  • This study was carried out to develop an automatic grafting system suitable for fruit-vegetable seedlings. The study consisted of two research sections: 1) development of a medium-sized, low-cost automatic grafting system for cooperative farms and commercial seedlings production company, and 2) commercializing research for prototype development based on the above concepts. The grafting robot developed in this research can be described as follows, a. Developed grafting robot can cover the whole operations for grafting scion and rootstock, only if operator provides scion and rootstock tray. b. Five seedlings can be grafted at one time, and about 1,200 seedlings can be grafted in one hour. c. The success ratio of mechanical grafting scion and rootstock with ceramic pin bonding provided by the holder is more than 95% when the conditions of seedlings are satisfied. d. The grafting efficiency has improved over 10 times compared with manual work, and the grafting cost generated 44% savings.

키위 쇠락증상 발생 및 습해 저항성 대목을 이용한 예방 (Occurrence of Kiwifruit Vine Decline Syndrome and Its Prevention Using Rootstock Tolerant to Waterlogging)

  • 키위산업은 키위나무가 급속하게 시들면서 1-2년 사이에 말라죽는 키위 쇠락증상의 출현으로 위협을 받고 있다. 쇠락증상의 주요 증상은 뿌리 피층 붕괴, 잎 괴사, 낙엽, 열매 표피 주름 및 잔가지 시들음이다. 키위 쇠락증상은 장마 후 한여름에 골드키위(Actinidia chinensis var. chinensis)와 그린키위(A. chinensis var. deliciosa)에서 모두 발생하였다. 키위 쇠락증상은 침수나 통기 불량으로 인해 수분이 많은 점질토양에서 심하게 발생하는 것으로 나타났다. 이 증상과 직접적인 관련이 있는 병원균은 없었다. 키위 쇠락증상은 침수와 같은 수분 스트레스에 강한 바운티대목(Bounty 71)을 사용하여 효율적으로 예방할 수 있을 것으로 전망된다.

'Fuji'/M.9-T337 묘목의 삭아접 시기, 대목절단 시기, 적엽 및 Promalin 처리가 나무의 생장과 측지발생에 미치는 효과 (Effects of Times of Chip Budding and Rootstock Removal, Leaf Removal Plus Promalin Application on the Tree Growth and Lateral Development for 'Fuji'/M.9-T337 Nursery Trees)

  • 대목재식 후 2년차에 측지가 10개 이상 발생 된 우량묘를 생산하기 위하여 삭아접시기 및 대목 절단시기에 따른 수체생육 특성과 측지발생에 미치는 적엽 및 promalin 처리 효과를 시험한 결과, 대목재식 후 약 90일 정도 경과한 6월중순경에 삭아접을 실시하는 것이 4월 중순에 삭아접을 하는 것보다 재식 1년차의 수체생육은 떨어지지만 접아고사주율이 평균 8% 정도 낮았다. 또한 M.9 대목 절단시기에 있어서는 삭아접전절단(ECCB)을 실시하는 것보다 삭아 접후절단을 할 경우, 접아고사율이 14% 정도 낮아 수체 생장의 균일도가 높으면서 접목효율이 높은 것으로 나타났다. 재식 2년차인 1998 년도에 접목시기 및 M.9 대목 절단시기에 따른 적엽(3회) 및 promalin(3회) 처리효과를 조사한 결과, 6월 중순 삭아접후절단(LCCB) 처리 구에서 측지발생 효과 및 화아형성 정도가 다른 처리에 비하여 가장 좋았다. 묘목 특성으로는 수고 209cm, 측지수 13.8개, 화아형성수 9.0개로 우량한 측지묘 조건에 부합한 묘목이 생산되었다.

사과 '서홍', '썸머드림', '홍금'의 생장 및 수량에 미치는 M.9 대목 노출길이의 영향 (Effect of the Exposed Length of Dwarf Rootstock M.9 on Growth and Yield of 'Seohong', 'Summer Dream' and 'Honggeum' Apples)

