로얄제리가 Streptozotocin 유발 당뇨 흰쥐의 치료 및 예방에 미치는 영향 (Effect of Royal Jelly on Therapy and Prevention of Streptozotocin Induced Diabetic Rats)

  • In order to investigate the preventive and therapeutic of royal jelly on diabetes, the levels of blood glucose and serum lipids as well as the number of blood cells were determined in streptozotocin(STZ) diabetic rats. Rats were divided into seven groups. The RJ group was administered royal jelly and the STZ group was treated with streptozotocin to induce diabetes. To determine the preventive effect, diabetes was induced after administration of royal jelly for 2 weeks in group RS1/RS2. In group SR1/SR2 diabetic rats were administered royal jelly for 2 weeks to investigate the therapeutic effect. After 3 weeks, the body weight was reduced in STZ and SR1 groups and food intake was increased in the STZ, RS1 and SR1 groups. The blood glucose level was similar to the control group in the RJ, RS1 and RS2 groups and there was no effect in the other groups. The total lipid and triglyceride level were lower in the SR1 group as compared to STZ, and the total cholesterol level was higher in the STZ group. The index of atherogenesis was lower in the RJ and SR1 groups compared to the normal group. The number of red blood cells and hemoglobin was higher in the RJ and SR1 groups and the number of white blood cells was higher in the RJ and SR2 groups.

  • 정상 마우스와 Cyclophosphamide(CY)로 처리된 마우스에 Royal Jell(RJ)를 투여하여 면역계에 미치는 영향을 실험한 결과 다음과 같은 결론을 얻었다. 1. 정상 마우스에 RJ를 투여하였을 때는 대조군에 비하여 마우스의 체중, 비장, 흉선 , WBC 수 , DNFB에 대한 접촉성 과민 반응으로 측정된 세포성 면역반응 및 면양 적혈구에 대한 응집가와 용혈가로 측정된 체액성 면역 반응이 투여일에 따라 증가 또는 감소되었고, RBC수와 간 중량은 영향을 받지 않았다. 2. 증가 작용을 보여준 투여일에 RJ를 병용 투여함으로써, CY 처리로 인한 마우스의 생존율, 비장의 중량 , WBC수 및 세포성 면역 반응의 감소가 약간 억제되었으나, CY를 감소된 흉선의 중량 및 체액성 면역반응에는 영향을 나타내지 않았다.

Royal Jelly 첨가가 동결융해 후 개 정자의 활력도 및 생존성에 미치는 영향 (Effect of the Addition of "Royal Jelly" on Post-thaw Viability and Longevity of Canine Spermatozoa)

  • This study was conducted to evaluate whether \"Royal jelly\" (RJ) added to Tris-buffer dilute contributed to supporting post-thaw viability and longevity of frozen canine spermatozoa. Two Japanese spitzs (2 to 4 years of age) were used as a semen donor. Semen was collected by manual masturbation and separated into 3 fractions. Only the sperm-rich fraction having sperm motility of more than 70%, containing sperm concentration of 2~4$\times$10$^{8}$ cells/ml and having dead or abnormal spermatozoa of less than 15% was used for the experiment. Each ejaculated semen was centrifuged at 400 $\times$ g for 5 min and then diluted in a Tris-buffer supplemented with 20 ml egg yolk (Ext I), 4% glycero1 and 1% Equex STM Paste (Ext II) or g1ycero1, Equex STM paste and RJ of various concentrations (Ext II-RJ). After freezing and thawing, viability of spermatozoa in Ext II -RJ containing 1% RJ immediately after thawing (67.5$\pm$9.6) was significantly lower than that of Ext II , Ext II -RJ containing 0.01 or 0.1% RJ (77.5$\pm$12.5, 78.7$\pm$8.2 and 80.0$\pm$6.3). However, Ext II-RJ containing 0.1% RJ yielded higher viability than Ext II, Ext II-RJ containing 0.01% at or 1% 1 h after thawing (69.5$\pm$8.1 vs. 55.0$\pm$12.9, 57.5$\pm$9.6 and 41.5$\pm$12.6; P<0.05). At 1 h after thawing, the viability of spermatozoa thawed in 7$0^{\circ}C$ (68.8$\pm$12.5) was significantly higher than that of spermatozoa thawed in 38$^{\circ}C$ (48.8$\pm$16.3), although there was no difference in the viability between both groups immediately after thawing (77.5$\pm$9.6 and 81.3$\pm$8.1). Post-thaw viability and longevity of post-thaw spermatozoa in Ext II-RJ containing 0.1% RJ was higher in those in Ext II at 1 h (65.0$\pm$12.9 vs. 42.5$\pm$12.6), 2 h (52.5$\pm$12.6 vs. 27.5$\pm$17.1) and 3 h (40.0$\pm$14.1 vs. 20.0$\pm$12.1) after thawing. These results indicated that addition of 0.1% af to Tris-buffer enhanced post-thaw viability and longevity of canine spermatozoa and this additive can be used for increasing the possibility of collision between spermatozoa and ova during insemination.emination.

