INORGANIC SELENIUM FOR SHEEP I. SELENIUM BALANCE AND SELENIUM LEVELS IN THE DIFFERENT RUMINAL FLUID FRACTIONS

  • The effects of inorganic selenium (Se), selenate and selenite on Se balance levels in the different ruminal fluid fractions were studied using Japanese Corriedale wethers with an average body weight of 47 kg. A $3{\times}3$ Latin square design was used with three animal, three periods and three treatments. In each period, there was 7 d dietary adjustment followed by 5 d total collection of urine and feces. Ruminal fluid samples were obtained at 0, 1, 3, 5 and 7 h postprandially on the final day of the collection period. The three dietary treatments were: (1) without Se supplementation (control); (2) with Se supplement as sodium selenate; and (3) sodium selenite at a rate of 0.2 mg Se/kg dietary DM. The basal diet was timothy hay (Phleum pratense L.) fed 2% of body weight/d. Results indicated that Se balance were higher (p < 0.05) for those animals under supplementation than those animals under control. Overall data gathered showed a similar digestion balance of selenate and selenite in sheep. Inorganic Se, both selenate and selenite produced positive Se contents of the ruminal feed particles and protozoa. Bacterial Se increased (p < 0.05) on the first three hours post-prandially in Se supplemented diets. Gross ruminal fluid fraction, although there was improvement on their Se content under the supplemented diets, the changes were insignificant over the control. free inorganic Se and Se in soluble protein of the ruminal fluid were not significantly different for selenate and selenite. Most of the Se in the ruminal fluids of the animals under supplementation were insoluble, indicating the influence of rumen environments on Se bioavaliability.

INFLUENCE OF DIETARY PROTEIN ON THE FRACTIONATION OF SELENIUM IN THE RUMEN OF SHEEP

  • The effects of diets differing in protein content through soybean meal supplementation on ruminal fractionation of selenium (Se) were studied. A $3{\times}3$ Latin square design was used with three Japanese Corriedale wethers (45 kg average body weight), three periods, and three dietary treatment. The three dietary treatments were : Diet 1, without soybean meal supplementation (14% crude protein, CP); Diet 2, with 10% soybean meal supplementation (16.5% CP); and Diet 3, with 20% soybean meal supplementation (19% CP). All the diets had a Se supplementation in the form of sodium selenite at 0.2 mg Se/kg dietary DM. The Se supplement and the concentrate mixture were fed only in the morning before the hay was given. Daily feeding schedule for gay was set at 09:00 and 17:00 h. On the final day of collection period, ruminal fluid samples were obtained at 0.5, 2, 6, 12 and 24 h post-feeding starting at 09:00 h. Total ruminal fluid Se was markedly higher (p<0.05) in Diet 3 than those in Diets 1 and 2 at almost all sampling time except at 24 h. The proportion of Se in soluble protein to the total ruminal Se was higher (p< 0.05) in Diet 3 (40%) followed by Diet 2 (28%) and Diet 1 (21%). The proportion of free inorganic Se to the total ruminal Se was the reverse, especially after two hours where Diet 1 (p<0.05) was higher than the other diets. Bacterial Se was lower (p < 0.05) in Diet 1 than those in Diets 2 and 3 at any sampling time. The highest was observed at 2 h postprandially in all diets with a value of 421, 556, $655{\mu}g/kg$ bacterial DM for Diet 1, 2 and 3, respectively. No differences (p>0.05) were observed on ruminal pH, ammonia and total nolatile fatty acids although increasing protein supplementation tended to decline the ruminal pH and increase ruminal ammonia. This study concludes that increasing dietary protein content by soybean meal supplementation can affect the ruminal Se metabolism.

INORGANIC SELENIUM FOR SHEEP II. ITS INFLUENCE ON RUMEN BACTERIAL YIELD, VOLATILE FATTY ACID PRODUCTION AND TOTAL TRACT DIGESTION OF TIMOTHY HAY

  • This study was conducted to determine the effect of inorganic selenium (Se) sources on rumen bacterial yield, ruminal volatile fatty acid (VFA) production and total tract digestion of timothy hay (Phlewm pratense L.) in Japanese Corriedale wethers. A $3{\times}3$ Latin square design was used with three wethers, three periods and three treatments. In each period, there was 7 d dietary adjustment followed by 5 d total collection of urine and feces. Ruminal fluid samples were obtained at 0, 1, 3, 5 and 7 h postprandially on the final day of the collection period. The three dietary treatments were: (1) without Se supplementation (control); (2) with Se supplementation as sodium selenate; and (3) sodium selenite at a rate 0.2 mg Se/kg dietary DM. The basal diet was timothy hay fed at 2% of body weight/d. Results indicated that there was slight decrease in rumen bacterial yield of animal supplement with inorganic Se, however, differences over the control were insignificant. It was found that Se content of ruminal fluid was negatively correlated (p < 0.05) to rumen bacterial yield. The various VFA contents and acetate and propionate ratio of the different ruminal fluid samples were insignificant across treatment means and the same manner was observed to the different digestibilities (DM, OM, CP, NDF, ADF and NDS). This study concludes that Se supplementation at 0.2 mg Se/kg dietary DM either from sodium selenate or sodium selenite could not significantly influence rumen bacterial functions.

