The production of therapeutic proteins for human diseases in plants results in many economic benefits, including reduced risk of animal virus contamination, high yields, and reduced production and storage costs. Human cytokines, interleukin-11 (hlL-11) and granulocyte-macrophage colony-stimulating factor (hGM-CSF), cDNAs were introduced into rice or tobacco, using either the maize ubiquitin promoter or the 35S promoter. The primary hIL-11 transgenic rice plants exhibited stunted growth and a sterile phenotype, whereas the hIL-11 transgenic tobacco plants did not. This suggests that hIL-11 expression in rice disrupts the normal growth and development of the plant. The regeneration efficiency of rice calli transformed with hGM-CSF was found to be approximately a quarter of that seen with the hIL-11, suggesting that hGM-CSF expression is more deleterious to the regeneration of rice calli than is hIL-11. However, the surviving hGM-CSF transgenic rice plants exhibited a normal phenotype of growth. Therefore, it appears that only those transgenic rice lines that expressed the human cytokines in small quantities were able to survive the selection process.
Flower buds of the mulberry(Morus alba L., Morus bombycis Koidz.) were cultured under different conditions such as basal media, and various concentrations of plant growth substances. Effects of the culture conditions on growth of the buds and organ regeneration were investigated and the result obtained are as follows: Murashige and Skoog(M.S.) medium was more effective on budding and growth of female(Keomseolppong) and male(Kaeryangppong) flower buds isolated directly from branches, compared to Greshoff & Doy(G.D.) and Wolter & Skog(W.S.) media. The growth of the female buds was promoted at higher concetration of benzyl amino purine(BAP) i.e., 2.0ppm. The female and male buds cultured after cuuting for seven days showed better growth than those without cutting treatment. The females and the males bloomed to form healthy stigmas and anthers, respectively, when cultured on M.S. media containing high Kinetin with low concentration of indole acetic acid(IAA).
Immature embryogenesis is a useful process in wheat tissue culture, including transgenic technology, because of its high regeneration efficiency compared to that in other tissues. However, it is a very labor-intensive and time-restrictive method, because the preparation of immature embryos is limited to the optimal time after flowering. In this experiment, 'Speed Breeding', a breeding technique that accelerates breeding generation advancement by extending the photoperiod, was applied to the wheat variety 'Bobwhite'. A controlled growth room was constructed by adjusting the photoperiod (22-hour light/2-hour dark) using LED lights at temperature of 22℃. After vernalization of the Bobwhite seeds at 4℃ for 4 weeks, the seedlings were grown in a controlled growth room and a greenhouse to compare the heading date. In both conditions, calli were induced from immature embryos on the 11th day after flowering. After 4 weeks, the calli were transferred to a regeneration medium. Regeneration efficiencies under greenhouse conditions and Speed Breeding conditions were determined as 45.05% and 43.18%, respectively. Antioxidant enzyme activity and reference gene expression analysis were performed to confirm the presence of stress due to an extremely long-day photoperiod. As a result, the antioxidant enzyme activity was not distinguished from that of the greenhouse condition. The reference gene expression analysis revealed that the PsaA and CDC genes were highly expressed under the Speed Breeding condition. However, expression of PsbA was similar expression in both conditions. These results will provide useful information for the application of immature embryogenesis to the wheat transformation system.

The purpose of this study is to examine the policy direction and execution plans of the Japanese central and local governments for the use of vacant houses after the enactment of the "Special Act on Vacant Houses." It also seeks to provide suggestions for establishing vacant house policies in Korea by identifying the characteristics of the Japanese policies, and to suggest policy alternatives that can contribute to environmental improvement and regional revitalization through the use of vacant houses. For this purpose, we examined Japan's "Special Act on Vacant Houses," the process of establishing vacant house policies in the Japanese central and local governments, and the contents and direction of these policies. Below are the results of this study. First, the Japanese government revised mid to long-term policies related to housing after the enactment of the "Special Act on Vacant Houses", thereby providing ground for local governments to establish vacant house policies. Second, the government actively supported the operation and funding of related associations so that the private sector rather than the public sector could address vacant house problems. Third, the government subsidized administrative expenses such as cost of finding vacant house owners to prevent neglect of vacant houses, and showed its will to address vacant house problems by revising taxes such as property taxes and transfer income taxes. Fourth, the government has enacted distinct ordinances for each local government and made it possible to perform customized policies such as allowing residents and local stakeholders to participate in the establishment of vacant house policies. Fifth, the government made it possible to manage vacant houses efficiently by allowing the vacant house bank to provide all information related to the use of vacant houses. It is necessary for Korea's central and local governments to refer to these precedent cases and create efforts to reduce trial and error when establishing vacant house policies.

Gwangju Metropolitan City has implemented a creative regeneration project of Gwangju Folly in the site of Gwangju-eup Fortress as part of the Gwangju Design Biennale by inviting well-known international architects. This study examined and analyzed the characteristics of location, place, and space through actual survey. The results were as follows. Gwangju Folly were mainly located at the four gates and corners of Gwangju-eup Fortress, main entrances of Asia Culture Complex, and historical places where the May 18 Democratization Movement occurred. The common place of Gwangju Folly was a footpath and common location types were the full location of footpath width and the partial location of footpath width. For the spatial types of Gwangju Folly, the practical type which people can stay was the most common(2/3). In the partial location of footpath width, the type which people take a rest and look out over the surroundings accounted for a half. In the full location of footpath width, the gate type which people pass accounted for 2/5. However, as footpath width was minimum for walking, both partial and full occupation types were narrow in place. It influenced the image of Gwangju Folly. Gwangju Folly did not play as a figure and show architects' intentions clearly because of their narrow locations. Therefore, it is very necessary to make a plan to maintain places so that Gwangju Folly do not have a cramped image and architects' intentions become clear with citizens' cultural competence. Also, urban property which creates the identity and attraction of Gwangju continuously should be settled down through helping citizens recognize the intention and value of artistic works.