Identification of royal jelly as a potential new drug to protect the ovarian reserve and uterus against cyclophosphamide in rats

  • Objective: The aim of this study was to investigate the effect of royal jelly (RJ), a powerful natural antioxidant, on cyclophosphamide-induced ovarian damage. Methods: Thirty-two Wistar albino rats were divided into four groups. Oral treatment was administered to all rats for 16 days after a single intraperitoneal injection. The control group received intraperitoneal and oral saline; the RJ group received intraperitoneal saline and 100 mg/kg/day oral RJ; the cyclophosphamide group received intraperitoneal 100 mg/kg cyclophosphamide and oral saline; and the treatment group received intraperitoneal 100 mg/kg cyclophosphamide and 100 mg/kg/day oral RJ. The groups were compared in terms of ovarian reserve tests and histopathological changes in the ovary and uterus. Results: All follicle counts were higher in the treatment group than in the cyclophosphamide group. The increase in the number of preantral follicles (p=0.001) and the decrease in the number of atretic follicles (p=0.004) were statistically significant. RJ treatment significantly improved follicular degeneration and cortical fibrosis in the ovary and epithelial and gland degeneration in the uterus due to cyclophosphamide toxicity. Conclusion: According to these results, RJ reduces cyclophosphamide-related ovarian and endometrial damage in rats. For this reason, it should be further investigated to determine its effects on reproductive function.

Antioxidant Activity of Royal Jelly Hydrolysates Obtained by Enzymatic Treatment

  • Recently, research on the processing of raw functional materials with the aim of improving various physiological activities has been conducted. In this study, we investigated the antioxidant activity of royal jelly (RJ) hydrolysates obtained from three commercial proteases. Enzyme-treated royal jelly (ERJ), in which the RJ hydrolysates were converted into easy-to-absorb shorter chain monomers through the removal of two known allergen proteins, showed no difference in the content of (E)-10-hydroxydec-2-enoicacid (10-HDA) or the freshness parameter and showed a significant increase in total free amino acid content. The antioxidant activity of ERJ was determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) and chemical assays. The ERJ showed about 80% DPPH-radical scavenging activity at same concentration of ascorbic acid. The antioxidant effect of ERJ was confirmed to be due to reduction of intracellular reactive oxidative species (ROS) and nitric oxide (NO) production in LPS-treated macrophages. Moreover, ERJ significantly increased the activity of the antioxidant enzyme superoxide dismutase (SOD) and the level of the antioxidant glutathione (GSH) in a dose-dependent manner. Interestingly, these antioxidant activities of ERJ were stronger than those of non-treated RJ. These findings indicate that ERJ has high potential as an antioxidant agent for use in human and animal diets.

Royal jelly enhances migration of human dermal fibroblasts and alters the levels of cholesterol and sphinganine in an in vitro wound healing model

  • Oral administration of royal jelly (RJ) promotes wound healing in diabetic mice. Concerns have arisen regarding the efficacy of RJ on the wound healing process of normal skin cells. In this study, a wound was created by scratching normal human dermal fibroblasts, one of the major cells involved in the wound healing process. The area was promptly treated with RJ at varying concentrations of 0.1, 1.0, or 5 mg/ml for up to 48 hrs and migration was analyzed by evaluating closure of the wound margins. Furthermore, altered levels of lipids, which were recently reported to participate in the wound healing process, were analyzed by HPTLC and HPLC. Migration of fibroblasts peaked at 24 hrs after wounding. RJ treatment significantly accelerated the migration of fibroblasts in a dose-dependent manner at 8 hrs. Although RJ also accelerated the migration of fibroblasts at both 20 hrs and 24 hrs after wounding, the efficacy was less potent than at 8 hrs. Among various lipid classes within fibroblasts, the level of cholesterol was significantly decreased at 8 hrs following administration of both 0.1 ug/ml and 5 mg/ml RJ. Despite a dose-dependent increase in sphinganines, the levels of sphingosines, ceramides, and glucosylceramides were not altered with any concentration of RJ. We demonstrated that RJ enhances the migration of fibroblasts and alters the levels of various lipids involved in the wound healing process.

