Expression of Luteinizing Hormone (LH) Subunit Genes in Mouse Testis

  • Gonadotropins are heterodimers consisting an alpha chain ($Cg{\alpha}$) and a beta chain. Interestingly, presence of complicated $LH-{\beta}$ transcripts in rat testis was accidently found; testicular $LH-{\beta}$ transcripts were confined in seminiferous tubules to spermatids, and the translated products were localized in the elongated spermatids. We hypothesized that mouse testis has potential to produce the tissue specific $LH-{\beta}$ with similar structure to the rat testicular forms. To verify our hypothesis, we examined the adult mouse (ICR) testis using RT-PCR and immunohistochemistry. The PCR revealed the presence of the identical products in the reactions for three LH subunit types. The expected product sizes for mouse $Cg{\alpha}$ and $LH-{\beta}$ known as pituitary type were 224 bp and 503 bp, respectively. The testicular type $LH-{\beta}$ products were produced by a primer set based on the rat sequences, with unexpected size of 800 bp. Sequencing revealed that the proximal and distal parts (2-82 and 661- 773 bp, respectively) were homologous to rat testicular $LH-{\beta}$ cDNA, and middle part (83-660 bp) was a unique mouse-specific region. Both $Cg{\alpha}$ and $LH-{\beta}$ positive signals were in the round and elongated spermatids and mature sperms, and the $LH-{\beta}$ signals were more intense. In conclusion, our study demonstrated that the presence and localization of the LH subunits in mouse testis. Further studies will be needed to understand the precise structure and function of mouse testicular LH.

  • 생식 세포는 한 세대의 유전 정보를 다음세대에 전달할 수 있는 유일한 세포로서 다양한 특성을 가지고 있다. 기능 유전체 연구를 통해서 새로운 유전자의 기능을 규명함으로써 그 유전자의 생물학적 의미와 상호작용을 설명할 수 있다. 본 실험의 목적은 생식세포에서 특이적으로 발현하는 유전자를 발굴하여 그 유전자가 생식세포의 발달과 분화에 중요한 역할을 수행하는 것을 증명하는 것이다. 이에 본 실험에서는 real-time quantitative RT-PCR 기법을 이용하여 정소에서 특이적으로 발현하는 5개(AGE1, AGE2, AGE3, AGE4, AGE5)를 선발하였고, in situ hybridization 실험을 통하여 정소 조직 내에서의 발현 양상을 확인하였다. AGE1, AGE2는 round spermatid에서 특이적으로 발현하고, AGE3, AGE4, AGE5는 spermatocyte에서 특이적으로 발현하는 것을 확인하였다. 본 실험을 통해 발굴한 유전자들은 닭의 생식선발달에 중요한 기능을 할 것으로 예상되며, 앞으로 닭의 유전육종분야에 큰 도움을 줄 수 있을 것이다.

Effect of rc Mutation on Semen Characteristics, Spermatogenic Tissues and Testosterone Profile in Blind Rhode Island Red Cockerels

  • Seven rc mutant and seven normal male birds (Rhode Island Red suie, RIR) were used in this study to determine the effects of rc mutation on semen characteristics, testosterone profile and spermatogenic tissues. All birds were randomly selected at week 12 of age and housed in individual cages and were fed and watered ad libitum. The birds were exposed to a 14L:10D light cycle during experiment. Semen were collected at weeks 22 to 23 from each bird twice a week and evaluated for semen volume (SV), sperm concentration (SC), total sperm count (TSC), percent of sperm motility (%SM), dead sperm (%DS), and sperm metabolic activity (SMA). To determine the testosterone concentration (TC) in plasma, blood was collected at weeks 12, 16 and 18. Testicular tissue were collected, processed and evaluated for semineferous tubule diameter (STD), round spermatid number (RSN), percent elongated sperm (%ES) and semineferous tubules length (STL). Body weight (BW), comb weight (CW) and testes weight (TW) were weighted at the end of experiment (week 23). The SV, TSC and %SM were significantly higher in normal birds but the %DS was higher in blind birds (p<0.05). The SC did not differ significantly between the two groups but its value was higher in normal birds. The sperm metabolic activity in the first h of collection did not differ significantly between the two groups but after 24 h, the level of SMA in normal group was significantly higher (p<0.05). The level of TC did not differ significantly between the two genotype groups but normal birds had higher TC in all collections except the last one. The STD, RSN, %ES and STL in normal birds were higher when compared to blind birds but the differences were insignificant except for ES percent. The BW, CW and TW between the two groups did not differ significantly but the weights were higher in normal group compared to blind birds. Statistical analysis of semen characteristics, testosterone profile and histological factors were indicated detrimental effects of rc mutation in prepubertal RIR blind male birds due to lack of light.