  • 사과나무의 수세는 품종 고유의 유전적 특성에 따라 결정되지만 환경 조건, 재배 관리, 대목 종류 및 노출 길이에 따라 달라질 수 있다. 따라서 품종 고유의 특성에 따라 적합한 대목 종류 및 적정 노출 길이를 선택하는 것이 나무의 생육, 생산성 및 수명에 바람직한 영향을 줄 수 있을 것이다. 본 연구는 국내에서 육성 된 '서홍', '썸머드림', '홍금'에 적합한 대목 노출길이에 대한 정보를 제공하기 위하여 M.9 자근대목에 각각 접목한 후 접목부가 지면에서 10, 20, 30 cm로 노출되게 재식하여 수체생장과 생산성을 조사하였다. 재식 6년차까지 '서홍', '썸머드림', '홍금'은 대목 노출이 짧을수록 TCA 증가가 컸다. 재식 6년차의 TCA는 10cm 처리구를 100으로 했을 때 20, 30cm 처리구는 각각 '서홍' 70, 60, '썸머드림' 88, 66, '홍금' 55, 41으로 현저히 낮아졌다. 또한 '서홍', '썸머드림', '홍금'의 대목 노출 길이가 짧을수록 수고, 수폭, 신초장, 정단신초장의 영양생장량이 더 우수하였다. 재식 6년차까지의 누적수량 역시 대목 노출 길이가 짧을 수록 많아지는 경향을 보였다. 재식 6년차 누적수량의 10cm 처리구를 100으로 했을 때 20, 30cm 처리구는 '서홍' 77, 63, '썸머드림' 85, 76, '홍금' 73, 58으로 조사되었다. 과실 특성에 있어서 대목 노출 길이에 따른 차이는 없었다. 따라서, 재식 6년차까지 '서홍', '썸머드림', '홍금'의 영양생장과 생산성을 고려했을 때 M.9 대목의 노출 길이는 10cm로 하는 것이 적당할 것으로 생각된다.

사과 왜성대목 무독묘 생산을 위한 바이러스 제거 방법 비교 (Comparison of Virus Elimination Methods for Disease-free Seedlings of the Apple Dwarfing Rootstock)

  • Apple (Malus domestica) is one of the most economically important fruits in Korea. But virus infection has decreased sustainable production of apple and caused the serious problems such as yield loss and poor fruit quality. Virus or viroid infection including Apple chlorotic leaf spot virus (ACLSV), Apple stem pitting virus (ASPV), Apple stem grooving virus (ASGV), Apple mosaic virus (ApMV) and Apple scar skin viroid (ASSVd) has been also reported in Korea. In many cases, apple is infected with virus and viroid with no specific symptoms, the damage caused by the virus are unaware significantly. In our research, we tried to eliminate viruses in the rootstock for the disease-free seedlings of the apple dwarfing rootstock M.9 and M.26. The method of virus elimination was meristem culture, heat($37^{\circ}C$, 6weeks) treatment and chemistry($Ribavirin^{(R)}$) treatment. The analytical methods commonly used for the detection of virus is Enzyme-linked Immuno-Sorbent Assay(ELlSA) and Reverse Transcription-polymerase Chain Reaction(RT-PCR). RT-PCR method was more 30% sensitive than ELISA method. Efficiency of method eliminate virus appeared meristem method > heat treatment > chemistry treatment. The higher acquisition rate of disease-free seedlings is 30~40% on meristem treatment. In meristem treatment, the apple dwarfing rootstock M.9 gained infection ratio of ACLSV, ASPV and ASGV were 45%, 60% and 50% respectively. In the apple dwarfing rootstock M.26, infection ratio of ACLSV, ASPV and ASGV were 40%, 55%, 55%, respectively. Based on our results, it was found that most effective method of disease-free seedlings apple dwarfing rootstocks was by meristem treatment than heat method and chemistry treatment.

대추나무 빗자루병의 발생량 변화 (Fluctuations in the Outbreak of Jujube(Ziziphus jujuba Miller) Witches'-broom Disease)

  • Infection rate of witches'-broom disease (WB) was observed yearly in transplanted jujube seedlings for 7 years. Seedlings of‘Hongan’grafted either on the rootstock from seed (RS) or on the rootstock from rooting (RR) were planted in separate farms. Occurrence of the disease was determined 4 years after transplanting with the witches'-broom symptom. Only 1 seedling of RS was infected with WB, while 56 seedlings of RR showed WB. Yearly infection rates of WB were observed in several varieties of jujube including‘Boeun’,‘Keumsung’,‘Moodeung’,‘Bokjo’,‘Koori’(10 trees for each in Cheongjoo), and‘Hongan’(165 trees in Boeun). In most varieties, WB first appeared in the third year from transplanting, and in the seventh year, more than 80% of the total tree showed WB. The yearly infection rate of newly infected trees was almost. stable during the first years. However, the ratio sharply increased from the sixth year. The accumulated infection rate of WB increased double, year by year from the third to seventh year. Between Cheongjoo and Boeun, no difference in infection rates was found.