폐경기 노화 유도 전후의 로얄제리 식이공급이 백서 표피의 필라그린과 유리아미노산 함량 및 관련 대사 효소의 단백질 발현 변화에 미치는 영향 (The Dietary Effect of Royal Jelly Supplementation on Epidermal Levels of Filaggrin and Free Amino Acids during Menopause in Rats)

  • 본 연구에서는 난소절제를 통하여 폐경기가 유도된 백서 모델에서 생산지가 다른 국내산 로얄제리 식이와 이소플라본을 식이로 공급한 후 표피의 필라그린 및 유리아미노산의 함량과 관련 대사 효소의 발현 변화를 정상대조군인 SHAM군 및 난소절제군인 OVX군과 비교 분석하였다. Profilaggrin, filaggrin 및 total filaggrin의 발현은 SHAM군> RJ2군> OVX군=IF군> RJ1군 순으로 RJ1군의 profilaggrin, filaggrin의 발현량이 가장 낮게 나타났다. Profilaggrin에서 filaggrin으로, filaggrin에서 유리아미노산으로의 분해하는 과정에 관여하는 여러 효소 중에 PAD3의 발현은 RJ2군에서 가장 높았으나 SHAM군을 비롯하여 OVX군, RJ1군, IF군에서는 군간 유의성이 없었다. 전체 유리아미노산의 함량은 SHAM군에 비해 OVX군에서 다소 감소하였으나 RJ1군 및 IF군에서 SHAM군보다 높게 나타났다. 특히 RJ1군에서 aspartate와 proline의 함량이 SHAM군에 비해 현저히 높아졌다. 결론적으로 RJ1군에서는 filaggrin의 함량 감소가 있었지만 표피 보습 유지기능이 있는 aspartate 및 proline을 비롯한 전체 유리아미노산의 함량이 높아졌다. RJ1군의 로얄제리가 폐경으로 인해 감소되는 표피의 보습인자 중 궁극적으로 피부 보습에 관여하는 천연보습인자의 유리아미노산의 발현을 증가시켜 손상된 표피의 장벽기능 보완에 긍정적 영향을 보였다. RJ2군은 profilaggrin, filaggrin을 포함한 total filaggrin의 함량이 증가되었으나 그 분해산물인 유리 아미노산의 함량은 SHAM군과 유사하였다. RJ2군의 로얄제리가 유리아미노산의 전구단백질의 발현 증가를 야기하나 유리아미노산으로의 분해는 촉진시키지 못하였음을 시사한다. 이는 산지별 로얄제리의 피부 보습 기능에 미치는 영향이 다르게 나타날 수 있음을 의미하며 추후 각 산지별 유효성분의 분석과 함께 관련 효능에 대한 연구가 이루어질 필요성이 있다. 본 연구에서 국내산 로얄제리의 식이 공급이 피부 보습 관련 건강기능식품 개발 소재로서의 가능성이 있을 것으로 사료된다.

자외선 조사와 병행된 로얄제리 식이 공급이 무모 생쥐의 표피 보습과 필라그린, 유리아미노산 함량 및 관련 대사 효소의 발현 변화에 미치는 영향 (Dietary effect of royal jelly supplementation on epidermal levels of hydration, filaggrins, free amino acids and the related enzyme expression in UV irradiated hairless mice)