  • Effects of oral administration of electrolyte solutions were studied in experimentally dehydrated adult sheep. By the latin square method five ruminal fistulated sheep were examined and dehydrated by deprivation of feed and water for 72 hours. Tap water, physiological saline, 0.45% NaCl+120 mM/L glucose and 0.9% NaCl+1% propylene glycol solution were orally administrated after dehydration, respectively. Rehydration effect and modification of the rumen function were compared. 1. After 72 hours of deprivation of feed and water, sheep were hypertonic dehydrated and blood acid-base parameters were not significantly changed. And there was marked increase in ruminal pH and decrease in ruminal total volatile fatty acid(VFA) concentration. 2. After the fluids administration the changes in blood acid-base parameters were not significant in all groups. 3. Although glucose fermentation in the rumen was observed, 0.45% NaCl+120 mM/L glucose was more effective in rehydration than physiological saline and tap water. But it was difficult to know the rehydration effect of 0.9% NaCl+1% propylene glycol solution exactly because of excessive increase in plasma osmolality. 4. After refeeding, total concentration and proportions of ruminal volatile fatty acid(VFA) were not significantly different among groups and recovered to normal concentration but not in proportions after 2 days in all groups. 5. In vitro cultured ruminal protozoa were susceptible to the decrease of the pH and osmolality.

Influence of Wheat Straw Pelletizing and Inclusion Rate in Dry Rolled or Steam-flaked Corn-based Finishing Diets on Characteristics of Digestion for Feedlot Cattle

  • Eight Holstein steers ($216{\pm}48kg$ body weight) fitted with ruminal and duodenal cannulas were used to evaluate effects of wheat straw processing (ground vs pelleted) at two straw inclusion rates (7% and 14%; dry matter basis) in dry rolled or steam-flaked corn-based finishing diets on characteristics of digestion. The experimental design was a split plot consisting of two simultaneous $4{\times}4$ Latin squares. Increasing straw level reduced ruminal (p<0.01) and total tract (p = 0.03) organic matter (OM) digestion. As expected, increasing wheat straw level from 7% to 14% decreased (p<0.05) ruminal and total tract digestion of OM. Digestion of neutral detergent fiber (NDF) and starch, per se, were not affected (p>0.10) by wheat straw level. Likewise, straw level did not influence ruminal acetate and propionate molar proportions or estimated methane production (p>0.10). Pelleting straw did not affect ($p{\geq}0.48$) ruminal digestion of OM, NDF, and starch, or microbial efficiency. Ruminal feed N digestion was greater (7.4%; p = 0.02) for ground than for pelleted wheat straw diets. Although ruminal starch digestion was not affected by straw processing, post-ruminal (p<0.01), and total-tract starch (p = 0.05) digestion were greater for ground than for pelleted wheat straw diets, resulting in a tendency for increased post-ruminal (p = 0.06) and total tract (p = 0.07) OM digestion. Pelleting wheat straw decreased (p<0.01) ruminal pH, although ruminal volatile fatty acids (VFA) concentration and estimated methane were not affected ($p{\geq}0.27$). Ruminal digestion of OM and starch, and post-ruminal and total tract digestion of OM, starch and N were greater (p<0.01) for steam-flaked than for dry rolled corn-based diets. Ruminal NDF digestion was greater (p = 0.02) for dry rolled than for steam-flaked corn, although total tract NDF digestion was unaffected (p = 0.94). Ruminal microbial efficiency and ruminal degradation of feed N were not affected (p>0.14) by corn processing. However, microbial N flow to the small intestine and ruminal N efficiency (non-ammonia N flow to the small intestine/N intake) were greater (p<0.01) for steam-flaked than for dry rolled corn-based diets. Ruminal pH and total VFA concentration were not affected ($p{\geq}0.16$) by corn processing method. Compared with dry rolled corn, steam-flaked corn-based diets resulted in decreased acetate:propionate molar ratio (p = 0.02). It is concluded that at 7% or 14% straw inclusion rate, changes in physical characteristics of wheat straw brought about by pelleting negatively impact OM digestion of both steam-flaked and dry-rolled corn-based finishing diets. This effect is due to decreased post-ruminal starch digestion. Replacement of ground straw with pelleted straw also may decrease ruminal pH.

