로부터 식물체 재분화 및 비정형체로부터 새로운 배발생캘러스 재생)

  • Long-term subcultured rose embryogenic calluses, which had been maintained for more than 5 to 6 years since the first embryogenesis from calluses induced from in vitro roots of rose, were identified as potential material for the development of transgenic plants. The first embryogenic calluses from 'Sweet Yellow' and two breeding lines (KR056002 and KR056006) were obtained in 2007 and 2009, respectively. Subsequently, we found that plants regenerated from long-term embryogenic calluses (LEC). Whereas the LEC from 'Sweet Yellow' takes 3 to 4 months to regenerate plants, those of the two breeding lines take 4 to 5 months. This period of time is the same as that taken for plants to regenerate from the first embryogenic callus. New embryogenesis was observed from atypical bodies (ABs) that appeared during the process of long-term subculture. We found that it is possible to use the AB as a material for new embryogenesis.

A Revitalize Rural Hub Project in Hwayang-eup by Introducing the Concept of Place Marketing (장소마케팅 개념을 도입한 화양읍 농촌중심지 활성화 사업 계획)

  • In a situation in which rural areas are declining, local governments are seeking to revitalize areas by place marketing. Place marketing, defined by various efforts to promote the image of a place, has been used as an economic tool. As a result, the image has been over-promoted and marketing has been driven in a perfunctory manner, so individual residents' lives and experiences have been ignored. Thus, in addition to the traditional types of cultural place marketing and economic place marketing, this study established a 'project for rural revitalization of Hwayang-eup' so that it could be applied to political place marketing aimed at inducing internal investment and improving the welfare of local residents. To implement this project, the concept was set up as building network organization, sustainable development and symbiotic relationship, and various H/W and S/W plans were developed. First of all, in terms of political place marketing, the Hwaeyang Oulim Center was constructed to strengthen the capacity of local autonomous organizations. In terms of cultural place marketing, we explored cultural resources at the village level and created a small community space. In terms of economic place marketing, the landscape around the main street and the township was reorganized to create a cultural business space for urban and rural exchanges. The reinterpretation of place marketing seen through this project was first, it was more process-oriented than results, second, it was important to induce the community-participating village-making project, and finally, the role of experts was important to expand the community movement.

Protoplast Regeneration, Reversion and Isolation of Auxotrophic Mutants in Flammulina velutipes (팽이버섯 (Flammulina velutipes)원형질체(原形質體)의 재생(再生), 환원(還元) 및 영양요구성(營養要求性) 균주선발(菌株選拔))

  • Factors affecting the regeneration, reversion of protoplasts from mycelium of F. velutipes were investigated and the selection of auxotrophic mutants from protoplasts of F. velutipes was performed. PDP medium stabilized with 0.6M sucrose was suitable for the regeneration of protoplasts, and regeneration frequency was 0.47-1.32. The regeneration frequency of protoplasts was increased when nutrients were added to the regeneration medium. Especially, yeast extract was the most effective to regeneration of protoplasts. Regeneration pattern of protoplasts was formation of germ tubes from bud-like cells. 13-18% of monokaryotic strains was appeared from reverted protoplasts. Five of auxotrophic mutants were isolated from strains showed survival frequency of 1.9-16.

A Study on the Characteristics of Rural Convention in Geongsang Region - Focused on the Rural Development Projects, Hierarchical Structure and Living Area - (경상지역 농촌협약 특징 - 일반 농산어촌개발사업, 계층구조, 생활권 설정을 중심으로 -)

  • The purpose of this paper is to provide basic data to Si and Gun preparing for rural agreements in the future through consideration and case analysis for rural conventions, such things as living standards, hierarchical analysis methods, and general rural development projects. In the 1970s, sporadic and one-off projects focused on individual projects were repeatedly carried out in rural areas, which were undergoing dramatic changes in population movement, land use, landscape, economic growth and living. The keyword of the government's policy has changed to a way that existing rural development projects are planned and promoted by local governments themselves in conjunction with the issue of autonomy and decentralization. In addition, with the introduction of Rural Convention, the central and local governments are striving to achieve common policy goals. However, due to the lack of understanding of the Rural Convention introduced as a pilot project in 2020, the lack of living areas setting-standards, unclear classification of hierarchical structure, and excessive goal consciousness for general agricultural and fishing village development projects remain, fading the original purpose of the Rural Convention. Therefore, this study analyzed the seven reports of the Gyeongsang region selected in the 2021 Rural Convention to identify the characteristics of living area, hierarchical structure analysis, and detailed projects (rural center revitalization project, basic living base development project) and provided them as basic data. As a result of the study, first, according to the results of the hierarchical structure analysis, the first class was derived from 6 out of 7 Si and Gun. This is judged to be the result of the current rural population, social, and economic problems. Second, according to the results of the division of living areas, it was confirmed that except for one local government in seven cities and counties, the remaining six local governments were divided into three single living areas and three two living areas. This is the result obtained through a comprehensive review by overlapping the results of various analyses such as hierarchical analysis and resident surveys and OD data, and shows a different form from the existing upper or related plans. It is judged that this is the result of including qualitative contents such as residents' opinions along with various analyses when determining the living area. Third, the proportion of the budget for rural center revitalization and basic living base development projects to the total project cost subject to the Rural Convention was 59.6%, which was relatively higher than that of other general agricultural and fishing village development projects. It is judged that it is preferred because the unit project cost is relatively high in Si and Gun, and that quantitative results can be visually shown after the project is terminated.

An Analysis of Abandoned House in Gyeongsangnam-do - Comparison of Urban and Rural Areas - (경상남도 빈집실태 분석 - 도시와 농촌지역 비교 연구 -)

  • There are approximately 1.06 million vacant homes in Korea, accounting for 6% of all housing, and approximately 98 thousand vacant homes are in Gyeongsangnam-do, taking up 8.7% of the housing in the province. In particular, the number of abandoned house in Gyeongsangnam-do is rapidly increasing from 6.7% in 2010 to 8.7% in 2015. This study examined the current state of abandoned house in Gyeongsangnam-do according to the characteristics of cities and rural areas, causes for vacancies, and demand for use of such abandoned house. The study areas were selected based on case studies in Korea and overseas, and a survey was conducted among residents in the selected areas. The survey was on awareness of abandoned house in the neighborhood, causes for vacancies, awareness of owners, the influence of the abandoned house, plans to deal with abandoned house, and plans to utilize abandoned house. The results showed that most residents were aware of abandoned house, especially the ones in rural areas, who had detailed perceptions about abandoned house nearby such as when the homes became empty and whether there are owners of the homes. As for the influence of abandoned house on the surrounding environment, both rural areas and cities responded that abandoned house mostly spoil the beauty of the neighborhood, followed by psychological anxiety such as fear of crimes. Regarding plans to utilize abandoned house nearby, at least half of the residents wanted to use them as rental housing after repairing them. A survey on the actual condition must be conducted at the provincial level in order to establish an integrated system to manage abandoned house in Gyeongsangnam-do. Moreover, it is necessary to come up with application measures according to categories and standards to classify the state of abandoned house in order to use them as resources for community regeneration.

  • This study organize the concept of sustainable development by compiling New-urbanism, sustainable urban regeneration and theory of development. And the framework of analysis about the planning characteristics of New Urbanistic Waterfront and urban development was derived by compiling theory of Waterfront and Waterfront city and considering prior study. This study will analyze the advanced cases at home and abroad through the analysis of the urban development and architectural characteristics analysis system of the New Urbanism Waterfront and Waterfront City Development by analyzing the advanced cases of the domestic and foreign countries through the analysis.
