Characterization of Microsomal $Ca^{2+}$ Uptake in Tomato Root Tissues (토마토 뿌리조직에서 분리한 마이크로솜의 $Ca^{2+}$ 흡수 특성)

  • In order to characterize the property of $Ca^{2+}$ transport in plant cell, microsomes were prepared from the roots of tomato and microsomal $^{45}Ca^{2+}$ uptake was measured. When 1 mM vanadate, a selective inhibitor of P-type ATPases, 50 mM $NO_3^-$, a specific inhibitor of vacuolar $H^{+}-ATPase$, and both of these inhibitors were treated, the microsomal $^{45}Ca^{2+}$ uptakes were inhibited by 20, 33 and 47%, respectively. The inhibitory effects of these two inhibitors were investigated by using a protonophore, gramicidin. When the chemical gradient of $H^{+}$ was relieved by gramicidin, the uptake was decreased by 30%, implying the presence of $Ca^{2+}/H^+$ antiporter in the microsomal membrane. In the $^{45}Ca^{2+}$ uptake experiment, the effect of gramicidin was independent of vanadate-induced inhibition. However, when the activity of vacuolar $H^{+}-ATPase$ was inhibited by $NO_3^-$, the effect of gramicidin was severely decreased. Meanwhile, thapsigargin, a specific antagonist of ER/SR-type $Ca^{2+}-ATPase$, inhibited the microsomal $^{45}Ca^{2+}$ uptake and the maximum inhibitory effect was obtained at $10\;{\mu}M$. The effect of thapsigargin was blocked by $NO_3^-$ and gramicidin, but not by vanadate. These results imply that vanadate directly inhibits the activity of $Ca^{2+}-ATPase$; however, $NO_3^-$ and thapsigargin block the activity of $Ca^{2+}/H^+$ antiporter by inhibiting the vacuolar $H^{+}-ATPase$. In conclusion, the microsomal $^{45}Ca^{2+}$ uptakes are mediated by two major enzymes, $Ca^{2+}-ATPase$ and $Ca^{2+}/H^+$ antiporter in tomato root tissue.

  • This study was conducted to evaluate the effects of two amino acid fertilizers on the growth of creeping bentgrass and N uptake. Fertilizer treatments were designed as follows; non-fertilizer (NF), control (CF), recommended amount (ALF), double amount (2ALF) of amino acid liquid fertilizer (AaLF), recommended amount (ASLF) and double amount (2ASLF) of amino acid liquid fertilizer contained with saponin (AaSLF). Turf quality of treatments of AaLF and AaSLF such as turf color index, chlorophyll index and root length was similar to the treatment of CF. Dry weight and content, uptake and availability of N were investigated highest in the 2ALF and 2ASLF. These results suggested that foliar application of AaLF and AaSLF was enhanced turf quality and growth of creeping bentgrass by stimulating uptake and availability of N.

  • Habitat observation, cultural experience of old and present plantation, weather factors in relation to crop stand and water physiology of root and leaf were reviewed. According to habitat observation ginseng plants love water but plate wit talus well grow at drained place with high moisture content in air and soil while ginseng plants were not found in dry or wet place. According to cultivation experience ginseng plants require abundant water in nursery and main field but most old planters believe that ginseng plaints are draught-loving thus require little water. The experience that rain especially in summer i.e unfavorable might be due to mechanical damage of leaves arid leaf disease infection, or severe leaf fall which is caused by high air temperature and coinsided with rain. According to crop stand observation in relation to weather factors abunsant water increased each root weight but decreased total yield indicating tile increase of missing root rate. Rain in summer was unfavorable too. Though rain in June was favorable for high yield general experience that cloudy day and rain were unfavorable might be due to low light intensity under shade. Present leading planters also do loot consider the importance of water in main field. Water content is higher in top than in root and highest in central portion of root and in stem of top. For seedling the heavier the weight of root is tile higher the water content while it reveries from two years old. Water potential of intact root appeared to be -2.89 bar suggesting high sensitivity to water environment. Under water stress water content severly decreased only in leaf. Water content of leaf appeared to be 78% for optimum, below 72% for functional damage and 68% for perm anent wilting. Transpiration or curs Principally through stomata in lower side of leaf thus contribution of upper side transpiration decreased with the increase of intensity. Transpiration is greater in the leaves grown under high light intensity. Thus water content is lower with high light inte nsity under field condition indicating that light is probable cause of water stress in field. Transpiration reached maximum at 10K1ut The decrease of transpiration at higher temperature seems to be due to the decrease of stomata aperture caused by water stress. Severe decrease of photosynthesis under water stress seems to be principally due to functional damage which is not caused by high temperature and Partly due to poor CO2 supply. Water potential of leaf appeared to be -16.8 bar suggesting weakness in draught tolerance. Ginseng leaves absorb water under high humidity. Water free space of leaf disc is %mailer than that of soybean leaf and water uptake appears to be more than two steps.