Effects of Dietary Addition of Surfactant Tween 80 on Ruminal Fermentation and Nutrient Digestibility of Hanwoo Steers

  • A non-ionic surfactant, Tween 80 has been known to exert a number of positive effects on degradative enzymes in in vitro aerobic and anaerobic microbial cultures. An experiment was conducted to examine effects of supplementation of Tween 80 on ruminal fermentation and nutrient digestibility of Hanwoo steers. The experiment was designed as a $3{\times}3$ Latin square with duplication and six Hanwoo steers with rumen cannulae, average weight 497 (SE 61.1) kg. For the experiment the animals were given a basal diet consisting of rice straw and compound feed mixed at 4:6 ratio. The three experimental treatments were (1) the basal diet, supplemented with (2) 5 g/d Tween 80 and (3) 10 g/d Tween 80. Ruminal pH was significantly (p<0.05) affected by Tween 80 supplementation at 6 h after feeding. Increasing supplementation levels of Tween 80 linearly increased the total VFA concentration. CMCase activity by the 10g/d supplementation of Tween 80 were significantly increased (p<0.05) by 24.4% compared with that of control. Digestibility of crude fiber was significantly increased (p<0.05) in Hanwoo steers fed the diet supplemented with 10 g/d Tween 80 compared with those of control, whilst digestibility of ether extract (EE) was linearly increased by increasing Tween 80 supplementation level (p<0.05). In other nutrient components, their digestibilities of Hanwoo fed diets supplemented with Tween 80 tended to increase. It is concluded that Tween 80 has a potential for industrial application as a feed additive to improve ruminant production.

추청벼 총체 사일리지의 사료가치 및 부위별 In situ 분해율에 관한 연구 (Study on Nutritive Value and In Situ Ruminal Degradability of Whole Crop Rice Silage Prepared Using Chucheongbyeo)

  • 본 연구는 추청벼를 이용하여 총체 벼 사일리지를 조제하였을 때 총체 벼 사일리지의 사료가치 및 부위별 소화율을 조사하여 총체 벼의 이용성을 검정하기 위하여 수행되었다. 총체 벼 사일리지의 무기물 함량은 망간이 가장 높았으며 탄소, 철, 아연, 구리 등 순으로 나타났다. 그리고 마그네슘 함량이 가장 낮게 나타났다. 추청벼 총체 사일리지의 아미노산은 glutamic acid 함량이 가장 높았으며 leucine, asparagine, alanine, valine, arginine 순으로 나타났다. 그리고 methionine 함량이 가장 낮았다. 추청벼 총체 사일리지 부위(총체 벼, 낟알을 제거한 줄기, 왕겨를 포함한 낟알, 쌀)에 따른 시간별 반추위내 건물 소화율은 쌀이 가장 높았으며 왕겨를 포함한 낟알, 총체 벼, 낟알을 제거한 줄기 등 순으로 나타났다. 따라서 본 연구에서는 추청벼 총체 사일리지 소화율은 부위에 따라 약간의 차이를 보였으며 무기물 중 망간 그리고 아미노산 중 glutamic acid가 가장 높은 함량을 보였다.

Pretreatments of Broussonetia papyrifera: in vitro assessment on gas and methane production, fermentation characteristic, and methanogenic archaea profile

  • Objective: The present study was conducted to examine the gas production, fermentation characteristics, nutrient degradation, and methanogenic community composition of a rumen fluid culture with Broussonetia papyrifera (B. papyrifera) subjected to ensiling or steam explosion (SE) pretreatment. Methods: Fresh B. papyrifera was collected and pretreated by ensiling or SE, which was then fermented with ruminal fluids as ensiled B. papyrifera group, steam-exploded B. papyrifera group, and untreated B. papyrifera group. The gas and methane production, fermentation characteristics, nutrient degradation, and methanogenic community were determined during the fermentation. Results: Cumulative methane production was significantly improved with SE pretreatment compared with ensiled or untreated biomass accompanied with more volatile fatty acids production. After 72 h incubation, SE and ensiling pretreatments decreased the acid detergent fiber contents by 39.4% and 22.9%, and neutral detergent fiber contents by 10.6% and 47.2%, respectively. Changes of methanogenic diversity and abundance of methanogenic archaea corresponded to the variations in fermentation pattern and methane production. Conclusion: Compared with ensiling pretreatment, SE can be a promising technique for the efficient utilization of B. papyrifera, which would contribute to sustainable livestock production systems.