Testicular Lesion in the Sprague-Dawley Rats Treated with High of 2-Bromopropane (고용량의 2-Bromopropane 투여가 Sprague-Dawley 랫트의 고환에 미치는 영향)

  • This study was carried out to investigate the testicular toxicity of environmental toxicant, 2-bromopropane(2-BP) recently caused occupational intoxication in Korea by light microscopy and electron microscopy. To evaluate the effect on spermatogenesis and find target germ cell 10 weeks old male Sprague-Dawley rats were treated with 5g/10m ℓ/kg/day of 2-bromopropane for 3 consecutive days orally and observed on day 1 or day 7 after treatment. 2-BP induced depletion of spermatogonia and early spermatocytes on stages I-IX or extensive degeneration of germ cells on the other stages on day 1. But extensive degeneration of germ cells without stage specificity was observed and round spermatid formed multinucleated giant cells in the lumen of seminiferous tubules on day 7. Electron microscopically Sertoli cells showed irregular shape of nucleus and cytoplasmic vauolation. And spermatocyte showed a extensive heterochromatin and cytoplasmic vacuolation. But there was no histopathological changes in the interstial cells. On the base of the results the target germ cell was spermatogonia in the early of the study but Srtoli cells also effected by high-dosed 2-BP in the late of the study.

Spermatogenesis of the Spiny Top Shell, Batillus cornutus (Lightfoot, 1786) (Gastropoda: Turbinidae) (소라, Batillus cornutus의 정자형성과정)

  • This study describes the spermatogenesis and sperm ultrastructure of the spiny top shell, Batillus cornutus using light and electron microscopy. The spiny top shells were collected by divers in the coastal water of Wandogun, Cheollanamdo, Korea(N $34^{\circ}13'$, E $126^{\circ}47'$) at May 2003. Spiny top shells of $60.0{\sim}69.9\;mm$ in shell height were used in this study. The testis comprises many spermatogenic follicles which contains germ cells in different developmental stages. The primary spermatocytes in the pachytene stage were characterized by synaptonemal complexes. The early spermatids were characterized by appearance of Golgi complex, increased karyoplasmic electron density and tubular mitochondria. In early spermatid the mass of proacrosomal granules consists of numerous heterogeneous granules with high electron density. From the mid-stage of spermiogenesis the well-developed mitochondria aggregate posterior to the nucleus, and surround the proximal and distal centrioles. In this stage, proacrosomal granules are condensed and form a acrosome with thin envelope. During the late spermiogenesis, the acrosome begins to elongate and then became conical. The sperm consists of head, mid-piece and tail. The head comprises a round nucleus and a conical acrosome. Acrosomal rod of microfibrous is observed between nucleus and acrosome. Five mitochondria observed in mid-piece. And tail has the typical "9+2" microtubular system originates from the centrioles.

  • The morphological study on different types of cells of reproductive organ including spermatogenesis in the adult planaria was performed to observe their cytochemical and ultrastructural characteristics. 1. Spermatogenesis The circular luminated material appears immediately inside the nuclear envelope of early spermatid and is found also in the nucleus of sperm, but typical acrosomal structures cannot be observed. Approximately ten of small-sized mitochondria occur around the nucleus in the transitional phase from primary spermatocyte to secondary spermatocyte, but in sperm a long mitochondrion is closely associated with nucleus, parellel to long axis of it. The sperm has a relatively long head connected with two tails via hollow neck. 2. Reproductive organ The penis bulb and the bursa stalk were observed. (1) Penis bulb The cells constituted penis bulb are classified into six types on the basis of ultrastructure of the cells and cytochemistry of the cytoplasmic granules. 1) A-type cells: These cells exhibiting low electron density are mainly occupied by large nucleus. These cells possess two different types of granules: highly electron-dense round granules with an average size of $0.9{\mu}m$, and electron-dense granules exhibit PAS-positive reaction. 2) B-type cells contain PAS-positive granules with the size of about $0.4{\mu}m$. They are rich in free ribosomes and mitochondria. 3) C-type cells are found to be dark cells due to high electron-density. These cells are largely occupied by large nucleus. 4) D-type cells: These cells are seen as light cells which have poorly developed cell organelles. 5) E-type tells: These cells contain a large number of glycogen granules which occupy most of cell. 6) F-type cells: These arc parietal epidermal cells surrounding the genital antrum. These cells are characterized by their finger-like shapes and the presence of a number of electron-dense, irregularly-shaped structures inside cells. The relatively large electron-lucent granules can be also found. The F-type cells possess numerous microvilli on their free surfaces. (2) Bursa stalk The cells constituted bursa stalk are classified into 3 types on the basis of cell shapes and presences of electron-dense or electron-lucent granules. 7) G-type cells with a long cytoplasmic process. They have large nuclei and poorly developed cell organelles. 8) H-type cells: These cells are characterized by the presence of a long cytoplasmic process and relatively highly electron-dense cytoplasmic profile. They have poorly developed cell organelles. 9) I-type cells contain large electron-lucent granules which exhibit negative reactions with three kinds of cytochemical staining methods used in this experiment. The fine electron-dense structures can be found inside these granules.