The study aims to find regional characteristics of Olle and Olle-Dam the old town of Jeju city, "Mu-geun-sung," through researches of the status of the spatial structure in the area. The research was mainly conducted through literature survey and field survey. Literature survey was conducted for the typification of alleys and stone walls. Field surveys were conducted to reveal the location, length, and materials of the stone walls on alleys. As a result, this study can grasp the spatial characteristics of the alleys and the typological characteristics of the stone walls in the old urban areas of Jeju City. The unit alley located in the old city area of Jeju was severely damaged. Also, the stone walls could reveal that there are few areas where the original shape is preserved. The management of the community has not been properly carried out in the use of alleys. In addition, there is concern about the damage caused by securing a road in the future. On the other hand, many of the damaged stone walls had basically the original shape. There were many stone walls that retained their original shape in the old alley. For this reason, there is a high possibility that the alleys and stone walls located in the old urban areas of Jeju are utilized as assets of the area. Based on the results of the above study, community recovery using old alleys of Jeju is required.
Cellulose, chitin, chitosan and hyaluronic acid are well known as polysaccharides. These polysaccharides have many effects on cell growth and differentiation. Cell activation increases with increasing the polysaccharides concentration. In this study, gelatin scaffold containing microorganism fermented cellulose, citrus gel were prepared by using irradiation technique. Physical properties of the scaffolds were investigated as a function of the concentrations of gelatin and citrus gel and the cell attachment, cell morphology and inflammation of the scaffolds also were characterized for regeneration of skin tissue.

A community-centered smart village process model was developed through the theoretical review of the rural field forum. By analyzing the difference in technology demand according to the digital capabilities of leaders by community type, village types were classified and detailed technologies were defined. The smart village process was proposed to enable residents to operate autonomously by inducing continuous interest and participation of local residents through the conception stage, planning stage, implementation and self-reliance stage, and allowing them to cooperate together. The business model canvas was reconstructed to be used in the workshop. It was applied to the village of Yesan-gun. As a result of running a resident workshop using the business model Cambus, the lack of resident awareness and illegal garbage dumping were presented as the first problems to be solved. The value of the village was set as 'a village that is clean and clean with a sense of residents, and a good place to live', and users were expressed as 'family' and 'outsiders'. It was suggested that we meet frequently to convey the value of the village by using broadcasting and announcements as a channel to convey the value. Core activities were to cultivate residents' consciousness, such as implementing a campaign against illegal garbage dumping, and to establish and guide separate collection sites. When a garbage collection center is installed, it was estimated that around 2 million won per month for management costs, and it was investigated that it was possible to spend an hour or so twice a month to solve the problem of illegal dumping. If a method to derive village projects based on the derived business model canvas is developed in the future, it will be more practical.

Urban flood management(UFM) strategy ought to consider the connections and interactions between existing and new infrastructures to manage stormwater and improve the capacity to treat water. It is also important to demonstrate strategies that can be implemented to reduce the flow at flooding sources and minimize flood risk at critical locations. Although the general theory of spatial impact is popular, modeling guidelines that can provide information for implementation in real-world plans are still lacking. Under such background, this study conducted a modeling research based on an actual target site to confirm the hypothesis that it is appropriate to install green infrastructure(GI) in the source area and to take structural protection measures in the impact area, as summarized in previous studies. The results of the study proved the hypothesis, but the results were different from the hypothesis depending on which hydrological performance indicators were targeted. This study will contribute to demonstrating the effectiveness of strategies that can be implemented to reduce the flow at flooding sources and minimize the risk of flooding in critical locations in terms of spatial planning and regeneration.

The urban park plays important roles in protecting the urban landscape and improving citizens' health, recreation, and the emotional life. Above and beyond these roles, the urban park is expected to rearrange urban spatial structures as a kind of urban system. The purpose of this paper is to identify empirically to change urban spatial structures by construction of the urban park. This study regards gentrification around the urban park as a process to change urban spatial structures. The gentrification means the regeneration and upgrading of deteriorated urban property by the middle class or commercial developers. The site of case analysis is the Seoul Forestry Park in Sungdong-gu, Seoul. The Seoul Forestry Park is regarded as a representative urban park of Seoul, and caused gentrification around park after the 2005 opening. This study operationally defines the gentrification index and the accessibility index from an urban park and offers an empirical analysis of relation among the urban park, the gentrification and urban spatial structure in a statistic district which is the minimum unit of Korean statistic data in 2000, 2005, and 2010, using Difference-in-Difference method and linear probability model. The results of this empirical study show that the Seoul Forestry Park changes urban spatial structures by gentrification. It reverses a trend of migration of gentrifiers before and after construction of the Seoul Forestry Park. It suggests urban park construction as an alternative method for urban regeneration by inducing the middle class into the inner city of Seoul.