The Effects of Dietary Urea on Microbial Populations in the Rumen of Sheep

  • Two experiments were conducted to examine the effects of a range of concentrations of ruminal fluid ammonia ($NH_3$-N) on forage digestibility, microbial growth efficiency and the mix of microbial species. Urea was either continuously infused directly into the rumen of sheep fed 33.3 glh of oaten chaff (Exp. I) or sprayed onto the oaten chaff (750 g/d) given once daily (Exp. 2). Concentrations of $NH_3$-N increased with incremental addition of urea (p < 0.01). Volatile fatty acids (VFA) concentrations and 24 h in sacco organic matter digestibility in the rumen were higher when supplemental urea was given (p < 0.01). The (C2 + C4) : C3 VFA ratio was lower (p < 0.05) when $NH_3$-N was above 200 mgN/I. The fungal sporangia appearing on oat leaf blades were significantly higher when urea was supplemented, indicating that $NH_3$-N was a growthlimiting nutrient for fungi at levels of $NH_3$-N below 30 mgN/l. The density of protozoa was highest when $NH_3$-N concentrations were adjusted to 30 mgN/I for continuously fed ($4.4{\times}10^5/ml$) and to 168 mgN/1 for once daily feeding ($2.9{\times}10^5/ml$). Thereafter increasing concentrations of $NH_3$-N, were associated with a concomitant decline in protozoal densities. At the concentration of $NH_3$-N above 200 mgN/l, the density of protozoa was similar to the density of protozoa in ruminal fluid of the control sheep ($1.8{\times}10^5/ml$). The efficiency of net microbial protein synthesis in the rumen calculated from purine excretion was 17-47% higher when the level of $NH_3$-N was above 200 mgN/1. The possibilities are that 1) there is less bacterial cell lysis in the rumen because of the concomitant decrease in the protozoal pool and/or 2) microbial growth per se in the rumen is more efficient with increasing $NH_3$-N concentrations.

식물원료 첨가가 In vitro 반추위 메탄가스 발생에 미치는 영향 (The Effect of Vegetable Sources Supplementation on In vitro Ruminal Methane Gas Production)

  • 각 시판되고 있는 식물을 2개의 그룹으로 나눠 첨가 후 각 시간별 배양을 실시한 후 pH, $NH_3$, VFA 발생량, 총 가스발생량, $H_2$, $CO_2$, $CH_4$ 발생량을 조사하였다. 그룹 1은 상추와 대파첨가구에서 암모니아 농도가 높았고 고추첨가구에서 낮았다 (P<0.05), 그룹 2는 마늘첨가구에서 암모니아 농도가 낮았고 (P<0.05) 깻잎, 무순과 부추첨가 구에서 높았다 (P<0.05). 마늘과 고추첨가구에서 단백질분해과정 중 아미노산에서 암모니아로 분해되는 과정에 영향을 미쳤을 것으로 사료된다. 총 VFA 농도는 대파첨가구에서 유의적으로 높았으나 (P<0.05) 그 외 시험구에서는 대조구와 차이가 없었다. Acetate와 propionate의 비율에서 볼 때 마늘첨가구가 propionate의 량이 상대적으로 증가한 것을 알 수 있었다. 모든 처리구가 대조구에 비해 가스발생량이 대체로 높았으며, 대파와 양파 첨가구가 유의적으로 높았다 (P<0.05). 마늘과 깻잎 첨가구를 제외한 모든 처리구에서 가스 발생량이 유의적으로 높았다 (P<0.05). 생강목에 속하는 강황과 생강첨가구는 pH와 총 가스발생량에서 비슷한 결과를 나타냈으며 in vitro 발효를 높였다. 마늘첨가구는 반추위내 총 가스발생량이 대조구와 유의적인 차이가 없었다 (P<0.05). 모든 시험구에서 총 가스발생량과 $CO_2$ 발생량은 대체로 유사한 경향을 보였다. 마늘첨가구는 총 가스발생량은 대조구와 차이가 없었으나 $CO_2$ 발생량은 대조구보다 유의적으로 높았다 (P<0.05). 마늘첨가구의 $CH_4$ 발생량은 배양시간과 관계없이 매우 낮은 수준을 유지했으며 48시간 배양 후 대조구의 약 1/3 수준으로 낮았다 (P<0.05). $H_2$는 거의 모든 시험구에서 미량 발생되었는데, 대조적으로 마늘첨가구에서만 매우 높은 수준으로 검출되었다. 마늘첨가구에서 g단위 DM 당 $CH_4$ 발생량이 현저히 낮았으며 다른 처리구는 대체로 대조구에 비해 높았다. 본 시험에서는 allium속의 마늘, 부추, 양파, 파를 포함한 시판 중인 채소를 이용하여 in vitro 발효조절시험을 실시하였는데 특정 식물들은 in vitro 발효 대사에 영향을 주었으며 특히 마늘 첨가는 $CH_4$ 생성에 직접적으로 영향을 준 것으로 사료된다.
