Objective: This study aims to identify the relationship between odd- and branched-chain fatty acids (OBCFAs) and microbial nucleic acid bases in the rumen, and to establish a model to accurately predict microbial protein flow by using OBCFA. Methods: To develop the regression equations, data on the rumen contents of individual cows were obtained from 2 feeding experiments. In the first experiment, 3 rumen-fistulated dry dairy cows arranged in a $3{\times}3$ Latin square were fed diets of differing forage to concentration ratios (F:C). The second experiment consisted of 9 lactating Holstein dairy cows of similar body weights at the same stage of pregnancy. For each lactation stage, 3 cows with similar milk production were selected. The rumen contents were sampled at 4 time points of every two hours after morning feeding 6 h, and then to analyse the concentrations of OBCFA and microbial nucleic acid bases in the rumen samples. Results: The ruminal bacteria nucleic acid bases were significantly influenced by feeding diets of differing forge to concentration ratios and lactation stages of dairy cows (p<0.05). The concentrations of OBCFAs, especially odd-chain fatty acids and C15:0 isomers, strongly correlated with the microbial nucleic acid bases in the rumen (p<0.05). The equations of ruminal microbial nucleic acid bases established by ruminal OBCFAs contents showed a good predictive capacity, as indicated by reasonably low standard errors and high R-squared values. Conclusion: This finding suggests that the rumen OBCFA composition could be used as an internal marker of rumen microbial matter.
The effect of three varying ratios (high, medium and low) of Rumen Degradable Protein (RDP) to Undegradable Dietary Protein (UDP) of 37:63, 52:48 and 70:30 in iso-nitrogenous and iso-caloric concentrate mixtures on rumen fermentation profile was studied using rumen fistulated Jersey crossbred cows. Rumen pH and ammonia nitrogen concentration were found to be lower with a concentrate mixture containing a higher UDP level of 63.38% when compared with those having medium and low UDP levels of 47.55 and 29.75%, respectively, at all post feeding intervals. Total volatile fatty acid concentration as well as concentrations of individual fatty acids viz., acetate, propionate and butyrate were also found higher in animals fed concentrate mixture with the highest UDP level.
The study evaluated different synthetic and semisynthetic media for maximal recovery of rumen bacteria and expression of their antibacterial activity. Rumen Glucose Cellobiose Agar (RGCA) medium was found to be the best for recovery of rumen bacteria. However, L-10 medium was the best for expression of antibacterial activity of ruminal isolates followed by Easy, M-10, RGCA and M-98-5 medium. The present study recommends the use of L-10 medium as the medium of choice for screening of antibacterial activity of ruminal isolates. Comparative evaluation of bacterial counts on different dietary regimes indicated significant difference between different growth media on a specific diet and between diets on specific growth media within a species. However, there is no overall significant difference between total bacterial counts obtained from rumen liquor of cattle and buffalo with respect to either the feeding regime or growth media. Feeding straw based diet to the animal is the best for high recovery of rumen bacteria.

Two strains of rumen fungi (Piromyces rhizinflata B157, Orpinomyces joyonii SG4) and three strains of rumen cellulolytic bacteria (Ruminococcus albus B199, Ruminococcus flavefaciens FD1 and Fibrobacter succinogenes S85) were used as mono-cultures or combinationally arranged as co- and sequential-cultures to assess the relative contributions and interactions between rumen fungi and cellulolytic bacteria on rice straw degradation. The rates of dry matter degradation of co-cultures were similar to those of corresponding bacterial mono-cultures. Compared to corresponding sequential-cultures, the degradation of rice straw was reduced in all co-cultures (P<0.01). Regardless of the microbial species, the cellulolytic bacteria seemed to inhibit the degradation of rice straw by rumen fungi. The high efficiency of fungal cellulolysis seems to affect bacterial degradation rates.
The paper presented here is aimed at increasing knowledge on purine metabolism in ruminants and hence the quantification of microbial cells entering the small intestine from urinaη excretion of purine derivatives. Nucleic acid metabolisms of micro-organisms in the rumen, digestion and absorption of nucleic acids entering the intestines, metabolisms of absorbed and endogenous purines involving de novo synthesis of nucleic acids in the ruminants host, and the relationship between absorbed and excreted purines are reviewed. Principal concerns about an amount of purine derivatives excreted in urine in relation to a change in purine-N: total-N ratios in rumen microbes that leave the rumen are discussed. The use of urinary excretion of purine derivatives as an indicator of the amount of microbial biomass leaving the rumen has to be done with some caution since it may be impossible to get a representative sample of microbes entering the intestine and thus yield estimates are relative rather than absolute.

A series of in vitro experiments was conducted to investigate response of rumen bacterial deamination to the ionophore salinomycin. Addition of salinomycin to the inoculum, strained rumen fluid, depressed ammonia production from casein, while increased accumulation of ${\alpha}$-amino acids. This suggests an inhibitory effect of salinomycin on ruminal deamination. When the effect in washed bacterial suspension was monitored with individual amino acid, aspartic acid degradation was markedly inhibited by salinomycin. This inhibition was not observed when the mixed rumen bacteria were ultrasonically disrupted and used as the enzyme source. Extent of the inhibition tended to be higher in the bacteria source from sheep on a high roughage diet. From these results it was speculated that the inhibition of deamination with salinomycin is caused by a decreased transport of amino acid into the bacterial cells as well as a decreased proportion of deaminating bacteria in the rumen.
This study reports light and electron microscope examination of rumen fungal colonization of alkali-treated roughage feeds incubated in decron bags in the rumen of cannulated sheep for varying time intervals. Six roughages, pre-treated with ammonium hydroxide or sodium hydroxide at 4% (w/w) level were examined together with untreated control samples. Alkali pre-treatment was associated with an earlier and more pronounced fungal colonization than all control roughages. Sodium hydroxide pre-treatment was significantly more effective than ammonium hydroxide in improving the susceptibility of roughages to rumen fungal colonization and studies by SEM showed that the pre-treatment permitted greater penetration of feeds by fungi. Sodium hydroxide pre-treatment also significantly increased dry matter disappearance from feed held in Dacron bags in the rumen with all feeds except Lucerne stem. It is not known to what extent fungal activity contributed to increased breakdown of the feeds.
Rumen liquor characteristics and disappearance rate of dry matter were studied in Kedah-Kelantan cattle and swamp buffaloes fed grass of rice straw-based diet. Cobalt-EDTA and chromium mordented fibres prepared from the faecal material were used to determine the liquid and solid particles movement in both animal species fed with rice straw. Swamp buffaloes showed a more intense rumen fermentation activity than Kedah-Kelantan cattle when both species were fed straw-based diet. The buffaloes also demonstrated faster rates of grass and straw degradation in situ. The fluid outflow rate from the rumen of buffalo ($1.06{\pm}0.19l/h$) was observed to be slower than that of cattle ($1.55{\pm}0.01l/h$). No significant differences between cattle and buffaloes were observed in rumen fluid volume and passage rate of small particles from the rumen.