박과채소용 단근합접 접목시스템 개발(1) -작업공정 분석과 시스템 설정 - (Development of a Root-Removed Splice Grafting System for Cucurbitaceous Vegetables (1) - Analysis of Grafting Process and System Setting -)

  • This study was conducted to develop a root-removed splice grafting system for cucurbitaceous vegetables, mainly watermelon and cucumber seedlings, for the seedlings factories where currently most of seedlings grafting works are carried out by manual works. The major results of the study are as follows. The dimensions of rootstocks and scions, except cotyledon width, of root- removed splice grafting of watermelon and cucumber were shown to be varied within the 2.5-fold range. The growth status of seedlings were not consistent in terms of cotyledon sprouting direction and angle which were considered as one of the important factors for in root-removed splice grafting. The grafting work of root-removed splice for grafted watermelon and cucumber could be divided by four sub-operations: seedling supplying, cutting, clipping and potting, while a part or all root of the rootstock was removed in the seedlings supplying operation. The cutting angles of the rootstock and scion were $34-45^{\circ}$ and $20-45^{\circ}$, respectively, while the stem length of the scion varied from 6 mm to 12 mm. The splices of rootstock and scion were heaped up in parallel and then fixed by a clip. It indicated that the ideal grafting system, adopting conventional grafting processes of seedlings specifications as well as conventionally manual root-removed splice grafting method, performed very well for seedlings gripping and transporting, supplying clip, clipping and discharging grafted seedlings while workers supplied seedlings to the semi-automatical system.

생장조절제 및 Potassium Humate가 사과대목 M.26 기내 증식에 미치는 영향 (Influence of Growth Regulators and Potassium Humate on in Vitro Multiplication of Apple Rootstock M.26)

  • 본 실험은 식물생장조절제인 TDZ, BA, TAh, IBA, zeatin, GA$_3$가 사과대목 M.25의 기내증식에 미치는 영향을 구명하기 위해서 실시하였다. 또한 시토카닌과 오옥신의 역할을 동시에 하는 것으로 알려진 potassium humate가 뿌리형성에 미치는 효과를 알아보기 위해서 몇가지 농도로 배지에 첨가했다. 생장점 배양으로부터 건전한 산소를 얻기 위해서 BA, TDB, zeatin을 단독으로 기본배지에 첨가한 결과, zeatin 1.0 mg/L 첨가한 배지에서 100% 생존을을 나타냈고 신초생장 또한 양정하였다. 생장점 배양을 통해서 얻은 신초의 증식을 위해서 여러 종류의 생장조절제를 다양한 농도로 처리했을때 NAA 0.5 + TDZ 0.2 mg/L이 첨가된 배지에서 증식된 신소재가 평균 12.1개로 증식율이 가장 높았다. 형성된 신초로부터 발근유도는 IBA 0.6 mg/L이 첨가된 배지에서 가장 효과적이었다. 이러한 발근유도 배지에 potassium humate 250 mg/L이 첨가된 처리구에서 발근율과 부정근 돈는 IBA 0.5 mg/L 단독 처리구보다 각각 30%와 350% 이상 증가되었다.

Breeding of the native vegetables using the biotechnology

  • For breeding of a new rootstock for eggplant production, somatic hybrids between two species, Solanum integrifolium and S. sanitwongsei were obtained through protoplast fusion. The former species has been commonly used for rootstock for eggplant production in Japan. Eggplants on these rootstocks are more productive than ungrafted plants, but are susceptible to bacterial wilt caused Ralstonia solanacearum. While the latter species is resistant, the growth of eggplants on this rootstock is rather slow and low yield. Protoplast of both species were isolated from cotyledons, and inactivated with iodoacetamide or UV-irradiation, then fused electrically. The fused products were then cultured. Regenerated plantlets were then transplanted on soil then maintained in a green house. The plants were classified into four groups. Those in the first group showed morphological characters intermediate of the parentalspecies. The plants bore fruit with viable seeds. The plants showed a chromosome number of 2n=48, the sum of those of the parental species, and are suggested to be symmetric fusion products. While plants in the other groupswas less vigorous and showed chromosome number 2n= 68 to 72 suggesting asymmetric fusion products by genomic in situ hybridization(GISH). Isozyme pattern of shikimate dehydrogenase (SKDH; EC 1.1.1.25), isocitrate dehydrogenase (IDH; EC 1.1.1.41) and phosphoglucomutase (PGM; EC 2.7.5.1) showed that 24 regenerated plants in three groups were somatic hybrids. Analysis of random amplified polymorphic DNA (RAPD) showed that 43 S. integrifolium-specific and 57 S. sanitwongsei-specific bands were all found in 24 plants. Both somatic hybrids and its S1 plants were found to be resistant to bacterial wilt, and eggplant grafted these plants using for rootstocks were more productive than grafted mother plants. Now, S1 progenies are used for commercial eggplant production in Osaka Prefecture.