Effect of Root Medium Formulations on Growth and Nutrient Uptake of Hot Pepper Plug Seedlings (혼합상토의 조성이 고추 플러그 묘의 생육 및 무기원소 흡수에 미치는 영향)

  • This research was conducted to evaluate the effect of root medium formulations on growth and nutrient uptake of hot pepper 'Knockgwang' in 72-cell plug trays. To achieve this, the nine root media were formulated by adjusting blending rate of perlite (PL) to coir (CO), peatmoss (PM) or coir+peatmoss (5:5, v/v, COPM). Then, the growth characteristics and tissue nutrient contents were determined at 35 and 70 days after sowing. The elevated blending rate of PL to CO increased fresh and dry weight of hot pepper at 35 days after sowing. The treatments of 20% in blending rate of PL to PM or that of 0% to COPM produced the highest fresh and dry weight among perlite treatments of PM or COPM. The results of crop growth at 70 days after sowing also showed similar trends to those of 35 days after sowing. The elevated blending rate of PL to CO or PM decreased tissue $P_2O_5$ and K contents and increased Ca and Mg contents at 35 days after sowing, With the equal blending rate of perlite, the plant tissue grown in CO had higher K contents and lower N, Ca and Mg contents than those in PM and COPM. The elevated blending rate of perlite in three organic matter also decreased tissue $P_2O_5$ and K contents at 70 days after sowing, but Ca and Mg contents were the highest in the treatment of 20% PL in CO, 40% PL in PM and 40% PL in COPM among perlite treatments in each organic matter.

Influence of Boron and Iron Toxicity on the Physiological Status, Growth, and Mineral Uptake of Ginseng in Hydroponic Culture (인삼 수경재배 시 붕소와 철 과잉 농도가 인삼의 생리장해 증상, 생육 및 무기원소 흡수에 미치는 영향)

  • Background: Ginseng is a perennial crop grown for more than four years in the same place. Therefore, it is highly affected by the soil environment, especially nutrients in the soil. The present study was carried out to investigate to the influence of boron and iron concentrations on the physiological status, growth, and mineral uptake of ginseng to obtain the basic information for diagnosing a physiological disorder in ginseng plants. Methods and Results: The boron and iron concentrations were controlled at 3, 30, 150, 300 and 2, 20, 100, $200mg/{\ell}$, respectively. When treated with $150mg/{\ell}$ of boron, the ginseng plants showed yellowing or necrosis symptoms at the edge or end of their leaves. Compared with the $3mg/{\ell}$ treatment, the root weight decreased by 13 and 24% in the 150 and $300mg/{\ell}$ treatments, respectively. When treated with $20mg/{\ell}$ of iron, the ginseng plants showed yellowing between the veins of the leaves followed by the formation of brown spots. The root weight gradually decreased with increasing iron concentration. Approximately 55% decrease in root weight was observed upon treatment with $200mg/{\ell}$ of iron. Conclusions: The boron toxicity occurs in the leaves of ginseng at the boron concentration of approximately 1,900 mg/kg or more. The iron toxicity occurs at the iron concentration of approximately 120 mg/kg for leaves and 270 mg/kg for roots.

Seedling Growth and Nutrients Uptake in Hybrid Rice (벼 1대잡종의 묘생육과 양분흡수)

  • This study was carried out to investigate the characteristics of the seedling growth and nutrients uptake at 45 day seedlings in cytoplasmic genetic male sterile lines(HR7028A, IR54756A), their restore line (Yongmoonbyeo) and check variety (Changsungbyeo). Hybrid rices had longer and more roots, heavier dry weight and lower shoot /root ratio than their parents and inbred check variety, showing heterobeltiosis in absorbed amount of inorganic elements, content of total sugar, starch and chlorophyll in plant. Different magnitude of heterobeltiosis were shown between F$_1$ hybrid rices with the same restore line; HR7028A/Yongmoonbyeo had higher heterobeltiosis than IR54756A/Yongmoonbyeo did. Hybrid rices had more number and higher $\alpha$-naphthylamine oxidizing ability of newly-grown roots in 10 day sand culture in ground water with root-cut 45 day-seedling than parental lines and inbred variety. Nunber of the newly-grown roots were positively and significantly correlated with content of total sugar, starch and absorbed amount of inorganic elements in the shoot, and content of chlorophyll in the leaf.