  • 본 연구에서는 자외선 조사에 의한 피부 광노화 유도와 함께 산지 별 국내산 및 중국산 로얄제리 식이를 6주간 공급한 무모 생쥐 표피의 필라그린과 유리 아미노산 함량 및 관련 대사 효소의 발현 변화를 정상대조군인 UV - 군 및 자외선 조사군인 UV + 군과 비교 분석하였고 그 결과는 다음과 같다. 1) UV + 군의 필라그린과 중간 생성물 및 profilaggrin을 포함한 총 필라그린 함량은 UV - 군에 비하여 유의적으로 감소하였다. RJ1군의 총 필라그린 함량이 UV + 군에 비하여 유의적으로 높게 나타났고 RJ2, RJ3 및 RJ4군에서는 UV + 군과 유사하거나 낮았다. 2) PAD3 발현은 UV + 군의 발현이 UV - 군에 비하여 유의적으로 감소하였고 로얄제리 공급군의 PAD3 발현은 모두 UV + 군에 비하여 유의적으로 높았다. 다만 각 로얄제리를 섭취시킨 군 사이에서 군간 유의성은 없었다. 3) 총 유리 아미노산 함량은 군간 유의적인 차이는 보이지 않았다. 그러나 개별 유리 아미노산 함량 변화를 분석한 결과, UV + 군에서 glutamate와 serine의 함량이 UV - 군에 비해 낮은 경향을 보였다. 반면 RJ1군의 glutamate와 serine의 함량은 UV + 군에 비해 유의적으로 높았으며 RJ2와 RJ3군은 serine의 함량이 UV + 군에 비해 다소 높았다. 결론적으로 RJ1의 식이는 표피의 필라그린 함량과 관련 대사 효소의 발현을 높이고 그에 따라 표피에서 천연보습인자로서 보습 증진에 크게 기여하는 glutamate와 serine의 함량을 증가시킴으로써 자외선 조사로 인해 저하된 피부 보습 능력을 회복시키는 역할을 하는 것으로 여겨진다.

알러지 유발 단백질이 제거된 수용성 로열젤리의 피부미백 효과 (Whitening Efficacy of Water Soluble Royal Jelly Removed Allergenic Protein)

  • Royal jelly composes of many components, especially protein. Protein is a major factor which cause allergy. We focused on water soluble royal jelly (WSRJ) that was removed allergy - inducing protein. 10-hyroxy-2-decenoic acid content of WSRJ is 2.42 g/100 g, which is double compared to that of lypophilized RJ. To further access WSRJ as a cosmetic ingredient and potential external treatment for topical use, we investigated its ability to inhibit tyrosinase activity and melanin biosynthesis on melanogenesis in B16F1 melanoma cells. We found that WSRJ increased the cell viability in B16F1 melanoma cell and WSRJ (1~10 mg/ml) inhibited melanin synthesis in with 10 nM ${\alpha}$-melanocyte-stimulating hormone (${\alpha}$-MSH) for 48 h. WSRJ inhibited direct tyrosinase activity, which decreased melanin synthesis in ${\alpha}$-MSH stimulated B16F1 melanoma cells. Thease findings suggest that WSRJ induces the down regulation of melanogenesis by inhibiting tyrosinase activation.

Development of a Novel Short Synthetic Antibacterial Peptide Derived from the Swallowtail Butterfly Papilio xuthus Larvae

  • Insects possess biological defense systems that can effectively combat the invasion of external microorganisms and viruses, thereby supporting their survival in diverse environments. Antimicrobial peptides (AMPs) represent a fast-acting weapon against invading pathogens, including various bacterial or fungal strains. A 37-residue antimicrobial peptide, papiliocin, derived from the swallowtail butterfly Papilio xuthus larvae, showed significant antimicrobial activities against several human pathogenic bacterial and fungal strains. Jelleines, isolated as novel antibacterial peptides from the Royal Jelly (RJ) of bees, exhibit broad-spectrum protection against microbial infections. In this study, we developed a novel antimicrobial peptide, PAJE (RWKIFKKPFKISIHL-NH2), which is a hybrid peptide prepared by combining 1-7 amino acid residues (RWKIFKK-NH2) of papiliocin and 1-8 amino acid residues (PFKISIHL-NH2) of Jelleine-1 to alter length, charge distribution, net charge, volume, amphipaticity, and improve bacterial membrane interactions. This novel peptide exhibited increased hydrophobicity and net positive charge for binding effectively to the negatively charged membrane. PAJE demonstrated antimicrobial activity against both gram-negative and gram-positive bacteria, with very low toxicity to eukaryotic cells and an inexpensive process of synthesis. Collectively, these findings suggest that this novel peptide possesses great potential as an antimicrobial agent.