Studies on the Nitrogen Effect in Red Discoloration of Rice IR 667 (수도 IR 667의 적고현상에 미치는 질소의 영향에 관한 연구)

  • Pot and paddy field tests were conducted to study red discoloration of rice var. IR 667 leaves with reference to the leading Korean native variety Jinhung and Paldal, with the following results: 1. Minor elements such as Mn, Fe, B, Al, Ca and Si had no influence on the discoloration, but a supply of various soluble nitrogen compounds completely restricted it. The more prosperous the growth of IR 667 with nitrogen, the more severe the discoloration appears when nitrogen absorptin becomes limited. 2. Chlorotic pigments extracted from both IR 667 and Jinhung were compared spectrophotometrically, and found to have different spectral peaks. IR 667 had peak closer to red than Jinhung, indicating the characteristic of the variety. IR 667 was observed to be more sensitive to nitrogen deficiency than Jinhung or the other japonica variety. 3. It was concluded that all the factors limiting nitrogen supply for IR 667 growth, such as low nitrogen application, restriction of root respiration (low temperature, poor drainage, toxic gases or substances in the root zone, etc.) and pest injuries, would result in the appearance of the so-called red discoloration, because of the reduction in nitrogen uptake. Since, the discoloration of IR 667 is varietal characteristic when grown in Korea, control of it may be beneficial cultural practice in increasing grain yield, although the increased succeptibility to pests and a drop in the rate of maturity due to relatively high nitrogen level in the leaves may result in an unexpected drop in yield. It is anticipated that further exploration conducted from practical point of view will establish the relatioknships between the extent of red discoloration, nitrogen availability and grain yield in IR 667.

Mobilization of Heavy Metals induced by Button Mushroom Compost in Sunflower

  • This study focused on evaluating the phytoextraction of heavy metals (Co, Pb, and Zn) induced by bioaugmentation of button mushroom compost (BMC) in Helianthus annuus (sunflower). When the potential ability of BMC to solubilize heavy metals was assessed in a batch experiment, the inoculation with BMC could increase more the concentrations of water-soluble Co, Pb, and Cd by 35, 25, and 45% respectively, compared to those of non-inoculated soils. BMC-assisted growth promotion and metal uptake in H. annuus was also evaluated in a pot experiment. In comparison with non-inoculated seedlings, the inoculation led to an increase in the growth of H. annuus by 27, 25, and 28% in Co-, Pb-, and Zn-contaminated soils, respectively. Moreover, enhanced accumulation of Co, Pb, and Zn in the shoot and root systems was observed in inoculated plants, where metal the translocation from root to the above-ground tissues was also found to be enhanced by the BMC. Evidently, these results suggest that the BMC could be effectively employed in enhancing the phytoextraction of Co, Pb, and Zn from contaminated soils.

Ethylenediamine as a Promising and Biodegradable Chelating Agent in Growth of Plant Under Zinc Stress (아연 스트레스를 받는 식물의 성장을 위한 생분해되는 킬레이트로서 에틸렌디아민)

  • Zinc (Zn) is an essential element required for growth and development of plants. However, Zn can be toxic to plants when it presents excessive amount. Phytoextraction is an economic and environment-friendly technique using plants to clean-up metal-contaminated soils. However, the technique cannot be applied in highly metal-contaminated areas because plants will not normally grow in such conditions. Therefore, this research focuses on identifying chelating agents which are biodegradable and applicable to highly metalcontaminated areas. Zn as a target metal and cysteine (Cys), histidine (His), malate, citrate oxalate, succinate, and ethylenediamine (EDA) as biodegradable chelating agents were selected. Plants were grown on agar media containing various chelating agents with Zn to analyze the effect on plant growth. Malate and His slightly increased the inhibitory effect of Zn on root growth of plants, whereas Cys, citrate, oxalate, and succinate did not show significant effects. However, EDA strongly diminished the inhibitory effect of Zn on root growth. The effect of EDA is correlated with decreased Zn uptake into the plants. In conclusion, as biodegradable chelating agents, EDA is a good candidate for growth of plants in highly Zn-contaminated areas.

Effect of Nutrient Concentrations and Leaching Percentage on Growth and Nutrient Uptake by Perilla Frutesens Britton var. Japonica Hara in Plug Culture (잎들깨의 플러그육모에서 용탈률 및 시비농도가 생육 및 무기원소 흡수에 미치는 영향)

  • Effect of Nutrient Concentrations, fertigation frequency, and learching percentage on crop growth and nutrient concentrations in root media were evaluated. The treatment of each irrigation with $50 mg.L^{-1}$ of nitrogen in stage 2 and increase to $80 mg.L^{-1}$ nitrogen in stage 3 had the highest crop growth at 34 days after sowing among treatments tested. Feeding with low nutrient concentrations and elevated frequency decreased crop growth. In treatments of each leaching percentage, feeding with low nutrient concentrations and elevated frequency resulted in increased tissue nutrient contents. The less tissue potassium content and higher calcium and magnesium contents were observed in treatment of 50% leach than those in 0% leach. All treatment tested had soil solution pH higher than 6.8. Electrical conductivity in treatments of 50% leach were lower than those of 0% leach. Feeding with low nutrient concentrations and elevated feeding frequency in each leaching percentage resulted in increased electrical conductivity in soil solution of root media. Trends of medium nutrient concentrations were similar to those of electrical conductivity.
